    Bioautography of a novel antibiotic produced by fermentation process of Streptomyces griseus ATCC 10137 had been investigated by using streptomycin standard. High Performance Thin Layer Chromatography had been carried out by using KH2PO4 5% solution as a solvent, and Bacillus subtilis PCI 219 as a bacterial test. Bioautogram showed that Rf value of spot derived from the antibiotic extract isolated from the fermentation filtrate is 0.65, which has significant different with Rf value of streptomycin standard 0.15. It was found that the antibiotic and streptomycin standard inhibited growth of Bacillus subtilis PCI 21

    of tofu refuse needed for the growth of Streptomyces griseus A TCC 10137, in media which contain 0,5%, 1% and 2% the extract of tofu refuse. The optimum medium obtained from this optimation was then being tested again to find the optimum pH for the growth of Streptomyces griseus ATCC 10137 among various pH 7,0; pH 7,3 and pH 7,5. The result of this optimation was then compared to the growth medium containing the powder oftofu refuse. One Ose of Streptomyces griseus ATCC 10137 culture from PDA medium was inoculated into 250 mL of the seed medium, ISP medium No.4, and incubated at room temperature for 2-4 days on rotary shaker at 200 rpm. Three mL of this seed culture was transferred into 30 mL the growth medium containing tofu refuse. Ten mL of sample was taken from the 30 mL seed culture at every 12 hours. This sam~le was filtered by filtering paper whatman No 4.1, then dried in oven at 105 C until constant Dry-cell Weight was obtained. This study showed that medium with 2% the extract of tofu refuse gave the highest Dry-cell Weight, followed by 1 % and 0,5% concentration. The pH gave highest Dry-cell Weight was pH = 7. Based on the curve obtained from this research the optimum medium for the growth of Streptomyces grise us ATCC 10137 was the medium contains 2% the extract of tofu refuse and pH 7,0. The other research found that the optimum medium for the growth of Streptomyces grise us ATCC 10137 was the medium contains 0,5% the powder of tofu refuse and pH 7,3. There was no significant difference between both optimum media mentioned above

    Streptomyces griseus merupakan bakteri mesofil yang dapat tumbuh pada suhu 20oe-40oe dan pH 7,0-7,5. Mikroba ini memiliki kondisi suhu dan pH optimum tertentu untuk pertumbuhannya, kondisi tersebut dapat berubah pada media yang berbeda. Hal ini terkait dengan tersedianya nutrisi pada media tersebut. Pada penelitian ini dilakukan optimasi suhu dan pH pertumbuhan Streptomyces griseus ATee 10137 dalam media yang mengandung ampas tahu. Berdasarkan penelitian pendahuluan, media optimal untuk pertumbuhan Streptomyces griseus ATee 10137 adalah media B, yaitu media ISP-4 tanpa Ammonium Sulfat yang mengandung ampas tahu 0,5%. Optimasi suhu dan pH pertumbuhan Streptomyces griseus ATee 10137 dalam media B dilakukan pada suhu 28°e, 30oe, dan 35°e, serta pH 7,0; 7,3 dan 7,5. Dari masing-masing kondisi di atas dilakukan pengamatan pertumbuhan Streptomyces griselL'I ATee 10 137 dengan cara penghitungan jumlah sel metode hitungan cawan. Hasil yang diperoleh menunjukkan bahwa Streptomyces griseus ATee 10137 dapat tumbuh pada suhu 28°e dan 300e baik pada pH 7,0: 7,3 maupun 7,5. Pertumbuhan optimal dicapai pada suhu 30oe, pH 7,3, sedangkan pada suhu 35°e Streptomyces griseus ATee 10137 tidak dapat tumbuh pada ketiga pH penelitian di atas

    Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed

    “Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship

    Appropriate Similarity Measures for Author Cocitation Analysis

    We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature.     Ampas tabu pada umumnya masih mengandung protein kasar 21-29% bib bahan kering (Hennana, 1985). HasH penelitian pendahuluan menunjukkan bahwa ampas tabu mengandung 16-18 macam asam amino dengan kadar bervariasi. Namun selama ini arnpas tahu hanya digunakan sebagai campuran pakan ternak dan makanan tradisional, sebingga pernanfaatannya rnasih perlu dikernbangkan lagi. Streptomyces griseus ATCC 10137 merupakan bak.teri aerob farnili Streptomycetaceae dari ordo Actinomycetes yang merniliki karakteristik mampu mernproduksi antlbiotika pada media fennentasi yang mengandung surnber nitrogen dan karbohidrat yang mernenulri persyaratan nisbah CIN. Berdasarkan kedua hal di atas, maka dapat dilakukan upaya untuk meningkatkan nilai ekonorni arnpas tahu dengan memanfaatkannya sebagai sumber nitrogen altematif dalarn media produksi antibiotikanya. Tujuan penelitian ini adalah untuk mernbuktikan apakah ampas tahu dapat digunakan sebagai sumber nitrogen pada media produksi anbbiotika basil biakan Streptomyces griseus A TCC 10137 dan apakah antibiotika yang dihasilkan adalah streptornisin. Penelitian dilakukan dengan pernbiakan Streptomyces griseus ATCC 10137 pada media PDA padat lalu dipindahkan ke media pertumbuhan ISP-4 cairo Setelah dari media pertumbuban kemudian dipindahkan ke dalam media produksi yang mengandung ampas tabu dengan konsentrasi 2%. Kaldu ferrnentasi yang diperoleh diuji potensi dengan Bacillus subtilis ATCC 9466 sebagai bakteri uji. Isolasi antibiotika dalam kaldu ferrnentasi menggunakan adsorben arang aktif, dielusi dengan methanol-HCl 0,5 N· dan diserapkan pada kromatografi kolom penukar kation CG-50 (NI-I/) bingga didapatkan fraksi eluat yang kernudian diuji dengan reak.si warna. Fraksi eluat positif yang didapatkan dikurnpulkan, dipekatkan dan dinetralkan. Selanjutnya, ekstrak yang diperoleb dianalisis kualitatif secara KLT -Densitometri dan bioautografi antibiotika kernudian dibsndingkan dengan baku streptomisi

    Dispelling the Myths Behind First-author Citation Counts

    We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more sophisticated methods
