    Mise en évidence et suivi par échographie ovarienne transrectale de nouveaux critères pour identifier le follicule dominant et déterminer l'imminence de l'ovulation chez la jument en oestrus (Comparaison de différents échographes)

    Les marqueurs échographiques couramment utilisés chez la jument souffrent d un manque de fiabilité pour la détermination du moment idéal de la mise à la reproduction. Nous nous proposons ici de poursuivre l étude de 2 marqueurs échographiques récemment décrits : un liseré anéchogène apparaissant entre la thèque interne et la granulosa des follicules dominants, et une zone crénelée enserrant la granulosa à la base du follicule dans les heures précédant l ovulation. Vingt juments du troupeau pédagogique d Oniris ont été suivies sur 2 cycles consécutifs par échographie transrectale. Le liseré anéchogène est apparu en moyenne 6,0 jours avant l ovulation avec un maximum d intensité atteint en moyenne 3,0 jours plus tard. Tous les follicules qui ont ovulé ont présenté un liseré anéchogène, mais la réciproque n est pas vraie. Aucun des follicules dépourvus de liseré anéchogène n a ovulé. Tous les follicules ayant présenté une zone crénelée ont ensuite ovulé dans un délai de 12 à 48 heures. La probabilité de visualisation de ces 2 marqueurs avec l échographe Aloka prosound 2 est apparue significativement plus élevée qu avec les échographes Merlin 1101 et Exago 1.09.NANTES-Ecole Nat.Vétérinaire (441092302) / SudocSudocFranceF

    Introduction : L'accouchement prématuré reste la principale cause de morbidité et de mortalité infantile dans le monde. La MAP sc situe au 1 er rang des complications de la grossesse, elIe représente le principal motif des tnmsferts in utero. La naissance et la prise en charge des grands prématurés dans un centre de niveau 3 restent une priorité. Le but de cette étude est de développer un modèle prédictif d'accouchement prématuré à l'aide de facteurs de risque étudiés afin d'aider les obstétriciens dans la prise de décision d'un transrert in utero pour MAP dans un centre de niveau adapté. Matériel et méthode : Il s'agit d'une étude rétrospective observationnelle sur le devenir des patientes transfrérées pour MAP via le CROP entre 2005 et 20 11 dans la région Languedoc Roussillon. Dans le but d'aider le clinicien dans sa décision de trans rert in utero, nous allons tenter de créer un modèle prédictif d'accouchcment en étudiant et en comparant les accouchements dans les 48h d' une part et les accouchements il moins de 37 SA d'autre part.Résultats: Dans notre travail, le taux d'accouchement avant 37 SA est de 50%. Le taux d'accouchement dans les 48h est de 20%. Deux modèles prédicfirs d'accouchement prématuré ont été élaborés. Les critères significatifs dans le modèle prédictir d'accouchcment avant 37 SA sont la longueur du col, la gémellité, les antécédents de MAP ou d'accouchement prématuré. Les crilères significatifs dans le modèle prédictif d'accouchement dans les 48 heures sont l'àge gestationnel au moment du transfert la longueur du col. Discussion : Nous avons pu établir un risque individuel par patiente d'absence d'accouchement dans les 48 heures. A partir de ce score de risque et de l'éloignement géographique nous avons établi un organigramme de prise en charge et de décision de transfert in utero afin de limiter ces transferts et de prendre en charge les patientes dans leur maternité d'origine. Conclusion : Nous avons élaboré un modèle prédictif d'absence d'accouchement dans les 48 heures il partir de plusieurs facteurs. Le facteur principal et déterminant dans ce risque d'accouchement eslla longueur du col utérin.     Différentiation des cellules T CD8 pendant la réponse immunitaire

    Les lymphocytes T CD8 ont un rôle essentiel dans la protection contre les agents pathogènes intracellulaires et la progression tumorale. Ainsi, la compréhension de la diversité des mécanismes de différenciation des lymphocytes T CD8 naïfs en cellules effectrices, ainsi qu en cellules mémoires compétentes, est fondamentale pour le développement efficace de vaccins à cellules T. Dans ce travail de thèse, nous avons abordé deux questions centrales : (1)Très tôt après l activation des cellules T CD8, quels sont les mécanismes par lesquels les cellules T effectrices agissent comme effecteurs pro-inflammatoires en recrutant d autres cellules? Et quel est leur rôle dans la réponse immunitaire? (2) Quel est le rôle du contexte infectieux dans le programme de différenciation des lymphocytes T CD8 ? Est-il responsable de l hétérogénéité des cellules répondeuses et a-t-il un rôle dans les différents effets protecteurs des cellules mémoires? Afin de répondre à ces questions, nous avons choisit d utiliser des cellules T CD8 exprimant un récepteur pour l antigène transgéniques (TCR-Tg) pour suivre la différentiation in vivo des lymphocytes T CD8. De plus, la méthode de RT-PCR sur des séries de cellules uniques, nous a permis d analyser la co-expression des ARNm dans ces cellules. Comme l utilisation à haute fréquence de cellules TCR-Tg a été fortement critiquée, nous avons comparé la différenciation de ces cellules avec celle des cellules endogènes (non transgéniques et rares). Dans ce premier manuscrit nous avons observé un comportement similaire, ce qui a renforcé l'avantage d'utiliser des cellules TCR Tg pour étudier les réponses immunitaires des lymphocytes T CD8. De plus, nous avons conclu que la diversité des réponses immunitaires des lymphocytes T CD8 n est pas conditionnée par la fréquence de cellules naïves. Dans un deuxième manuscrit, nous avons comparé la réponse des cellules OT1 TCR-Tg (spécifiques de l antigène OVA) à l'infection bactérienne LM-OVA (Listeria Monocytogènes exprimant OVA) avec la réponse des cellules P14 TCR-Tg (spécifiques de l épitope GP33) à l infection par le virus LCMV. Nous avons montré que les cellules OT1, stimulées par l OVA dans un contexte bactérien (LM-OVA), présentent un profil d expression génique distinct de celui des cellules P14 stimulées par le GP33 dans un contexte viral (LCMV). Nous avons également co-stimulé les cellules P14 et OT1 dans une même souris suivant le même contexte bactérien avec LM-GP33 et LM-OVA. Dans ce cas, nous n avons pas observé de différence dans le profil d expression génique. L ensemble des résultats démontrent que les stimulations spécifiques des cellules T CD8 par différents agents pathogènes génèrent des cellules T CD8 présentant des caractéristiques différentes qui ne sont pas déterminées par la spécificité du TCR mais plutôt par le contexte infectieux. De plus, nous avons montré que les cellules mémoires endogènes résultant de la stimulation des CD8 en présence de LCMV ont été plus efficaces après une deuxième réponse immunitaire que des cellules mémoires générées après stimulation avec LM-GP33 (bactérie). Nous avons également observé que la protection plus efficace dans le contexte viral est associée à des cellules T CD8 qui présentent un phénotype de cellules T mémoires effectrices (TEM) tandis que les cellules T CD8 générées dans un contexte bactérien ont plutôt un phénotype associé aux cellules T mémoires centrales (TCM). Ces résultats démontrent que différents pathogènes induisent différents profils de différentiation des cellules T CD8 et que malgré l élimination efficace des différents pathogènes dans une réponse primaire, la qualité des cellules mémoires générées au cours de cette réponse peut être différente. Dans un troisième manuscrit, nous avons étudié les mécanismes de recrutement d autres cellules par les lymphocytes T CD8 activés à un temps précoce de la réponse immunitaire. (...)CD8 T cells are essential for the elimination of intracellular pathogens and tumor cells. Understanding how naïve CD8 T cells differentiate into effector cells capable of eliminating pathogens and to generate adequate memory cells during immune responses is fundamental for optimal T cell vaccine design. In this PhD thesis work we addressed two central questions: 1) What are the mechanisms by which early effector T cells could act as pro-inflammatory effectors? And what is their role in the immune response? 2) How heterogeneous are CD8 responses? Could different pathogens modulate CD8 T cell differentiation programs and be responsible for CD8 cell-to-cell heterogeneity? Could they also generate memory cells with different protection capacities? To address these questions related to the diversity of CD8 T cell differentiation during immune responses, we used the single cell RT-PCR technique to detect ex vivo expression of mRNA in each individual cell, and Brefeldin A injected mice to detect ex vivo intracellular proteins. As experimental system to evaluate in vivo cell activation we used T cell receptor transgenic (TCR-Tg) CD8 T cells. Since the use of TCR-Tg cells to study immune responses has been subjected to criticism (due to high frequency of naïve-precursor transfers), in a first Ms. we compared the behavior of TCR-Tg and endogenous (non-transgenic and present at low frequency) cells in the same mouse. We found fully overlapping behavior between these two cell populations, which reinforced the advantage of using TCR-Tg cells to study CD8 immune responses. In addition, we concluded that the frequency of naïve-precursors do not induce diversity on CD8 T cell differentiation patterns. In a second Ms. we evaluated the impact of different pathogens in the diversity of CD8 T cell properties during two different immune responses: OT1 TCR-Tg cells (specific for OVA antigen) in the response to LM-OVA (Listeria Monocytogenes expressing OVA) infection; and P14 TCR-Tg cells (specific for GP33 epitope) in the response to Lymphocytic choriomeningitis vírus (LCMV) infection. We found that OT1 and P14 cells had different properties. As this difference could also be attributed to the different TCR avidity between OT1 and P14 cells, we then compared the behavior of P14 and OT-1 cells in the same mouse, co-injected with LM-OVA and LM-GP33. Since no differences were then detected, these results demonstrated that priming with different pathogens generates CD8 T cells with different characteristics that are not determined by TCR usage, but rather by the infection context. In addition, when looking for the protection capacity of endogenous CD8 memory cells generated in bacterial or viral context, we found that memory cells generated after LCMV priming were more efficient in responding to a second challenge, than memory cells generated after LM-GP33 priming. We also found that this better protection is associated with a T cell effector memory (TEM) phenotype associated with the LCMV infection, in contrast with a T cell central memory (TCM) phenotype generated after LM-OVA infection. These results demonstrate that different pathogens are responsible for diversity of CD8 T cell differentiation patterns and that even when distinct pathogens are efficiently eliminated during the primary immune response the quality of the memory generated may differ. In a third Ms. we studied the mechanisms by which effector CD8 T cells attracted other cell types in the early days of an immune response. We used two experimental systems: the response of OT1 TCR-Tg cells to LM-OVA infection; and the response of anti-HY TCR-Tg cells to male cells ( sterile -non infectious context). In both cases we found that immediately after activation, CD8 T cells expressed high levels of pro-inflammatory cytokines and chemokines (such as TNFa, XCL1, CCL3 and CCL4).     The open access principle requires that scientific information be made widely and readily available to society. Defined in 2003 as a “comprehensive source of human knowledge and cultural heritage that has been approved by the scientific community”, open access implies that content be openly accessible and this needs the active commitment of each and every individual producer of scientific knowledge. Yet, in spite of the growing success of the open access initiative, a significant part of scientific and technical information remains unavailable on the web or circulates with restrictions. Even in institutional repositories (IRs) created to provide access to the scientific output of an academic institution, more or less important sectors of the scientific production are missing. This is because of lack of awareness, embargo, deposit of metadata without full text, confidential content etc. This problem concerns in particular electronic theses and dissertations (ETDs) that are disseminated with different status – some are freely available, others are under embargo, confidential, restricted to campus access (encrypted or not) or not available at all. While other papers may be available through alternative channels (journals, monographs etc.), ETDs most often are not. Our paper describes a new and unexpected effect of the development of digital libraries and open access, as a paradoxical practice of hiding information from the scientific community and society, while partly sharing it with a restricted population (campus). The study builds on a review of recent papers on ETDs in IRs and evaluates the availability of ETDs in a small panel of European and American academic IRs and networks. It provides empirical evidence on the reality of restricted access and proposes a model of independent variables affecting decisions on embargo and on-campus access, together with a table of different degrees of (non) open access to ETDs in IRs.Includes: Conference preprint, Powerpoint presentation, Abstract and Biographical notesXAInternationa

    Digital Preservation at International Nuclear Information System (INIS)

    Since its creation in 1970 until 1996, the International Nuclear Information System (INIS) collected and converted to microfiche over 312 000 non-conventional literature (NCL) reports received from IAEA member states and international organizations. The microfiche collection contains over 1 million items, with an estimated total of 25 million pages of full-texts. In 1997, the INIS Secretariat replaced the microfiche-based production system with an imaging system to process and to disseminate all NCL documents in electronic format. That marked the beginning of digital preservation efforts that still continue today. This paper provides an overview of the digital preservation practices and the technical infrastructure of the International Nuclear Information System (INIS).     For better or for worse: Knowledge output 1944-2001 and effects of the Legal Deposit Act no. 20/2002 and e-publishing on access to GL in Iceland

    Two studies are presented in this paper. The former was a study on the amount of knowledge output in Iceland, from 1944 to 2001. It covered all publications issued in Iceland in the sampling years, in the following publication genres: all market publications and GL published in Iceland on paper and catalogued in Gegnir, the national union catalogue, international peer-reviewed journal articles bibliographically accessible in Web of Science (WoS), in addition to patent applications, and standards in Iceland. The findings indicated considerable changes in the knowledge output, in particular increases in GL, international peer reviewed journal articles, and patent applications. They also indicated that GL might increasingly have been produced in-house, and published electronically as the 20th century drew to a close; a trend that continued in the 21st century. Publications produced inhouse were often not delivered on legal deposit and the e-medium was not covered by the Legal Deposit Act no. 43/1977 and therefore many a time missing in Gegnir. The latter study was aimed at finding how access to GL developed at the beginning of the 21st century. In particular to find what the effects of the Legal Deposit Act no. 20/2002, which covered e-publications, were on legal deposit in the National and University Library of Iceland (NULI).     Cette thèse se se compose de trois parties. La première partie est consacrée à l'étude des points périodiques des applications birationnelles des surfaces projectives. Nous montrons que toute application birationnelle de surface dont la croissance des degrés est exponentielle admet un ensemble de points périodiques Zariski dense. Dans la seconde partie, nous démontrons la conjecture de Mordell-Lang dynamique pour toute application polynomiale birationnelle du plan affine définie sur un corps de caractéristique nulle. Notre approche donne une nouvelle démonstration de cette conjecture pour les automorphismes polynomiaux du plan. Enfin la troisième partie porte sur un problème de géométrie affine inspiré par la généralisation au cas de toutes les applications polynomiales du plan affine de la conjecture de Mordell-Lang dynamique. Etant donné un ensemble fini S de valuations sur l'anneau de polynomes k[x,y] sur un corps algébriquement clos k triviales sur k, nous donnons une condition nécessaire et suffisante pour que le corps des fractions de l'intersection des anneaux de valuations de S avec k[x,y] soit de degré de transcendance 2 sur k.This thesis contains three parts. The first one is devoted to the study of the set of periodic points for birational surface maps. We prove that any birational transformation of a smooth projective surface whose degree growth is exponential admits a Zariski-dense set of periodic orbits. In the second part, we prove the dynamical Mordell-Lang conjecture for all polynomial birational transformations of the affine plane defined over a field of characteristic zero. Our approach gives a new proof of this conjecture for polynomial automorphisms of the affine plane. The last part is concerned with a problem in affine geometry that was inspired by the generalization to any polynomial map of the dynamical Mordell-Lang conjecture.     Utilisation des modèles dynamiques pour l'évaluation des traitements de l'hépatite C

    L'introduction depuis 2011 des trithérapies associant un inhibiteur de protéase (IP), telaprevir ou boceprevir, à l'interféron pégylé et la ribavirine (Peg-IFN/RBV), constitue un tournant majeur dans la lutte contre le virus de l'hépatite C (VHC). Cependant la réponse au traitement chez les patients cirrhotiques reste mal connue et pose de sérieux problèmes de tolérance. Les modèles de cinétique virale, analysés par modèles non-linéaires à effets mixtes (MNLEM), constituent un outil puissant pour la compréhension de la réponse au traitement. Nous avons démontré par simulation la puissance du test de Wald dans le cadre des MNLEM pour détecter une différence d'efficacité antivirale entre deux traitements. Cette approche, même pour un nombre réduit de prélèvements, s'est révélée systématiquement supérieure à l'approche standard reposant sur un test de Wilcoxon sur le déclin de charge virale. Cependant nous avons mis en évidence une inflation de l'erreur de type I de ce test lorsque le nombre de patients est faible et avons proposé une correction par permutation. Nous avons appliqué cette approche pour estimer l'efficacité antivirale des trithérapies chez des patients cirrhotiques en échec thérapeutique (ANRS MODCUPIC). Nous avons démontré que plus de 99% de la réplication virale était bloqué par les IP, avec une efficacité sighificativement supérieure du telaprevir par rapport au boceprevir, et une efficacité sous-optimale de Peg-IFN/RBV. Enfin, en modélisant la cinétique des paramètres sanguins, nous avons démontré que les concentrations de Peg-IFN et de RBV sont directement associées, respectivement, è l'anémie et la thrombopénie fréquemment observées chez ces patients.Since 2011, the introduction of triple therapies combining a protease inhibitor (PI), telaprevir or boceprevir, with the pegylated-interferon and the ribavirin (Peg-IFN/RBV) marked a milestone for anti hepatitis C virus (HCV) therapy. However, the response to treatment in cirrhotic patients remains unclear and poses serious safety problems. Vira kinetic models, analyzed by nonlinear mixed effect models (NLMEM), are a powerful tool for understanding the response to treatment. We have demonstrated, by means of clinical trial simulation, the power of the Wald tes1 within NLMEM to detect a HCV drug's antiviral effectiveness difference. This approach, even for a small number of samples, provided consistently higher power to the standard approach based on a non parametric Wilcoxon test on the viral load decay. However, we have highlighted an inflation of the type I error with the Wald test when the number of patients is low and have proposed a permutation correction. We applied this approach to estimate the antiviral effectiveness of the triple therapies in cirrhotic patients and non-responders to a prior therapy (ANRS MODCUPIC). We hav( shown that over 99% of the viral replication was blocked by PI, with a significantly superior effectiveness of telaprevir compared to boceprevir and suboptimal effectiveness of Peg-IFN/RBV. Finally, by modeling the kinetics of blood parameters, we demonstrated that the concentrations of Peg-IFN and RBV are associated with, respectively, anemia and thrombocytopenia which are frequently observed in these patients.PARIS7-Bibliothèque centrale (751132105) / SudocSudocFranceF
