Studies involving the influence of ophidic proteins to different coagulaction factors

    No full text
Accident involving snakes from Bothrops and Crotalus genera can be considered a public health and social problem in Brazil. Different local and systemic deleterious effects are directly caused by proteins that compose the venoms, such as phospholipases A2 and their variations, proteinases and L-amino acid oxidases. Some of these effects include myonecrosis and haemostatic alterations (such as hemorrhage), which can lead to permanent tissue loss, amputations and death. This project proposes studies involving proteins from the venom and components can act on blood clotting, using the molecular basis for a better understanding of how ophidic proteins can influence the clotting process and additionally help in the search for new peptide-based drugs or therapies. For these purposes, biophysical assays (crystallography, dynamic light scattering and affinity assays), biochemical assays (clotting and enzymatic assays) and bioinformatics assays (docking and molecular dynamics simulations) are proposed. These assays can shed light on the interaction of ophidic proteins and clotting factors, including the interaction details, affinity and haemostasis alterations. The data produced here can help improve the conventional treatment for snakebite victims, reducing sequelae and providing a better quality of life.Outra05/2024/PROPe - Programa Unesp de Pós-Doutorad

Projeto Lucinda Ferreira: perfis de microcomunidades surdas no Brasil e tipologia de Línguas de Sinais

    No full text
Esthetic rehabilitation using adhesive ceramics has stood out due to its longevity, high strength, and excellent optical properties, with the ability to mimic natural teeth. In modern dentistry, the digital workflow stands out by offering greater precision, better communication with the patient, greater case predictability, and highly efficient guidance of the laboratory phase, which is essential for achieving a satisfactory final result. This work presents a clinical case report of esthetic correction associated with the correction of vestibularization of tooth 13, in which 12 porcelain laminate veneers made of leucite-reinforced glass-ceramic (Empress CAD) were fabricated for teeth 16 to 26, and minimally invasive preparations were performed guided by the mock-up, aiming to preserve tooth structure. The digital workflow was used from the planning stage, through the CAD phase, to the milling of the restorations (CAM phase). The obtained results demonstrated that the interaction between digital dentistry and concepts of minimally invasive rehabilitation was able to optimize outcomes with precision, efficiency, and speed

This thesis explores the intersections between Machine Learning and Quantum Information, investigating how artificial intelligence tools can be applied to solve fundamental problems in complex quantum systems. The work focuses on two main challenges: quantum phase classification and quantum synchronization prediction. In quantum phase classification, we present three complementary approaches. The first explores model independence, demonstrating how a classifier can be trained on data from one Hamiltonian model and used to classify phases in a distinct model. The second approach employs Quantum Machine Learning (QML) algorithms with feature selection based on the SHAP (Shapley Additive Explanations) technique, optimizing the choice of relevant features to improve classifier accuracy. Finally, we introduce the use of Generative Adversarial Networks (GANs) to create synthetic data, enabling the expansion and diversification of datasets and, consequently, increasing the efficiency and accuracy of classifications. For the second challenge, the prediction of quantum synchronization, we apply a supervised learning algorithm to predict the occurrence of synchronization or antisynchronization in quantum systems. Using initial data from the system’s temporal evolution, we demonstrate how to identify patterns that indicate the future dynamics of these phenomena with high reliability. This thesis contributes by presenting innovative solutions and a comprehensive set of tools that connect artificial intelligence to the understanding of quantum systems. The results provide valuable insights into the comprehension and manipulation of these systems.Coordenação de Aperfeiçoamento de Pessoal de Nível Superior (CAPES)88887.607339/2021-0

Intrinsically synthesized melatonin in mitochondria and factors controlling its production

    No full text
The percentage of the total amount of melatonin produced in vertebrates that comes from the pineal is small (likely <5%) but, nevertheless, functionally highly noteworthy. The significance of pineal melatonin is that it is secreted cyclically such that it has a critical function in influencing not only the suprachiasmatic nucleus but clock genes that reside in perhaps every cell throughout the organism. Extrapineal melatonin, which may be synthesized in the mitochondria of all other cells in much larger amounts than that in the pineal gland has a different function than that derived from the pineal gland. Its synthesis is not circadian and it is not directly impacted by the photoperiodic environment. Also, melatonin from the extrapineal sites is not normally secreted into the blood stream; rather, it acts locally in its cell of synthesis or, possibly via paracrine mechanisms, on immediately adjacent cells. The functions of extrapineal melatonin include central roles in maintaining molecular and redox homeostasis and actions in resisting pathological processes due to its ability to directly or indirectly detoxify free radicals. The vast majority of organisms that exist on Earth lack a pineal gland so pineal-derived melatonin is unique to vertebrates. Evidence suggests that all invertebrates, protists and plants synthesized melatonin and they have no pineal homolog; thus, the production of melatonin by extrapineal cells in vertebrates should not be unexpected. While the factors that control pineal melatonin synthesis are well documented, the processes that regulate extrapineal melatonin production are undefined.Department of Cell Systems and Anatomy UT Health San Antonio Long School of MedicineApplied Biomedical Sciences University of the Incarnate Word School of Osteopathic MedicineDepartment of Structural and Functional Biology Institute of Biosciences of BotucatuDepartment of Biology Universidade Estadual Paulista (UNESP)Department of Neurosciences Centro de Investigacion Biomedica de Occidente Instituto Mexicano del Seguro SocialUniversidade Federal de Mato Grosso Do Sul, Campus de Três Lagoas, Três Lagoas, Mato Grosso Do SulDepartment of Biology Universidade Estadual Paulista (UNESP

Antibiofilm properties, cytotoxicity, and effect on protease activity of antibiotics and EGCG-based medications for endodontic purposes

    No full text
Objective: To evaluate the effect of two intracanal medications (IM) containing epigallocatechin-3-gallate (EGCG) with fosfomycin (FOSFO) and a triantibiotic combination of metronidazole, ciprofloxacin and fosfomycin (TRI), compared to controls calcium hydroxide (CH), all dissolved in polyethylene glycol 400 (PEG) on multispecies biofilms, fibroblast toxicity and on collagenolytic and gelatinolytic activities detected in radicular dentin. Methods: The antibiofilm effect and cytotoxicity of medications containing EGCG + FOSFO, TRI or CH were evaluated on multispecies biofilms formed in bovine root dentin specimens by confocal microscopy and on fibroblasts by resazurin assays, respectively. The inhibition of protease activity of each IM was evaluated by measuring collagenolytic enzyme activity by ELISA (Enzyme-linked immunosorbent assay) and gelatinolytic activity by metalloproteinases (MMPs) using in situ zymography in radicular dentin specimens. Results: PEG containing EGCG+FOSFO, PEG+TRI, and PEG+CH significantly reduced multispecies biofilms in radicular dentin tubules. At the concentrations tested, those IM were not toxic to fibroblasts. Additionally, all IM presented anti-collagenolytic activity by reducing telopeptide fragments released from radicular dentin compared to PEG carrier and water controls. In situ gelatinolytic activity, assessed via fluorescence levels, was significantly lower in radicular dentin adjacent to PEG containing CH, EGCG+FOSFO, or TRI compared to PEG and water controls. Conclusion: EGCG+FOSFO and TRI in PEG-400 exhibited antibiofilm, anti-collagenolytic and anti-gelatinolytic properties at concentrations that were non-toxic to fibroblasts, making them feasible intracanal medications for endodontic applications. Clinical significance: EGCG-based medications enhance the efficacy of endodontic treatment by providing antibiofilm, anti-collagenolytic, and anti-gelatinolytic properties, contributing to the preservation of root structure and improved treatment outcomes.Coordenação de Aperfeiçoamento de Pessoal de Nível Superior (CAPES)Canadian Institutes of Health ResearchNatural Sciences and Engineering Research Council of CanadaSão Paulo State University (UNESP) Department of Preventive and Restorative Dentistry School of Dentistry, SPUniversity of Toronto Department of Clinical Sciences Faculty of DentistryOregon Health & Science University (OHSU) Division of Biomaterials and Biomechanics Department of Restorative DentistryNova Southeastern University College of DentistryUniversity of Toronto Department of Cell and Systems Biology Faculty of Arts and ScienceUniversidade Católica Portuguesa (UCP) Faculty of Dental Medicine Centre for Interdisciplinary Research in Health (CIIS)São Paulo State University (UNESP) Department of Preventive and Restorative Dentistry School of Dentistry, SPCAPES: 001Canadian Institutes of Health Research: PJT-178272Natural Sciences and Engineering Research Council of Canada: RGPIN-2020-0588

Proposing a short version of the Unesp-Botucatu pig acute pain scale using a novel application of machine learning technique

    No full text
Surgical castration of males is carried out on a large scale in the US swine industry and the pain resulting from this procedure can be assessed using the Unesp-Botucatu pig composite acute pain scale (UPAPS). We aim to propose a short version of UPAPS based on the behaviors best-ranked by a random forest algorithm. We used behavioral observations from databases of surgically castrated pre-weaned and weaned pigs. We trained a random forest algorithm using the pain-free (pre-castration) and painful (post-castration) conditions as target variable and the 17 UPAPS pain-altered behaviors as feature variables. We ranked the behaviors by their importance in diagnosing pain. The algorithm was refined using a backward step-up procedure, establishing the Short UPAPS. The predictive capacity of the original and short version of the UPAPS was estimated by the area under the curve (AUC). In refinement, the algorithm with the five best-ranked behaviors had the lowest complexity and predictive capacity equivalent to the algorithm with all behaviors. The AUC of Short UPAPS (89.62%) was statistically equivalent (p = 0.6828) to that of UPAPS (90.58%). In conclusion, the proposed Short UPAPS might facilitate the implementation of a standard operating procedure to monitor and diagnose acute pain post-castration in large-scale systems.Coordenação de Aperfeiçoamento de Pessoal de Nível Superior (CAPES)Laboratory of Applied Artificial Intelligence in Health Department of Anesthesiology Botucatu Medical School São Paulo State University (Unesp), São PauloDepartment of Veterinary Surgery and Animal Reproduction School of Veterinary Medicine and Animal Science São Paulo State University (Unesp), São PauloGlobal Production Animal Welfare Laboratory Department of Population Health and Pathobiology College of Veterinary Medicine North Carolina State University (NCSU)Department of Large Animal Clinical Sciences College of Veterinary Medicine Michigan State University (MSU)Laboratory of Applied Artificial Intelligence in Health Department of Anesthesiology Botucatu Medical School São Paulo State University (Unesp), São PauloDepartment of Veterinary Surgery and Animal Reproduction School of Veterinary Medicine and Animal Science São Paulo State University (Unesp), São PauloCAPES: 001CAPES: 00

Integrative taxonomy of Calcarea (Porifera) from Espírito Santo, Eastern Brazil

    No full text
Despite the biogeographical and ecological importance of the Eastern Brazil ecoregion as a transitional zone, this ecoregion still represents a taxonomic and distributional gap to the knowledge on macrobenthic invertebrates. Therefore, our aim was to narrow this gap studying the calcareous sponge diversity of this important marine ecoregion. Using morphological and molecular tools, 12 species were identified, four of them new to science: Ascandra polejaeffi sp. nov., Clathrina albata sp. nov., Clathrina capixaba sp. nov., and Vosmaeropsis anomala sp. nov., with new diagnostic proposals and an amendment of the genus Vosmaeropsis. One species known from the Eastern Caribbean ecoregion and from the Fernando de Noronha and Atoll das Rocas ecoregion is being registered for the Eastern Brazil ecoregion for the first time: Clathrina delicata. Seven species previously recorded for the Eastern Brazil ecoregion were recollected in this study, corroborating their occurrence and expanding their geographic distribution within it: Borojevia brasiliensis, Borojevia trispinata, Clathrina aurea, Clathrina soluta, Grantia kempfi, Paraleucilla magna and Sycettusa hastifera. These last two species are probably invasive in Brazil and are recorded in its northernmost location in the Southwestern Atlantic. Considering the total of 43 species registered in the Eastern Brazil ecoregion, seven are shared with the Amazonia, two with São Pedro e São Paulo Islands, seven with Fernando de Noronha and Atoll das Rocas, 13 with Northeastern Brazil, one with Trindade e Martin Vaz Islands, and 14 with Southeastern Brazil ecoregion. Eastern Brazil is the best-known ecoregion in the Brazilian coast and its role as a transitional zone between tropical and subtropical faunas is discussed.Central European UniversityComissão Nacional de Energia NuclearOffice of Diversity and InclusionBiodelFundação Grupo Boticário de Proteção à NaturezaEuropean CommissionFundação de Amparo à Pesquisa do Estado de São Paulo (FAPESP)Conselho Nacional de Desenvolvimento Científico e Tecnológico (CNPq)Coordenação de Aperfeiçoamento de Pessoal de Nível Superior (CAPES)Fundação de Amparo à Pesquisa do Estado do Rio de Janeiro (FAPERJ)Federal University of Rio de Janeiro Biology Institute Zoology Department TaxoN Laboratory, Av. Core-Shell UCNP@MOF Nanoplatforms for Dual Stimuli-Responsive Doxorubicin Release

    No full text
Nanocarrier systems with multifunctional capabilities hold great potential for targeted cancer therapy, particularly for breast cancer treatment. Metal-organic frameworks (MOFs) are notable for their high porosity and, in some cases, biocompatibility, with ZIF-8 being particularly advantageous due to its pH-sensitive degradability, enabling selective drug release in tumor environments. Additionally, lanthanide-doped upconversion nanoparticles (UCNPs) offer unique optical properties that enhance both imaging and therapeutic applications. In this study, NaYF4/Yb3+Er3+ UCNPs were synthesized via a hydrothermal method, subsequently coated with poly(acrylic acid) (PAA) and encapsulated within a ZIF-8 shell, forming of UCNP@ZIF-8 core-shell nanocomposites. This system was designed to leverage stimulation by a 980 nm laser and acidic pH to facilitate drug release. When exposed to this specific laser wavelength, the nanocomposites demonstrated significantly enhanced drug release, achieving up to 90% release of the incorporated antitumor drug, doxorubicin (DOX), in acidic environments. In vitro studies indicated selective cytotoxicity, with MCF-7 tumor cell viability decreasing from 85.7% to 20% following laser activation, while showing minimal toxicity toward healthy cells. These findings underscore the potential of the UCNP@ZIF-8 nanocarrier system as a pH and laser-responsive platform for improved cancer therapy, enabling precise control over drug delivery while minimizing side effects on surrounding healthy tissues.Institute of Chemistry São Paulo State University, São PauloFederal University of ABC UFABC, São PauloInstitute of Chemistry São Paulo State University, São Paul
