Microgeographical Variation in Dirofilaria immitis Prevalence in Dogs in Suburban and Urban Areas of Rio De Janeiro, Brazil
Dirofilariosis, a mosquito-borne disease caused by Dirofilaria immitis and Dirofilaria repens, affects canids, felids and occasionally humans. Recent evidence suggests that D. immitis prevalence is rising in the canine populations in several areas of Brazil, even those historically considered to be non-endemic, highlighting the need for ongoing surveillance. However, prevalence studies are frequently based on inference from single diagnostic methods, and it is acknowledged that this may lead to biases and an underestimation of the disease situation. Thus, this cross-sectional study aimed to assess the prevalence of Dirofilaria spp. infection in dogs and cats across suburban and urban areas in Rio de Janeiro, Brazil. The investigation was conducted using a multi-diagnostic approach with binomial generalised linear models (GLMs) applied to investigate the association between the occurrence of infection and potential disease determinants. Blood samples from 497 dogs and 107 cats, collected at a veterinary mobile neutering unit, underwent parasitological, serological and molecular testing to detect Dirofilaria spp. infections. Among the dog samples, an overall 7.44% tested positive for D. immitis infection, while D. repens was not detected. In contrast, no cat samples tested positive for either parasite species. Prevalence was notably higher in suburban areas (14.47%) than in the urban area of Rio de Janeiro (6.17%) (p = 0.003). Dogs aged over two years had a higher risk of being infected (p < 0.001), while features such as breed, sex, size, coat type, use of macrocyclic lactones or access to outdoor spaces were not found to be predictive factors. This study reinforces the evidence that the prevalence of D. immitis is increasing in areas previously considered to have low prevalence or to be non-endemic. It also suggests that environmental and socioeconomic factors may play a key role in determining the current epidemiological status of this parasite. Perspectives on telecollaboration In English language: a case study in the language course
A pandemia da COVID-19 intensificou a necessidade do uso das Tecnologias Digitais de Informação e Comunicação (TDICs) no contexto educacional, levando à reavaliação das práticas pedagógicas com foco na utilização de recursos digitais. As TDICs permitem a superação de barreiras geográficas e temporais a um custo acessível, oferecendo oportunidades para a internacionalização em casa (Salomão; Júnior, 2020) e o enriquecimento das práticas pedagógicas. Nesse cenário, a telecolaboração, colaboração on-line, intercâmbio virtual, termos intercambiáveis, aqui, emergem como uma estratégia viável, utilizando ferramentas de comunicação online para conectar estudantes de línguas em diferentes países, visando ao desenvolvimento de projetos colaborativos e ao intercâmbio intercultural (O'Dowd; Ritter, 2006). Nesta pesquisa, estudantes brasileiros e aprendizes de diversas partes do mundo utilizam a língua inglesa como língua franca para discutir temas variados na construção conjunta de conhecimento. O objetivo deste estudo foi investigar o impacto da telecolaboração na formação de professores de língua inglesa, especialmente entre alunos do último ano do curso de Letras da Unesp, campus de Assis. Utilizando-se da metodologia de estudo de caso proposta por Yin (2015), foi possível identificar que os participantes expressaram suas inseguranças mas, também, colocaram em prática seus conhecimentos linguísticos e culturais. A coleta de dados foi realizada por meio de questionários no Google Formulários, do diário reflexivo da pesquisadora e de gravações feitas pelos participantes acerca de suas impressões e aprendizagens ao longo das interações. Este estudo pretende contribuir para o avanço das práticas de ensino nos cursos de graduação em Letras, com vistas a um processo de formação mais integrador e que contemple demandas atuais, minimizando custos e ampliando as perspectivas docentes.The Covid-19 pandemic intensified the need for the use of Digital Information and Communication Technologies (DICTs) in the educational context, leading to the reassessment of the pedagogical practices focused on the use of digital resources. DICTs allow the overcoming of geographical and time barriers at an affordable cost, offering opportunities for at-home internationalization (Salomão; Júnior, 2020) and pedagogical practices’ enrichment. In this scenario, telecollaboration, online collaboration, virtual exchange, interchangeable terms, here, emerge as a viable strategy, using online communication tools to connect language students from different countries, aiming for the development of collaborative projects and intercultural exchange. In this research, Brazilian students and learners from all around the world use the English language as lingua franca to discuss several themes on the joint construction of knowledge. The objective of this study was to investigate the impact of telecollaboration on the training of English language teachers, especially among final-year students of the Language and Literature program from Unesp, Assis campus. Using the qualitative case study methodology proposed by Yin (2015), it was possible to identify that participants expressed their insecurities but also put into practice their linguistic and cultural knowledge. Data collection was carried out through questionnaires on Google Forms, from the researcher’s reflective journal and recordings made by the participants about their impressions and learning throughout the interactions. A novel GLUT-4 electrochemical immunosensor based on a poly(thionine)-gold nanoparticle nanocomposite: Combining complex capacitance and dissolved oxygen to obtain an analytical signal
Detection of glucose transporter 4 (GLUT4) is essential for understanding various physiological and pathological processes. This work reports the development of a novel electrochemical immunosensor for the direct detection of GLUT4, employing dissolved oxygen as a redox probe. This molecular oxygen-sensitive response is mediated by a redox-conductive polymer based on thionine. The sensor platform was fabricated via a one-step electropolymerization of thionine and gold nanoparticles (AuNPs) onto a platinum screen-printed electrode (Olean-Oliveira et al., 2022a). The immunosensor was then constructed by physical adsorption of a GLUT4 antibody onto the poly(thionine)-AuNP composite surface. This label-free approach eliminates the need for secondary antibodies or enzymes. The immunosensor performance was evaluated using electrochemical impedance spectroscopy (EIS). The sensing mechanism relies on impedance changes; increasing GLUT4 concentrations lead to increased impedance due to enhanced surface blocking upon GLUT4-antibody binding. This interaction impedes oxygen diffusion to the polymer redox sites, resulting in increased electrical resistance. Development of a natural rubber latex-based biodevice with mesenchymal stem cells as a potential treatment for skeletal muscle regeneration in gestational diabetes-induced myopathy
Women with gestational diabetes mellitus show a high risk of developing Gestational Diabetes Induced Myopathy (GDiM). GDiM is characterized by significant pelvic floor skeletal muscle atrophy and urinary incontinence. This study aimed to develop a natural rubber latex (NRL) based biodevice with mesenchymal/stromal stem cells (MSCs) for skeletal muscle regeneration for women with GDiM. NRL showed porosity, roughness, biocompatibility, and bioactivity. MSCs adhesion on the NRL scaffold surface was assessed by scanning electron microscopy (SEM), confocal microscopy, and zymography. The scaffold's physicochemical and biological properties were carried out by Fourier transform infrared spectroscopy (FTIR), swelling and degradation studies, hemolytic activity, and antioxidant activity (AA), using Electronic Paramagnetic Resonance (EPR). MSCs in culture expressed CD90, adhered to plastic, differentiated, and produced fibroblast colonies. A high rate of cell proliferation was seen in MSCs on the NRL scaffold. FTIR analysis confirmed protein structures and polyisoprene in the scaffold. Swelling and degradation showed low water uptake and weight loss. Furthermore, NRL presented a hemolytic rate of 2.90 ± 0.26 % for 24 h, and EPR revealed the scaffold's strong AA. Potential of xenogeneic and synthetic biomaterials associated with absorbable membrane for bone augmentation in rat calvaria: prospective study in rats
As propriedades e respostas clínicas dos biomateriais usados na regeneração óssea guiada tem ganhado cada vez mais destaque na literatura. Os diferentes padrões de reabsorção, osteocondução e bioatividade associados ao tipo de reconstrução desejada influenciam a escolha do substituto ósseo. Este estudo comparou a neoformação óssea vertical e a topografia entre biomaterial xenógeno e sintético. Trinta e dois ratos machos Wistar foram divididos aleatoriamente em dois grupos e submetidos à reconstrução óssea vertical em calvária com biomaterial xenógeno (Bio-Oss®/GBO) ou sintético (GenPhos XP®/GGP), ambos recobertos com membrana absorvível (Techgraft®). A eutanásia foi realizada após 60 e 90 dias da cirurgia, seguida por análise microarquitetural, topográfica e histométrica. Comparação estatística inferencial por Teste T (histometria) e Mann-Whitney (microarquitetura) foi realizada adotando significância de 5%. Parâmetros semelhantes de microarquitetura óssea foram observados (p>0,05), com maior neoformação, trabéculas mais espessas e menos espaçadas e menor porosidade para o grupo GGP. Na caracterização topográfica, o grupo GGP apresentou superfície mais rugosa e complexa, com visualização de trabéculas em maior aumento. A avaliação histométrica indicou maior neoformação no grupo GGP (p=0,001) e menor remanescente de biomaterial (p=0,003). O grupo GBO apresentou diminuição do osso neoformado entre os tempos (p=0,013) e maior redução da altura do volume reconstruído (p=0,03). Ambos os biomateriais demonstraram papel osteocondutor, com padrões de neoformação e reabsorção diferentes. Apesar da maior absorção do biomaterial sintético, esse apresentou neoformação óssea otimizada e manutenção do volume em avaliação de 90 dias.The properties and clinical responses of biomaterials used in guided bone regeneration have gained increasing prominence in literature. Differences in resorption, osteoconduction, and bioactivity, along the bone reconstruction desired, influence the choice of bone substitute. This study compared vertical bone formation and surface topography between xenogenic and synthetic biomaterials. Thirty-two male Wistar rats were randomly divided into two groups and underwent vertical bone reconstruction in the calvaria using either a xenogeneic (Bio-Oss®/GBO) or a synthetic biomaterial (GenPhos XP®/GGP), both covered with an absorbable membrane (Techgraft®). Euthanasia was performed at 60 and 90 days post-surgery, followed by microarchitectural, topographic, and histometric analyses. Inferential statistical analysis was performed using the T-test (histometry) and Mann-Whitney test (microarchitecture), adopting a 5% significance level. Similar bone microarchitectural parameters were observed between groups (p>0.05). GGP group show greater bone formation, thicker and less spaced trabeculae, and lower porosity. Topographic characterization of GGP group exhibited a rougher and more complex surface, with visualization of trabeculae at higher magnification. Histometric analysis indicated greater bone formation in the GGP group (p=0.001) and lower residual biomaterial (p=0.003). The GBO group exhibited a decrease in the newly formed bone (p=0.013) and in the reconstructed volume height (p=0.03) over time. Both biomaterials demonstrated osteoconductive potential, with distinct patterns of bone formation and resorption. Beta-lactam and vancomycin resistance profile in Staphylococcus epidermidis isolated from blood cultures
Staphylococcus epidermidis integra a microbiota humana saudável, mas pode se manifestar como um patógeno oportunista, especialmente em pacientes idosos, neonatos e imunossuprimidos, sendo frequentemente associado ao uso de dispositivos médicos invasivos. A alta resistência aos antimicrobianos e sua capacidade de atuar como reservatório de genes de resistência, com grande potencial de disseminação, são fatores que contribuem para sua persistência no ambiente hospitalar. Além disso, o rápido surgimento de resistência à meticilina em espécies de estafilococos nas últimas décadas tem representado um desafio significativo, prejudicando o tratamento eficaz das infecções. Sendo assim, esse estudo teve como objetivo caracterizar o perfil de resistência antimicrobiana, realizar análises fenotípicas e genotípicas de amostras clínicas de S. epidermidis isoladas de hemoculturas, além de utilizar técnicas epidemiológicas para identificar linhagens multirresistentes em um período de treze anos, em enfermarias de um hospital universitário brasileiro. Foram analisadas 405 amostras de S. epidermidis isoladas de hemoculturas de pacientes internados em um hospital universitário brasileiro entre 2009 e 2022. A detecção do gene mecA foi realizada para determinar a resistência à meticilina. A suscetibilidade antimicrobiana foi avaliada por métodos fenotípicos, e a tipagem molecular dos isolados foi conduzida para caracterização dos elementos SCCmec e investigação de genes de resistência. Dos 405 isolados analisados, 340 (84%) apresentaram resistência à meticilina pela presença do gene mecA, enquanto 65 (16%) foram sensíveis. A análise comparativa entre os discos de oxacilina e cefoxitina evidenciou que o potencial de detecção da resistência aos beta-lactâmicos difere entre eles. A oxacilina demonstrou maior potencial discriminatório, sendo mais eficaz na identificação de isolados resistentes. O perfil de suscetibilidade antimicrobiana revelou altas taxas de resistência aos discos de oxacilina (78,5%), cefoxitina (65,4%), eritromicina (72,8%) e ciprofloxacino (45,1%). Treze (3,2%) isolados foram suscetíveis dependentes da dose à ceftarolina, e onze (2,7%) apresentaram heterorresistência à vancomicina. Nenhum isolado apresentou resistência à linezolida. A taxa de multirresistência foi de 58,8%. A tipagem de SCCmec mais prevalente foi o tipo I (53,5%), comumente associada ao ambiente hospitalar, seguida do tipo IV (14,7%). Observou-se uma grande variabilidade genética em relação ao SCCmec, com isolados apresentando tipagens SCCmec coexistentes e elementos pouco frequentes, como o tipo IX. Um isolado multirresistente do pronto-socorro apresentou o gene vanA, de resistência à vancomicina. Destacou-se, ainda, a diversidade genética significativa desses isolados, com a presença de muitos clones em diferentes enfermarias. Esses achados reforçam a necessidade de vigilância molecular contínua e de estratégias rigorosas de controle de infecção para conter e mitigar o impacto de S. epidermidis na saúde pública.Staphylococcus epidermidis is part of the healthy human microbiota, but it can manifest itself as an opportunistic pathogen, especially in elderly patients, neonates, and immunosuppressed individuals, and is frequently associated with the use of invasive medical devices. Its high resistance to antimicrobials and its ability to act as a reservoir of resistance genes, with great potential for dissemination, are factors that contribute to its persistence in the hospital environment. In addition, the rapid emergence of methicillin resistance in staphylococcal species in recent decades has represented a significant challenge, hindering the effective treatment of infections. Therefore, this study aimed to characterize the antimicrobial resistance profile, perform phenotypic and genotypic analyses of clinical samples of S. epidermidis isolated from blood cultures, and use epidemiological techniques to identify multidrug-resistant strains over a period of thirteen years in wards of a Brazilian university hospital. A total of 405 S. epidermidis samples isolated from blood cultures of patients admitted to a Brazilian university hospital between 2009 and 2022 were analyzed. Detection of the mecA gene was performed to determine methicillin resistance. Antimicrobial susceptibility was assessed by phenotypic methods, and molecular typing of the isolates was conducted to characterize SCCmec elements and investigate resistance genes. Of the 405 isolates analyzed, 340 (84%) showed resistance to methicillin due to the presence of the mecA gene, while 65 (16%) were sensitive. Comparative analysis between oxacillin and cefoxitin discs showed that the potential for detecting beta-lactam resistance differs between them. Oxacillin demonstrated greater discriminatory potential, being more effective in identifying resistant isolates. The antimicrobial susceptibility profile revealed high rates of resistance to oxacillin (78.5%), cefoxitin (65.4%), erythromycin (72.8%), and ciprofloxacin (45.1%) disks. Thirteen (3.2%) isolates were dose-dependently susceptible to ceftaroline, and eleven (2.7%) showed heteroresistance to vancomycin. No isolates showed resistance to linezolid. The multidrug resistance rate was 58.8%. The most prevalent SCCmec typing was type I (53.5%), commonly associated with the hospital environment, followed by type IV (14.7%). A great genetic variability was observed in relation to SCCmec, with isolates presenting coexisting SCCmec typings and infrequent elements, such as type IX. One multidrug-resistant isolate from the emergency room presented the vanA gene, resistance to vancomycin. The significant genetic diversity of these isolates was also highlighted, with the presence of many clones in different wards. Comportamentos interativos entre mães e seus bebês com risco para deficiência auditiva
Objective In the present study, we intended to describe and compare the interaction between mothers and infants at risk for hearing loss (Group 1) and mothers and infants who could hear (Group 2). Method A total of 25 dyads participated in each group and were filmed while free playing. The films were examined using the observation adapted protocol Mother-Child Interaction Coding System, for later comparison. Positive interactive behaviors were significantly more frequent among infants in Group 2. Results Infants in Group 1 showed more non-interactive behaviors than the other group. Mothers in Group 2 showed more positive sensitive and less non-responsive behaviors than mothers in Group 1. A Volume-Stable Collagen Matrix (VCMX) to Treat Single Gingival Recessions Associated with Partially Restored Noncarious Cervical Lesions: A Case Series
Collagen matrices have been developed as possible connective tissue graft substitutes to overcome patient discomfort, undesired palatal healing, and the limited amount of donor tissue. The aim of this case series is to assess a coronally advanced flap (CAF) with a volume-stable collagen matrix (VCMX) to treat single gingival recessions (GRs) associated with partially restored noncarious cervical lesions (NCCLs). Twelve patients diagnosed with single GRs (type 1; RT1) + NCCL (B+) were included in this study and received a restorative approach (partial composite resin with an apical margin 1 mm beyond the estimated cementoenamel junction [CEJ]) and a surgical approach (CAF+VCMX). Clinical and patient-centered assessments were recorded from 10 patients at baseline and 6 months postoperatively. Significant recession reduction (2.1 mm), clinical attachment level gain (1.34 mm), and combined defect coverage (51.67%) were observed at 6 months. The estimated root coverage was 69.48%, obtained using the estimated CEJ. No difference in keratinized tissue width was observed over time. A 0.42-mm gain in gingival thickness was observed after 6 months (P = .002) as well as an improvement in patient satisfaction due to better esthetics (P < .001). Synthesis and Characterization of Self-Healing Polymers Obtained from Polyphenols and Cyclic Carbonates of Amide Derivative of Macaw Palm Oil
In this work, the Brazilian biomass – macaw palm oil, was reacted with tris(2-aminoethyl)amine to give a fatty amide derivative, which was further modified to obtain an epoxidised derivative. After that, the last derivative reacted with CO2, providing a new carbonated compound. The vegetable oil-based products were analysed using spectroscopic techniques and showed fluorescence when exposed to UV light. The carbonated compound was polymerised with four different polyphenols (quercetin, curcumin, tannic acid, and gallic acid). This resulted in dark brown, thermosetting poly-hydroxycarbonates with a shiny and brittle appearance. Modified vegetable oil compounds and final polymers present bio-based and bio-based carbon contents above 88%, indicating their renewability. Polymers made with quercetin and gallic acid were hydrophobic (water-repellent). Thermal analysis confirmed that all polymers could withstand temperatures up to 195 °C before decomposing. In addition, they have resistance against acid solutions but are fully hydrolysed after 24 h in an alkaline solution. Expression of MHC I Isoforms in Bovine Placentomes: Impact of Cloning
Major histocompatibility complex class I (MHC-I) gene expression in the placenta is modulated to tailor the maternal immune response to fetal antigens during pregnancy. This study evaluated MHC-I expression through immunohistochemistry (IHC) using an anti-mouse preimplantation embryo development (PED) clone Qa-2 and anti-bovine leukocyte antigen I (BoLA) monoclonal antibody clone IL-A88 (n = 23), as well as RT-qPCR (n = 17) for classical and non-classical (BoLA-NC) genes in control and cloned bovine placentomes during early and near-term gestation. Control samples showed minimal Qa-2 protein expression in early gestation, with intense labeling in trophoblasts and the maternal uterine epithelium near term. In contrast, cloned samples exhibited intense Qa-2 labeling in both maternal and trophoblastic epithelia at both stages, while trophoblast giant cells (TGCs), located apposed to the maternal epithelium, showed no labeling. Control samples exhibited intense IL-A88 labeling in the maternal epithelium at both stages. In cloned samples, weak to no labeling was observed in early gestation, with intense labeling in maternal and fetal epithelium near term. RT-qPCR revealed significant upregulation of BoLA-NC3 in early gestation, with sustained elevated expression in cloned samples in the near term.
