Sustainable Water Management for Steam Generation in Sugarcane Biorefineries: Applying PCA and MST Clustering in Sample Analysis
Our study on sustainable water management in sugarcane biorefineries, which utilizes water as a primary resource for generating bioenergy through steam production, has employed a novel approach. High water quality is crucial for optimal efficiency, particularly in boiler operations. We have utilized unsupervised methods, such as principal component analysis (PCA) and minimum spanning tree (MST), alongside instrumental analysis data, to assess water quality in steam production. The PCA exploratory analysis identified three distinct clusters, with the relevant variables being conductivity and SiO2 content, to differentiate the purity of a dataset of 120 samples. MST-based clustering corroborated the PCA findings, forming three clusters: sample 1 represented the purest water, while samples 3 and 6 were in different clusters, indicating less purity in boiler feedwater. These unsupervised methods are highly effective, providing accurate and reliable data analysis and significantly benefiting sugarcane biorefineries by eliminating subjective biases. Euclid preparation: LXIV. The Cosmic Dawn Survey (DAWN) of the Euclid Deep and Auxiliary Fields
Euclid will provide deep near-infrared (NIR) imaging to ∼26.5 AB magnitude over ∼59 deg2 in its deep and auxiliary fields. The Cosmic DAWN survey combines dedicated and archival UV- NIR observations to provide matched depth multiwavelength imaging of the Euclid deep and auxiliary fields. The DAWN survey will provide consistently measured Euclid NIR-selected photometric catalogues, accurate photometric redshifts, and measurements of galaxy properties to a redshift of z ∼ 10. The DAWN catalogues include Spitzer IRAC data that are critical for stellar mass measurements at z ≳ 2.5 and high-z science. These catalogues complement the standard Euclid catalogues, which will not include Spitzer IRAC data. Identification of a novel GP60 subtype family of Cryptosporidium wrairi from capybaras (Hydrochoerus hydrochaeris) inhabiting urban areas in the state of Mato Grosso do Sul, Brazil
Capybaras (Hydrochoerus hydrochaeris) are hosts for several parasites of public health importance, including Cryptosporidium spp. Therefore, this study aimed to perform the molecular characterization of Cryptosporidium spp. in fecal samples from capybaras inhabiting urban areas. We analyzed 401 fecal samples from capybaras in two municipalities of the state of Mato Grosso do Sul, Brazil. Fecal samples were purified using centrifugal sedimentation with ethyl acetate. They were then screened for Cryptosporidium spp. by malachite green negative staining and a nested PCR protocol targeting the 18S rRNA gene. Samples positive by microscopy or PCR were examined by PCR protocols targeting the actin, HSP-70, and GP60 genes. Amplicons from all PCR protocols were subjected to genetic sequencing. Microscopic screening and 18S rRNA gene-targeted PCR identified 0.25 % (1/401) and 0.5 % (2/401) of samples, respectively, as positive for Cryptosporidium spp. The genetic sequences of the 18S rRNA, HSP-70, actin, and GP60 genes showed genetic similarity to Cryptosporidium wrairi sequences of 99.07 %, 99.69 %, 99.57 %, and 91.51 %, respectively. Genetic sequencing and phylogenetic analyses identified the novel GP60 subtype family VIIbA13 of C. wrairi. In conclusion, we report in this study a low prevalence of the novel GP60 subtype family VIIbA13 of C. wrairi in free-living capybaras from urban areas of the state of Mato Grosso do Sul, Brazil. Genetic characterization of Araucaria angustifolia (Bertol.) kuntze populations across phytophysiognomies in the mixed ombrophilous forest
Araucaria angustifolia (Bertol.) Kuntze, símbolo da Floresta Ombrófila Mista (FOM), apresenta distribuição heterogênea, ocupando distintas fitofisionomias ao longo dessa formação vegetal. A intensa exploração madeireira no século XIX e a fragmentação do habitat colocaram a espécie em risco crítico de extinção. Assim, conservar os remanescentes é essencial, visto que estudos apontam significativa diversidade genética que deve ser explorada e preservada. Este estudo, realizado em parceria entre a Embrapa Florestas e a empresa Engie S.A., teve como objetivo mensurar a variabilidade genética e fenotípica de populações naturais de Araucaria angustifolia em diferentes fitofisionomias da FOM no estado do Paraná. Foram amostradas árvores para mensuração de caracteres fenotípicos relacionados ao crescimento e à qualidade da madeira, além da coleta de material genético para análises moleculares com marcadores microssatélites e amostras de madeira para estudo dos anéis de crescimento. Cada conjunto de dados foi analisado com softwares específicos para o cálculo dos parâmetros necessários. A partir dos resultados, foram estimadas as distâncias genéticas, fenotípicas e geográficas entre as populações, investigando possíveis relações entre estas por meio do coeficiente de correlação de Pearson e do teste de Mantel. Os caracteres fenotípicos apresentaram heterogeneidade intra e interpopulacional, com a altura sendo o caráter de maior divergência fenotípica (PST). O estudo de anéis revelou idades variando entre 54 e 158 anos, com incremento médio anual semelhante entre populações (0,57 cm/ano). Os parâmetros genéticos revelaram alta variabilidade e elevado fluxo gênico elevado entre e dentro das populações, sem diferenças significativas na estrutura genética entre as distintas fisionomias. O agrupamento por cluster e a baixa distância genética apontaram um único grupo de populações, indicando ausência de diferenciação genética entre elas, apesar da fragmentação. No entanto, a distância fenotípica separou a população de Ipiranga, sugerindo possível adaptação local desta a condições ambientais específicas, reforçada pelo alto PST em relação ao coeficiente de diferenciação (FST). Portanto, há a presença de variabilidade fenotípica nas populações, independentemente do tipo de fitofisionomia. E uma alta diversidade genética foi encontrada em todas as populações, com destaque para a riqueza alélica. Isso destaca a importância de conservar a espécie, considerando tanto sua diversidade quanto seu papel ecológico. A combinação desses dados genéticos e fenotípicos oferece uma base sólida para criar estratégias eficazes de conservação, melhoramento genético e uso sustentável da espécie.Araucaria angustifolia (Bertol.) Intense logging in the 19th century and habitat fragmentation placed the species at critical risk of extinction. Therefore, conserving the remaining populations is essential, as studies indicate significant genetic diversity that must be explored and preserved. This study, conducted in partnership between Embrapa Florestas and the company Engie S.A., aimed to measure the genetic and phenotypic variability of natural populations of Araucaria angustifolia in different phytophysiognomies of the FOM in the state of Paraná, Brazil. Trees were sampled to measure phenotypic traits related to growth and wood quality, along with the collection of genetic material for molecular analyses using microsatellite markers, and wood samples for dendrochronological studies. Each dataset was analyzed using specific software to calculate the necessary parameters. Based on the results, genetic, phenotypic, and geographic distances between populations were calculated, and potential relationships among them were investigated using Pearson's correlation coefficient and the Mantel test. Phenotypic traits showed heterogeneity within and between populations, with height presenting the highest and most significant phenotypic divergence (PST). The growth ring study revealed tree ages ranging from 54 to 158 years, with a similar mean annual increment among populations (0.57 cm/year). Genetic parameters indicated high variability and elevated gene flow within and among populations, with no significant differences in genetic structure even across distinct phytophysiognomies. Cluster analysis and low genetic distance values pointed to a single population group, indicating an absence of genetic differentiation despite fragmentation. However, phenotypic distance separated the Ipiranga population, suggesting possible local adaptation to specific environmental conditions, reinforced by its high PST relative to the genetic differentiation coefficient (FST). There is clear phenotypic variability present in the populations, regardless of phytophysiognomy type. Furthermore, high genetic diversity was found in all populations, with particular emphasis on allelic richness. This highlights the crucial importance of conserving the species, considering both its diversity and its ecological role. ANÁLISE DE PALAVRAS-CHAVE DA PRODUÇÃO CIENTÍFICA DE PESQUISADORES: O AUTOR COMO INDEXADOR
Introduction: With the advent of the digital and web era, the keyword has become an essential representation product in open access information storage and retrieval systems. It is used in the extraction functions for the purpose of scientific identification of authors, bibliometric analysis, indicators of scientific impact, development of controlled vocabularies and other systems of knowledge organization. The attribution of keywords by the author in scientific publications is a practice of representing the content carried out when filling in metadata. This assignment goes through personalized subject analysis and depends on the author's specialty vocabulary, which, in general, has no guidance on standardization and vocabulary control. On the other hand, such subject metadata that receives the keyword does not undergo professional validation. Objective: Analysis of keywords assigned by researchers to submit articles from journals indexed in Scopus and in the Portal Professores Unesp regarding the standardization and vocabulary control for different functions in information storage and retrieval systems. Methodology: For this purpose, an exploratory research was carried out with an observation study and analysis of keywords attributed by researchers from Portal Docentes Unesp based on data from the CNPq Lattes Curriculum compared to keywords attributed to journal articles. Results: The results demonstrate an absence of standardization in the keywords of the journal articles of researchers registered in the Lattes Curriculum, both at syntactic and semantic level. Regarding the assessment of indexation, low levels of consistency are observed when compared to the original articles, both in the rigid index and in the relaxed / flexible index. Simulating graphene-based single-electron transistor: incoherent current effects due to the presence of electron-electron interaction
Carbon-based nanostructures have unparalleled electronic properties. At the same time, using an allotrope of carbon as the contacts can yield better device control and reproducibility. In this work, we simulate a single-electron transistor composed of a segment of a graphene nanoribbon coupled to carbon nanotubes electrodes. Using the non-equilibrium Green’s function formalism we atomistically describe the electronic transport properties of the system including electron-electron interactions. Using this methodology we are able to recover experimentally observed phenomena, such as the Coulomb blockade, as well as the corresponding Coulomb diamonds. Phenol-free in-house kit for RNA extraction with applicability to SARS-CoV-2 genomic sequencing studies: A contribution to biotechnological sovereignty in Colombia
During the COVID-19 pandemic, reagents for SARS-CoV-2 detection were scarce or sold at high prices, particularly in Latin America. In this study, a significant step towards self-sufficiency was achieved through the development of an in-house extraction kit for detecting SARS-CoV-2 from nasopharyngeal swab samples. The purity and concentration of the RNA extracted using the in-house kit were compared to those obtained using the GeneJET RNA Purification Kit (Thermo-Scientific®) as a reference. The applicability of the RNA extracted using the kit was evaluated using four samples positive for SARS-CoV-2 by NGS sequencing with Illumina®. There were no significant differences between the results obtained with the in-house kit and those obtained with the commercial kit. These findings confirm that the in-house protocol demonstrated satisfactory diagnostic accuracy for detecting the virus in patients with COVID-19. The in-house extraction kit works effectively, providing optimal RNA extraction for genomic characterization and lineage assignment of SARS-CoV-2 within the four positive samples analyzed. This phenol-free kit represents a local design and production achievement, offering an effective solution for RNA extraction and detection and sequencing of SARS-CoV-2 from nasopharyngeal swabs. Smart Textile Electrochemical Capacitive Biosensor for Real-Time Monkeypox Virus Detection
The latest outbreak caused by monkeypox virus (MPXV) has turned into an international public health emergency, underscoring the urgent need for rapid, large-scale, and sensitive diagnostic tests for MPXV. Here, a capacitive biosensor for detecting MPXV was developed using a laser-scribed graphene (LSG) sensor manufactured on synthetic aramid fiber. The aramid-LSG sensor was modified with monoclonal antibodies for detecting MPXV through electrochemical capacitance measurements (Cμ-). The electrochemical detection was performed using a system of two interdigitated electrodes, providing excellent reproducibility and without cross-reactivity in the presence of other poxviruses and nonpoxviruses. Also, the wearable textile biosensor achieved a LOD of 7.5 × 10-1 PFU mL-1 and a LOQ of 2.4 × 100 PFU mL-1, enabling its application in plasma, saliva, and PBS samples (simulating application to human skin containing the virus). Furthermore, cytotoxicity assay studies demonstrated that the device is safe to use, according to the in vitro studies employing 3T3 cell cultures. Francisca Senhorinha da Motta Diniz: the state of knowledge in academic research between 1982 and 2023
Objetivou-se elaborar o estado do conhecimento das pesquisas sobre a vida e a obra de Francisca Senhorinha da Motta Diniz a partir do conjunto de referências e resumos, com datas de produção e divulgação entre 1982 e 2023, que foram localizados, recuperados, reunidos e ordenados no instrumento de pesquisa e analisados neste texto. Tendo em vista os referenciais teóricos de Belloto (1979), Duarte (2017), Ferreira (1999), Mortatti (1999, 2021), Morosini (2021) e Soares (2000), analisaram-se 12 referências de textos e seus respectivos resumos, quando houver, de trabalhos acadêmicos com fins de titulação e avaliação compreendendo duas teses, cinco dissertações e cinco monografias de trabalhos de conclusão de curso, mediante a metodologia de análise da configuração textual aplicada ao conjunto das referências reunidas. Apresentam-se, neste texto, os antecedentes da pesquisa, o conjunto de procedimentos de elaboração do instrumento de pesquisa, os aspectos da vida e obra de Diniz, a análise das temáticas, os objetivos, as metodologias, os autores, as instituições e os períodos de produção das pesquisas selecionadas, bem como as perspectivas e as contribuições desses trabalhos. Nesse sentido, compreendeu-se que o estado do conhecimento alcançado por tais pesquisas destacou a importância da imprensa feminina oitocentista, o conceito de emancipação veiculado pela autora no jornal e as estratégias argumentativas de Diniz para compor seu discurso. A análise, portanto, evidenciou Diniz como uma figura histórica de importância para a aquisição de direitos para mulheres no Brasil, principalmente no acesso à educação.The objective was to develop the state of knowledge regarding research on the life and work of Francisca Senhorinha da Motta Diniz, based on a set of references and abstracts produced and disseminated between 1982 and 2023. These references were located, retrieved, compiled, and organized in the research instrument and analyzed in this text. Considering the theoretical frameworks of Belloto (1979), Duarte (2017), Ferreira (1999), Mortatti (1999, 2021), Morosini (2021), and Soares (2000), 12 references of texts and their respective abstracts (when available) were analyzed. These references include academic works aimed at obtaining degrees and evaluations, comprising two theses, five dissertations, and five undergraduate monographs. The methodology applied was the análise da configuração textual to the collected references. This text presents the background of the research, the set of procedures used in the development of the research instrument, aspects of Diniz’s life and work, the analysis of themes, objectives, methodologies, authors, institutions, and periods of production of the selected studies, as well as the perspectives and contributions of these works. In this regard, it was understood that the state of knowledge achieved by these studies highlighted the importance of the 19th-century women’s press, the concept of emancipation conveyed by the author in the newspaper, and Diniz’s argumentative strategies in shaping her discourse. The analysis, therefore, revealed Diniz as a historical figure of great significance for the advancement of women's rights in Brazil, particularly in access to education
Head morphology and diet of whiptail catfishes (Loricariidae: Loricariinae)
Adaptations in the head structures of fishes can influence food item selection and consumption in aquatic ecosystems. In this study, we tested the relationship between the morphology of the bony structure of the head (suspensorium, mandibular, hyoid, and branchial arches) and the consumption of food items in Loricariinae species. We hypothesize that osteological specializations in the head structures (i.e., mouth) can lead to differential consumption of resources. Despite the relatively high trophic similarity, food item consumption was not uniform among Loricariinae species. The phylogenetic reconstruction recovered morphological patterns already expected for Loricariinae, such as the sister taxa Hemiodontichthys and Reganella. Phylogenetic characters associated with shape, width, and structures in the suspensorium, and branchial arches were correlated with differences in detritus, sediment, and invertebrates consumption. Specifically, branchial arches characters were associated with the consumption of aquatic and terrestrial invertebrates, while suspensorium characters were linked to the consumption of sediment, superior plants, detritus, and aquatic invertebrates. These findings suggest that trophic specialization in loricariines is strongly influenced by the morphological differentiation of these structural features.Adaptações nas estruturas bucais dos peixes podem influenciar a seleção e o consumo de itens alimentares nos ecossistemas aquáticos. Neste estudo, testamos a relação entre a morfologia de estruturas ósseas do crânio (suspensório, arco mandibular, arco hioide e arcos branquiais) e o consumo de itens alimentares em Loricariinae. Nossa hipótese é que especializações osteológicas nas estruturas do crânio (i.e., boca) podem levar a um consumo diferenciado de recursos alimentares. Apesar da similaridade trófica relativamente alta, o padrão de consumo de itens alimentares não foi uniforme entre as espécies de Loricariinae. A reconstrução filogenética recuperou padrões morfológicos já esperados para Loricariinae, como os táxons irmãos Hemiodontichthys e Reganella. Caracteres filogenéticos associados à forma, largura e estruturas no suspensório e nos arcos branquiais foram correlacionados com diferenças no consumo de detritos, sedimentos e invertebrados. Em particular, os caracteres dos arcos branquiais foram associados ao consumo de invertebrados aquáticos e terrestres, enquanto os caracteres do suspensório foram associados ao consumo de sedimentos, plantas superiores, detritos e invertebrados aquáticos.
