    Ciliated muconodular papillary tumor masked by COVID-19 infection and aspergilloma

    Novel coronavirus disease-2019 (COVID-19) continues to pose a threat all over the world with the effect of new variants. The frequency of other secondary infections such as aspergilloma in patients with COVID-19 disease is not uncommon. Extremely rare lesions such as ciliated muconodular papillary tumor can be masked by radiological images caused by COVID-19 infection or secondary pathologies associated with it, leading to difficulties in diagnosis. Herein, we report a case in whom a diagnosis of aspergilloma and ciliated muconodular papillary tumor after six months of COVID-19 infection was made.4OCT635-6403

    In Vivo Renal Lipid Quantification by Accelerated Magnetic Resonance Spectroscopic Imaging at 3T: Feasibility and Reliability Study

    A reliable and practical renal-lipid quantification and imaging method is needed. Here, the feasibility of an accelerated MRSI method to map renal fat fractions (FF) at 3T and its repeatability were investigated. A 2D density-weighted concentric-ring-trajectory MRSI was used for accelerating the acquisition of 48 x 48 voxels (each of 0.25 mL spatial resolution) without respiratory navigation implementations. The data were collected over 512 complex-FID timepoints with a 1250 Hz spectral bandwidth. The MRSI sequence was designed with a metabolite-cycling technique for lipid-water separation. The in vivo repeatability performance of the sequence was assessed by conducting a test-reposition-retest study within healthy subjects. The coefficient of variation (CV) in the estimated FF from the test-retest measurements showed a high degree of repeatability of MRSI-FF (CV = 4.3 +/- 2.5\%). Additionally, the matching level of the spectral signature within the same anatomical region was also investigated, and their intrasubject repeatability was also high, with a small standard deviation (8.1 +/- 6.4\%). The MRSI acquisition duration was similar to 3 min only. The proposed MRSI technique can be a reliable technique to quantify and map renal metabolites within a clinically acceptable scan time at 3T that supports the future application of this technique for the non-invasive characterization of heterogeneous renal diseases and tumors.5MAY1

    Targeting mitochondrial DNA polymerase gamma for selective inhibition of MLH1 deficient colon cancer growth

    Synthetic lethality in DNA repair pathways is an important strategy for the selective treatment of cancer cells without harming healthy cells and developing cancer-specific drugs. The synthetic lethal interaction between the mismatch repair (MMR) protein, MutL homolog 1 (MLH1), and the mitochondrial base excision repair protein, DNA polymerasey (Poly) was used in this study for the selective treatment of MLH1 deficient cancers. Germline mutations in the MLH1 gene and aberrant MLH1 promoter methylation result in an increased risk of developing many cancers, including nonpolyposis colorectal and endometrial cancers. Because the inhibition of Poly in MLH1 deficient cancer cells provides the synthetic lethal selectivity, we conducted a comprehensive small molecule screening from various databases and chemical drug library molecules for novel Poly inhibitors that selectively kill MLH1 deficient cancer cells. We characterized these Poly inhibitor molecules in vitro and in vivo, and identified 3,3'-{[}(1,1'-Bipheny1)4',4'-diyl)bis(azo)]bis{[}4-amino-1-naphthalenesu lfonic acid] (congo redCRZinc 03830554) as a high-affinity binder to the Pol y protein and potent inhibitor of the Pol y strand displacement and one-nucleotide incorporation DNA synthesis activities in vitro and in vivo. CR reduced the cell proliferation of MLH1 deficient HCT116 human colon cancer cells and suppressed HCT116 xenograft tumor growth whereas it did not affect the MLH1 proficient cell proliferation and xenograft tumor growth. CR caused mitochondrial dysfunction and cell death by inhibiting Poly activity and oxidative mtDNA damage repair, increasing the production of reactive oxygen species and oxidative mtDNA damage in MLH1 deficient cells. This study suggests that the Pol y inhibitor, CR may be further evaluated for the MLH1 deficient cancers' therapy.61

    PEG4000 modified liposomes enhance the solubility of quercetin and improve the liposome functionality: in vitro characterization and the cellular efficacy

    Quercetin, a multifunctional therapeutic agent, is used in various types of cancer. However, its therapeutic effect is limited by virtue of poorly aqueous solubility and instability in the physiological medium. To overcome these limitations, we aimed (i) to design quercetin loaded Liposomes with unlinked-PEG4000 with regard to not only surface modification but also solubility enhancement, and (ii) to investigate the antineoplastic effects on HeLa cells. PEG4000 increased the quercetin solubility 2.2 fold. PEG4000 modified Liposomes displayed small particle size (254 +/- 69 nm), low polydispersity index (0.236 +/- 0.018), favorable zeta potential (-35.4 +/- 0.6 mV), high quercetin encapsulation efficiency (87.6 +/- 5.6\%), and drug loading (22.2 +/- 6.9\%). The homogeneity and particle size of stable PEGylated liposomes were proved by transmission electron microscopy. The drug release was reached up to 65.1 +/- 3.8\% in 6 h. The IC50 value of quercetin loaded PEGylated liposomes was 16.3 mu g/mL on HeLa cells, while that of quercetin was 88.3 mu g/mL. PEGylated liposomes remarkably hampered the adherence and colony formation ability of cells according to crystal violet staining tests. The convenience of PEGylated liposomes for the parenteral application was stated by the hemolysis assay. The high-throughput screening assays based on AO/PI staining proved the drastic decrease of viable cell count. Moreover, qPCR tests based on gene expression levels revealed that the quercetin loaded PEGylated liposomes treatment could be more effective than free quercetin on the mitochondrial apoptosis of lieLa cells. These promising results allow considering further in vivo studies for efficient cancer treatment with quercetin loaded PEG4000 modified liposomes.41011-10234

    Background: This study evaluates the clinical outcome of patients with robotic mitral valve replacement (MVR).Methods: Between January 2010 and April 2022, 117 consecutive patients underwent robotic MVR with or without additional cardiac procedures. All procedures were completed by a single surgical team with Da Vinci Robotic Systems. Perioperative variables and early clinical outcomes were recorded.Results: Mean age and EuroScore II of the patients were 57.1 +/- 12.9 and 5.1 +/- 5.7, respectively. Isolated MVR was performed in 55 (47.0\%) patients and combined cardiac procedures were performed in 62 (53.0\%) patients. Additional procedures included: ablation for atrial fibrillation, tricuspid valve replacement, tricuspid valve repair, left atrial appendix ligation, patent foramen ovale closure, left atrial thrombectomy and septal myectomy for hypertrophic obstructive cardiomyopathy. Mean cardiopulmonary bypass time and cross clamp time were 143 +/- 54 and 93 +/- 37 minutes, respectively. Mean intensive care unit stay time was 26.5 +/- 26.0 hours. Postoperative stroke was observed in one (0.9\%) patient and new onset renal failure was observed in two (1.7\%) patients. Perioperative and postoperative early mortality was observed in three (2.6\%) patients, which was lower than expected.Conclusions: Robotic MVR is feasible and can be performed with good early postoperative outcomes.     Generating ad creatives using deep learning for search advertising

    We generated advertisement creatives programmatically using deep neural networks. A landing page contains relevant text data, which can be used for generating advertisement creatives, i.e. ads. We treated the ad generation task as a text summarization problem and built a sequence to sequence model. In order to assess the validity of our approach, we conducted experiments on four datasets. Our empirical results showed that our model generated relevant ads on a template-based dataset with moderate hyperparameters. Training the model with more content increased the performance of the model, which we attributed to rigorous hyperparameter tune-up. The choice of word embedding used in the representation of the input altered the model's performance. When the source and the target shared common sequences during training, the model produced the best results.51882-18963

    Clinical and radiological features of COVID-19 infection in pediatric hematology-oncology and transplant patients

    Purpose: The goal of this study is to explain the characteristics of COVID-19 in this demographic, as well as the impact it had on pediatric cancer care during the pandemic. Materials and Methods: Fifteen COVID-19 patients diagnosed were studied retrospectively. Clinical, laboratory and radiological data were collected in relation to 15 patients under the age of 18 who tested positive for SarsCoV-2. Results: Of the 12117 pediatric hemato-oncological patients tested for COVID-19, 1125 patients showed symptoms, and 14 tested positive. The majority of cases suffered minor illnesses. A total of ten children required inpatient care, five needed oxygen support, and four required mechanical ventilation and later died. It was noted that the duration of real-time polymerase chain reaction (RT-PCR) positivity was prolonged in patients receiving intensive chemotherapy and/or immunosuppressive therapy. Patients' chemotherapy was delayed in all 15 (100\%) Sars-CoV-2-positive patients. Although changes visible on chest computed tomography (CT) imaging of children were mainly milder than in adults, radiological findings were more severe in patients who received relatively intensive cancer treatment. Conclusion: Children who have been treated for cancer or who have undergone a hematopoietic stem cell transplantation (HSCT) may be at greater risk of severe COVID-19 and should be under constant observation.1377-3884

    Allergic diseases and immunodeficiencies in children, lessons learnt from COVID-19 pandemic by 2022: A statement from the EAACI-section on pediatrics

    By the April 12, 2022, the COVID-19 pandemic had resulted in over half a billion people being infected worldwide. There have been 6.1 million deaths directly due to the infection, but the pandemic has had many more short- and long-term pervasive effects on the physical and mental health of the population. Allergic diseases are among the most prevalent noncommunicable chronic diseases in the pediatric population, and health-care professionals and researchers were seeking answers since the beginning of pandemic. Children are at lower risk of developing severe COVID-19 or dying from infection. Allergic diseases are not associated with a higher COVID-19 severity and mortality, apart from severe/poorly controlled asthma. The pandemic disrupted routine health care, but many mitigation strategies, including but not limited to telemedicine, were successfully implemented to continue delivery of high-standard care. Although children faced a multitude of pandemic-related issues, allergic conditions were effectively treated remotely while reduction in air pollution and lack of contact with outdoor allergens resulted in improvement, particularly respiratory allergies. There is no evidence to recommend substantial changes to usual management modalities of allergic conditions in children, including allergen immunotherapy and use of biologicals. Allergic children are not at greater risk of multisystem inflammatory syndrome development, but some associations with Long COVID were reported, although the data are limited, and further research is needed. This statement of the EAACI Section on Pediatrics provides recommendations based on the lessons learnt from the pandemic, as available evidence.10OCT3
