Identifying Highly Effective Fludarabine-Based Novel Target Cancer Therapy Agents By İn Silico Studies
Experimental Biomedical ResearchAim: To develop an alternative drug molecule design to fludarabine which is commonly used in chronic lymphocytic leukemia. Methods: The molecular properties and biological activities of the drug molecules were determined using Molinspiration software. We investigated the biological activity and drug properties of fludarabine by changing the positions of bioisosteres on the molecular structure. Results: In our studies of derivatives of the fludarabine drug molecule, we obtained data by adding different structures to the Y part without changing the X structure (F) of fludarabine. We have used the abbreviation 'M' to refer to the molecules in these experiments. We predict that the M6 derivative of fludarabine will have higher ion channel modulator, kinase and protease activity compared to fludarabine. We predict that the M15 derivative of fludarabine will have higher G- protein coupled receptors, ion channel modulator, kinase, and protease and enzyme inhibition activity compared to fludarabine. In our experiments with fludarabine derivatives, we have experimented by binding different molecules to both the X and Y structures of fludarabine at the same time. We have used the abbreviation 'C' to refer to the molecules in these experiments. In these experiments, we did not achieve higher biological activity than fludarabine. Conclusions: The results suggest that this newly designed M15 derivative of fludarabine molecule may be a better antileukemic drug molecules in the future and may be useful for further drug molecule development research in medical biochemistry, chemistry and pharmacology
The Relationship Between Anxiety Levels And Menstruation Symptoms In Adolescents
Balıkesir Sağlık Bilimleri Dergisi (BSBD)Objective: The aim of the study is to determine the relationship between anxiety levels and menstrual symptoms in adolescents. Material and Methods: The research is a descriptive and relationship-seeking research. 602 adolescents studying in two schools were invited to the study. The sample group of the study consisted of 544 adolescents who agreed to participate in the research. Data was collected using a personal information form, Menstrual Symptom Questionnaire, and Beck Anxiety Inventory. The relationship between two continuous variables was evaluated with the Pearson Correlation Coefficient. Statistical significance level was considered as p<0.05. Results: The BAI Subjective Anxiety subscale mean score was 28.49±8.28, the Somatic Symptom subscale mean score was 17.16±5.60, and the total mean score was 45.65±13.07. MSQ Negative effects/somatic complaints subscale mean score 33.75±10.59, pain subscale mean score 20.92±6.74, coping subscale mean score 62.64±18.59, and total mean score was 117.36±34.44. Conclusion: There is a positive, moderate level and statistically significant relationship between Menstrual Symptom Questionnaire total score and Beck Anxiety Inventory total score. Anxiety level does affect menstrual symptoms in adolescents
Postnatal Sense Of Security Of Primiparous And Multiparous Mothers And Affecting Factors
CURRENT PSYCHOLOGYThis study aims to determine the postnatal sense of security of primiparous and multiparous mothers and to evaluate the affecting factors. This is a descriptive comparative study, in which 484 mothers (242 primiparous and 242 multiparous) were included. Data were collected using a Questionnaire Form and the Mothers' Postnatal Sense of Security Scale. The average age of primiparous mothers is 25.52 /- 3.75, and the average age of multiparous mothers is 34.57 /- 5.17. The overall average score of the Mothers' Postnatal Sense of Security Scale is 52.02 /- 7.18 for primiparous mothers and 52.16 /- 7.77 for multiparous mothers. A statistically significant difference was found between postnatal sense of security and marriage age, educational level, employment status, educational level of spouse, income status, pregnancy desire, making routine pregnancy check-ups done, hospitalization during pregnancy, support from spouse and family members during pregnancy, having postnatal education, postnatal support from spouse and family members for primiparous mothers; while between postnatal sense of security and educational level, income status perception, pregnancy desire, making routine pregnancy check-ups done, having education during pregnancy, involvement of spouse in education during pregnancy, support from spouse during pregnancy, support from family members during pregnancy, involvement of spouse in postnatal education, postnatal support from spouse and family members for multiparous mothers. It was found that the average score of postnatal sense of security scale was high in primiparous and multiparous mothers who had pregnancy desire, made routine pregnancy check-ups done, were not hospitalized during pregnancy, and had no health problem during pregnancy
Development And Psychometric Evaluation Of The Paternal Support Scale Of Breastfeeding
JOURNAL OF PEDIATRIC NURSING-NURSING CARE OF CHILDREN & FAMILIESBackground: Social support positively affects the adaptation process of the woman to the maternal role during pregnancy and the postpartum period and increases her sensitivity to her baby. It is known that the support pro-vided by fathers to their wives during this process positively affects their decision to breastfeed and continue. Purpose: The aim of this study is to contribute to the literature by the Paternal Support Scale of Breastfeeding, testing its validity, reliability and psychometric properties. Methods: The study, which included 203 fathers with babies 0-6 months of age who were actively breastfed be-tween January and June 2022, was completed in a Medical Faculty Hospital at Konya in Turkey. The psychometric properties of the scale were evaluated with exploratory factor analysis and confirmatory factor analysis. In addi-tion, number/percentage, t-test for dependent and independent groups and correlation analysis were used in the evaluation of the data. Results: Total item correlation coefficient of this scale was found to vary between 0.63 and 0.81. According to the confirmatory factor analysis results the goodness-of-fit index values of the scale indicated that the model has an acceptable fitness, and the 21-item one-dimensional scale has confirmed validity. Conclusion: Our findings showed that this scale is a good reliable measurement tool that can be used to evaluate levels of paternal support in breastfeeding. The scale can be adapted to different cultures, and cross-cultural comparisons can be planned in future studies. Practice implications. Spouse and family support increases breastfeeding success. Due to their active role in the decisions made within the family, fathers positively influence mothers' breastfeeding decisions and increase mothers' motivation to continue breastfeeding. This measurement tool, developed to measure partner support in breastfeeding, helps nurses, to determine fathers' support levels in breastfeeding. In this way, nurses can contribute to increasing the duration of breastfeeding by making effective interventions for the solution of partner support problems related to breastfeeding. Molecular Investigations On T Cell Subsets In Patients Affected By Hypomorphic Dclre1C Mutation
EXPERT REVIEW OF CLINICAL IMMUNOLOGYObjectiveIn this study, we explored the expression of transcription factors, cytokines, and co-stimulatory molecules within the helper T (Th) cell subsets (Th1, Th2, Th17, and Treg) of patients with hypomorphic DCLRE1C gene mutations. MethodsThe study comprised eight patients and five controls. Transcription factor and cytokine expressions of Th subsets and co-stimulatory molecules were investigated by qPCR and flow cytometric following T cell stimulation. The findings were compared between patients (non-HSCT) and with hematopoietic stem cell transplantation (HSCT). ResultsFlow cytometric analyses; while the Treg rate was significantly lower in non-HSCT than in controls (p 0.010), the IFN-gamma rate was significantly higher in patients than in the control and HSCT groups (p 0.016, p 0.022, respectively). Co-stimulatory molecule expressions were significantly lower in non-HSCT than in control (p < 0.001), and there was a significant improvement after HSCT. Post-stimulation qPCR analysis, significant changes were detected in non-HSCT/control, non-HSCT/HSCT, and HSCT/control comparisons. ConclusionsOur study is the first study to molecularly investigate Th cell subsets in hypomorphic DCLRE1C patients. It was determined that abnormalities in Th cell subsets still persisted despite HSCT. There are still many conditions to be explained in these patients, and we believe that our study may shed light on future studies
Totally Implantable Venous Access Device Care Practices And Experiences Of Pediatric And Adult Oncology Nurses
JOURNAL OF INFUSION NURSINGThis study aimed to determine the practices of nurses working in pediatric and adult oncology clinics regarding totally implantable venous access device (TIVAD) care. The descriptive study was conducted with 227 oncology nurse members of the Oncology Nursing Association. The data were collected online with a survey form, which included questions about the participants' sociodemographic characteristics, professional experience, and TIVAD implementation practices. Descriptive statistics and chi(2) tests were used for the analysis of the data. It was determined that 44.1 of the nurses used 0.9 NaCl for active TIVAD flushing; 15.9 of them used a positive-pressure 0.9 NaCl-filled syringe; 12.3 used antireflux connectors; 85.5 used manual positive pressure technique; and 53.7 used the pulsatile technique. A statistical difference was found between nurses' training on TIVAD care and TIVAD occlusion rate in the clinic, TIVAD infection rate, following the guidelines, and using the pulsatile technique (P < .05). This study revealed that there are differences in the practices for TIVAD care and that the recommendations in the literature/guidelines are not implemented at the desired level to ensure continuity and prevent complications
The Intermediary Role Of Searching For Meaning In Young Adults In The Relationship Between Loneliness And Digital Game Addiction
ADDICTA-THE TURKISH JOURNAL ON ADDICTIONSIn the present study, the intermediary role of searching for meaning in the relationship between loneliness and digital game addiction in young adults was examined. The study was designed as a correlational survey and model testing using path analysisThe sampling for this study, consisted of a total of 1476 participants- of which 1152 were female and 324 were male. The Personal Information Form, Game Addiction Scale, Meaning of Life Scale, and Social -Emotional Loneliness Scale were used as the data collection tools. The results of structural equation modeling show that the data fit perfectly with the model. The model that was designed was validated. It was found that social loneliness and emotional loneliness predict the meaning and purpose of life at a negative and significant level. Also, the meaning and purpose of life predicted excessive focus and suspense, deprivation and seeking, emotional change, and contemplation at a negative and signifi - cant level. It is recommended to conduct awareness studies and value education regarding the meaning and purpose of life in the fight against digital addiction for young adults and adolescents

Before it is enabled, online shopping is a very expensive and challenging process for businesses to manage, especially since customers can set a time window for their products. Customers may lose business and incur expenditures if the products they requested are not timely and of the appropriate quality supplied to them. Due to this, it is crucial to handle this difficult process well, and for this to happen, both in- and out-of-warehouse procedures must be carried out simultaneously. Order batching, a branch of the order picking process inside the warehouse, and vehicle routing, a distribution problem outside the warehouse, are these processes. The Combined Order Batching and Vehicle Routing Problem arises if these two procedures are considered as being integrated. The linked problem becomes the Integrated Order Batching and Vehicle Routing Problem with Time Window (OB_VRP_TW) when the time component is added. This study categorized and analyzed papers in the literature for OB_VRP_TW and created a novel mixed integer nonlinear programming model. Since just a part of the small data set can be handled by packet encoding programs and this developed model is linearized under specific circumstances, a metaheuristic search algorithm called the genetic algorithm is offered to solve that problem. The test data of the sub-problems were searched for, and the test data for the problem were added to the literature by combining the test data in the literature and some data of a company whose problems were applied, as there were no test data for the current problem in the relevant area.Online alışveriş, Covid-19 pandemi döneminde oldukça yaygınlaşmış ve sonrasında ise kalitesi arttırılarak kalıcı hale getirilmiştir. Online alışveriş süreci, özellikle müşterilerin, siparişlerinin ulaştırılması için zaman penceresi belirttiği hali ile, şirketler için etkin bir hale getirilene kadar yönetilmesi oldukça maliyetli ve zor bir süreçtir. Müşterilerin sipariş etmiş olduğu ürünler müşterilere zamanında ve doğru kalitede ulaştırılmadığı takdirde hem müşteri kaybı hem de maliyete neden olmaktadır. Bu yüzden, bu zorlu sürecin verimli bir şekilde yönetilmesi oldukça önemlidir ve doğru bir yönetim için depo içi ve depo dışı süreçlerin eş zamanlı olarak yürütülmesi gerekmektedir. Bu süreçler depo içinde sipariş toplama sürecinin bir alt dalı olan sipariş gruplama, depo dışında ise bir dağıtım problemi olan araç rotalama süreci olmaktadır. Bu iki süreç entegre olarak düşünülürse, bütünleşik Sipariş Gruplama ve Araç Rotalama Problemi oluşmaktadır. İlgili probleme zaman faktörü eklenmesi ile problem bütünleşik Sipariş Gruplama ve Zaman Pencereli Araç Rotalama Problemi (SG_ZP_ARP)’ ne dönüşmektedir. Bu çalışmada, SG_ZP_ARP için literatürdeki çalışmalar sınıflandırılarak incelenmiş ve yeni bir karma tamsayılı doğrusal olmayan programlama modeli geliştirilmiştir. Geliştirilen bu model, belirli koşullar altında doğrusallaştırılmış ve paket çözücü programlar ile yalnızca küçük boyutlu veri setinin bir kısmı çözülebildiğinden dolayı, problemin çözümü için bir metasezgisel arama algoritması olan Genetik Algoritma yaklaşımı önerilmiştir. İlgili alanda mevcut problem için test verileri olmadığından dolayı alt problemlerin test verileri araştırılmış ve bulunan literatürdeki test verileri ile problemleri problemin uygulaması yapılan bir firmanın bazı verileri entegre edilerek problem için test verileri literatüre eklenmiştir
Constituents Of Alexander's Celery (Smyrnium Olusatrum) Extracts and Their Antioxidant, Enzyme Inhibitory and Anticancer Effects Based on in Vitro, In Silico and Network Pharmacology Methods
Journal Of Molecular LiquidsThis research delves into the medicinal significance of Smyrnium olusatrum by investigating ethyl acetate, ethanol, ethanol/water, and infusion extracts. The chemical composition analysed through LC-MS-qTOF metabolomic analysis, determine total phenolic and flavonoid contents, evaluate antioxidant potential using six in vitro tests (DPPH, ABTS, FRAP, CUPRAC, phosphomolybdenum assay (PBD), and metal chelating assay (MCA)), assess enzyme inhibition activity against acetylcholinesterase (AChE), butyrylcholinesterase (BChE), tyrosinase, alpha-amylase, and alpha-glucosidase, and explore their anticancer effects on HEp-2 cells. Additionally, qPCR analysis is conducted on HEp-2 larynx cancer cells to examine the impact of S. olusatrum on self-renewal and apoptosis pathways, along with the expression levels of key genes associated with these pathways. The findings indicated that the infusion extraction method demonstrated the highest levels, with a recorded TPC of 35.02 mg GAE/g and a TFC of 15.08 mg RE/g. All four extracts of S. olusatrum exhibited a total of 328 entities. The most significant metabolites, primarily comprising polyphenolics, flavonoids, non-structural carbohydrates, and amino acids in negative ionization mode, as well as sesquiterpene lactone and amino acids in positive ionization, were identified. Infusion exhibited the highest antioxidant effect among the extracts, with peak values ranged from 55.55 mg TE/g (DPPH) to 100.07 mg TE/g (ABTS). The extracts exhibited variable enzyme activities. Notably, ethyl acetate also demonstrated superior alpha-Amylase inhibition (0.69 mmol ACAE/g), while the ethanol extract exhibited higher alpha-Glucosidase inhibition (1.70 mmol ACAE/g). The HEp-2 cells demonstrated the quickest attainment of half maximal inhibitory concentration values with ethanol and ethanol/water extracts, yielding IC50 values of 250 mu g/mL (24 h) and 125 mu g/mL (24 h), respectively, among the applied extracts. The qPCR results revealed that S. olusatrum inhibited all self-renewal pathways and activated the apoptotic pathway. The ethanol and ethanol/water extracts of S. olusatrum significantly suppressed WNT1, APC, LEF1, and TCF7 genes. Furthermore, the downregulation of NOTCH1, SHH, and SMO gene expressions in all extracts suggests the activation of the Type 1 non-canonical hedgehog pathway in laryngeal cancer. The findings not only underscore the therapeutic potential of these extracts but also open the way for further exploration of their applications in combating oxidative stress, enzyme-related disorders, and potential anti-cancer effects through modulation of crucial cellular pathways
Impact of High-fructose Diet and Metformin on Histomorphological and Molecular Parameters of Reproductive Organs and Vaginal Microbiota of Female Rat
There are limited data on the effects of a high-fructose diet on the female reproductive system. Although metformin has some functional effects on female fertility, its reproductive outcome on high fructose diet-induced metabolic syndrome is unclear. The aim of the present study is to evaluate the impact of a high fructose diet on histomorphological and molecular parameters of the reproductive organs and vaginal microbiota as well as the treatment potential of metformin. Wistar albino rats were used in the study. The metabolic syndrome model was induced by a high-fructose diet in rats for 15 weeks. Metformin was orally administered once a day for the last 6 weeks. The high-fructose diet increased blood glucose, triglycerides, insulin, and ovarian testosterone levels; however, it reduced ovarian aromatase levels and follicle numbers and caused uterine inflammation. The high-fructose diet-induced molecular abnormalities on ovarian tissue were demonstrated by the downregulation of ovarian insulin signaling pathway proteins and dysregulation of ovarian mitogenic and apoptotic pathway proteins. A high-fructose diet caused vaginal dysbiosis, metformin increased probiotic bacteria in the vaginal microbiota. Our results revealed that metformin improves ovarian impairments by modulating hormonal balance, insulin level, mapk, and apoptotic signaling molecules, as well as regulating the vaginal microbiota. © 2024.
