    Investigation of miR221 and miR222 as biomarkers in non-small Cell lung cancer

    Abstract Background/Aim: Acute liver injury is the hallmark of organ failure in sepsis. Enteral nutrition (EN) is an important clinical therapeutic measure in septic patients. However, the therapeutic effect of EN alone is not obvious. Here, we investigated whether octanoic acid (OA)-rich EN alleviated acute liver injury through PPARγ/STAT-1/MyD88 pathway in endotoxemic rats. Materials and Methods: First, rats were randomly divided into four groups: Sham, Lipopolysaccharide (LPS), LPS+EN and LPS+EN+OA groups to investigate the effect of OA-rich EN on LPS-induced acute liver injury in endotoxemic rats. Then rats were randomly divided into five groups: Sham, LPS, LPS+EN+OA, LPS+EN+OA+SR202 (SR) and LPS+ pioglitazone (PI) groups to examine whether OA-rich EN alleviated acute liver injury through the PPARγ/STAT-1/MyD88 pathway. Rats received nutrition support via a gastric tube for 3 days. We evaluated the liver histology, apoptosis, liver enzymes and inflammatory cytokine levels in the liver and serum. PPARγ/STAT-1/MyD88 pathway was also measured. Results: OA-rich EN inhibited the phosphorylation of STAT-1 and the activity of MyD88 by activating PPARγ and alleviating LPS-induced acute liver injury more effectively than EN alone in endotoxemic rats. The use of SR counteracted the effect of OA-rich EN on acute liver injury. Meanwhile, PI showed effects similar to OA-rich EN in endotoxemic rats. Conclusion: OA-rich EN alleviated acute liver injury through PPARγ/STAT-1/MyD88 pathway in endotoxemic rats

    A clinical guide to rare male sexual disorders

    Abstract Conditions referred to as ‘male sexual dysfunctions’ usually include erectile dysfunction, ejaculatory disorders and male hypogonadism. However, some less common male sexual disorders exist, which are under-recognized and under-treated, leading to considerable morbidity, with adverse effects on individuals’ sexual health and relationships. Such conditions include post-finasteride syndrome, restless genital syndrome, post-orgasmic illness syndrome, post-selective serotonin reuptake inhibitor (SSRI) sexual dysfunction, hard–flaccid syndrome, sleep-related painful erections and post-retinoid sexual dysfunction. Information about these disorders usually originates from case–control trials or small case series; thus, the published literature is scarce. As the aetiology of these diseases has not been fully elucidated, the optimal investigational work-up and therapy are not well defined, and the available options cannot, therefore, adequately address patients’ sexual problems and implement appropriate treatment. Thus, larger-scale studies — including prospective trials and comprehensive case registries — are crucial to better understand the aetiology, prevalence and clinical characteristics of these conditions. Furthermore, collaborative efforts among researchers, health-care professionals and patient advocacy groups will be essential in order to develop evidence-based guidelines and novel therapeutic approaches that can effectively address these disorders. By advancing our understanding and refining treatment strategies, we can strive towards improving the quality of life and fostering healthier sexual relationships for individuals suffering from these rare sexual disorders

    ÖZET : Günümüzde, günlük fiziksel aktivite ortalamamızın önceki yüzyıllara göre önemli ölçüde düştüğü görülmektedir. Teknolojik gelişmelere paralel olarak, yapılan işlerin büyük çoğunluğunun oturarak ve bilgisayar kullanılarak yapılır hale gelmesinin insanların daha az aktif olmasında önemli ölçüde etkisi olduğu vurgulanmaktadır. Bu bağlamda araştırma, banka sektöründe çalışan bireylerin fiziksel aktivite düzeylerini ve etkileyen faktörleri incelemek amacıyla tek bir bankada gerçekleştirildi. Araştırma evrenini, çalışmanın yürütüldüğü bankada çalışan 252 birey oluşturdu. Örneklem sayısı belirlenerek; çalışmaya katılmayı kabul eden ve veri toplama formlarını eksiksiz dolduran 252 banka çalışanı ile araştırma tamamlandı. Veriler “Tanıtıcı Bilgi Formu” ve “Uluslararası Fiziksel Aktivite Anketi” kullanılarak elde edildi. İstatistiksel analizler için SPSS 22.0 programı ve Bağımsız gruplar T-Testi, Anova Testi, Pearson Korelasyon analizleri kullanıldı. Araştırma sonucunda; banka çalışanlarının %67,9’unun kadın ve %32,1’inin erkek olduğu belirlendi. Katılımcıların fiziksel aktivite düzeyi ortalama 1937,406±1401,094 MET-dk/hafta (Min=45; Maks=8010) olarak bulundu. Fiziksel aktivite düzeylerinin cinsiyete, medeni duruma ve çocuk sahibi olma durumuna göre anlamlı fark gösterdiği bulundu. Katılımcılardan erkek olanların kadın olanlara göre fiziksel aktivite düzeylerinin daha yüksek olduğu sonucu bulundu. Bekarların da evli olanlara göre fiziksel aktivite düzeylerinin daha fazla olduğu sonucu bulundu. Çocuğu olmayanların çocuğu olanlara göre de fiziksel aktivite düzeylerinin daha yüksek olduğu sonucu elde edildi. Çocuğu olmayanların olanlara kıyasla orta şiddetli, yürüme ve toplam puanları daha fazladır. Eğitim düzeyi, aile tipi, kurumda çalıştıkları yıl, sigara ve alkol kullanma durumu, kronik hastalık varlığı, egzersiz ve yürüyüş yapma durumu, yeterli ve dengeli beslenme durumu gibi diğer değişkenlerin ise, fiziksel aktivite düzeyi üzerine anlamlı düzeyde etkisi olmadığı belirlendi.ABSTRACT : Today, it is seen that our daily physical activity average has decreased significantly compared to previous centuries. In parallel with technological developments, it is emphasized that the fact that most of the work is done sitting and using the computer has a significant effect on people being less active. In this context, the research was carried out in a single bank in order to examine the physical activity levels of individuals working in the banking sector and the factors affecting them. The population of the research consisted of 252 individuals working in the bank where the study was conducted. By determining the number of samples; The research was completed with 252 bank employees who agreed to participate in the study and filled out the data collection forms completely. Data were obtained by using the “Descriptive Information Form” and “International Physical Activity Questionnaire”. SPSS 22.0 program and Independent samples T-Test, Anova Test, Pearson Correlation analyzes were used for statistical analysis. As a result of the research; It was determined that 67.9% of the bank employees were female and 32.1% were male. The mean physical activity level of the participants was found to be 1937,406±1401,094. It was found that physical activity levels differed significantly according to gender, marital status and having children. It was found that the physical activity levels of the male participants were higher than the female participants. It was found that singles had higher physical activity levels than married ones. It was concluded that the physical activity levels of those who did not have children were higher than those who had children. Compared to those who do not have children, moderate severity, walking and total scores are higher. It was determined that other variables such as education level, family type, years of employment in the institution, smoking and alcohol use, presence of chronic diseases, exercise and walking status, and adequate and balanced nutrition did not have a significant effect on the level of physical activity

    Diş hekimliğinde multidisipliner yaklaşımların çok önemli olduğu ve bu disiplinler arası sinerjinin sağlandığı oral diagnoz, diş hekimliği ile tıp hekimliği arasında köprü görevini üstlenmektedir. Uzun yıllar bu prensipler çerçevesinde genç hekim arkadaşlarımıza, asistanlarımıza ve uzmanlık öğrencilerimize bu alanda rehber olduğumuza inanıyoruz. Bu rehberliğin oluşturduğu ekolle diş hekimliğinde semptomlar, ayırıcı tanı yöntemleri, tedavi planlamaları, sistemik hastalıklar ve acil durumlardaki değerlendirmelerde didaktik bir yaklaşımla pratik uygulamalı bir bakış açısı geliştirilmiştir. Düşüncelerimiz doğrultusunda daha önceki yıllarda editörlüğünü üstlendiğim “Oral Diagnoz”(1995) ve “Sistemik Yaklaşımlarla Oral Diagnoz” (2007) isimli mevcut kitaplarımızdan sonra gün geçtikçe baş döndürücü bir hızla değişen bilgiler ve klinik yaklaşımların diş hekimliğinde de tezahür etmesiyle güncel kalma ihtiyacı tüm gerçekliğiyle ortadadır. Oral diagnoz alanında yine editörlüğünü üstlendiğim yeni kitabım bu anlayışla yola çıkılarak düzenlenmiştir. Uzun soluklu bu yolculukta, akademik merakı ve etik sorumluluğu yüksek çok değerli hocalarımız olduğu gibi mesleğin başındaki genç hekim arkadaşlarımız da bizlere eşlik etmiştir. Kitabımızda, oral diagnoza giriş ile başlayarak tüm sistemik hastalıkları ve klinikte bu sistemik hastalıklara diş hekimliği yönünden nasıl yaklaşılması gerektiğini de içine alacak şekilde çok geniş yelpazede bilgi sunulmaktadır. Bu bilgi paylaşımları sayesinde ayırıcı tanılarda farklı bakış açıları kazandırmak amaçlanmış olup hem akademik ortamda hem de diş hekimliği pratiğinde meslektaşlarımızla olan etkileşimimiz bizi ayrıca çok mutlu etmektedir.     The relationship between the level of soluble CD40 ligand and angiographic extent and severity of coronary artery disease

    ABSTRACT Objectives: Previous studies have suggested that atherosclerosis is a chronic infl ammatory disease and implicate components of the immune system in atherogenesis. The CD40 and CD40 ligand system regulates multiple phases of the humoral and cellular immune response. Soluble CD40 (sCD40) ligand especially releases from activated platelets. Platelets are involved not only in thrombosis, but also in atherosclerotic lesion formation and restenosis processes. The aim of our study is to evaluate whether there is a relationship between the level of sCD40 ligand and angiographic extent and severity of coronary artery disease (CAD) and to compare other cardiovascular risk factors. Methods: 102 participants were included in our study. Among them, 37 patients constituted acute coronary syndrome (ACS) group, 41 patients were diagnosed as stable angina pectoris group (SAP), while 24 subjects served as control group. Peripheral venous blood samples were drawn at the Coronary Care Unit admission of ACS group patients. From SAP group patients and controls who have normal coronary arteries, blood samples collected after coronary angiography.     Abstract: The paper presents soliton solutions to the nonlinear Schrödinger’s equation, considering chromatic dispersion and cross spatio-dispersive effects, while incorporating a self-phase modulation proposed by Kudryashov with quadrupled power-law forms. Two approaches are employed to obtain these soliton solutions. These significant findings contribute to the field of quantum optics and have implications for the advancement of modern telecommunication engineering sciences. Furthermore, these results pave the way for exploring additional advanced topics, such as extending the model to include perturbation terms and addressing fractional temporal evolution. The outcomes of these ongoing research activities will be disseminated in the near future

    A rare coexistence: Hammock mitral valve and aortopulmonary window

    Abstract Congenital mitral stenosis is a broad-spectrum pathology in which blood flow to the left ventricle is obstructed both functionally and anatomically. Hammock mitral valve, also known as anomalous mitral arcade, is a rare congenital anomaly particularly in infants and children. Hammock mitral valve may not be suitable for repair regarding the advanced dysplastic mitral valve structure. Aortopulmonary window is an unusual cardiac anomaly which is defined as a communication between the main pulmonary artery and the ascending aorta. As a result of the excessive left-to-right shunt, early intervention and surgical closure deemed mandatory to avoid development of severe pulmonary hypertension and its consequences. All patients with an aortopulmonary window necessitates prompt repair immediately. In this brief report, mitral valve replacement with a mechanical valve and repair of aortopulmonary window with a Dacron patch were performed simultaneously in a 5-month-old patient with a hammock mitral valve and accompanying aortopulmonary window

    Compatibility of digital and analog methods in assessmentof gingival zeniths

    Abstract Objective: The compatibility of digital and analog methods assessing the positions of gingival zenith (GZ) points has not been evaluated before. Therefore, the aim of this study was to investigate the accuracy of a digital scanner and a three-dimensional (3D) measuring tool in specifying positions of GZs by comparing it to the conventional method, an analog plaster cast and a caliper. Materials and methods: Both analog and digital impressions were obtained from 32 individuals. Following measurements were conducted in the anterior maxillary region from canine to canine on both the plaster casts and 3D images: (1) The distance from GZ to the corresponding vertical midline in each tooth (GZVM), (2) The distance from GZ of lateral incisor to the corresponding tangent that connects GZs of central incisor and canine (GZLI). Results: The intraclass correlation coefficients (ICC) of GZVM in each tooth varied between 0.366 and 0.755, demonstrating moderate to good compatibility between the two methods. The ICC of analog and digital GZLI for the right and left lateral incisors was 0.788 and 0.395, respectively. Conclusion: The digital and analog methods used in this study are compatible in determining the positions of GZs. Therefore, within the limitations of the present study, the choice of method can be based on the available equipment, time, comfort, and personal preference of the researcher. Clinical significance: Both digital and analog methods have the capability of determining the positions of GZs in compatible way

    Impact of silencing eEF2K expression on the malignant properties of chordoma

    Background Eukaryotic elongation factor 2 kinase (eukaryotic elongation factor 2 kinase, eEF2K) is a calcium calmodulin dependent protein kinase that keeps the highest energy consuming cellular process of protein synthesis under check through negative regulation. eEF2K pauses global protein synthesis rates at the translational elongation step by phosphorylating its only kown substrate elongation factor 2 (eEF2), a unique translocase activity in ekaryotic cells enabling the polypeptide chain elongation. Therefore, eEF2K is thought to preserve cellular energy pools particularly upon acute development of cellular stress conditions such as nutrient deprivation, hypoxia, or infections. Recently, high expression of this enzyme has been associated with poor prognosis in an array of solid tumor types. Therefore, in a growing number of studies tremendous efort is being directed to the development of treatment methods aiming to suppress eEF2K as a novel therapeutic approach in the fght against cancer. Methods In our study, we aimed to investigate the changes in the tumorigenicity of chordoma cells in presence of gene silencing for eEF2K. Taking a transient gene silencing approach using siRNA particles, eEF2K gene expression was suppressed in chordoma cells. Results Silencing eEF2K expression was associated with a slight increase in cellular proliferation and a decrease in death rates. Furthermore, no alteration in the sensitivity of chordoma cells to chemotherapy was detected in response to the decrease in eEF2K expression which intriguingly promoted suppression of cell migratory and invasion related properties. Conclusion Our fndings indicate that the loss of eEF2K expression in chordoma cell lines results in the reduction of metastatic capacity
