Two New Records of Wild Bananas (Musa balbisiana and Musa itinerans) from Sulawesi
Abstract The diversity of wild banana species in Sulawesi was investigated based on the morphological characteristics of herbarium specimens collected in Sulawesi and deposited in the Herbarium Bogoriense, Bogor, Indonesia (BO). New specimens were collected from Central, North, South, and Southeast Sulawesi, and digital type specimens were also used in this study. The aim of this study was to describe the diversity of wild banana species in Sulawesi as most Musaceae specimens stored at BO have not been identified. By examinating 110 sheets of herbarium specimens, five species of Musa, including two infraspecific taxa of M. acuminata housed in Sulawesi, were identified. M. acuminata, M. celebica, and M. textilis were previously reported from Sulawesi. However, M. balbisiana and M. itinerans are two new records of wild banana species in Sulawesi. Identification keys, descriptions, distribution maps, and line-drawing illustrations of these two species are provided. Abstrak Dua Catatan Baru Pisang Liar (Musa balbisiana dan Musa itinerans) dari Sulawesi. Studi keanekaragaman pisang-pisang liar di Sulawesi telah dilakukan menggunakan karakter morfologi spesimen herbarium yang dikoleksi dari Sulawesi dan disimpan di Herbarium Bogoriense, Bogor Indonesia (BO). Spesimen baru yang dikoleksi dari Sulawesi Tengah, Sulawesi Utara, Sulawesi Selatan, dan Sulawesi Tenggara dan gambar digital spesimen tipe juga digunakan dalam studi ini. Studi yang dilakukan bertujuan untuk mengetahui keanekaragaman pisang liar di Sulawesi mengingat sebagian besar specimens Musaceae yang tersimpan di BO belum teridentifikasi. Berdasarkan pengamatan terhadap 110 lembar spesimen herbarium, lima jenis dari marga Musa termasuk didalamnya dua intraspesifik taksa dari M. acuminata yang tumbuh di Sulawesi berhasil diketahui melalui studi ini. Musa acuminata, M. celebica, dan M. textilis telah dilaporkan sebelumnya tumbuh secara liar di Sulawesi. Sementara itu, Musa balbisiana dan M. itinerans merupakan dua catatan baru pisang liar di Sulawesi. Kunci identifikasi, deskripsi, peta distribusi, dan gambar ilustrasi dari kedua jenis tersebut didiskripsikan pada makalah ini
Suspension Stability and Characterization of Chitosan Nanoparticle–Coated Ketoprofen Based on Surfactants Oleic Acid and Poloxamer 188
In this research, ketoprofen was used as a drug model in the preparation of chitosan nanoparticles as a potential drug delivery system through the ionic gelation process with tripolyphosphate (TPP). The particle size analysis (PSA) revealed that the average particle size, polydispersity index (PI), and entrapment efficiency of chitosan nanoparticles prepared with oleic acid were 253.7 nm and 0.375 with drug entrapment efficiency of 73.30%. Those prepared with poloxamer 188 were 242.94 nm and 0.302 with drug entrapment efficiency of 87.89%. Scanning electron microscopy (SEM) analysis showed that the shapes of the nanoparticles, both prepared with oleic acid and poloxamer 188, were intact and spherical. Fourier transform infrared spectroscopy (FTIR) indicated several differences between the spectra of chitosan- and ketoprofen-loaded chitosan nanoparticles; for example, a new peak at the wavenumber 1409/cm indicated the presence of electrostatic interaction between the carboxyl group of ketoprofen and the amino group of chitosan. The chitosan nanoparticle suspension prepared with poloxamer 188 showed smaller increases in turbidity and viscosity than that prepared with oleic acid after 34 d of storage
Model of Steady State Cyclohexane Oxidation for Ketone-Alcohol (K-A) Oil Production
Cylohexane oxidation is of great industrial importance in the production of intermediates for the manufacture of nylon-6 and nylon-6,6. Most cyclohexane is commercially converted into a cyclohexanone–cyclohexanol mixture (known as K-A oil) by catalytic oxidation. K-A (Ketone-Alcohol) oil is then used as a raw material for adipic acid and caprolactam production. Practically, if the cyclohexanol content of KA oil is higher than that of cyclohexanone, it is more profitable to convert it into adipic acid; otherwise caprolactam production is more likely. The steady state cyclohexane oxidation reaction model in a stirred tank reactor for K-A oil production is presented and solved in this paper. The model was derived based on the mass balance and mass transfer equations using the kinetic equation. The set of algebraic equations was solved using non linear programming. The advantage of this method is that the relationship among variables can be better understood and an appropriate solution to the equation set can be obtained more quickly. Simulation results are particularly useful for process design such as in determining reactor dimensions and operating conditions
Phase Diagrams and Current Density Profiles of the Totally Asymmetric Simple Exclusion Process in Two Dimensions, for a Three-Way Junction
This study explores a dynamical model called the totally asymmetric simple exclusion process (TASEP) in two dimensions (2D). An open boundary condition is specified for the model, and sequential updating dynamics are used as the dynamical rule. The system studied is a discrete 2D system of lattice sites, which are modified into a three-way junction. Two cases are considered: a three-way junction with two entrances and one exit, and a three-way junction with one entrance and two exits. The density and current density of the system are determined numerically, such that a phase diagram is obtained. The continuity equation describing the dynamics of particles in the system is solved by using a simple Euler method. The results show that the density and current density profiles, as functions of the lattice sites, are determined by the input and output rates at their boundaries. Moreover, the density phases obtained are combinations of the density phases of the TASEP, which yield a rich phase diagram
Development of Immunochromatographic Strip Tests for Selective and Quantitative Detection of Melamine
An immunochromatographic strip test based on the complex reaction of antigen-antibody (melamine-antimelamine) was developed for quantitative detection of melamine. Gold nanoparticles (AuNP) were used to form AuNP-labeled antibody, which then acted as a biosensor. Melamine quantification was performed by the determination of AuNP using anodic stripping voltammetry technique with a boron-doped diamond as the working electrode. With sample volume of 100 µL and immunoreaction time of 7 min, the developed immunochromatographic strip test produced a linear calibration curve for melamine concentration range of 0–0.6 mg/L, with detection limit of 0.1 mg/L and RSD of ~5%. Furthermore, negative results were obtained for samples containing cyanuric acid and urea, indicating that the developed immunochromatographic strip test has potential for selective and quantitative detection of melamine
Effectiveness of Powder and Solid Ragi (Starter) from Dago (Bandung, West Java, Indonesia) Traditional Market on Production of Bioethanol from Rice Straw Hydrolyzate
Abstract Rice straw is one of the most abundant forms of agricultural wastes in Indonesia. Rice straw contains polysaccharide in the form of cellulose and hemicellulose, which can be used as raw materials in the production of bioethanol. This study aims to examine the effectiveness of bioethanol production from rice straw’s hydrolyzate. Research was carried out by fermenting the sample using two types of starters (Baker's yeast, known as powder starter (F); and Tapai’s starter, known as solid starter (S) with a control of pure yeast, Saccharomyces cerevisiae. Glucose levels were measured using a glucometer, and bioethanol levels were analyzed using high-performance liquid chromatography (HPLC). In this study, split plot design was used as a data analyzer with two treatment factors: starter’s inoculum (R) and time of fermentation (T). This study shows that there is no significant difference between the starters’ levels of bioethanol production. However, S’s treatment produced higher levels (5,1 g/l) of bioethanol compared to F’s (4,8 g/l); the rate of bioethanol production in S’s treatment is also higher (1,3 g/l.h) than that in F’s (1,2 g/l.h). This study concludes that Tapai’s starter is more effective in producing bioethanol from rice straw’s hydrolyzate. Abstrak Efektivitas Produksi Bioetanol dari Hirolisat Jerami Padi Menggunakan Ragi Kering dan Padat yang Diperoleh dari Pasar Tradisional Dago, Bandung. Jerami padi merupakan salah satu limbah pertanian yang sangat melimpah di Indonesia. Jerami padi mengandung polisakarida dalam bentuk selulosa dan hemiselulosa, yang dapat dimanfaatkan sebagai bahan baku dalam produksi bioetanol. Penelitian ini bertujuan untuk melihat efektivitas produksi bioetanol dari sampel hidrolisat jerami padi dengan menggunakan ragi roti (ragi kering – F) dan ragi tapai (ragi padat – S). Penelitian dilakukan dengan memfermentasikan sampel menggunakan kedua jenis ragi tersebut dan isolat murni khamir Saccharomyces cerevisiae sebagai kontrol. Kadar glukosa diukur menggunakan glucometer dan kadar bioetanol dianalisis menggunakan high-performance liquid chromatography (HPLC). Penelitian menggunakan split plot design dengan dua faktor perlakuan; pemberian ragi (R) dan waktu fermentasi (T). Hasil penelitian menunjukkan bahwa kedua jenis ragi pada produksi kadar bioetanol dari sampel memberikan pengaruh yang tidak berbeda nyata. Namun perlakuan S menghasilkan kadar bioetanol yang lebih tinggi (5.1 g/l) dibandingkan dengan perlakuan F (4.8 g/l); laju produksi bioetanol pada perlakuan S juga lebih tinggi (1.3 g/l.j) dibandingkan dengan laju produksi bioetanol pada perlakuan F (1.2 g/l.j). Kesimpulan dari penelitian adalah perlakuan ragi Tapai lebih efektif dalam memproduksi bioetanol dari hidrolisat jerami padi
Morphological and Molecular Analysis and Flowering Time of T2 Generation Transgenic Rice cv. Nipponbare Carrying CONSTANS (CO) Gene
Abstract Flowering is an important process that initiates plant productivity. Rice (Oryza sativa cv. Nipponbare) was genetically engineered by introducing a CONSTANS (CO) gene from Arabidopsis thaliana (AtCO) to improve rice productivity. The CO gene is a gene responsible for inducing early flowering. The aims of the experiment were to use morphological and molecular analysis to study the integration of the AtCO gene and its influence on agronomic traits in the T2 generation of Nipponbare transgenic AtCO rice. Morphological observations showed that agronomic traits tended to be better in the transgenic plants than in the wild type (controls), although the transgenic plants flowered later than the wild type. Polymerase Chain Reaction (PCR) analysis showed that 169 out of 227 transgenic rice plants showed hpt and CO gene integration. Abstrak Analisis Morfologi dan Molekular Padi Transgenik cv. Nipponbare yang Membawa Gen CONSTANS (CO) Terkait Umur Pembungaan. Pembungaan merupakan proses penting yang mengawali produktivitas pada tanaman. Metode rekayasa genetika dengan mengintroduksi gen CONSTANS dari tanaman Arabidopsis thaliana (AtCO) ke dalam tanaman padi kultivar Nipponbare digunakan sebagai upaya peningkatan produksi padi. Gen CO diketahui mampu menginduksi terjadinya pembungaan yang lebih awal pada tanaman. Penelitian bertujuan untuk mengetahui integrasi gen AtCO pada genom padi serta pengaruh gen tersebut terhadap waktu pembungaan dan karakter agronomi pada tanaman padi Nipponbare transgenik generasi T2. Hasil pengamatan morfologi menunjukkan bahwa tanaman padi transgenik cenderung memiliki karakter agronomi yang lebih baik dibandingkan dengan tanaman padi kontrol, namun tidak menunjukkan waktu pembungaan yang lebih cepat. Photocatalytic Decolorization of Acid Red 4 Azo Dye by Using Immobilized TiO2 Microparticle and Nanoparticle Catalysts
This research studied the synthesis of immobilized TiO2 microparticle and nanoparticle catalyst materials and evaluated its photocatalytic activity on the decolorization of Acid Red 4 (AR4) azo dye solution. TiO2 microparticles were immobilized on an acrylic plate, while TiO2 nanoparticles were immobilised on a glass plate. TiO2 nanoparticles were embedded in a nanofiber support layer to enhance photocatalytic performance, forming a nanofiber-nanoparticle composite on the glass plate. In the catalyst load 2.0 g/l, the performance of the photocatalytic layer of immobilized catalyst (k '= 0.013 min-1) is better than that of the suspended catalyst (k' = 0.008 min-1). Photocatalytic decolorization efficiency of TiO2 immobilized catalysts of nanoparticle sized reached 82.3% in irradiation time of 2 hours, which is higher than microparticle sized catalysts (77.8%). The technique of TiO2 nanofiber–nanoparticle composite showed improved photocatalytic kinetics (k’= 0.018 min-1) compared to a single layer of nanoparticle catalyst (k'= 0.015 min-1). Vertical Distribution and Flux of Nutrients in the Sediments of the Mangrove Reclamation Region of Muara Angke Kapuk, Jakarta
The reclaimed mangrove estuary in Muara Angke Kapuk is a reclaimed area that has not evaded the impacted of pollution and waste in the areas surrounding Cengkareng, Jakarta. This is apparent from the fact that almost all sediments under the mangrove trees are buried under heaps of plastic trash. However, the reclaimed region still has variety of organism, which indicating that the region still has an internal carrying capacity, especially nutrients from sediment. The purpose of this research was to examine the condition of sediment nutrients in this mangrove reclamation region. The research was conducted by taking water samples using a modification of the stratified cup at a sediment depth of 0-15 cm with depth intervals of 2.5 cm, and taking sediment samples using the sediment ring. Pore water samples were measured for dissolved oxygen (DO) and concentrations of ammonia, nitrite, nitrate, and phosphate. Sediment samples were used to obtain porosity values. The data obtained is used to make vertical concentration profiles and analysis of vertical nutrient flux. Vertical nutrient flux analysis was performed with the aid of QUAL2K software version 2.11. The results showed different vertical distributions and flux of nutrients, where influx for ammonia and phosphate and an increase in line with increasing sediment depth, while nitrate efflux and a decreased concentration. The flux calculation of nitrite as transitory nutrient was not done, but the concentration decreased after a depth of 2.5 cm. This indicates that the high contamination on the surface does not prevent the natural chemical processes so the reclaimed region can still provide nutritional support for its organism. Effect of a Combination of Extract of Centella asiatica L. Leaves and Extract of Green Coffee (Coffea canephora robusta P.) Beans in a Cream Preparation for Grade 1-3 Cellulite and Slimming
An accumulation of fat in the subcutaneous tissue causes cellulite and dimpling on the surface of the skin. Although not related to obesity, obesity worsens cellulite. There are abundant topical anticellulite creams on the market, but the efficacy of these creams has not been scientifically proven. A combination of Centella asiatica L. leaves extract and green coffee (Coffea canephora robusta P.) bean extract in a cream preparation was clinically tested in 30 women for 84 days in the absence of diet and exercise. The descriptive-true experimental before (T-0)-after (T-84) method was used to classify the cellulite (grade 1 3) and to determine the slimming effect. The cellulite appearance and the body circumferences (abdominal and thigh) perimeters were photographed 5cm and 10cm below the navel and below the gluteal fold. The results of before and after the treatment showed that grade 1 cellulite lower (p < 0.000), the measurement of abdominal circumference showed reduction (p < 0.013), but the measurement of both thigh circumference showed insignificantly reduction (p < 0.512). The combination of both extracts reduce cellulite and fat deposits in the abdominal area, making the volunteers look slimmer. Keywords: cellulite, Centella asiatica, Coffea canephora robust
