    Characterization of Bacillus thuringiensis isolat from several areas in Java and South Sulawesi for biological control of myasis, Chrysomya bezziana

    Bacillus thuringiensis is a species of bacteria which has potential use for biological control of parasite. The aim of the study is to characterize the local isolate of B. thuringiensis using PCR technique. Primer of cry gen was used in the study. Two hundred and twenty seven soil samples were collected from several areas in Java Island and South Sulawesi Province. Samples were obtained from surrounding soils of animal housing and paddocks. Samples were inoculated in Nutrient Agar and the suspect of bacilli were then cultured in Luria Bertani acetic buffer. Seventy-three pure isolates of Bacillus thuringiensis were obtained. Thirty-five isolates from Bogor, Yogyakarta and Majalengka regencies were amplified using PCR and consisted of a B. thuringiensis sporulation gene which has a 850 bp band in an agarose gel. Amplification using cryIVA,B primers demonstrated four isolates contain a faint band of 800 bp and 12 isolates consist of a distinctive band of 300-400 bp. The results showed that several B. thuringiensis isolates containing a cry IV gen and those isolates are needed to be tested as a biological control agent for Chrysomya bezziana.     Effect of biofertilizer and chemical fertilizer on growth and production of Panicum maximum

    A glass-house experiment was conducted to study the effect of biofertilizer, chemical fertilizer and their combinations on the growth and production of Panicum maximum. Four cultivars of Panicum maximum were used, namely cv. Riversdale, cv. Petrie, cv. Natsukaze and cv. Natsuyukata. The grasses were planted in Latosol soil. The treatments with 3 replicates were: (1) control, (2) urea 100 kg N/ha, (3) biofertilizer 10 l/ha and (4) urea 50 kg N/ha + biofertilizer 10 l/ha. The grass was harvested every 40 days for 10 times and data was polled. The average production of grass for cv. Riversdale was 20.0 pols, height was 77.2 cm, fresh weight was 40.5 g and dry weight was 9.0 g. The average production of grass for cv. Petrie was 36.4 pols, height was 63.9 cm, fresh weight was 42 g and dry weight was 9.2 g. The average production of grass for cv. Natsukaze was 28.6 pols, height was 67.2 cm, fresh weight was 40.7 g and dry weight was 9.2 g. The average production of grass for cv. Natsuyukata was 36.9 pols, height was 74.9 cm, fresh weight was 45.3 g and dry weight was 9.7 g. The average production of treatment (I) were 17.2 pols, height 59.7 cm, fresh 11.4 g and dry weight 2.6 g. The average production of treatment (2) were 22.8 pols, height 61.1 cm, fresh 19.3 g and dry weight 4.2 g. The average production of treatment (3) were 40.3 pols, height 82.5 cm, fresh 71.6 g and dry weight 14.9 g. The average production of treatment (4) were 41.4 pols, height 79.9 cm, fresh weight 66.1 g and dry weight 14.5 g.     Effect of mixing banana stem juice and subsequent heating on rumen degradability of soy bean meal

    The study was conducted to evaluate the rumen degradability of soy bean meal when it was mixed with banana stem juice and was subsequently heated.  Banana stem juice was used as source of tanin to protect protein in soy bean meal from degradation in the rumen. The banana stem juice (in volume) was mixed with soy bean meal at three ratio, which were 1 : 1, 1:2 and 1 : 3 (w/v). These mixing was then heated at 60oC and 90oC. The heating was done in an oven. Sample from each treatment was then filled into polyester bag with porosity of 45 mm and the size of 5 x 9 cm. The bags were incubated in the rumen of fistulated sheep for 0, 2, 4, 8, 16, 24 and 48 hours. The experiment was carried out in factorial 3 x 2, with three levels of banana stem juice and 2 levels of heating and was arranged in completely randomized design. Untreated soy bean meal was also incubated used as  control. Degradation data of dry matter and crude protein at each incubation time was fitted into exponential equation,  P = a + b (1 - ect). Results shows that there was a significant interaction factor of stem juice ratio and heating on water soluble substrate (constant A) where heating at 90oC with ratio 1 : 2 (w/v) resulted in the highest a. The insoluble but fermentable substrate (B) was decreased by the increasing concentration of banana stem juice, while the rate of protein degradation of b fraction was lower compared to control. In conclusion, banana stem juice can reduce the rate of degradation of soy bean meal in the rumen. Key words: Degradation, Crude Protein, Soy Bean Meal, Banana Stem Juic

    Genetic relationships of Kampung, Pelung, Sentul and Black Kedu Chicken using Microsatellite DNA Markers: I. Lingkage group of macro chromosome

    Genetic relationships of Kampung, Pelung, Sentul and Black Kedu chickens were studied on the basis of microsatellite DNA polymorphism. DNA samples were analyzed using nine microsatellite markers which chosen from linkage group of macrochromosome (chromosome 1-8) such as, locus ABR 258, ABR359, ABR 297, ABR 339, ABR 75, ABR 209, ABR 28, ABR 419 and ABR 604. Analyses of amplified DNA fragments were performed using Gene Mapper 2.0 software (PE, Applied Biosystems). The allele frequencies in each breed estimated by direct counting. Since all nine microsatellite markers were polymorphic, genetic distance between the breeds could be calculated based on the frequencies of alleles of the microsatellite. Genetic relationships between the breeds could be constructed. The results indicated that a total of 73 allele were detected while typing all the four breeds of local chicken and one breed of White Leghorn as outgroup breed acrossed nine loci. The number of alleles was observed in all of the breed ranged 3-17 alleles according to the microsatellite under scrutiny. Highest observed number of alleles was found in Kampung Chicken 60 alleles (82.2%). The UPGMA method for dendogram based on Nei genetic distances indicated that the local chickens have the same of ancestor, while Kampung and Sentul chicken have the same cluster followed by Black Kedu and Pelung Chicken.   Key words: Local chicken, microsatellite, macrochromosome, genetic distanc

    The growth of Java bulls fed rice straw and concentrates containing different levels of protein

    This study was performed to determine the growth of Java bulls fed rice straw and concentrates at different levels of protein. Twelve heads of Java bulls, one and a half years old, with initial body weight ranging from 113.83-191 kg were used in this experiment. They were grouped into four replications based on the initial body weight. The rice straw (30%) and concentrates (70%) diet containing three different levels of protein (8.27; 11.03 and 14.43%) were fed during nine weeks. Data gathered were the average daily gain and the average body measurements (chest girth, shoulder height and body length). Result of this study showed that the increase of protein levels from 8.27 to 14,43% did not significantly influence the average daily gain, shoulder height, and body length of Java bulls (P 0.05), but it influenced daily chest girth (P 0.05). The average daily gain (ADG), shoulder height and body length were 0.633 kg, 0.08 cm, and 0.09 cm, respectively. The highest average of daily chest girth of Java bulls was T2 = 0.19 cm, followed by T3 = 0.15 cm, and T1 = 0.12 cm. It is concluded that the increase levels of protein from 8.27 to 14.43% did not affect the average daily gain, shoulder height and body length of Java cattle.     Effect of humid acid on performance of broiler chicken

    An experiment had been carried out to study the effect of supplementation of humic acid via drinking water as natural feed additive on the performance of broiler chickens, in order to find the replacement of antibiotic, which recently known had negative effect on the consumen. Three hundred and twenty of day old broilers were divided into 4 treatment groups: (I) control (without humic acid); (II), (III) and (IV) with 100, 200 and 300 mg l-1 humic acid in the drinking water, respectively. Each group consisted of 4 replications with 20 birds each replication. The drinking water contained 2 mg l-1 probiotic. The birds were kept in litter system; feed and water were given ad libitum for 5 weeks. The parameter measured were body weight gain, feed convertion ratio (FCR) and percentage of carcass. The treatments had a significant effect on both body weight gain (P0.05) and FCR (P0.05). Body weight gain of control (I) 1839 g head-1 5 weeks-1, with FCR 1.66 were poorer than that of the group received humic acid: 1882 g head-1 5 weeks-1, 1.66; 1881 g head-1 5 weeks-1, 1.64; and 1874 g head-1 5 weeks-1, 1.59 for treatments II, III and IV, respectively. There were no significant different (P0.05) between treatment II, III and IV neither on body weight gain nor FCR values.The treatments had no significant effect on carcass yield, 66.28 ± 1.78; 67.06 ± 3.11; 67.63 ± 2.06 and 67.68 ± 2.21% for treatments I, II, III and IV, respectively. It is concluded that humic acid has a potential effect as natural feed additive, which could improve growth and feed efficiency without affecting carcass yield.     Protection of avian influenza (AI) vaccines for poultry against infection of field isolates A/Chicken/West Java/Smi-Pat/2006 and A/Chicken/West Java/Smi-Mae/2008 under laboratory condition

    The aim of this research was to study level of protection of avian influenza (AI) commercial vaccines available in Indonesia (subtipe H5N1, H5N2 and H5N9) against infection of HPAI field isolates of A/Chicken/West Java/Smi-Pat/2006 and A/Chicken/West Java/Smi-Mae/2008. There were 7 commercial vaccines used in this study, the each vaccines were injected in to 3 weeks old of layer chichickenen intramuscularly. At 3 weeks after vaccination, ten chichickenens from each group were challenged separately with the A/Chicken/West Java/Smi-Pat/2006 and A/Chicken/West Java/Smi-Mae/2008 isolates intranasaly with dose 106 ELD50 per 0,1 ml per chicken. Ten unvaccinated chicken were included in the challenge test as control. The study demonstrate that the AI vaccines with subtipe H5N1 protected chicken (100%) against virus of A/Chicken/West Java/Smi-Pat/2006 and 90-100% against virus A/Chicken/West Java/Smi-Mae/2008. Viral shedding were not seen by 2 days post challenge. The AI vaccines with subtipe H5N2 protected chicken at 20-30% against virus of A/Chicken/West Java/Smi-Pat/2006 and protected chicken at 70-100% against virus of A/Chicken/West Java/Smi-Mae/2008. Viral shedding still detected at 8 days post challenge. The AI vaccines AI with subtipe H5N9 did not protect chicken (0%) against virus A/Chicken/West Java/Smi-Pat/2006 and protected chicken at 50% against virus A/Chicken/West Java/Smi-Mae/2008. Viral shedding still detected by 8 days post challenge. This study concluded that AI vaccines with subtipe H5N1 are better than other AI subtipe vaccines in preventing HPAI virus A/Chicken/West Java/Smi-Pat/2006 dan A/Chicken/West Java/Smi-Mae/2008 infections under laboratory condition.     Forage production systems in upland agriculture of Jratunseluna watershed

    Study on forage potency at upland area of Jratunseluna watershed was conducted in three different agroecosystems. The locations represent deep soil area (volcanic) and shallow soil area (sediment) and three climate zones, namely zone B-2, C-2 and C-3. Data on forage production and food crops by-product were collected every month in a year. The result showed that there was different forage production between the deep soil and the shallow soil area. The forage production was also influenced by climate zone. The forage balance at each location shows that data collected once in a month was more meaningful on determination of surplus or deficit of forages compare to the one time data collection only. On the forage production, in dry matter as well as TDN, any single animal in volcanic area and B-3 climate zone got more forages compare to other location. On the other hand ruminants in sediment area with C-3 climate zone consumed forages below the demand line.     Morphology growth curve of female cross breed duck between Pekin and White Mojosari

    Morphological growth is a very dynamic process. It is reflected the changes of body structure affected by genetic and environmental responses. Each parameter had different growth curve sigmoid pattern those expressed different characteristics. Phenotypic growth data were collected from female ducks resulted from crossbreed between Pekin male and White Mojosari. The hatchers were twicely collected, the first collection was 69 heads and then they were 89 heads. Equation of non-linier regression is determined by Gompertz model using the Marquardt method. Result showed that fast growth happened from the start to 30 days old. Estimation values of circumstance and depth brest length variables were 30 and 83 mm respectively. In the first derivation, the maximum size of back and brest were observed on 16 weeks old. In the second derivation the maximum sizes of wide hip, wide and depth brestes were reached for longer time, those were 22 to 25 weeks. Meanwhile, the correlation between parameter A (body mature) and k (rate of mature) was negative. Every variable had different oscillation slopes, the slope of two infexion points of the original curve or they were variative.     Key words: Duck, growth curve, morphology, crossbree

    Pre-weaning growth of Boer x Peranakan Etawah goats

    An experiment was conducted to study the growth patterns of Boer x Peranakan Etawah (PE) crossbred during pre-weaning period, at the Indonesian Research Institute for Animal Production, Ciawi-Bogor. Sixty-one kids were used in this experiment. They were reared with their mothers in group pen (3 x 4 m), with each pen contained 6-7 does. Does were fed of 2.5 kg freshlychopped King Grass and 0.7 kg concentrate head-1 day-1. Research results showed that birth weight of Boer x PE kids (group A) were significantly higher than those of PE kids (group B) (4.29 ± 0.63 vs 3.71 ± 0.89 kg/head, P0.05). However, average preweaning daily weight gain (ADG) (116.40 ± 49.95 vs 105.29 ± 28.36 g/head, P0.05) and weaning weight (14.64 ± 4.56 vs 13.30 ± 2.71 kg/head, P0.05) were not significantly different between the groups. Pre-weaning mortality of the kids was relatively high in both group A (25%) and B (21,21%). While sex ratio (male : female) was 57.14 : 42.86% in group A and 51,52 : 48,48% in group B.     Key Words: Growth, PE, Boer Goat, Pre-Weanin
