Detection of antibody responses by using haemagglutination inhibiton test and the protection titer of avian influenza virus H5N1 subtype
Study on the detection of antibody responses using haemagglutination inhibition (HI) test and the protection titer to Avian influenza (AI) virus H5N1 subtype local isolate has been conducted at the Research Institute for Veterinary Science (RIVS). A total number of 50 village chicken (10 chicken served as un-injected controls) and 30 quail were injected intramuscularly with inactivated virus of AI H5N1 subtype local isolate. Serum samples were collected 3 weeks after injection and were tested using haemagglutination inhibition tests. The correlation between antibody titer and its protection to AI virus H5N1 local isolate were measured by challenging the birds with AI virus H5N1 local isolate The HI test was then used to determine field serum samples. A total number of 48 village chicken from three (3) Districts (Bekasi, Tangerang and Bogor) and 96 quails from two (2) farms in District of Sukabumi which were all vaccinated with commercial AI adjuvant vaccine were sampled. The study revealed that village chicken and quails showed antibody responses after 3 weeks vaccination and that titer of ≥ 3 log 2 was able to protect chicken and quails when they were challenged with local isolate virus. Based on this result, village chicken field samples from Districts of Tangerang, Bekasi and Bogor showed antibody titer which will protect 50, 100 and 85% of the flocks respectively. While quail field samples from Farm I and Farm II in District of Sukabumi showed antibody titer which will protect 60-100% and 0-80% of the flocks respectively. It is concluded that the study has successfully measured antibody titer to AI virus H5N1 subtype which protect village chicken and quails from local isolate virus challenge so that the results will be used to analyze field serum samples after vaccination program to eradicate AI from Indonesia. In vitro expression of native H5 and N1 genes of avian influenza virus by using Green Fluorescent Protein as reporter
The hemagglutinin and neuraminidase are important immunogen of avian influenza virus that are suitable for recombinant experimentation. However, both genes have been experienced rapid mutation resulting in diverse variety of genotypes. Hence, gene expression in recombinant systems will be difficult to predict. The objective of the study was to examine expression level of H5 and N1 genes from a field isolate by cloning the genes into expression vector pEGFP-C1. Two clones respresenting fulllength of H5 and N1 gene in plasmid pEGFP-C1 were transfected into chicken embryo fibroblasts (CEF), rabbit kidney (RK13) and African green monkey kidney (VERO) cells using Lipofectamine ‘Plus’ reagent. The experiment showed level of gene expression in the VERO cell was higher than in the RK13 and CEF cells. Observations using fluorescent microscopy and Western blotting revealed that the N1 gene was expressed better in all cells compared to the H5 gene. Genomic DNA restriction endonuclease from Pasteurella multocida isolated from Indonesia, katha strain and reference strains and analysed by PFGE
Pasteurella multocida strains are the causative disease agents of wide range of domestic and wild animals in Indonesia. The most important serotypes are associated with Hemorrhagic septicaemic (HS) diseases in cattle and buffaloes, cholera in ducks and chickens. The HS disease associated with P. multocia in large ruminants in Indonesia is controled by killed whole cell vaccines produced by the use of P. multocida Katha strains. There is no discriminatory data of the molecular biology technique has been applied to investigate P. multocida isolates from different geographic locations in Indonesia. The purpose of this studies were to observe the genetic diversity among P. multocida isolated from various geograpic locations and compared with Katha vaccine strain and other reference strains. A total samples of 38 isolates and strains of P. multocida were analysed by means of pulsed-field gel electrophoresis (PFGE). Each sample was grown in nutrient broth, cells were separeted by centrifugation. Whole cell pellet was mixed with agarose and then prepared agarose plugs. The genomic DNA of each sample was digested in situ (plug) with either restriction endonuclease of ApaI and/or BamHI. The digested genomic DNA of each sample was analysed by PFGE, the genomic DNA restricted profile of each sample was compared with others. The use of ApaI restriction endonuclease digestion and analysed by PFGE, demonstrated that 34 out of 38 P. multocia samples could be differentiated into 16 ApaI types, whereas based on the BamHI digestion of these samples were differentiated into 20 BamHI types. Genomic DNA restriction pattern of Indonesian P. multocida isolates originated from cattle and buffaloes associated with haemorrhagic septicaemic diseases demonstrated different pattern to those of vaccine Katha strain, poultry strains as well as the reference strains currenly kept at Balitvet Culture Collection (BCC) unit. Two P. multocida isolates derived from ducks with cholera disease showed similar pattern support the previous findings which exhibited similar pathogecity and vaccine protection. The majority of HS causing P. multocida isolates from some provinces in Indonesia belong to ApaI type 1 (7 isolates), six of these isolates belong to BamHI type 1. The BamHI restriction endonuclease digestion demonstrated higher discriminatory than that of ApaI. These restrection endonucleases digestion combined with PFGE analysis were effective for molecullar defferentiation of P. multocida strains and could be applied to other bacterial veterinary pathogens as well as for local isolate vaccine sellection. Ovulation rates and twinning birth following Follicle Stimulating Hormone (FSH) treatment at differents stages of estrus cycle
Twinning rate could be increased through genetically approach or through hormonal induction approach. The aim of this study was to observe the effect of first injection of FSH initiated on the ovulation rate. The experiment design was completely randomized with 3 different time of FSH was initiated as treatment using 12 lactating dairy cattle. Injection of FSH was initiated at day 2 (Treatment I), day 10 (Treatment II) and at day 18 (Treatment III) of estrus cycle. A total of 6 ml Folltropin (Equivalent 120 mg FSH) was intramussculary injected in decreasing dossis method twice a day with 12 hours interval for 4 days. Blood was collected on day 12 of estrus cycle for progesterone level. Data recorded were the diameter of ovary (DO), total corpus luteum (TCL), concentration of progesterone (P), percentage of pregnancy and number of birth. The means DO, TCL and P were significantly (P 0.05) higher after injection (2.0 cm; 2.1 and 1.6 ng/ml) than those before injection (1.4 cm, 1.0 dan 0.6 ng/ml). The time of first injection of FSH was initiated, significantly affect the ovulation rate. The mean DO, TCL and P were significantly higher (P 0.01) in treatment II (2.6 cm; 4.0 and 2.9 ng/ml) than those in treatments I (1.9 cm; 1.3 and 0.9 ng/ml) or in treatment III (1.6 cm; 1.3 and 0.9 ng/ml). There was no significant diference between treatment I and III. The percentage of pregnant were 25.0; 75.0 and 25.0 for treatments I; II and III respectively. One twin birth and 1 single birth were obtained in treatments II but only one single birth for each treatment I and III. Number of CL were positively correlated with the concentration of progesterone but were not fully useful for prediction number of birth. In conclusion, the dairy cattle gave a better response to exogenous gonadotropin hormone when the first injection was initiated at day 10 of estrus cycle. Conformation and component parts of the carcass of Philippine native goat
Only limited work has been done to assess the potential of Philippines native goat in terms of conformation and component parts of the carcass. Thus, an experiment has been conducted at Institute of Animal Science, University of the Philippines os Baños, Philippines to determine carcass and lean-fat-bone yield of different cuts of chevon from Philippine native goat. Result showed that among the wholesale cuts of chevon, shoulder had significantly higher separable lean content (8.80% of LW) than leg, loin, rib, and neck. Based on % wholesale cuts (WC), however, the leg had significantly higher value (69.18%) than the other chevon cuts. Loin had significantly higher separable fat (1.67% of LW) than the other chevon cuts. Shoulder had significantly higher separable bone (4.62% of LW). Based on %WC, however, rib had significantly higher separable bone (53.36%) than the other cuts. The shoulder had significantly higher boneless recovery (9.39% of LW) than other chevon cuts. The effectiveness of garlic extract (Allium sativum linn) in controlling aflatoxicosis in laying hens
Aflatoxicosis is a disease generated as the consequence of aflatoxin contamination in foods and feeds. Garlic (Allium sativum Linn.), a natural spice known to reduce the toxicity of aflatoxins in poultry. Twenty five laying hens were used to study the effectiveness of garlic extract in reducing aflatoxicosis. The animals were divided into 5 groups i.e (1) control group, (2) group treated with 0.4 mg/kg BH, (3) group treated with 0.4 mg/kg BH and 4% garlic extract in feed, (4) group treated with 5 mg/kg BH, and (5) group treated with 5.0 mg/kg BH and 4% garlic extract in feed. Body weight gains, eggs production, and the activity of glutamate oxaloacetate transaminase (GOT), glutamate pyruvate transaminase (GPT), and γ-glutamytransferase (γ- GT) enzymes were observed every week. The residue levels of the aflatoxin and metabolites were measured in the eggs using high performance liquid chromatography (HPLC). The results indicated that group treated with 0.4 mg AFB1/kg BH dan 5.0 mg AFB1/kg BH resulted in decreasing body weight gains, egg production, and increasing the level of GOT, GPT and γ-GT. The addition of 4% garlic extract in the feed was effective to improve the body weight gain and egg production only in the group of chicken treated with 0.4 mg AFB1/kg BH. However, it decreased the enzymes activities of the GOT, GPT, and γ-GT, as well as reduced the aflatoxin residues and metabolites in the groups at both aflatoxin levels (0.4 and 5.0 mg/kg BH). Aflatoxin residues decreased up to 42.2% for the group treated with the low dose of AFB1 (0.4 mg/kg) and 49.0% for the group treated with the high dose of AFB1 (5 mg/kg). Growth hormone gene family (GH, GHR, GHRH and Pit-1) polymorphisms and its association with superovulation response, ovulation rate and embryo quality in Embryo Transfer Station (BET) of Cipelang
The decrease in fertility is considered to be the main cause of reproductive loss in dairy cattle and beef industry. Many candidate genes that play an important role in fertility and embryonic development. The purpose of this study was to detect genetic variations of the growth hormone gene family (GH|MspI, GH|AluI, GHR| AluI, GHRH|HaeIII and Pit-1|HinfI) and its association with superovulation response, ovulation, fertilization and transferable embryos rate. A total of 45 blood samples taken from cows that have been superovulated Angus, Brahman, HF, Limousin and Simmental. DNA was extracted with phenol-chloroform protocol followed by polymerase chain reaction technique (PCR) using specific primers for GH, GHR, GHRH and Pit-1 gene. PCR product was cut with restriction enzyme MspI, AluI, HaeIII and HinfI and electrophoresed on agarose gel and stained with ethidium bromide (EtBr). Superovulation is done by injecting a totally of 20 ml FSH for 4 days. Injecting the prostaglandin hormone (PGF2α) was performed on the eleventh day of CIDR implantation. Artificial insemination (AI) performed two or three days after the injection of PGF2α and Flushing was done on the seventh day after the AI. The results showed that the Angus, Limousin, Brahman and Simental GH loci diversity of GH|MspI, GH|AluI, GHR|AluI, GHRH|HaeIII and Pit-1|HinfI was not associated with superovulation response, ovulation, fertilization and transferable embryo rate. In HF dairy cattle, genotype on Pit-1|HinfI AA has higher percentage of superovulation response (P 0.05) when compared to AB genotype, but did not differ to BB genotype. Dairy cattle HF AA genotype also had higher ovulation rate (P 0.05) when compared to AB and BB genotypes, but AB and BB have the same ovulation rate. Estimation of genetic parameters for body weight of Alabio and Mojosari ducks at starter period
A selection program is one of many important tools in livestock breeding in improving the quality of breeding stock. The choice of an effective selection method requires some information on the value of genetic parameters for some economically important traits, such as heritabilities and genetic correlation coefiicients. This experiment used 25 drakes and 100 ducks of each Alabio and Mojosari ducks, mated at random 1 drake to 4 ducks. A number of ducklings were obtained from each mating in each population, and their body weights were observed from hatching to 8 weeks old. Results showed that the heritability estimation for body weight to 8 weeks old were generally low either in Alabio or in Mojosari, ranging between 0.061 to 0.227. The highest heritability estimation was obtained for 6-week body weight 0.151 for Alabio and 0.227 for Mojosari ducks. The estimates of genetic correlation among body weights varied widely but generally high. It is concluded that 6-week body weight can be considered as a selection criterion depending on the selection objective in the local Indonesian ducks. Rice bran inclusion in the fruit and vegetable waste-based diets for fryer rabbits
The aim of the study is to investigate the effect of rice bran inclusion in the fruit and vegetable waste (FVW)-based diets on the growth performance of fryer rabbits. Thirty-six growing rabbits (New Zealand x Flemish Giant) of about 1070.8 g initial body-weight were housed individually in the wire mesh cages and assigned to either one of a FVW-based diet containing 20% rice bran (RB 20%), 10% rice bran (RB 10%) or zero rice bran (RB 0%). Thus, each treatment consists of 12 replicates. Data were collected for 28 days. Results showed that inclusion of rice bran in the diet significantly decreased (P0.05) dry matter intake (2888, 2830 and 3095 g, for Diets RB 20%, RB 10%, and RB 0%, respectively). Average daily weight gain of the rabbits consuming RB 20% (23 g), RB 10% (25 g) and RB 0% (33 g) was significantly different (P0.05). Inclusion of rice bran in the FVW-based diet significantly (P0.05) affected the feed conversion ratio of the diets (3.4 versus 4.4 and 4.1, for RB 0% versus RB 20% and RB 10%, respectively). Consistently, the average carcass weight of rabbits consuming RB 0% (1140g) was superior (P0.05) to the carcass weight of rabbits fed RB 20% (1022 g) or RB 10% (1046 g). Overall, inclusion of rice bran in the FVW based diet is not necessary. Simultaneously, use of FVW for rabbit is promising to avoid the accumulated fermenting FVW problem and produce healthy meat instantly for food. The effect of estrus and pregnant sheep serum on in vitro ovine embryo production
The aim of this research is to observe the influence of estrus sheep serum (ESS) and pregnant sheep serum (PSS) on in vitrous ovine embryo development. The research was carried out in Animal Reproduction Laboratory, Faculty of Animal Husbandry, Padjadjaran university. Oocyte and ovary of local sheep were collected from slaughter house. Maturation, fertilization and embryo culture media were supplemented with 10, 15 and 20% ESS or PSS respectively. Results show that supplementation of 20% ESS had significantly (P0.05) better maturation rate than those of 10-20% PSS (79.98% vs 58.89-68.97%). However, increasing ESS into 15-20% did not affect the maturation rate (71.86-74.98%). Therefore, 10% estrus sheep serum (ESS) can be used as an alternative serum in the ovine maturation media. The supplementation of ESS or PSS did not significantly increas the fertilization rate and in vitrous ovine embryo development, however, it was suggested to add 10% pregnant sheep serum (PSS) at in vitrous ovine embryo culture.
