Microbes of fermented kefir-like using combination of kefir grains and Bifidobacterium longum
The objectives of research were to find out physico-chemical characters and to detect flavor volatile compound of kefir-like. Material used was skim milk TS 9.5% which was heated at 85oC for 30 minutes and cooled at 22oC before innoculation of the starter. Microorganisms used were (a) Lactobacillus acidophilus P155110, (b) Lactobacillus delbrueckii subsp. Bulgaricus NCIMB 11778, (c) Lactococcus lactis P155610, (d) Leuconostoc mesenteroides subsp. dextranicum NCIMB 3350, (e) Acetobacter aceti P154810, (f) Bifidobacterium longum BF1, and (g) Saccharomyces cerevisiae P156252. The treatments consist of P1 = without (b); P2 = without (a); and P3= used (a) until (g). The physico-chemical characters identified were lactic acid and lactose percentages, pH, viscosity, organoleptic test for intensity of kefir-like sensory attributes. Results indicated that B. longum was potential bacterium use for starter combination on kefir-like making. The use starter P1 combination has high acidity and viscosity, low pH and lactose percentage, and high intensity on attribute creamy-white color, soft and curdle consistency, and kefir specific aroma on kefir-like. Volatile compound acid group were dominate by high acidity character on kefir-like resulted from starter P1 combination. Compound of 3-hydroxi-2-butanone (acetoin) was affecting butter-like of P3 character. This compound resulted from which is a character of fermented milk flavor was not detected on P1 kefir-like. The qualitative variation of Katingan cattle
The characterization is main activities in order to identify important trait of Katingan cattle. Information of its phenotypic characteristic is not available yet. The aim of the study is to describe the qualitative characteristic of Katingan cattle. Three location of the study were sub-district of Tewah Sanggalang Garing (Pendahara Village), sub-district of Pulau Malan (Buntut Bali Village), and sub-district of Katingan Tengah (Tumbang Lahang Village). The samples of Katingan cattle were taken as many as possible based on the field conditions to observe pattern of colour, growth of horn and gibbosity appear among two horns. General characteristic of Katingan cattle was various in colours of coat, having horns, humped and dewlop. The main characteristics were expressed at female of Katingan cattle. There were six variations of horn shape at female, however the growth of arching forwards was dominant (78.4%). Horn shape of male was generally grown upside (98.3%). Gibbosity was founded among horn only at female cattle (92.6%). There were 9 combinations of coat colour of female Katingan Cattle, those were brown reddish (27%), brown whitish (14.1%), brown like colour of Bali cattle (13.8%), black (12.5%), brown dull (9.6%), brown sorrel (9.3%), blackish (7.1%), white brownish (5.5%) and white grayish (4.5%). Male of Katingan cattle had eight colour combinations, those were brown whitish (14.8%), brown whitish and reddish (14.8%), brown reddish (13.1%), blackish (12.3%), brown whiteish with black withers (10.7%), brown sorrel (9.8%) and brown sorrel with black withers (7.8%). Based on qualitative analysis, the Katingan cattle had variations in coat of colour, growth of horn and the gibbosity. Stallion semen cryopreservation using different cryoprotective agents on the skim milk trehalosa extender
Cryoprotective agents (CPAs), protect the sperm during cryopreservation. There are two general classes of CPAs, at first penetrating cryoprotectants, these pass through the sperm membrane and act both intracellular and extracellularly, and the second non-penetrating cryoprotectants, these act only extracellularly. The objective of this study was to evaluate the addition of different CPAs namely glycerol (G), combination of ethylene glycol with glycerol (EG) and dimethilformamide (DMF) using skim milk trehalosa extender. The semen collected from 3 stallions using artificial vagina twice aweeks. The semen was evaluated, centrifugated and diluted in skim milk extender supplemented with 50 mM trehalose and three different CPAs with the concentration of sperm were 200x106 ml-1. Extended semen was then packed into minitub 0.3 ml and equilibrated at 4oC for 2 hours, freeze in the liquid nitrogen vapor for 10 minutes and stored in liquid nitrogen container -196oC. After 24 hours, the semen was thawed at 37oC for 30 second. Results of this experiment indicated that the percentage of motile and viable sperm in skim trehalosa extender using DMF and glycerol better than combination of ethylene glycol and glycerol. Effect of Morinda citrifolia leaf as saponin sources on fermentation characteristic, protozoa defaunated, gas and methane production of ruminal fluid in vitro
Many studies have reported that the Morinda citrifolia (pace plant) was a useful material for human health. However the exploration of this plant on rumen fermentation is still needed. Therefore, a research was done to study the effect of M. citrifolia leaf on fermentation characteristics of rumen fluid consisted of protozoa defaunated process, VFA composition, NH3 content, rumen microbial protein content, gas and methane production using in vitro techniques. Rumen fluid obtained from two fistulated Ongole crossbreed cattle fed with forage and concentrate feed ration (70 : 30). The fluid was incubated at 39ºC for 48 hours. The treatment on the rumen fluid consisted of control treatment: 100% (200 mg DM) kolonjono forage substrate (Penisetum purpureum) and M. citrifolia treatments: kolonjono forage plus M. citrifolia (equivalent saponin) 3; 6; 9; and 12 mg DM, respectively. The treatment of M. citrifolia leaf addition showed declined patterns in the number of protozoa population (P 0.05) compared to control. There was no effect of M. citrifolia treatment on VFA composition and NH3 content (P 0.05). Microbial protein content in rumen fluid increased (P 0.05) by M. citrifolia treatments compared to control. Effect of M. citrifolia treatments on the gas production was not significantly different (P 0.05) compared to control, while M. citrifolia treatments reduced the methane gas production of (P 0.05) compared to control. Effect the administration of human chorionic gonadotrophin (hCG) hormone following superovulation treatment in buffalo
A numbers of studies had been reported to evaluate the effect of hormone for superovulation treatment in buffaloes, however the numbers of embryo recovered is still very small. One of the limiting factors which affect the numbers of embryo recovered is preovulatery LH concentration. The experiment was carried out to study the effect of hCG following superovulation treatments in buffaloes as an effort to increase the embryo recovered. Ten (10) buffaloes in three different genotypes (Riverine, swamp and its crosses) were superovulated using follicle stimulating hormone (FSH). A total of 12 ml FSH (Folltropin) was injected intramuscularry twice a day with 12 hours interval in a decreasing doses for 4 days (2.5, 2.5; 2.0, 2.0; 1.0, 1.0 and 0.5, 0.5 ml). Prostaglandin was injected intramuscularry 2 days after the first injection of FSH followed by administration of 500 IU hCG two days later. At the same days of administration of hCG, artificial insemination (AI) using frozen semen was conducted and repeated again after 12 and 24 hours. Blood collecting was conducted 3 times a week for obtaining the progesteron level of plasma blood. Embryo was collected by non-surgically technique on day 6 of estrus cycle by flushing each horn of uterus with 500 ml Dubelco's Phosphat Buffer Saline (DBPS). Parameter recorded were diameter of ovary (DO), total corpus luteum (TCL), number of embryo collected (NE), percentage of recovery rate (%RR) and peak progesteron level (PP). Administration of hCG significantly increase the response of buffalo to superovulation treatment. The mean of DO (cm), TCL, NE and RR (%) was 4.0, 6.3, 2.1 and 37.2 and 4.5, 7.5, 3.9 and 48.1 for controll and 500 IU hCG respectively. The mean NE was significantly higher (P0.05) in hCG than those control. The peak progesterone concentration was highly significant higher (P0.01) in hCG (8.9 ng/ml) than those control (6.8 ng/ml). Both left and right ovary gave a similar response to superovulation treatments. In conclusion the admission of hCG following superovulation treatments using folltrophin increase the number of embryos recovered. Key Words: hCG, Superovulation, Buffalo, Embry
The effect on infection of irradiated Fasciola gigantica (liver flukes) on goats (Capra hircus Linn) blood values
This experiment was carried out to study the effect of infection Fasciola gigantica irradiated by using 60Co to goat’s blood values. Twenty local male goats, ten to twelve old months, 15-20 kg of body weight were used for this study. The animals were divided into 5 groups consisting of four replications. Three groups received irradiated metacercariae of Fasciola gigantica using 45, 55 and 65 Gy respectively. One group was the positive control (goats were infected by non-irradiated metacercariae) and another one was the negative control (goats were not infected by metacercariae). Each goat received 350 metacercariae. Body weight, Eosinofil, PCV and Hb were measured every weeks for 20 weeks. The result indicated that pathogenicity of irradiated Fasciola gigantica was reduced. The dry matter digestibility characteristics of tropical tree legumes using menke in vitro gas production technique
The amount of gas released when a feed was incubated in vitro with rumen fluid, was closely related to the digestibility of the feed. The experiment using Menke in vitro gas production was conducted to determine the dry matter digestibility of tropical tree legumes; Leucaena leucocephala, Flemingia macrophyla, and Gliricida sepium. The samples were collected from Institute of Animal Science farms of the University of the Philippines at Los Banos. The amount of gas produced were recorded at 3, 6, 12, 48 and 72 hours of incubation. To estimate the volume of gas, the equation of Tuah was adopted; while to describe the course of gas production, the equation of MCDONALD was used. The washing loss was not measured and assumed to be zero. Results showed that leucaena at 12 hours was the highest in the volume of gas produced (1.217 ml/g DM). There was a similarity of highly increase of the gas produced between gliricidia and flemingia at 24 hours of incubation i.e 1.60 ml/g DM and 1.60ml/g DM. The potential extent gas production at 72 hours (b value) and the gas production rate (c value) of three legumes were no significantly different. According to the logarithmic-curve of gas volume produced, leucaena tended to be the highest of increase in potential extent and the gas production rate. The different rate of gas production in related to incubation reflected the dry matter digestibility of the three legumes in the rumen. Key words: In vitro gas production, dry matter digestibilit
Comparison of fermentation kinetics (in vitro) of grass and shrub legume leaves: The pattern of gas production, organic matter degradation, pH and NH3 production
Rumen microbes convert carbohydrate in a feed into useful product such as VFA, microbial protein and B-vitamin; and some waste products such as CH4 and CO2. The amount and pattern of each product depend on the type of feed consumed by the animal. High fiber diet produces high gas and VFA, while high protein diet produce high NH3. Two types of feeds, grass and legume, have different cell structure and content, thus might have different pattern of rumen fermentation and product. In vitro method developed by Theodorou and Brooks was used to determine the pattern of rumen fermentation end product of two types of feeds, namely grass and legume. Result shows that legume has higher amount of OM degraded and produce higher amount of NH3 compared to grass. On the basis of OM degraded, the grass yielded higher volume of gas than that of legume. The two types of feeds did not change the pH of rumen fluid
Physico-chemical properties and metabolizable energi value of protein concentrate from palm kernel meal in broiler.
Protein concentrate from palm kernel meal (BIS PRO) was produced using combination method of grinding and ethanol extraction. The experiment was conducted to evaluate physico-chemical properties and metabolizable energy value of BIS PRO in broiler. The metabolizable energy experiment was conducted in Completely Randomized Design using 19 broilers Ross strain with average body weight of 1.79 ± 0.11 kg. The treatment diets were: R1(90 % basal diet + 10% palm kernel meal), R2 (90% basal diet + 10% BIS PRO) and R3 (90% basal diet + 10% soybean meal).The result showed that BIS PRO had higher specific density (0.723 g/ml), compacted specific density (0.885 g/ml), specific gravity (1.596 g/ml), and angle of repose (28.320) compared to those of palm kernel meal and soybean meal. Total solubility of BIS PRO (70.22%) were higher than that of the others. BIS PRO had metabolizable energy of 1.94 – 2.66 time higher compared to metabolizable energy of palm kernel meal, but there was no difference compared to metabolizable energy of soybean meal. It is concluded that BIS PRO has physicochemical properties and metabolizable value better than that of palm kernel meal and were equal to that of soybean meal. The potential of sugar cane juice as the liquid supplement and phytase enzyme carrier for poultry by in vitro
Most of the components of poultry feed (80%) of grains and meal that contains phytic acid which has anti-nutritional factor because it can bind minerals and reduce its availability. Phytic acid can be hydrolyzed by the enzyme phytase. Phytase enzyme naturally found in sugar cane juice, but its use as poultry feed supplements have not been done. The study was conducted using sugar cane juice PS 851 from Jatiroto PTPN XI, Lumajang, East Java in order to get the information potential of sugar cane juice as a liquid supplement and phytase enzyme carrier for poultry viewed from the aspect of nutrient content of sugarcane juice and phytase activity in the release rate of phosphorus. Research conducted at the Faculty of Animal IPB for 10 months. The rate of hydrolysis of phytase on P was tested using rice bran as a substrate. Sugar cane juice is added to the 2.5% level, using 4-level incubation (1, 2, 3 and 4 hours), each level consisting of 37°C and 42°C; pH 2; pH 4.5 and pH 5 with three replications. Study using a Two Factors Experiments in Completely Randomized Design and it was continued by DMRT test. P release rate was measured by spectrophotometry. The results showed that the sugar cane juice has a phytase activity of 0.0766 U / ml, brix level of 22.15%, containing water 73.03%, protein 0.47%, crude fiber 6.43%, minerals Ca 0.03%, P 0,02%, Co 0.14 mg / l, Fe 1.8 mg/l, Mn 1.55 mg/l, Zn 1.37 mg/ l, Cu 0.19 mg/ l, Se 12.63 mcg/100 g, vitamins B3 5.26 mg/100 g, C 0.72 mg/100 g, E 0.08 mg/100 g, sucrose 32.42%, fructose 2.41%, galactose 2% and glucose 1.58%. Supplementation of 2.5% sugar cane juice can increase the P release rate of 112-235% at optimum conditions of pH 5, at 37°C with a long incubation period of 1-4 hours.
