The Manipulation effectivity of cell co-cultures in 5% CO2 incubation system to increase in vitro cattle embryo production
The purpose of this research is to determine the effectivity of various cell co-cultures of cattle embryo production by in vitro CO2 5% incubation system and get the best cell co-culture. Cell co-culture which are used in the synthesis is the oviduct cells, isthmus cells, ampulla cells, follicle cells and without cells. Data were analyzed based on completely randomized desiggn. The average growth rate/ cleavage in various cell culture was: the oviduct cell 59.24%, ampulla cell 58.69%, isthmus cell 58.25%, follicle cell 52.24% and without cells 47.76%. The average growth of 8-16 cells embryos to various cell co-culture was: the oviduct cell 46.02%, ampulla cell 45.45%, isthmus cell 45.15%, follicle cell 43.07%, and without cell 38.50%. The mean percentage of morula in various cell co-culture treatment was: the oviduct cell 20.59%, ampulla cell 20.48%, isthmus cell 20.30%, follicle cell 16.96% and without cell 12.58%. The average percentage of embryonic growth (cleavage, 8-16 cells and morula) was not significantly different (P 0.05). The treatment of a variety of cell co-culture increased significantly (P0.05), blastocysts production, namely: the oviduct cell 3.28%, ampulla cell 3.22%, isthmus cell 3.08%, follicle cell 2.45% and without cell 1.97%. In conclusion, the treatment of various cell co-culture in 5%CO2 incubation system can increace the growth of cattle embryos in vitro. In vitro study of larvacidal effect of Chrysomya bezziana between atsiri oil (Piper betle leaf) from Sri Lanka and Bogor
Treatment of myiasis caused by the larvae of Chrysomya bezziana resulted variation results. The aim of the experiment are to compare larvacidal effect of atsiri oil of Piper betle leaf originated from Sri Lanka and Bogor agains Chrysomya bezziana larvae and to identify atsiri oil using Thin Layer Chromatography (TLC). Silica gel plate GF254 with toluena and ethyl ecetate as diluents was used in TLC. L1 and L2 of Chrysomya bezziana larvae were used for in vitro assay using agar plate contained atsiri oil of three concentrations ie. 2, 3 and 4%. Asuntol 1% and aquadest sterile were used as positive and negative control, respectively. Ten larvae were used in five replicates. The larvae were observed every 30 minutes for four hours. Larval mortality were counted and probit analysed using POLO-PC software, therefore the lethal concentration (LC50 and LC95) and lethal time (LT50 and LT95) were defined. The results showed there was a difference on migration distance between atsiri oil from Sri Lanka and Bogor on nRf 79-88 analysed using TLC. In vitro assay showed that both lethal concentration and lethal time of atsiri oil from Sri Lanka was higher than from Bogor. This results indicated that larvacidal effect of atsiri oil from Bogor was stronger than atsiri oil from Sri Lanka in in vitro both on L1 and L2. Growth rate of rex, satin rabbits and their cross fed with lactosym@ in an intensive raising system
An experiment was conducted to study the effect of breed, level of lactosym and their interaction on the growth performance of three breeds of rabbit, i.e.: Rex, Satin and their Cross. Thirty three-six week old (weaning) rabbits were contribute in a 3 x 3 factorial used of completely randomized design having 3-5 individual replication and fed one out of three levels of lactosym (0, 0.25 and 0.50 ml/head/week). The two factors were: breed and level of lactosym. Result showed that there were no significant differences between breed (P0,05) for body weight gain, feed intake and feed conversion. Body weight was affected by the level of lactosym (P0,05). Increase of lactosym level decreased body weight gain. Feed conversion were not affected by the level of lactosym (P0,05). Interaction between breed and level of lactosym did not affect body weight gain and feed conversion (P0,05), but affected feed intake (P0,05). Key words: Productivity, rabbits, lactosy
Development inactivated vaccine prototype of avian influenza (AI) H5N1 local isolate and its application at laboratory level
A preliminary study related on vaccine safety and vaccination effectivity for controlling avian influenza (AI) subtype H5N1 was carried out at Virology Laboratorium, Indonesian Veteriner Institute, Bogor. A Prototype of inactivated vaccine was made using AI H5N1 local isolate (A/Chicken/West Java/67-2/2003). The vaccine was then tested for safety and protection in DOC of layers. Antibody response, protection and shedding virus challenge were observed in the experiment. Result showed that the vaccine was saved and protected against virulent viral challenge. Efective vaccination was achieved at 3 weeks chicken old started with low level of antibody. Antibody titre increased gradually and reached the top at 8 weeks post vaccination. Challenge test using AI virulent at the age of 4 and 8 weeks post vaccination showed that the vaccine gave high protection (90%). Viral shedding was not longer expressed than 7 days after challenge. It is concluded that this prototype is a satisfied AI vaccine in laboratory level. Key Words: Vaccine, Avian Influenza, H5N1, HPA
Comparison fermentation kinetics (in vitro) of grass and shrub legume leaves: The pattern of VFA concentration, estimated CH4 and microbial biomass production
In the process of fermentation, rumen microbes normally convert major fractions of carbohydrates and proteins in a feed to useful end-products (i.e. VFA, microbial protein and B-vitamins) and some waste products (i.e. CH4 and CO2). The pattern of these end-products depend largely on the fraction contained in the feed eaten by the animal. Two types of feeds, namely grass and shrub legume, leucaena have different fraction proportions. Grass contains more fibre but less protein compared to shrub legumes. Thus in the rumen they might be fermented to produce different pattern of end products. The experiment was conducted in order to examine the pattern of VFA, CH4 and microbial protein products of the two types of feeds when fermented in the rumen. In vitro method was used to determine the pattern of these end products. Results showed that the grass produced more total VFA/mg organic matter degraded (0.0229 mM/mg vs 0.0075 mM/mg) and CH4 gas (0.20 mole/mg vs 0.09 mole/mg) but less propionate in partial and less microbial protein (2646 g vs 2656 g ) compared to the legume. Approximately 32% less CH4 (per mg OM degraded) would be produced from leucaena compared to that produced from grass, which mean that there will be less energy loss as CH4 thus more energy for animal production. Quality of canine epididymal spermatozoa during storage at 4C
The aim of this present study was to investigate the quality of canine epididymal spermatozoa during storage at 4°C. Spermatozoa was collected by flushing technique with physiological saline (NaCl 0.9% w/v) and diluted in modified Tris based extender containing 20% (v/v) of egg yolk for three days. The result showed that mean concentration of spermatozoa from cauda epididymal was 95.29.106 spz/ml. The percentage of progressive motility and membrane integrity of spermatozoa on time of collection was 70.71% and 72.85%, respectively. Quality of epididymal spermatozoa was decreased significantly (P0,05) during storage at 4°C. The percentage of progressive motility during storage were 70.71% on day 0 (H-0, after diluted), 60.71% (H-1), 45.71% (H-2), and 33.57% (H-3). The percentage of membrane integrity during storage were 72.85, 68.88, 61.06 and 47.47% on H-0, H-1, H-2 and H-3, respectivelly. In conclusion, quality of canine epididymal spermatozoa was decreased during three days of storage at 4°C. Key Words: Epididymal Sperm, Storage, Canin
The macroscopic and microscopic patology changes on broiler infected with local reovirus isolate
Local reovirus isolate is the virus which could be isolated from runting and stunting syndrome. The ability of local reovirus isolate to induce runting and stunting syndrome in broilers chicken was investigated. The day old chicks (doc) were infected with local reovirus isolate assessed clinically and pathologically at 1, 2 and 3 weeks post inoculation. A total of 40 DOC were divided into two groups. The first group (20 doc) was orally infected with ± 2 x 103 local reovirus isolate particle and the other used as negative control group. The results showed that the isolate caused wet droppings, stunting, enteritis, pancreatitis, malabsorbtion, bursal atrophy and spleenic hypertrophy, which similar to runting and stunting syndrome (RSS) disease in chicken. The body weight was reduced to 14.7% on the inoculated group at 4 weeks post inoculation. Key Words: Broiler, Local Reovirus Isolate, Runting and Stunting, Malabsorptio
Quality evaluation of shrimp by-product complete ration pellets
This research was done to evaluate the physical characteristic and chemical quality of the complete ration in pellet form that contain shrimp by-product. The evaluation was done on several variables namely: moisture content, water activity, particle size, average collision endurance, friction endurance and angle of repose. Data obtained was analyzed based on Completely randomized design. The treatment was: R0 (complete ration without shrimps by-product), R1 (complete ration with 10% shrimps by-product), R2 (complete ration with 20% shrimps by-product) and R3 (complete ration with 30% shrimps by-product). The results showed that physical characteristic of the complete ration pellet that contain 20% shrimps by-product had the lowest moisture (13.07%) and the water activity (0.45). Gambaran seroepidemiologi dan histopatologi infeksi virus parainfluenza tipe 3 pada sapi
A study to gain seroepidemiological feature and histopathological changes in order to obtain a viral causative agent had been conducted against parainfluenza virus type 3 (PI-3) in infection cattle in Indonesia. Serological survey was conducted in different areas in Indonesia and from serum Bank to gain the information on the distribution of parainfluenza type 3 (PI-3) in large ruminants. A total of 1334 sera had been tested using serum neutralization test, and the result indicated that prevalence of reactors was varied from 0 to 60 %. The highest prevalence was 60% in sera detected from Bogor abbatoir. Reactors were also found in other areas such as West Java, Central Java, East Java, NTT and Papua. Titration results indicated that the distribution of titre was varied from 4 to 256, and titre of 8 to 32 was the most common. Titre of 128 and 256 was only found in each of 1 sera only. Isolation results indicated that no isolate was obtained from 237 samples processed. Histological examination showed that more than 60% had interstitial pneumonia, which indicated vairal infection had been occurred. The effectiveness of Aloe vera barbadensis bioactives on laying hens on commercial farmers
A field trial was conducted to study the effectiveness of dry gel Aloe vera (DG) as a feed additive for laying hens in commercial farms. The trial was consisted of two treatments, one was control, commonly used farmer ration containing antibiotic of zinc bacitracin at 0.5 g/kg and the second feed containing DG equal to 1.0 g/kg. Every treatment had two replicates with 504 Loghman laying hens. The hen day production (% HD), egg weight, feed consumption, feed conversion ratio (FCR), egg quality comprising yolk colour, albumin and yolk weights, egg shell eight and thikness, and mortality were observed for 24 weeks. The results showed that feed consumption, % HD, egg weight, FCR, yolk colour, albumin weight, yolk weight, egg shell weight, and egg shell thickness were not significantly different (P0.05) between the control and DG treatment, except for the Haugh unit (HU). Thus, it can be concluded that Aloe vera bioactives has the same effectiveness as antibiotic as a feed additive at the level of commercial farms.
