    Exogenous progesterone treatment during pregnancy for increasing milk production and growth of kids of Etawa grade goat

    Naturally, progesterone in ruminant is mainly produced by corpus luteum and it is reponsible for maintaining pregnancy, and affecting udder development and milk production. Exogenous progesterone treatment is expected to give similar positive effect on milk production as the endogenous progesterone does. Fourty mature Etawa grade (PE) does were synchronized for oestrus using Controlled Internal Drug Release (CIDR) followed by natural mating. Does then were treated with CIDR intravaginally, as a source of exogenous progesterone, at day 15-75 after mating (Group A), day 75-135 after mating (Group B), and without exogenous progesterone treatment (Control). Blood samples were taken before and after exogenous progesterone treatment for determination of progesterone concentrations. Soon after birth, kids were separated from their respective dams and they were milk fed through milk bottle. Feed consumption and milk production were measured daily, while bodyweight was measured forthnightly. Results showed that response of does following exogenous progesterone treatment was not expressed by an increase in progesterone concentration in the blood plasm. However, treatment group showed kid with birthweight of 10-13% higher compared to Control Group. In relation to milk yield, birthweight and pre-weaning growth of kids, the positive responses of exogenous progesterone treatment were not significant, though there was a substantial decrease (71.2%) in pre-weaning kid’s mortality.     Productivity of Indigofera sp. at different drought stress level and defoliations interval

    This study was aimed at determining the interaction between drought stress and defoliation interval on the productivity of Indigofera sp. This study was done based on Complete Randomized Design (CRD) 3x2 factorial, the first factor was without drought stress (100% FC), medium drought stress (50% FC), and severe drought stress (25% FC), and the second factor was the defoliation interval of 60 day and 90 days, each treatment was repeated 5 times. Observed variables were biomass production, and leaf/stem ratio. Results showed that there were significant interactions (P 0.05) between drought stress and defoliations interval on the productivity of Indigofera sp. The highest biomass production of Indigofera sp. (424.47 g/plant) was obtained from treatment of wihtout dought stress (100% FC) combined with defoliations interval of 90 days, while the lowest (184.55 g/plant) was reached at treatment of severe drought stress (25% FC) and depoliations interval of 60 days. The highest leaf/stem ratio (1.47) was reahced at treatment severe drouht stress (25% FC) and depoliations interval of 60 days and the lowest (1.05) was reached at combination treatment of drought stress and depoliations interval of 90 days.     Verification for the control of kappa-casein gene on milk protein yield of friesian-holstein in dairy central region of West Java

    Genotipe of BB kappa kasein (ĸ-kasein) gene positively influences high milk protein yield. The aim of this research was to verify the use of the к-casein gene as marker of assisted selecion (MAS) of high protein yield in domestic Holstein-Friesian (HF) cattle. Genotyping blood samples was conducted for lactating cows from BPPT-SP Cikole (82 head) and a number of small holders in KPSBU Lembang (190 head). Genotyping the к-casein gene was also conducted for active and non active AI bulls (HF) from BIB Lembang (25 head) and BBIB Singosari (32 head). Varian genotipes were identified trough DNA migration, whereas frequencies of alleles were calculated by Nei Method (1987). The effects of variant genotypes on protein yield and other milk component yields were studied in lactating cows (56 head in BPPT-SP Cikole and 111 head in KPSBU Lembang) by applying Least Square Means (LSM) method of the General Linear Model (GLM). Genotyping the ĸ-kasein gene resulted in three genotypes, namely AA, AB and BB, with two alleles, namely A and B. It was identified that a very limited number of lactating cows with BB genotype (0-9%). These cows produced higher protein yield around 3.37-3.84% than that of AA cows, while AB cows produced protein yield in between, though these differences were not statistically significant (P0,05). In contrast, AA cows tended to produced higher milk fat yield than BB cows. Cows BB and AB seemingly produced higher dry matter, while both specific density and pH were not affected by variant genotypes of the к-casein gene.  The results showed that BB genotype of the к-casein gene tended to be quitely consistent in controlling high protein yield, so it would be a good oppurtinity to be used as MAS of milk protein yield in HF domestic.     Supplementing energy and protein source at different rate of degradability to mixture of corn waste and coffee pod as basal diet on rumen fermentation kinetic of beef cattle

    The use of corn waste and coffee pod as basal diet needs energy and protein supplementation in order to optimize rumen microbial growth. A research was done to study the appropriate supplement which is suitable based on the result of rumen fermentation kinetics. Four ruminally canulated cows, (205-224 kg of live weight) were placed in individual cages. The basal diet (BD) offered were corn waste and coffee pod mixture (80:20). Source of high degradable energy (HDE) was cassava pomace; while the low degradable energy (LDE) was arenga piñata waste. The high degradable protein (HDP) was mixed-concentrate while the low degradable protein (LDP) was leucaena leave meal. The supplementation of energy and protein to basal diet was in ratio of 50:50 based on dry matter. Feed offered were basal diet and the supplement at ratio of 60: 40 (3 % of LW). Observation was carried out for 4 periods (10 days/period). In each period, animal was fed one of the following diets:  A = BD, B = BD+ HDE+LDP, C = BD+LDE+LDP, and D = BD+HDE+HDP. Rumen kinetics observed were: pH, and VFA, NH3 and rumen microbial protein concentrations. Rumen fluid was taken at the end of each period gradually along the course of 24 hours fermentation. The results showed that the diets gave significant effect (P 0.05) on rumen pH. The lowest rumen pH (5.76) was observed on diet D  at 2:00, 12 hours after feeding. Meanwhile, the highest pH (7.22) was found in animal fed diet A at 16:00. The total VFA on diet D of periode:1, 3 and 4 showed the highest level: 68.1 mmol/l; 75.37 mmol/l and 85.14 mmol/l respectively. The highest NH3 concentration was found in diet D followed  by diet C, B and A. At 12:00 observation or at 4 h after morning feeding the highest NH3 was observed from diet D (41.94 mg/100 ml). It is concluded that diet D resulted in the best rumen fermentation kinetic, therefore it could be used in feed formulation in cattle diets.     Utilization of complete rumen modifier on sheep fed high fibrous forages.

    A research to improve livestock productivity and lower enteric methane production on ruminant was conducted by manipulation approach on rumen system using a complete rumen modifier (CRM). An in vivo experiment was carried out using twenty four sheep ( mean weight 18 kg) which were distributed into 3 treatment groups of feed additive: I. control ( without treatment: K); II: K + CRM-LG; III: K + CRM-EL. Diet given consisted of fermented rice straw (ad libitum) + concentrate containing 16 % protein (400 g/head/day), and drinking water was given ad libitum. The experiment was conducted for 14 weeks based on completely randomized design. By the end of the experiment, animals were placed in the metabolism cages for 2 weeks (ie. 1 week for adaptation and 1 week for data collection). Rumen liquid of each treated animal was taken for the measurement of rumen characteristics. Parameter measuremed were: total gas production; gas composition of CO2 and CH4; in vitro DMD; NH3 and VFA contents; pH; bacterial and protozoal counts; consumption/ DMI; in vivo DMD; ADG and FCR. The results showed that productivity of sheep was improved by CRM treatments followed by lowered enteric methane production. The ADG values of CRM treatments (71.4 to 73.5 g) were significantly higher (P 0.05) than that of control (50 g). The improvement of average daily gain was followed by a better feed conversion (P 0.05) (ie. 10.6 vs. 12.8). The CRM treatments lowered the percentage of CH4 by 24% compared to Control (P 0.05). The total and composition of VFA of CRM-treated rumen liquor were significantly different (P0.05) compared to that of rumen liquor of Control (ie. the total VFA: 85.3 vs 73.5 mM and the percentage of acetic acid: 67.8 vs 60.3%). It is concluded that CRM treatment resulted in positive effects on growth of ruminant fed high fibrous forages such as rice straw and could lower enteric methane production.     The study aimed to investigate the effects of offering different levels of ensiled  or fresh Indigofera arrecta in completete diets on the feed consumption, ADG, ruminal fermentation characteristics and blood metabolites concentrations in growing goats. The I. arrecta/concentrate ratios of the complete diets were set at 85/100,75/100 and 65/100. Thirty weaned-male goats were used in the study, and were randomly allocated to one of the six treatments (5 heads per treatment). The diets were offered at 4% BW and the animals were weighed weekly. The study was arranged  as a completed randomized design. Feed intake, average daily gain, feed efficiecy ratio, ruminal fermentation (pH, NH3 and VFA characteristics and blod metabolites (glucose and urea) were analysed. Feed intake of goats receiving ensiled Indigofera arrecta were lower (P 0.01) compared to those receiving fresh I. arrecta. ADG were higher (P 0.01) when goats were fed diets with lower level inclusion of Indigofera arrecta. Efficiency of feed utilization decreased (P 0.01) when ensiled Indigofera arrecta was used in the diets, but it was not affected (P 0.01) by the inclusion level of I. arrecta in the diets. Ruminal pH was not affected by ensiling process, but ruminal NH3 concentrations were greater (P 0.01) in goats receiving fresh Indigofera arrecta. Ruminal VFA levels were also greater (P 0.05) in the group offered fresh Indigofera arrecta, but the differences were not significant (P 0.05) when diets consisted of 65% Indigofera arrecta in the complete diets. Increased  rate of Indigofera arrecta inclusion in the diets resulted in lower (P 0.01) counts of ruminal microbial population. The concentration of plasma urea nitrogen were neither affected by the ensiling process nor by the levels of I. arrecta inclusion in the diet (P 0.05), however, the plasma glucose level decreased (P 0.05) as the level of I. arrecta inclusion in diets increased. It is concluded that I. arrecta could be used as the sole fresh or ensiled foliage in complete diets, although the performances of goats were better when  fresh I. arrecta was offered. The level of I. arrecta inclusion in complete diets was recommended at not greater than 65%. Key Words: I. arrecta, Silage, Complete Feed, Goa

    Utilization of plant bioactives as feed additives for poultry: The effect of Aloe vera gel and its extract on performance of broilers

    Feed additives are commonly added in poultry feed as a growth promotant or to improve feed efficiency. The most common feed additive used is antibiotic at sub-therapheutic doses, although there is a controversy on its impact on human health. Previous results showed that Aloe vera gel could improve feed efficiency in broilers and an in vitro study showed that the extract have an antibacterial effect. Therefore, a further experiment was designed to study the response of broilers to Aloe vera gel or its extract as feed additives. Aloe vera was prepared in dry gel or chloroform-extract and included in the diet at levels of 0.25; 0.50 and 1.00 g/kg (equal to dry gel). Standard diets with or without antibiotic were also formulated as control and a commercial diet was included for comparison. The diets were fed to broilers from day old to 5 weeks. Each treatment has 9 replicates and 6chicks/replicate. Parameters observed were: feed consumption, weight gain and feed convertion ratios. Carcass yield, abdominal fat levels, relative weight of liver, gizzard, tractus digestivus and length of tractus digestivus were also measured at the end of feeding trial. The results showed that Aloe gel and its extract did not influence body weight gain and feed consumption of broilers significantly (P0.05), but improved feed convertion slightly (3.50%). The response in this trial was similar as thosecommercial diet and diet added with antibiotic. There was no significant (P0.05) effect of Aloe vera bioactives on carcass yield, abdominal fat level and relative weight of liver. However, Aloe vera gel and its extract tend to increase gizzard weight, gastro intestinal weight and length. The Aloe vera gel and its extract also reduced the total count of aerobic bacteria in the digesta of tractus digestivus. It is concluded that the Aloe vera gel improve feed efficiency in broilers by increasing the size of tractus digestivus and reducing the total count of aerobic bacteria in the gastro intestinal tract.   Key words: Aloe vera, feed additives, broilers, anti-bacteri

    Response of broilers on the diet containing fermented palm oil sludge stored at various length of time

    An experiment was conducted to study the response of broilers on the diet containing fermented palm oil sludge with Aspergillus niger stored at different length of time. The research was carried out in Research Institute for Animal Production Ciawi, Bogor. The experiment was conducted based on completely randomized design using 245 day old broiler chicks with 5 replicates. They were allotted to 7 diets containing one control without palm oil sludge (R0), 5 and 10% of fresh-fermented palm oil sludge (LSF) (R1 and R2), 5 and 10% of LSF stored at room temperature for 2 months (R3 and R4) and stored for 3 months (R5 and R6). Diets were fed ad libitum for 5 weeks. Feed intake, weight gain, feed conversion ratio, and carcass yields were observed as parameters. Results showed that no parameters were significantly affected by the treatments. It is concluded that 10% of fermented palm oil sludge could be included in diet of broilers even after three months storage at room temperature.   Key words: Broiler, fermented palm oil sludge, storage perio

    In vitro slow-release urea contained in rice straw-based diets to increase efficiency of rumen microbial protein synthesis

    Effect of slow-release urea on efficiency of rumen microbial protein synthesis (EMPS) was examined using an in vitro technique. The objective of this experiment was to reveal the in vitro slow-release urea characteristics of zinc-urea, zeolites-urea, and zeolites-zinc-urea in relation to EMPS observed in different incubation time. The experimental design employed was randomized block design with 4 x 3 factorial plus a control treatment, and conducted in two replications. Factors were various urea sources (urea, zinc-urea, zeolites-urea, and zeolites-zinc-urea) and molasses concentrations (0%, 6%, and 12%) in rice straw-based diets. The control treatment was the rice straw-based diets containing neither urea nor molasses. Diets consisting of 45% rice straw and 55% concentrates (DM basis) were formulated to have similar N and TDN levels. Responses of parameters measured were subjected to MANOVA using the GLM procedure of SPSS 16.00 and differences among mean values, if applicable, were examined using HSD-test. Orthogonal comparisons were used to determine the effects of the control treatment vs. various urea sources. Results indicated that treatment of UZ combined with 6% of molasses showed the highest microbial biomass production (2.71 mg/l) at 24 hours fermentation period with its peak production estimation (3.2 mg/l) reached at 33.5 hours of fermentation period. Moreover, UZ treatment resulted in the highest microbial protein synthesis (1,381.45 ± 77.1 mg/l) at 24 hours fermentation period with its peak microbial protein synthesis estimation (1,756.04 mg/l) reached at 33.7 hours of fermentation period.     Assessment of the purity and characteristics of rat splenic T cells isolated by one-step discontinuous gradient of percoll

    T cells isolated from spleen lymphocytes of rat (Rattus rattus) have been commonly used in an immunological research. However the T cells are still contaminated with other splenic cells.  Therefore a simple and reliable technique to isolate the T cells in one step process is needed. The aim of the present study was to assess whether T cells isolated from spleen lymphocytes of rat as a model by means of discontinuous density gradient of Percoll could give pure T cells with their activities as needed in an immunological research. Isolation was carried out in 5 density gradients of Percoll i.e. 1.052, 1.063, 1.075, 1.085, and 1.122 g/ml. The purity of the cell fractions was determined by immunocytochemistry and fluorescence-activated cell sorting (FACS) methods. The activity of each lymphocyte fraction was determined through cell proliferation assays by culturing the cells in the presence of phytohemaglutinin (PHA) and Concanavalin A (Con A) as T cells inducers. The FACS results show that fraction I and II were found to contain mostly B cells and macrophages. The fractions were obtained on the layers of gradient 1.052/1.063 and 1.063/1.075. Fraction III (gradient 1.075/1.085) contain T cells (43.2%) which consisted of CD4 (34.4%) and CD8 (18.6%), B cells (36.5%), and macrophages (10.0%). Fraction IV was strongly expressed T cell receptors (76.2%) which consisted of CD4 (63.6%) and CD 8 (20.0%), while B cells were found 20.0% and macrophages just about 0.6%. The activity of T cells as expressed by their respond to PHA and Con A shown that fraction III gave the most positive response, while fraction IV which contained the most ‘pure’ T cells gave less activity. From the present study it can be concluded that the discontinuous density gradient of Percoll method can be used to isolate and purify T cells in one step process. Furthermore, the present study indicated that T cells to proliferate in response to mitogen require other immune cells such as B cells and macrophages.
