    Utilization of organic chromium from tannery waste on reducing transportation stress and shortening recovery period at beef cattle

    Transportation increases stress that subtantially decreases body weight and feed comsumption even weight gain loss after arriving at the location of fattening. A research has been conducted to study the effects of organic chromium from tannery wastes on the level of transportation stress and recovery period in beef cattle fattening. Twenty Ongole crossbreed cattles were transported from the Wirasaba Feedlot at Purbalingga in Central Java to the Agro Citra Buana Semesta Feedlot in Malangbong-Garut in West Java for about 18 hours. Completely Randomized Design (CRD) was applied, wih four repetitions. The              Cr-organic was given seven days before and after transportation. Dose of Cr-organic used was: R0 = control diet without         Cr-organic, R1 = R0 + 1.5 ppm, R2 = R0 + 3.0 ppm, R3 = R0 + 4.5 ppm, R4 = R0 + 6.0 ppm. Results showed that 3.0 ppm organic chromium of the dry matter of ration tended to affect physiology and haematological conditions, as well as decreased weight loss, shortened recovery time, improved weight gain. It is concluded that organic chromium supplementation was able to lowered stress levels, shortened recovery time, and increased daily gain for one week recovery process, especially at dose of 3.0 ppm.     Detection of Mycobacterium avium subspecies paratuberculosis of dairy cows in Bogor

    Johne’s disease (JD) or partuberculosis is a chronic granulomatous enteritis in ruminants caused by infection of Mycobacterium avium paratuberculosis subspecies (MAP). The disease has been detected serologically in Indonesia. It’s potential to spread to other herds and could create great economic losses. The objectives of current study were to detect MAP in milk and faeces of dairy cows as well as to evaluate the association between farm management factors and presence of the bacteria in dairy cows in Bogor. The sample size was calculated using the formula to detect disease with the prevalence assumed to be 5% using 95% significant level. Milk and faeces samples were taken from 62 dairy cows which were suspected as suffering from MAP infection. Detection of MAP was done by isolation in Herrold’ egg yolk medium with mycobactin J (HEYMj), acid-fast bacilli Ziehl-Neelsen staining, PCR IS900 and F57. Biochemical test to confirm M. tuberculosis presence was also conducted. Fifteen isolates of Mycobacterium sp. were found from the faeces samples but not from the corresponding milk samples. However, conventional PCR conducted on the isolate as well as the milk samples, gave negative results. Biochemical test proved that all Mycobacterium sp. isolates were not M. tuberculosis. This study indicated the prevalence of MAP in Bogor was less than 5%. These findings should be continued by observational study to achieve the comprehensive information at the cattle and herd level.     The content of melamine in milk based products, and milk powders analyzed by liquid chromatography mass spectrometry

    Melamine is a white crystal of organic compound has a molecular weight of 126.12, difficult to solve in water. Cases of illegally adding melamine in milk powder is subjected to increase the nitrogen content of milk, because melamine contains high nitrogen (66%), so when milk is tested, seems contains high protein. This paper presented data the content of melamine in products based milk, and milk powders which entered and marketed in Indonesia. Melamine analysis is done by LC- MS 2010 EV, Shimadzu. Confirmation and validation tests indicate that melamine scanning found at m/z = 127, suitable system of analysis with relative standard deviation (RSD) given of 1,18% ( 2,0%). Accuracy test gave the average of 89.1% recovery, detection limit of 5 ppb and limit of quantition 7 ppb. About 91.3% samples (n = 46) collected from animal quarantine Tanjung Priok contained melamine in the range of 6.7 ppb to 61.5 ppb which is 1/49 to 1/16 times less than standard limit. Whereas about 40%, 14 out of 35 samples collected from Bandung and Jakarta supermarket was not detected of melamine, and 60% samples positive contain melamine in the range of 5,1 to 26,5 ppb (1/200 to 1/38 standard limit). However, all the samples analyzed contain melamine below the standard limit of 1 ppm determined by WHO/FAO. Key Words: Melamine, Milk Powder, Milk Based Products, LCM

    The influence of cage density on growth and behavior of Tangerang-Wareng Pullets

    One hundred and eight pullets of 13 weeks old white Tangerang-Wareng, were allocated to numbers of wire cages. Each cage had 4050 cm2 floor space. The treatments were three cage-densities (4, 6 and 8 pullets/cage) with six replications for each treatment for growth observation, while there were other three treatments (observation times), which were 07.00-08.00 (morning), 12.00-13.00 (noon) or 17.00-18.00 (afternoon), that were applied and replicated in three for behavior observation. Commercial ration containing 20.86% crude protein, 3.22 Ca, 0.87% total P and 2982 kkal metabolizable energy (ME)/kg and drinking water were provided ad libitum. Results showed that cage density neither significantly (P0.05) affected birds’ final bodyweight, bodyweight gain, feed consumption nor feed conversion ratio. Analysis of variance showed that there was no interaction effect of cage densites and observation times on every observed behavior variable, except on standing activity. Cage density did not significantly (P0,05) affect behavior, except behavior of cleanning, which increased with the increase of space allocation. The presentages of eating and pecking were significantly increase in the morning, while presentage of bird having a rest increased at noon. Drinking behavior was not affected by both cage density and observation time. The most birds showed the highest activities in the morning. In general the most cage density of 8 birds/4050 cm2 of floor space (506 cm2/bird), was more likely comfortable to the birds to live.     Effects of irradiation on the survival of bacterial contaminants in food

    The primary concern about microbial safety of irradiated food is the survival of pathogenic spore forming bacteria. Clostridium sporogenes was selected as the spore forming test organism for conducting inoculated pack studies for its similarities to the most toxigenic Cl. botulinum, in radiation resistance. Minimum radiation dose applied (45 kGy under cryogenic condition, in -790C) was determined to eliminate Cl. sporogenes spores and other bacterial contaminants in different kind of Indonesian chicken and beef dishes. In separate studies, irradiation doses of 3 – 7 kGy at cryogenic condition was used to improve the microbiological safety of a number chilled prepared meals. The dishes or ready to eat foods were packaged in air impermeable pouches. Irradiation process was carried out after inoculation on chicken and beef dishes with certain amounts of Cl. sporogenes spores. The evaluation of colony count differences between the irradiated and unirradiated foods revealed the effect of radiation on the survival of bacterial spores or other bacterial contaminants. It was demonstrated that a minimum dose of 45 kGy under cryogenic condition eliminated the spore of Cl. sporogenes, Bacillus spp and Staphylococcus spp. Irradiation at doses 5-7 kGy significantly reduced some potential pathogenic microorganisms in samples without affecting quality up to 3 months of storage at the refrigeration temperature.     In vitro slow-release urea characteristics under different molasses levels contained in rice straw based diets

    Slow-release urea characteristics of zinc-urea, zeolites-urea, and zeolites-zinc-urea were examined using in vitro techniques. The objective of this experiment was to study the in vitro slow-release urea characteristics of zinc-urea, zeolites-urea, and zeolites-zinc-urea under different molasses concentrations in relation to the ruminal fermentative changes observed in different incubation time. The experimental design employed was randomized block design with a 4 x 3 factorial arrangement plus a control treatment, and conducted in two replications. Factors were various urea sources (urea, zinc-urea, zeolites-urea, and zeolites-zinc-urea) and molasses concentrations (0%, 6%, and 12%) in rice straw based diets. The control treatment was rice straw based diet containing neither urea nor molasses. Diets consisted of 45% rice straw and 55% concentrates (DM basis) were formulated to have similar N and TDN levels. Responses of parameters measured were subjected to MANOVA using the GLM procedure of SPSS 16.00 and differences among mean values, if applicable, were examined using HSD-test. Orthogonal comparisons were used to determine the effects of control treatment vs. various urea sources following significance for the two-factor ANOVA model. Results indicated that zinc-urea, zeolites-urea, and zeolites-zinc-urea under different molasses concentrations contained in rice straw based diets decreased ruminal ammonia up to 48 hours incubation, controlled total VFA level and pH values revealed from lower NH3:VFA ratio, and improved both in vitro dry matter and organic matter degradabilities. The best impact of the in vitro slow-release urea characteristics of zinc-urea, zeolites-urea, and zeolites-zinc-urea on the ruminal fermentative changes (NH3, VFA, pH, DMD, OMD) was well attributed to the diets contained 6% molasses.     Polymorphism of Insulin-like growth factor-I (IGF-I) gene and their effect on growth traits in Indonesia native chicken

    The research was aimed is to detect Insulin-like growth factor-I (IGF-I) gene polymorphism and their effect on growth traits in Indonesia natives chicken. Seventy two Indonesian native chicken are going to be used in this research. The polymorphism of IGF-I gene was detected by PCR-RFLP/Pst-I. Four growth traits (body weight at 1, 2, 3, and 4 months) were recorded for analyzing the association between IGF-I gene polymorphism and growth performance.The results showed that allele A (621 bp) and allele B (364 and 257 bp) were found in this research. It was found that Indonesian native chicken carried high frequencies of allele A (0.82), and frequencies of IGF-I genotypes (AA, AB, BB) were 68.0, 27.8, and 4,2%, respectively. When compared to the IGF-I genotypes, the BB genotype had the highest body weight at 1, 2, 3, and 4 month (P0.05). The results showed that the B allele was positive of associated to a higher growth rate. Therefore, these results suggest that there is a possibility of IGF-I genotypes acting as a molecular marker for growth rate of Indonesia native.     Fixed Regression Test Day Model as a solution in predicting the genetic value of dairy cattle

    Genetic evaluation on milk yield production basically based on breeding value. This research was addressed to study milk production curve, for the most appropriate genetic evaluation, and the best methods in estimating genetic parameter and predicting breeding values. Data were collected from PT. Taurus Dairy Farm, consisting of 581 cows having 305 days records in first lactation (P305) and 542 cows having 305 days records in second lactation. Records used for Test Day (TD) evaluation were 5,373 for first lactation, and 4,925 for second lactation. Total records used for Test Day were 10,298. The results indicated that the most suitable curve, being able to be used for genetic evaluation was Ali-Schaeffer with the coefficient of correlation of 0.999, and the most appropriate model for estimation of genetic parameters and prediction of breeding values was fixed regression model with the regression of Ali-Schaeffer by taking account to the covariates. The genetic evaluation can therefore be conducted simultaneously by ignoring lactation levels. Key Words: Test Day, Milk Production Curve, Breeding Value, Dairy Cattl

    improving performance of laying hens

    A trial was conducted in order to study the effect of the supplementation of Avizyme 1500® (Danisco Animal Nutrition, Marlborough, UK) on the performance of laying hens for one year. A control diet based on corn – soybean meal was formulated to meet nutrient requirement of ISA Brown laying hens. Two treatments, the control diet (C) and C + 1000 g Avizyme/tonne diet were tested. Each diet was fed to 80 birds (20 replicates of 4 birds) from 20 to 72 weeks of age, and performances of birds (feed intake, egg production, egg size, egg mass, feed conversion ratio, and egg quality) were measured. All data were subject to analyses of variance following the t-test. Results showed that the addition of Avizyme 1500 to the feed reduced feed intake by 4% (P 0.01), mortality by 75 % or from 15% to 3.75% (P 0.01) and improved the feed conversion ratio by 3 % (P 0.05).  The high mortality of the control treatment (15%) is explained by an E.coli infection that was observed following the post-mortem examination of dead birds. The egg production (HD and HH), egg size and egg mass however were not significantly affected by the Avizyme supplementation. Egg quality (HU, yolk colour score, yolk weight and shell thickness) was not significantly affected by Avizyme supplementation. It can be concluded that the supplementation of 1000 g Avizyme /tonne of diet improved feed efficiency and this was mediated via a reduction in feed intake.     Response of sheep fed on concentrate containing feather meal and supplemented with mineral Chromium

    A study was conducted to evaluate the effect of substitution of protein concentrate with feather meal supplemented with organic chromium mineral on performance of lambs. Twenty five male lambs were fed basal feed of fresh chopped king grass ad libitum and were allotted to either one of five different supplements (five dietary treatments): Control (C); 10% of protein in concentrate was substituted by feather meal (FM); 10% of protein in concentrate was substituted by feather meal supplemented with Cr yeast at 1.5 mg (FMCrOrg); 10% of protein in concentrate was substituted by feather meal supplemented with Cr inorganic which equal to the amount of Cr bound in yeast (FMCr); Concentrate control supplemented with 1.5 mg Cr yeast (CCrOrg). Cr-organic was synthesized by incorporating CrCl3 in fermented rice flour by Rhizopus sp. The mineral is mixed with feather meal as a mineral carrier. Sheep in all treatments received iso protein concentrate. Parameters observed were body weight change, feed consumption and nutrient digestibility. Results shows that there was no significant effect of diet treatments on average daily gain (ADG), dry matter consumption and feed conversion, with the average value of 75.4 gr/day; 74.9 g/BW0.75 and 9.9 respectively, However diet treatment of organic chromium and protein substitution with feather meal (FMCrOrg) showed tendency of having higher ADG (83.57 g/h/d). Average nutrient digestibility of dry matter, organic matter and NDF were 68.7; 69.6 and 60.9%, respectively. However NDF digestibility of FMCrOrg tended to be higher than other treatment (67.0%). It is concluded that partial substitution of protein concentrate by feather meal and 1.5 mg Cr-organic supplementation did not affect sheep performance.
