    Synthesis of an oxytetracyline-tolidin-BSA immunogen and antibodies production of anti-oxytetracyline developed for oxytetracyline residue detection with enzyme-linked immunosorbent assays technique

    An oxytetracycline-tolidin-bovine serum albumin (OTC-tolidin-BSA)-conjugate was synthezed as immunogen for producing specific antibodies in immunized rabbits that would be used as reagent for development of OTC residue detection with enzym-linked immunoassays technique. The immunogen was prepared through diazotization tolidin and subsequently reacted with OTC. The red purple immunogen of OTC-tolidin-BSA absorbed at wave lengths of 277 nm and 488 nm under UV screening absorbances and confirmation with the high performance liquid chromatography (HPLC) showed the absence of peak at retention time of 3.46 minutes. Characaterized result with SDS-PAGE showed the molecular weight of the OTC-tolidin-BSA at 69.79 kDA. Subsequently, the immunogen was immunized into New Zealand rabbits in order to produce the polyclonal antibodies. The antibodies were purified using a protein A sepharose column. The OD optimum responses of 0.92 to 1.20 were obtained from the second fractionation at dilution of 1/1000 by titrating the antibodies and OTC-tolidin-BSA coating antigen at concentration of 10 µg/mL on several bleeding times.     Improving the nutritive values of solid heavy phase to substitute corn in laying hens diet

    Solid heavy phase (SHP), a by product material of palm oil factory obtained by ceramic filtration from liquid waste could be used as a feedstuff to replace corn in poultry diet. A series of experiment was carried out to improve nutrient value of the SHP by supplementation of enzymes and amino acids in order to increase the proportion of SHP to substitute corn in layer diet. There are three enzymes i.e.: Balitnak production (BS4), a commercial single enzyme (consist of mannanase) and comercial multienzymes were tested. All the enzymes were mixed with fresh SHP in different dose, dried and ground. The nutrient digestibility of these materials was measured in order to decide the optimum level of each enzyme. Based on this result, a feeding trial was carried out. Experimental diets were formulated to study the effect of substitusion of 25% or 50% corn with dried SHP or enzymes-treated SHP on the performances of the layers. The effect of methionine and lysine supplementation into diets contained high levels of SHP was also studied. Results showed that all enzymes studied could increase the energy (TME) of the SHP. BS4 enzyme and the commercial multienzimes, except single enzyme, also increase the true protein digestibility of the SHP. The optimum dose of each enzyme for each kg dry mater of SHP was 13.3 ml BS4, 2 g single enzyme and 3 g multienzymes. Substitution of 25% corn in layer diet with dried SHP or enzymes-treated SHP did not significantly impair the performances (hen-day egg production and FCR) of layers. However, substitution of 50% corn with SHP + multienzymes or SHP + single enzyme significantly impaired the performances of the layers. Addition of methionine and lysine amino acids restored the performance of the hens fed with SHP + commercial multienzyimes, but not those fed with high levels of SHP + commercial single enzyme. Substitution of 50% corn with SHP + BS4 enzime did not significantly impaire the performance of layers and therefore, addition of amino acids into the diet was not required. Substitution of 25% or 50% corn with dried SHP or enzymes-treated SHP did not affect the egg quality (HU, yolk color index and shell thickness). It is concluded that solid heavy phase (SHP) could be used to substitute 25 to 50% corn in layer diet, especially when the SHP was treated with BS4 enzyme.     Alleviation of oxidative stress, carcass fat and plasma cholesterol in broiler chickens consuming antanan (Centella asiatica) and vitamin C

    Oxidative stress (a condition where oxidant -free radical- activity dominates the antioxidant activity) in poultry may be caused by heat stress from high environmental temperature. In this research, antanan (Centella asiatica) and vitamin C was utilized as anti heat-stress agents for broilers. One hundred and twenty male broilers of 2 – 6 weeks of age were kept at 31.98 ± 1.940C poultry house temperature during the day and 27.36 ± 1.310C at night. The treatments consist of two kinds: two levels of vitamin C (0 and 500ppm) and three levels of antanan (0, 5 and 10% of diets). The data colected was analized for variance with a factorial in completely randomized design of 2 x 3 (2 levels of vitamin C, 3 levels of antanan) and continued with contrast-orthogonal test when applicable. The results indicated that the treatments of antanan 5% without vitamin C (A5C0), antanan 10% without vitamin C (A10C0), vitamin C 500ppm without antanan (A0C500), combination of A5 and C500 (A5C500), and combination of  A10 and C500 (A10C500) significantly (P0.05) decreased the level of liver malonaldehida (MDA), carcass lipid and plasma cholesterol.     Utilization of Passion fruit hulls (Passiflora edulis Sims f. edulis Deg) as component of complete feed for growing Kacang goats

    Processing of passion fruit (Passiflora edulis Sims f. edulis Deg) to produce passion fruit juice produce passion fruit hulls that is potential for feedstuff. Twenty male kacang goats (average initial body weight 17±1.24 kg) were used in an experiment to study the effect of utilization of passion fruit hulls as feed component in the complete pellet ration on their growth. The experiment was arranged in completely randomized design consisting of 4 diets and 5 replications. Animal were randomly allocated into 4 diets (0, 15, 30, 45% level of passion fruit hulls). Each diet contained 14% crude protein and 2550 Kcal kg-1 metabolism energy. The ration was offered at 3.8% of body weight based on dry matter. The result of the experiment showed that all variables observed were not affected by level of passion fruit hulls (P0.05). Average daily gain and feed efficiency tended to decrease with the increase level of passion fruit hulls. The highest dry matter, organic matter and N intake (768,78 g h-1 d-1; 687,37 g h-1 d-1 and 17,22 g h-1 d-1 respectively) were found from R1 treatment (15% level of passion fruit hulls). It was concluded that passion fruit hulls can be used till 45% level in the diet of Kacang goat. Key Words: Passion Fruit, Complete Pellet Ration, Kacang Goa

    Characteristics of Body Measurement and Shape of Garut Sheep and Its Crosses with Other Breeds

    It is important to know body measurement because it could be used to estimate body weight as well as to differentiate the chrateristic of body measurement and shape of animal due to different breed or environment. This research was carried out to study morphometric characteristic of body size and body shape from 78 of Garut sheep (GG), 29 HG sheep {crossbred between St. Croix (HH) and Garut sheep (GG)}, 36 MG sheep {crossbred between Mouton Charollais (MM) and Garut sheep (GG)}, 62 MHG sheep (MG x HG) and 38 HMG sheep (HG x MG). Body part measured were wither height (X1), rump height (x2), body length (X3), chest width (X4), chest depth (X5), hip width (X6), chest girth (X7), cannon circumference (X8) and hip length (X9). Data obeserved were analised using t test and Principle Components Analysis (PCA). Based on PCA it was showen that chest girth was the primary identity for body measurement on males and females of Garut, HG, MG, MHG and HMG with its Eigenvector value 0.689; 0.709; 0.689 and 0.681 respectively. The primary indentity for body shape of Garut sheep were chest girth and hip heigth with Eigenvector value -0.600 and 0.551 respectively. The primary indentity for body shape of HG sheep were body length with Eigenvector value -0.725. The primary indentity for body shape of MG sheep were chest girth, rump heigth, and wither heigth with Eigenvectors value: -0.600; 0.558 and 0.555 respectively. The primary indentity for body shape of MHG sheep was wither height with Eigenvector value 0.608. The primary indentity for body shape of HMG was body length with Eigenvector value 0.764. Body shape of HG and MG sheep is different than that of Garut sheep, but the body shape of MHG and HMG were close to Garut body shape. This result indicated that the adaptability to environment of HMG and MHG is close to that of Garut sheep. Key words: Sheep, Body Size, Body Shape

    The quality of Garut ram liquid semen in Tris egg yolk extender to the sucrose supplementation

    The successful program of artificial insemination in sheep is determined by the quality of sperm that are used. Therefore, it is important to maintain the quality of sperm during and after storage in low temperature. The research was conducted to study the influence of sucrose in the Tris-yolk extender in maintaining the quality of Garut ram sperm during preservation in reaction tube for four days at 5°C. Ejaculated sperm was collected once a week for five weeks using artificial vagina from same ram. Semen was divided into four groups of extenders, i.e. Tris egg yolk 20% (TKT), TKT + 0,1% sucrose, TKT + 0,3% sucrose and TKT + 0,5% w/v sucrose. The results showed that the percentage of motility on day four (D-4) of storage in TKT (41.00 ± 2.0%) was lower (P0.05) than that in TKT + 0.3% (48.00 ± 2.45%) and TKT + 0.5% (51.00 ± 3.74%), however there was no significant different (P0.05) from TKT + 0.1% (45.00 ± 3.16%). Whereas the percentage of live cells (%H) on D-4 in TKT, TKT + 0.1%, TKT + 0.3% and TKT + 0.5% were 55.00 ± 2.19%; 57.20 ± 2.79%; 59.20 ± 3.25% and 61.20 ± 3.60%, respectively. Meanwhile, the percentage of membrane integrity (MPU) in TKT (51.80 ± 1.94%) was significantly different (P0.05) from TKT + 0.3% (57.40 ± 2.65%) and TKT + 0.5% (59.20 ± 3.66%), however there was no significant different (P0.05) from TKT + 0.1% (54.80 ± 2.86%). In conclusion, the addition of sucrose 0.3% w/v into TKT extender could maintain the quality of Garut ram sperm more efficiently and better than TKT extender.     Ability of fiber bacteria isolated from buffalo rumen in digesting various sources of forage

    Local buffalo rumen fluid is a source of fiber digesting bacteria. Such bacteria presumably are well adapted to feed stuffs derived from agricultural byproducts with low quality. The purposes of this study were: (1) to isolate the fiber-digesting bacteria from buffalo rumen fluid, (2) to determine the fiber digesting characteristics and adaptability of such bacteria, and (3) to characterize the bacterial diversity.  Rumen fluids for the experiment were collected from buffaloes slaughtered at the Faculty of Animal Science, Bogor Agricultural University abattoir. Fiber-digesting bacteria were isolated using McDougall buffer supplemented with fibrous feed. A factorial experiment was conducted to study characteristics and adaptability of the bacteria using two factors: the type of bacteria and type of feed source of the fiber. Diversity among the bacteria was analyzed using the NTSys2.10 program.Results showed that nine bacterial isolates had a high adaptability to fiber feed based on CMCase. The highest CMCase activity bacteria for Pennisetum purpureum were A9 (11.36±1.70 unit/ml/h), A3 (11.22±0.60 unit/ml/h) and A42 (10.62±1.96 unit/ml/h). CMCase activity of fiber digesting bacteria from buffalo rumen fluid was not correlated with the number of bacteria in the culture.     The performance of Nagrak and Kampung chicken kept intensively in Cibadak Sukabumi, West Java

    A study on the performance of Nagrak and Kampung chicken under intensive management system was conducted in Cibadak District of Sukabumi West Java. As many as 200 hens of Nagrak and Kampung, each were placed in individual cages. The birds were given 90 g of diet daily, the diet was the mixture of layer commercial diet and ricebran at equal ratio, drinking water was given ad libitum. Artificial insemination was conducted every three days using semen collected from 20 cockerels of each local bird. Disease control was done by vaccination against Newcastle Disease and Infectious Bursal Disease. Sulfamix and antibiotics were given whenever needed. Eggs were collected twice a day, weighed and stored to be incubated or to be analyzed for their characteristics. Parameters observed were physical appearance, hen-day egg production, egg quality, egg fertility and hatchability, body weight, feed conversion and mortality. Results showed that Nagrak chicken physical appearance was different from that of Kampung chicken, yet their hen-day production were not significantly different (26.93 ± 14.10% vs 27.04 ± 16.20%). The egg characteristics of these birds (Nagrak vs Kampung) were as follows: egg weight (36.29 ± 6.50 g vs 35.55 ± 5.42 g); yolk weight (16.61 ± 1.34 g vs 16.22 ± 2.11 g); albumen weight (17.31 ± 2.64 g vs 16.87 ± 1.35 g); shell weight 37 ± 0.81 g vs 2.46 ± 0.54 g); yolk color (9.62 ± 1.81 g vs 9.67 ± 1.70 g); haugh unit (83.60 ± 5.41 vs 83.45 ± 6.10); shell thickness (24.0 ± 0.83 µm vs 24.4 ± 0.67 µm), however no significant difference was found. Neither the egg fertility, hatchability nor mortality rate of the birds was significantly different. At 12 weeks old the average body weight of male Nagrak chicken (1260.04 ± 57.33 g) was much higher (P0.01) than that of male Kampung chicken (750.68 ± 60.11 g) while the average body weight of female Nagrak chicken (980.37 ± 48.11 g) was much higher (P0.01) than that of female Kampung chicken (656.11 ± 58.47 g). The feed conversion ratio of male Nagrak chicken (4.21) was better (P0.05) than that of male Kampung chicken (5.62), the feed conversion ratio of female Nagrak chicken (5.15) was also better (P0.05) than that of female Kampung chicken (6.81). No significant difference was found on the feed conversion ratio of Nagrak hens (9.24) and Kampung hens (9.06). Key words: Performance, Nagrak, Kampung, Local Chicke

    Effects of inclusion Aspergillus niger fermented shrimp waste meal in broiler diets on live performance and digestive organ weight

    An experiment was conducted to evaluate the effect of the inclusion of different levels of shrimp waste meal fermented with Aspergillus niger (LUF) in diets on growth performance and digestive organ weight of broilers. A total of 75 d-old chicks were randomly allocated to 5 (five) treatments in 3 replication pens of 5 birds each. Treatments consisted of LUF inclusion of 0 (control), and 5, 7.5, 10 and 12,5% (P0, P1, P2, P3 and P4) in the diets. Birds were raised under standard condition and provided with feed and water ad-libitum. Feed and birds were weighed weekly up to 35 days to determine body weight, feed intake and feed conversion. At the end of experimental period, the birds were slaughatered and dressed up to determine carcass percentage and digestive organ weight. There was a significant negative linear response in body weight, feed consumption and feed conversion with increase of LUF more than 7.5% in the diets until 35 days of age, but % carcass was almost the same for all treatment. There was no significant response in digestive organ weight with increasing levels of LUF. The present result indicated that LUF could be considered as a potential feed ingredient as protein source of broiler but its inclusion should be limited until 7.5% of the diet to maintain growth performance and digestive organ weight.     Purification of neuraminidase from Influenza virus subtype H5N1

    Influenza-virus neuraminidase plays vital role in the survival of the organisms. Vaccination of animals with this glycoprotein confers immune responses so that enable it to protect the animals from incoming infection. Supplementation of conventional vaccines with this glycoprotein increases the protection and longevity of the vaccine. Purified neuraminidase can also be used to develop serological tests for differentiation of serologically positive animals due to infection or to vaccination. In this study purification of neuraminidase from influenza virus subtype H5N1 was described. Triton x-100 and Octyl β-D-glucopyranoside were used to extract and diluted the glycoprotein membrane. The enzymatic activity of the neuraminidase was assayed using a fluorochrome substrate, 4-methylumbelliferyl-a-D-N-acetyl neuraminic acid, which was found to be simple, sensitive and suitable for the purification purpose. The neuraminidase was absorbed selectively on an oxamic-acid agarose column. The purity of neuraminidase eluted from this affinity column was high. A higher purity of the neuraminidase was obtained by further separation with gel filtration on Superdex-200. The purified neuraminidase was enzymatically active and did not contain any detectable haemagglutinin, either by haemagglutination assay or by monospecific antibodies raised against H5N1 hemagglutinin.  The purified neuraminidase was recognized strongly by antibodies raised against an internal but only weakly by that against C-terminal regions of the neuraminidase protein of H5N1-influenza virus. The purified neuraminidase was in tetrameric forms but dissociated into monomeric form on reducing condition, or mostly dimeric form on non-reducing SDS-PAGE.
