    Influence of concentrate-rice straw ammoniated ratio and fattening period on body weight and meat physical quality of Pesisir cattle

    Pesisir cattle, an original local cattle of West Sumatera, have good genetic potential because they adapt very well to both the low-quality feed, and changes in ambient temperature. This research was conducted to determine body weight, feed consumption and conversion, daily gain and meat quality (tenderness, water holding capacity, cooking loss and pH) of Pesisir cattle in different ration composition and feedlot period. This experiment was done based on Randomized Block Design in 2 x 2 factors with 3 groups. The first factor (factor A) was two kinds of diet i.e. A1 = 50% concentrate + 50% ammoniated rice straw and A2 = 75% concentrate + 25% ammoniated rice straw. The second factor (factor B) was duration of fattening (B1) i.e. 3 months and (B2) 4 months. There was no interaction between the level of the diet with duration of fattening on body weight, daily gain, tenderness, water holding capacity and cooking loss on meat of Pesisir cattle (P 0.05). Body weight in A2 group was significantly higher than A1 beginning from second month of treatment. Factor A had significant effect on daily gain. The pH value of meat of cattle that received 75% concentrate was lower than that received 50% concentrate. The higher the percentage of concentrate in diet caused better body weight, daily gain, consumption, feed conversion and tenderness.     Cloning and Clone Analysis of GRA1 Gene from Local Isolate Toxoplasma gondii Tachyzoite

    The GRA1 gene of Toxoplasma gondii encoding protein called GRA1 protein. GRA1 protein known to be immunogenic and essentialy involved in modification of parasitophorus vacoule which has role in immune evasion and virulency of organism. The local isolate of T. gondii is successfuly isolated and known as highly pathogenic isolate similarly as its RH strain. Unfortunately, the homology sequence of GRA1 gene between those isolate still unknown. The purpose of the research are to clone the GRA1 gene and to analyze the homology from pathogenic T. gondii isolate and RH strain. Tachyzoite of T. gondii was grown in mice peritoneum by intraperitoneal injection. Then, total mRNA was isolated and purified. cDNA was synthesized from mRNA and then amplified using F1 dan R1 primers to get clone of GRA1 from local isolate. Homology analysis was perform using several bioinformatic softwares. The result showed that cDNA of GRA1 from local isolate has 84% homologs with RH strain of T.gondii. However, when subsequently editing performed to parts of suspected non coding sequence of cDNA GRA1 to get CDS of GRA1, the homology was increase to 100% compare to CDS of GRA1 of RH strain.     Impact of sheep stocking density and breed on behaviour of newly regrouped adult rams

    Placing animals in cages with certain density and good grouping were two important aspects needed in intensive livestock production system to produce optimal production and animal welfare. The objective of this study was to examine effect of stocking density, breed and elapse of time on behaviour of newly regrouped, unacquainted adult rams from three sheep breeds i.e. Barbados Blackbelly Cross, Local Garut and Composite Garut, as possible factor causing variation in welfare status. Instantaneous scan sampling was used for recording sheep behaviour at three different stocking densities. Thirty-six adult rams were used in this research and divided into three groups (n = 12) on the basis of breed. At each stocking density four rams of the same breed were observed during two consecutive days. The recorded behaviours were agonistic-, self-care-, exploratory-, aberrant-, mating-, locomotive- and standing behaviour. The results showed that during the entire experiment agonistic behaviour was observed at the highest frequency. Stocking density was found to have a significant effect on exploratory-, locomotive- and standing behaviour. The effect of breed was found to cause significant differences in agonistic-, self-care-, aberrant- and mating behaviour. Significant differences were also found between day 1 and day 2 of regrouping for agonistic-, exploratory, self-care- and mating behaviour. It is concluded that the three breeds do differ in their behavioural reactions to different stocking density levels and time needed for adaptation after regrouping.     The quality of spotted buffalo epididymal sperm with addition of raffinose as external cryoprotectant

    The aims of this research was to obtain the quality of Spotted buffalo epididymal sperm in different kind of extender in the three stages of cryopreservation (after dilution, post equilibration and post thawing). Spermatozoa was collected with combination of slicing and pressure method into the epididymal tissue in Andromedâextender. Soon after diluted and equilibrated, epididymal spermatozoa was cryopreserved in liquid nitrogen (-196°C).  The result showed that the percentage of motility after thawing in Andromedâ + raffinose 0.4% (47.0 ± 2.4%), was significantly different (P0.05) from that of control (41.0 ± 2.0%), but there was no significantly different (P0.05) from that of Andromedâ + raffinose 0.2% (46.0 ± 2.0%). The percentage of live sperm after thawing in control (52.2 ± 2.5%), was the lowest and significantly different (P0.05) from that of Andromedâ + raffinose 0.2% (59.2 ± 2.6%) and Andromedâ + raffinose 0.4% (58.8 ± 3.1%). Moreover, the percentage of membrane integrity after thawing in control, Andromedâ + raffinose 0.2% and Andromedâ + raffinose 0.4% was 68.0 ± 1.1%; 67.2 ± 1.6% and 67.6 ± 1.2%, respectively. There was no significantly different (P0.05) in the percentage of membrane integrity from all treatments. In conclusion, the addition of 0.2 and 0.4% raffinose into Andromedâ extender could improve the percentage of motility and viability of post thawing spotted buffalo epididymal spermatozoa.     Extraction and isolation of Ovine Pregnancy-Associated Glycoprotein (ovPAG) from cotyledon placenta of Garut sheep

    Pregnancy-associated glycoprotein (PAG) structurally related to aspartic protease, expressed in the outer epithelial cell layer (trophectoderm) of ungulate placenta. Ovine PAG (ovPAG) synthesized by mono- and binucleic trophoblast before complete implantation at Day 14-15. Of this, ovPAG could be used as a marker for early pregnancy. The objective of study was to extract and isolate PAG from placenta of Garut Sheep collected at term and to characterize their molecular weight. The procedures included extraction of protein at neutral pH (cotyledon was thawed, minced, added PBS, blended and centrifuged), acidic (H3PO41M, pH 4,5; centrifuged) and ammonium sulfate (40% and 80% (NH4)SO4, centrifuged) precipitation; gel filtration (Sephadex-G75), anion exchange chromatography (DEAE- cellulose). Cotyledon extract was subjected to Sephadex-G75 and DEAE cellulose, and their fractions were measured their absorbances. Absorbances of Sephadex-G75 and DEAE fractions at peak were assayed for protein concentration (Bichinconinic protein assay). Continuously, these fractions were subjected to monogel SDS-PAGE and stained by Commassie Brilliant Blue. It was four different molecular weights isolating from cotyledon of Garut Sheep, namely 68.8, 36.04, 32.39 and 12.18  kDa. However, after chromatography anion exchange (DEAE-cellulose), only  three bands exist, those were  71,67;  33,64 and 30,86 kDa.     Inactivation of Bacillus anthracis by Gamma irradiation

    The use of Bacillus anthracis as a biological weapon heighlightened awareness of the need for validated methods for the inactivation of B. anthracis spores. Ionizing radiation is capable of causing a variety of chemical changes and biological effects on bacteria which can be due both to direct interactions with critical cell components and to indirect actions on bacteria by molecular entities formed as a result of radiolysis of other molecules in the bacterial cell. This study determined the gamma irradiation dose for inactivating B. anthracis spores and its biological effects on the bacterial characteristics. Gamma irradiation was conducted at the IRKA irradiator at the National Nuclear Energy Agency, Jakarta and cobalt-60 was used as the source of ionizing radiation (capacity of ca. 134,044 Kci). Freeze dried culture of B. anthracis in glass ampoules was irradiated using   variable doses of 30, 20 and 10 KGy. Viability, biochemical and protease enzyme characteristics of B. anthracis were evaluated before and after irradiation.  The ability of B. anthracis to degrade gelatin, haemoglobin and bovine immunoglobulin G was also tested. The results showed that ionizing radiation  was able to inactivate or kill 11,05 x 108 cfu B. anthracis by 95.37%, 99.58% and 99.99 at respective doses of 10, 20 and 30 KGy. Bacterial spores appear to be less susceptible to irradiation than the vegetative cells, because of their specific structure. The survive spores irradiated at 30kGy shows some biochemical characteristic changes. The survivors failed to degrade methyl -D-glucopyranoside and arbutine. The ability of B. anthracis protease to degrade gelatin, haemoglobin and bovine immunoglobulin G was not affected by irradiation. These findings showed that a gamma irradiation at 30 KGy effectively inactivates B. anthracis spores without changing the protease activities.     Puberty of Katingan cow in realation to Cu mineral and the environtment

    The onset of puberty is an important role in order to optimize performance of Katingan cattle reproduction. The onset of puberty can be estimated from to blood progesterone concentration. In this study the onset of puberty was estimetid through analysis of progesterone hormone in various age of individual cattle. Thirty blood samples were obtained from 30 Katingan heifers of 13 months and 15 days to 23 months and 18 days old from Tumbang Lahang (10 samples) and Buntut Bali (20 sampels) to be analyzed for progesterone concentrations using RIA method. The same samples were also analyzed to find out information about Copper (Cu) concentration. The results showed that progesterone concentration varied narrowly from 0.008 to 0.184 ng/ml. The result indicated that the Katingan cattle in 23 months old was still in prepuberty category. Environment factor such as land pH was acid (pH6), grass quality and climate in term of temperature and humidity relatively high, may an important role to delay the oset of their puberty.     Nitrofurans residue in broiler chicken meat which analysed by an HPLC

    Furazolidone (FZD), furaltadone (FTD), nitrofurantoin (NFT) and nitrofurazone (NFZ) are veterinary drugs that belong to the nitrofurans (NFs) group and employed as feed additives for growth promotion and theurapetic treatment of gastrointestinal infections caused by Eschericia coli and Salmonella spp. The occurrence of NFs in animal products will end to cause health problem in human consumed such food. This research conducted to study the analysis of NF residues in chicken meat by a high performance liquid chromatography (HPLC) and to study the occurrence of NFs residues in samples collected from traditional markets and supermarkets in Bandung, Bogor and Depok. The results of validation method on several parameters for each NF showed that the average of the relative standard deviation (RSD) from the precision study were 2.15 to 2.38%, the R2 values of the linearity study were 0.9964 to 0.9995; recoveries were 75.90 % to 91.50 % and the detection limits were 12.01 to 37.25 ng/g. The residual level of NFs for 42 field samples showed that 2 samples positive for NFZ (9.09 and 10.74 ng/g), 1 positive for NFT (10.46 ng/g), 4 positive for FTD (16.44 up to 27.21 ng/g) and none positive for FZD. Present results showed that analysis of NFs in broiler chicken meat can be done using an HPLC and the analysis results from field samples showed that these types of drugs were being used for broiler chicken production both as single and/or combination drugs, therefore it is necessary to raise public awareness to monitor the use of NF in livestock production in Indonesia.     Effect of Supplementations of Comin+ and Zn-biocomplex on the performances of Ettawa Crossbred goats

    Digestibility of ruminant feeds is greatly determined by ruminal fermentation which in turn is greatly affected by micro-ecology of the rumen. Manipulation of micro-ecology such as supplementing with various yeasts or micro elements could improve digestibility of the feed. The effect of Comin+ (R1) and Zn-biocomplex (R2) alone or their combination (R3) on performance of Ettawa Crossbred kids, fed concentrate containing Aspergillus oryzae (R0) was studied. Twenty four Ettawa Crossbred kids (6-8 month of age; initial liveweights =16.63±2.10 kg) were divided into 4 groups. Feeds given were King grass (Pennicetum purpuroides) ad libitum and concentrate. The observations were carried out for 16 weeks with 2 weeks adaptation period and the experiment was conducted in a completely randomized design with 4 treatments and 6 replications. The results showed that supplementations significantly (P 0.05) increased total DM intakes from 685 g/d (R0) became 748, 711 and 858 g/d; and grass DM intakes from 304 g/d (R0) became 373, 331 and 479 g/d for R1, R2 and R3, respectively. Digestibilities of DM, NDF and DE/GE were not affected by supplementation, but supplementations significantly (P 0.05) affected digestibilities of ADF and CP. Supplementation also significantly improved (P 0.05) ADG from 65.18 g/d (R0) became 94.64, 83.04 and 90.77 g/d; and FCR from 10.51 (R0) became 7.82, 8.36 and 9.46 for R1, R2 and R3, respectively. It was concluded that supplementation of Comin+ (R1) gave the best growth performance of Ettawa Croosbred goats, where ADG and FCR improved by 45.20% and 34.40%.     Reproductive performances of Boe, Kacang and Boerka does

    Does reproduction performance could be expressed by her ability to give birth and to give milk to their kids during the pre-weaning period. This study was aimed to evaluate the reproductive performance of Boer, Kacang and Boerka does; and was carried out for two years at Research Institute for Goat Production, Sungei Putih. The materials used were goats owned by the institute. The parameters observed were: litter size, parity of does, preweaning mortality, kidding interval and sex ratio of kids. The rate of reproduction of the does was estimated using Amir and Knipscheer methods and were statistically analyzed using General linear model. Results showed that litter size and kidding interval of Boer goats were higher (P 0.05) then that of Kacang goats, while the Boerka goats was in between. The mortality at preweaning of Boer goats was lower (P 0.05) then that of Kacang goats, while the Boerka goats was in between. The percentage of kid sex ratio of three breeds were fluctuated. Parity of does had significant effect on all reproduction traits (P 0.05) except for sex ratio of kids. The does reproduction was smallest at the first parity, and increased with the increase of parity from one to four, and then decreased in subsequent parities. The highest does reproduction rate was found in the Boerka (1.82), followed by Boer (1.80) and Kacang (1.80). It is concluded that the reproductive performance of the three breed female goat was relatively the same. Key Words: Performance, Reproductive, Boer, Kacan
