    Viability of Bali bull epididymal spermatozoa preserved at 3–5oC in Tris extender with different lactose concentrations

    Cauda epididymal spermatozoa could be used as an alternative source of gamete in the application of various reproductive technologies, because the spermatozoa is motile and has ability for fertilizing the oocyte. The objective of this research was to examine the effectivity of lactose in maintaining viability of Bali bull epididymal spermatozoa preserved at 3–5oC. Five testis with epididymides of Bali bulls were obtained from slaughterhouse. Epididymal spermatozoa was collected by the combination of slicing, flushing and tissues pressure of cauda epididymides with physiological saline (0.9% NaCl). Collected-spermatozoa was divided in equal volume into three tubes and diluted with Tris extender containing 20% egg yolk (control), Tris extender + 0.3 g lactose/100 ml (L0.3), and Tris extender + 0.6 g lactose/100 ml (L0.6), respectively. Diluted-spermatozoa was stored in refrigerator at 3–5oC. Quality of diluted-spermatozoa including percentages of motile spermatozoa (MS), live spermatozoa (LS), and intact plasma membrane (IPM) were evaluated every day during storage at 3–5oC for six days. Data were analyzed using completely randomized design with three treatments and five replications. Results of this study showed that mean spermatozoa concentration, percentage of MS, percentage of LS, percentage of abnormal spermatozoa, percentage of cytoplasmic droplet, and percentage of IPM of Bali bull fresh epididymal spermatozoa were 11,222.5 million cell/ml, 75, 86.75, 10.5, 14, and 86.75%, respectively. At day-7 storage, percentages of MS, LS, and IPM for L0.3 (39, 50.6, and 51.6%) and L0.6 (39, 51.4, and 51.8%) were significantly (P0.05) higher than control (29, 41.8, and 42.4%). In conclusion, addition of lactose in Tris extender extended viability of Bali bull epididymal spermatozoa preserved at 3–5oC. Key Words: Lactose, Preservation, Epididymal Spermatozoa, Bali Bul

    Comparison of four diluents for the retriever dogs semen preservation

    The quality of chilled semen depends on the composition of diluent. The choice of the buffer, anti-cold shock and nutrition sources can be the first decision in order to choose appropriate diluents. Nowadays a lot of diluent are used for canine semen preservation such as Tris buffer and Citrate buffer. This study was aimed to observe the differences of diluent for preserving Retriever dog spermatozoa. The semen sample collected from four Retriever dogs with three times repetition. The semen was evaluated macro-and microscopically. The semen with 70% sperm motility was divided into four tubes and diluted with diluter 1 (P1), diluter: P2, P3 and P4 (modified P3). The diluted semen was divided into two tubes and each sample was stored at room and 50C temperature. The viability of chilled semen was observed every 3 hours at room temperature and 12 hours at 50C. The result showed that P2 keep the sperm viability better than the other diluents. On 50C at 24 hours storage P2 showed the highest motile and live sperm percentage (46.25 ± 0.22%; 57.11 ± 0.25%). In room temperature at 6 hours P2 showed the highest motile and live sperm percentage (40.94 ± 0.20%; 52.65 ± 0.23%). It is concluded that P2 can keep the sperm viability by 84 hours of 50C and 21 hours at room temperature. Key Words: Diluents, Dog Sperm, Retrieve

    Clostridial necrotic enteritis in chicken associated with growth rate depression

    Clostridium perfringens (C. perfringens) is a normal inhabitant of the intestinal tract of chickens as well as a potential pathogen causing necrotic enteritis. C. perfringens only causes necrotic enteritis when it transforms from non-toxin producing type to toxin producing type. The alpha toxin, (phospholipase C) is believed to be a key to the occurrence of Clostridial necrotic enteritis (CNE). The best known predisposing factor is mucosal damage, caused by coccidiosis that damages the intestinal lining, making the gut susceptible to infections including C. perfringens. The purpose of this study was to observe the chicken performance in experimental CNE and field cases of CNE. Diagnosis of CNE were made by latex agglutination test, isolation and identification of the agent. Pathological and histopathological changes were also observed. Experimentally, NE could be reproduced when Eimeria sp and C. perfringens spores are inoculated in chicken. Signs of an NE are wet litter and diarrhea, and an increase in mortality is not often obvious. The depression of growth rate and feed efficiency of chicken become noticeable by week 5 because of damage to the intestine and the subsequent reduction in digestion and absorption of food. Subclinical form of CNE was also frequently found in the field, leading to significant decreases in performance. Chicken gut samples examinations revealed that subclinical form of CNE causes damage to the intestinal mucosa caused by C. perfringens leads to decreased digestion and absorption, increased feed conversion ratio and reduced weight gain. Dual infection with C. perfringens and Eimeria sp. was frequently found in field. The results of these studies provide evidence for C. perfringens as a causative bacteria for growth depression.     Optimizing artificial insemination on swamp buffalo (Bubalus bubalis) through synchronization of estrus and ovulation

    Artificial insemination (AI) program in swamp buffalo will be more efficient by implementing synchronization of estrus and ovulation. By synchronizing of ovulation, AI can be done at a fixed time schedule without concerning to estrus detection. Gonadotropin Releasing Hormone (GnRH) and human chorionic gonadotropin (hCG) have been used in protocols of estrus synchronization to induce ovulation. A study of AI in swamp buffalo was conducted on 83 buffaloes to evaluate the impact of protocol of estrus synchronization on reproductive efficiency of swamp buffalo. The three protocols used were Ovsynch (GnRH-PGF2α-GnRH-AI), convensional (PGF2a-PGF2a-AI) and Select-Synch (GnRH-PGF2a-AI). Inducing of ovulation were done by administration of GnRH or hCG after prostaglandin (PGF2α) injection. AI was done at 18 and 24 hour after the second GnRH injection (66 hours and 72 hours after PGF2α injection) for Ovsynch method and 72 hours after the last PGF2α injection for convensional and Select-Synch methods. Parameters observed were percentage of estrus and pregnancy from the three estrus synchronization protocols and the differences were analysed by statistics. All of buffaloes (100%) in the three synchronization protocols showed estrus behavior prior to AI. The percentage of pregnancy was 64.71; 77.14 and 83.87% for the Ovsynch, convensional and Select-synch respectively and there was no significantly different (P 0.05) among the three protocols. hCG administration after the last PGF2α also did not affect pregnancy rate, ie: 76.47 vs 77.78% (with hCG vs without hCG) for the convensional and 88.24 vs 78.57% for the Select-Synch. It is concluded that the synchronization of estrus protocols in this study can synchronize the estrous and ovulation and AI can be done in a fixed-timed and could reach better pregnancy rate of swamp buffalo.     Effect of fermented Banana peel on Broiler Carcass

    This experiment was conducted to examine effect of inclusion of fermented banana peel by Rhyzopus oligosporus in diets on slaughter weight, carcass weight  and carcass percentage, weight and percentage abdominal fat of broiler. The experiment was done based on Completely Randomized Design with four treatments and four replications and each replication consisted of six chickens. The treatment were R0 = without banana peel fermented, R1 = 5% banana peel fermented, R2 = 10% banana peel fermented, R3 = 15% banana peel fermented. Data of the experiment were analyzed, using ANOVA and then continued with Duncan's Multiple Range Test. Result showed that level of fermented banana peel affected slaughter weight and carcass weight. However carcass persentage, weight and percentage of abdominal fat was not affected by treatment. Banana peel fermented by Rhizopus oligosporus could can be used maximally 10% in broiler ration.     Utilization of fermented rice straw as substitution of elephant grass in cow feed

    The objective of this research was to evaluate the use of fermented rice straw by probion on feed consumption, digestibility, daily gain and feed efficiency of Simmental cow. This study was carried out based on completely randomized design, with here dietary treatments and four replications for each treatment. The treatments were R1 (JP-15) = 40% elephant grass + 15% untreated rice straw + 45% concentrate; R2 (JPF-15) = 40% elephant grass + 15% fermented rice straw + 45% concentrate; and R3 (JPF-35) = 20% elephant grass + 35% fermented rice straw + 45% concentrate. Concentrate was given at around 08:00 while unfermented/fermented rice straw was given afterward at around 09:00. Chopped elephant grass was given twice a day at 11:00 and 16:00. Water was available through out the day. Observation was done for two months on feed consumption, digestibility, daily gain and feed efficiency. The results did not show significant differences on feed consumption, digestibility, daily gain and feed efficiency, except on digestibilities of cellulose and hemicelulose. The digestibilities of cellulose and hemicelulose of treatment R3 was higher than that of R1 and R2. It is concluded that fermented rice straw is suggested to be used as an alternative feed to substitute elephant grass in maintaining feed consumption, digestibility, daily gain and feed efficiency of Simmental cow.     Artificial neural networks simulation to define critical temperature of Fries Holland based on physiological responses

    Artificial Neural Networks (ANN) simulation for industrial engineering is used to define critical temperature of Fries Holland (FH) heifer based on physiological responses on models to predict heart rate and respiratory rate, using ambient temperature and humidity inputs. The research was conducted using six dairy cattles in Bogor and in Jakarta. The heifers were fed at 6 am and 3 pm daily. The environmental condition (Ta, Rh, THI, and Va) and physiological responses (heart rate and respiration rate) were then measured for 14 days in two months at 1 h intervals started from 5 am to 8 pm. By using this ANN simulation, the critical temperature for FH heifer were defined, from heart rate at Ta 24,5°C and Rh 78% at Bogor, and at Ta 23,5°C and Rh 88% at Jakarta, from respiratory rate at Ta 22,5°C and Rh 78% at Bogor, and at Ta 23,5°C and Rh 78% at Jakarta. The respiratory rate on FH heifer was more sensitive to stress due to Ta and Rh fluctuation than the heart rate.     Effect of Priangan ram seminal plasma on viability of Peranakan Etawah buck spermatozoa preserved at 3–5oC

    In processing of buck semen, seminal plasma is a problem because it contains a phospholipase A enzime produced by the Cowper gland. If this enzime interacts with egg yolk, it causes semen coagulation, and consequently death of spermatozoa. The purpose of this research was to examine the effect of Priangan ram seminal plasma on viability of Peranakan Etawah (PE) buck spermatozoa preserved at 3–5oC. Semen was collected using artificial vagina once a week. Fresh semen was divided into three tubes then centrifuged at 3,000 RPM for 30 min. Supernatant of the first tube was mixed again with Pasteur pipette (treatment A or control). Supernatant of the second tube was removed (treatment B or without seminal plasma). Supernatant of the third tube was removed and changed with Priangan ram seminal plasma in the same volume (treatment C). Semen was diluted with Tris extender containing 20% egg yolk and stored in refrigerator at 3–5oC. Quality of diluted-semen including percentages of motile spermatozoa (MS), live spermatozoa (LS), and intact plasma membrane (IPM) was evaluated every day during storage at 3–5oC for three days. Results of this study showed that mean volume, colour, consistency, pH, mass activity, spermatozoa concentration, MS, LS, spermatozoa abnormal, and IPM of PE buck fresh semen, respectively was 0.68 ml, cream, thick, 7, ++/+++, 4,148.57 million cell/ml, 70%, 83.89%, 7.12% and 84%. At day-4 of storage, percentages of MS, LS, and IPM for treatment C (40, 52.2 and 51.6%) was significantly (P0.05) higher than that of: treatment B (31, 44.8 and 45.2%) and treatment A (11, 15.6 and 14.8%). In conclusion, seminal plasma of Priangan ram could maintain the quality of PE buck semen preserved at 3–5oC for three days, and it prevent semen from coagulation.     Strategic control of acute diarrhea of newborn calves

    Economic performance of beef cattle operations can be severely hampered by acute calfhood diarrhea. Accordingly, a study was conducted at Bbalitvet to identify the causal agents, reduce clinical incidence, and increase body weight gain of newborn calves. One potential control is application of suitable vaccines to pregnant cows. The study was begun by identifying cases of diarrhea followed by isolation and identification of the causal agents in 12 beef cattle farms in West Java. A field trial was then designed for controlling calf diarrhea in such farms. Inactive vaccines Ecoli-Closvak polivalen were administered to pregnant cows to increase specific resistance of the newborn calves. At 2 months prepartum, 12 pregnant cows were assigned either to a vaccination or a placebo group, with a booster vaccination 3 weeks prior to parturition. Strict hygenic management was provided to both groups, and all calves were provided adequately with colostrum.  Subjects were observed for 5 months, starting from the time of initial vaccination until the calves were 3 months of age. In the initial farm surveys, entero-pathogenic bacteria such as Escherichia coli serotype K99 and Clostridium perfringens type A and C were isolated and identified in fecal samples from 4 beef cattle farms in 3 districts (Garut, Tasikmalaya, Ciamis) and 2 beef cattle farms in 2 districts (Tasikmalaya and Ciamis) of West Java. In the vaccination trial, good immune responses to E. coli and C. perfringens alpha toxin measured by ELISA were observed. Application of effective control of calf diarrhea including vaccination and good livestock management showed good results. No death or signs of diarrhea were found in the new born calves up to 3 months of age. The rate of body weight gain was significantly higher in calves of vaccinated dams than in calves of non-vaccinated dams. Key Words: Strategic control, Calf diarrhe

    Exploration and conservation of bacterial genetic resources as bacteriocin producing inhibitory microorganisms to pathogen bacteria in livestock

    Exploration and conservation of microorganisms producing bacteriocin was done as the primary study towards the collection of potential bacteria and its application in improving livestock health condition and inhibit food borne pathogens. Diferent kinds of samples such as beef cattle rectal swab, rumen fluids, cow’s milk, chicken gut content, goat’s milk were collected at Bogor cattle slaughter houses, poultry slaughter houses, dairy cattle and goat farms. A total of 452 bacterial isolates consisted of 73 Gram negative bacteria and 379 Gram positive bacteria were isolated from samples collected and screened for bacteriocin activity. Determination of bacteriocin activity with bioassay using agar spot tests were carried out on liquid and semisolid medium assessing 8 kins of indicators of pathogenic bacteria and food borne pathogens. A total of 51 bacteriocin producing strains were collected and some of the strains had high inhibitory zone such as Lactobacillus casei SS14C (26 mm), Enterobacter cloacae SRUT (24mm), Enterococcus faecalis SK39 (21mm) and Bifidobacterium dentium SS14T (20mm) respectively, to Salmonella typhimurium BCC B0046/ATCC 13311, E. coli O157 hemolytic BCC B2717, Listeria monocytogenes BCC B2767/ATCC 7764 and Escherichia coli VTEC O157 BCC B2687. Evaluation after conservation ex situ to all bacterocin producing strain at 5oC for 1 year in freeze drying ampoules in vacuum and dry condition revealed the decreasing viability starting from log 0.8 CFU/ml for Lactococcus and Leuconostoc to log 2.2. CFU/ml for Streptococcus. Result of the study showed that the bacteriocin producing strains obtained were offered a potential resource for preventing disease of livestock and food borne diseases.
