Utilization of palm oil sludge in poultry diet: 2. Dried palm oil sludge and its fermented product for growing drakes
Inclusion limit of palm oil sludge (POS) in poultry diet varies according to processing, and species or strain of the animal. Therefore, an experiment was conducted to study the utilization of fermented (FPOS) and non-fermented palm oil sludge (POS) for growing drakes. A number of 224 one week-old male ducklings were randomly distributed into 7 dietary treatments, with 4 replicates and 8 ducklings for each replicate. The dietary treatment consists of 2 kinds of feedstuffs (POS and FPOS) with 3 levels (5, 10, and 15%) and one control diet consisting neither POS nor FPOS. All diets were formulated with similar nutrient contents and meet the requirement of growing duckling and fed to 8 weeks old. The results showed that at the first week of the trial, feeding of POS or FPOS significantly (P0.05) depressed growth of the ducklings, although the feed consumption was higher than the control. However, overall performances (body weight, feed consumption and feed conversion) of the ducklings, carcass yield, liver weight, gizzard weight and abdominal fat weight measured at the end of the trial were not significantly (P0.05) affected by the dietary treatments. Comparison between antibody-elisa test and fecal egg count for detecting Fasciola gigantica infection in cattle
The comparison between antibody-ELISA test and fecal egg count for detection of natural infection of Fasciola gigantica in cattle was observed. One hundred and fifty samples of blood, feces and livers were collected from cattle slaughtered in the abattoir in Jakarta. Serum was collected from the blood samples and the level of antibody was determined by using antibody- ELISA test. The fecal samples were processed by using sedimentation technique in order to detect the present of F. Gigantica eggs. The livers were processed for liver flukes count. The result showed that from the liver examination, 38.7% cattle were negative flukes, 16% had 1-10 flukes, 34% cattle had 11-100 flukes and 11.3% cattle had more than 100 flukes. About 44.7% infected cattle had less than 100 eggs of F. gigantica per gram feces, however no eggs of F. gigantica were found in 13% infected cattle. The result of antibody-ELISA test showed that from 92 cattle infected with F. gigantica, 84 cattle had OD 0.38 (range from 0.38-1.77) and 8 cattle had OD 0.38 (range from 0.18-0.37). In contrast, from 58 cattle without flukes, 7 cattle had OD 0.38 (range from 0.38-1.95) and 51 cattle with the OD 0.38 (range from 0.1-0.33). The sensitivity of the fecal examination technique was 87% and the specificity was 100%. The sensitivity and specificity for antibody-ELISA test were 91 and 88% respectively. The use of Wonnolas in controlling gastrointestinal nematode infections in sheep under traditional grazing management in Indonesia
Blocks containing 3% phenothiazine in solidified molasses (Wonnolas, Animeal Australia Ltd .) were tested to control gastrointestinal nematode infections and the effect on mineral status in sheep in villages in Cirebon, Indonesia. Two hundred and thirteen Javanese Thin Tail sheep which were grazed during the day and housed at night were divided into two groups on the basis of location . One group was allowed in an access to Wonnolas blocks when penned for 24 weeks while the other group remained untreated. The bodyweight was recorded and faecal samples were collected for egg counts and larval culture every four weeks. Sera and saliva samples were collected from 20 adult sheep in each group at weeks 0, 12 and 24 for mineral analysis by AAS and colorimetry. The results indicated that the mean egg counts of the treated group decreased from 576 epg to 123 epg and the percentage of sheep producing viable larvae decreased from 50% to 24%. In contrast egg counts of the control group increased from 768 epg to 4,840 epg and the percentage of sheep producing viable larvae increased from 65% to 84% over the same period . In the treated group the number of Haenronchus larvae declined significantly (36% to 6 %) and at the end of the trial Tiichostrongylus larvae predominanted in larval cultures (80%) . Mineral analysis revealed deficiencies in sodium and copper, low levels of zinc and normal levels of potassium, calcium, magnesium and phosphorus . Wormolas had a significant effect on sodium and zinc status but not on copper although sufficient levels of this element were available. The improvement of rice straw quality by urea-molasses treatment
A study was conducted in Balai Penelitian Temak Bogor to evaluate the effect of urea-molasses treatment on the quality of rice straw . Five levels of urea were 0%, 0.5%, 1%, 1 .5% and 2% and 4 levels of molasses were 0%, I%, 2% and 3%. The study was conducted based on factorial completely randomized design with 3 replications . Urea and molasses were mixed with 5 kg chopped rice straw and stored for 21 days in plastic bag . Parameters observed were dry matter, organic matter, crude protein, crude fibre, and silica contents . Nutritive value of rice straw was measured by proximate analysis and its nutrient digestibility by in-vitro using rumen liquid of fistulated Ongole-crossed cattle . The results showed that urea and molasses treatment on rice straw decreased the dry matter and silica contents but increased the crude protein content of rice straw (P0 .01). Dry matter and organic matter digestibility of rice straw didn't show the significant different among urea treatment but highly significant increased (P0.01) by the molasses treatment. It was concluded that urea and molasses treatment on rice straw could improve the quality so that by the levels of 1 .5% urea and 3% molasses produced the similar quality with napier grass . Effects of hormonehCG following injection of estnanate of reproductive performances ofswamp buffalo (Bubalus bubalis)
Low reproductive performance of buffalo resulted the population growth in Indonesia was only slightly increase during the last 50 year. Reproductive performance can be improved by hormone treatment during synchronization . One study was conducted in Porsea and Siborong-borong to see the effects of hormone hCG on the reproductive performances of swamp buffalo . A total of 34 buffaloes were kept in field grazing and commercial concentrate 4 kg/head/day was given for supplementation in 27 buffaloes and the rest 7 buffaloes treated as control with no supplemeniation . Synchronization performed by two intromuscular (IM) injections of2 ml estrumate in 11 days interval . The buffaloes were divided into 2 groups of 16 buffaloes without hCG (Control group) and 18 buffaloes with hCG (Treated group). Treated group wereinjected with 500 IU hCG intro-muscularly (IM) 24-48 hours following second injection of estrumate . All buffaloes were artificially inseminated (AI) 48 and 72 hours after the second injection of estrumate. Results showed that injection of estrurnate twice in 11 days interval was very effective to synchronize oestrus of buffaloes. The persentage of buffalo showing oestrus was 67 .5 and 85 .3% following 48 and 72 Ins of thesecond injection of estrumate resfectively . The effectiveness of estrumate was significantly affected by body condition of buffaloes where the persentage of oestrus 48 and 72 hours following estrumate injection was significantly higher (P0.01) in buffaloes with good body condition than those in poor body condition (74.5 and 92.5% vs 42 .9 and 57 .1%). Injection ofhCG following estrumate increased the percentage of oestrus and pregnancy of buffalo. Percentages of oestrus of buffaloes 48 and 72 hours following estrumate injections was significantly higher (P0 .01) in hCG treatment (83 .3 and 100%) than those in control groups (68 .8 and 81 .3%) . The pregnancy rate of buffalo with good body condition was significantly higher (P0.01) in group of buffaloes with hCG than those control groups ( 86 .6 vs 50.0%) . Optimization of dairy based farming agricultural in critical land area of Yogyakarta Special Territory
A study has been done to provide a conceptual basis for the development of agricultural system in critical land area in Yogyakarta Special Teritory as the smallest production unit to (1) determine optimum dairy farming based agriculture for the area and income maximization on either farmer own inputs or with CAFlTAL aid provided by a funding party, (2) to develop a model for developing farming system based on dairy farming in critical land area in Yogyakarta Special Teritory.Methods being used multistage problem solving approach to the problem encountered in farming system as practiced in Glagaharjo Village, Sub District of Cangkringan, Sleman with Rapid Rural Appraisal (RRA) and Agro-ecosystem Analysis as diagnostic activities. Farm record keeping and survey to fourty respondents purposively selected were then conducted. Using Linear Programming (LPM), a normative solution, under the imposed constraints for each activity, was obtained to see development effectivity of dairy based farming agricultural system. The result indicated that land resources had not managed optimally. Out of an average possession of 1,5 ha dry land, only 0,6 ha was cultivated, leading to relativelly low yield and income. Optimum dairy farming agriculture migh be an option to develop the area. Dairy farm Cooperative Agency (Model of Scenario I) can play important role to improve farmer income. With an average of 1,5 ha dry land and an average of 3 Animal Unit raised would optimized the resources allocation and income of Rp.20,385,340 for a period of eight years. The income rnigh be further increased to Rp.36,176,070 for similar production period with soft loan provided by ventura fund (Model of Scenario ll) to optimize land resources and family labor force use and a farmer was able to raised an average of 4 Animal Unit. If as being planned - such as scheme is adopted by 1,400 farmer, 230 worker will be employed in the first years. Employement need is increasing with time and after eight year it may reach 1154 workers/year. It can then be concluded that dairy based farming agriculture may improved economic endurance of farmer in critical land area, and directed as well toward land conservation program. Solid state fermentation of palm kernel meal by using Aspergillus niger
The solid state fermentation technique on palm kernel meal by using Aspergillus niger wild type and NRRL 337 was studied. The fermentation was carried out at 30oC for 3 days continued with enzymatic process at room temperature and 40oC for 2 days. The result showed that at the third days of fermentation spores started to grow on the surface. The 3rd days fermentation can improve protein content and in vitro digestibility (IVDMD) with minimal loss of dry matter. The dry matter contents at 3 days fermentation were 48.88 and 48.83% for product using Aspergillus niger wild type and NRRL 337. Combination 3 days fermentation and 2 days enzymatic process at room temperature by using Aspergillus niger NRRL 337 type gave the best product with optimal IVDMD value and protein digestibility of 51.47 and 71.33%. Dairy farming production factors in Garut and Bogor
Dairy farming in West Java is still dominated by small scale. The research focussed on production factors of dairy fanning which are influencing the farmers income from selling milk . The research was carried out in 1993 via survey to 30 respondents in Garut and Bogor. The results showed that the profit earned per month from dairy farming was Rp 130,331 and Rp 118,449 in Garut and Bogor, respectively . Return to labor from dairy farming was Rp. 4.56 in Bogor and Rp. 4.38 in Garut. The production factors positively affecting the income of the farmers from selling milk were cost for barn, concentrate feed, animal health care and artificial insemination, labor, and number of lactating cows . In addition, several production factors such as cost for forages, retribution cost, and number of male calf were proven negatively affecting the income of the Canners from selling milk . Therefore, thrive related production factors should be considered in developing small scale dairy farming, not only from availability of inputs but also from institutional aspect
Development of alpha hemolytic verotoxigenic Escherichia coli vaccine: Verotoxic antibody responses in experimental animals, mice, rabbits and dairy cows
Escherichia coli isolates showing alpha hemolytic on blood agar were found in calves with bloody diarrhoea from Bandung, Sukabumi and Bogor. Three E. coli isolates designated as (B34c, B909, B910) were pathogenic for mice and selected for vaccine candidates. Crude supernatant of tryptic soy broth (TSB) cultures precipitated with saturated ammoniumsulphate caused cytopathic effect on vero cell line. For vaccine preparation the isolates were grown on tryptic soy agar (TSA). Whole cell inactive vaccine containing equal proportion of each isolate was prepared and emulsified in aluminum hydroxide gel at a final concentration of 1.5% and cell concentration equal to the tube number 10 of MacFarland standard. Mice and rabbits were injected subcutaneously of monovalent vaccine with the dose of 0.2 ml and 1 ml respectively. Four weeks later mice or rabbits were boostered with the same dose. Before injection each animal was bleed, subsequently every two weeks period up to 4 weeks after the second injection. The sera were separated and kept at -20oC up to serological assay with an enzyme linked immunoassay (ELISA) was done. Vaccinated mice with VTEC were challenged with homologous isolate did not die, where as 60% of unvaccinated mice died within 2 days post challenged. Pregnant cows were given 2 doses of three valent vaccine 6 weeks and again 2 weeks before expected date of calving. Calves born were given colostrum and milk of the own mother. The three day old calf was challenged with three isolates of verotoxigenic E. coli studied. Calves born from vaccinated cows did not show diarrhoea at post challenged, whereas calf from unvaccinated cow demonstrated bloody diarrhoea at post challenged. From the experimental animals (mice and calves) demonstrated the presence of protection against challenged. The antiverotoxic antibody probably involved in important role of immunoprotection. These were supported by the presence of high titer anti verotoxic antibody in serum of experimental animals (mice and cows), as well as in experimental rabbits, but none in the unvaccinated animals. An estimate of seasonality and intensity of infection with gastrointestinal nematodes in sheep and goats . in West Java
Tracer Inoodle tbin-tail sheep and Kacang goats were used to measure the seasonal changes in gastrointestinal nematodes parasitism i ~'und[, .xillapek .Fodd~ 'ofis in West Java . Each 3 months for 12 months worm-free male sheep (5) and goats (5) about 5 months of age were ~ $Jri~tgldI o% ef- farmer, and managed as part of their flock for 2 months . Animals were then returned to the laboratory and maintained on "`~^taaan-ftwAiet in elevated slatted pens for 3 weeks prior to slaughter. In all trials sheep had higher faecal egg counts than goats . Egg counts were significantly lower during the late dry-early wet season due mainly to lower burdens of Oesophagostomum spp. than at other times of the year. The predominant genera recovered from faecal larval cultures were Haemonchus and Trichostrongylus . At post mortem more than 94 percent of animals were infected with Trichostrongylus colubriformis, T. axei, Haemonchus contortus, Oesophagostomum columbianwn and Strongyloides papillosus . Other species found, in descending order of occurrence, were Cooperia curticei, Trichuris ovis, Bunostornum trigonocephalum, Oesophagostomumn asperum, Capillaria bovis and Gaigena pachycelis. It was concluded that intensity of exposure of both sheep and goats to H contortus, T. axei and C. curticei was similar throughout the year, but that availability of infectioe larvae of T. colubriformis was higher during the dry than the wet season and vise versa for O. columbianum .
