    Clinical responses of vaccinated Bali cattle against challenge with a field isolate of Bovine Herpesvirus- 1

    The aim of this study is to investigate the post challenge clinical responses of Bali cattle vaccinated with an inactivated vaccine of a field isolate Bovine Herpesvirus-1 after being challenged with the same virus. Twenty eight Bali cattle were divided into two groups of ten animals, one of which was vaccinated with a single dose and the other one was vaccinated with a booster dose of vaccination, and the rest of the animals were remained untreated as a control group. One month after the last vaccination, a half number of animals in each group were challenged intravenously with 5 x 108 TCID 50 pathogenic strain of BHV-1 local isolate. Clinical signs were observed from day 0 to day 14 after challenge. These observed included body temperature, mucosal membrane changes of nostril, vagina and palpebrae, hyperlacrimation and hypersecretion of nose and eyes. General clinical changes and mucous membrane lesions of nostril were scored from 0 to 3. The results showed that the non-vaccinated animals: elevated temperature (up to 40.1°C), hyperlacrimation, nasal discharge and mucosal hyperaemia of nostril and vagina after challenge. The duration of illness ranged from 3 to 7 days. Symptoms appeared after an incubation periode of 3 days and disappeared after 5 days (day 9). These clinical symptoms didn’t appear on the vaccinated animals with single or boostered vaccination. One month after vaccination, the antibody titre reached 211 (2048) based on serum neutralisation test (SNT).     Improving the nutritive value of palm oil sludge by fermentation : The effect of fungi strain, environmental temperature and enzymatic process

    An experiment was conducted to improve the nutritive value of palm oil sludge (POS) by fermentation process. Aspergillus niger BPT and NRRL 337 were used into fermenting POS for three days and followed by anaerobic enzymatic process. The experiment was arranged in 2x3 factorial design i.e., enzymatic incubation time (2, 3, and 4 days) and environmental temperature during enzymatic process (room temperature and 40°C). Changes on chemical compositions (crude protein, true protein, ADF and NDF), in vitro dry matter digestibility (IVDMD) and protein digestibility (IVPD) were measured. The results showed that both A. niger BPT and NRRL 337 grew well on POS media with the best result at 3 days. The fermentation process by both strains increased crude protein and true protein of POS. The A. niger NRRL reduced the fiber content (ADF and NDF) more than A. niger BPT the IVDMD and IVPD were not significantly affected by fermentation process without enzymatic process. The IVDMD increased significantly when the fermentation followed by enzymatic process. Overall results showed that the best fermentation of POS was obtained by using A. niger NRRL followed by enzymatic process in room temperature for two days.     kernel meal and its fermented product for growing ducks

    A series of experiment was carried out to evaluate the use of palm kernel meal and its fermented product in the ration of growing ducks. Treatments were arranged in a 2x3 factorial experiment, i.e. the combination between two processes (unfermented and fermented palm kernel meal) and three levels of ingredients in the diet (5, 10 and 15%). Four replicates were applied for each treatment, with 8 ducklings in each replicate. A control diet with no palm kernel meal was also formulated. All diets were formulated to have a similar protein and ME contents. Results of the experiment showed that fermentation process increased the nutrient composition (crude protein, true protein, phosphorus, and the metabolizable energy) and reduced its fibre and fat content. The feeding trial showed that neither the fermentation process, levels of ingredients nor the interactions of the two factors affected final body weight, feed consumption, feed conversion, carcass percentage and abdominal fat of the ducklings significantly. It is concluded that fermentation process improved the nutritional value of palm kernel meal and the inclusion of palm kernel meal (unfermented and fermented), up to 15% in the diet of ducks did not negatively affect the growthperformance and the carcass yield.   Key words : Ducks, palm kernel meal, fermentatio

    Extrahepatic bile duct ligation in broiler chickens: ultrastructural study of Ito cell

    The Ito cell (fat-storing cell) is a cell lying in perisinusoidal space of liver. The function of Ito cell is expanding from a site of fat-storing site to a center of extracellular matrix metabolism and mediator production in the liver. This study was performed in order to evaluate the Ito cells in cholestatic condition. The artificial cholestatic was conducted by ligation of extrahepatic bile ducts (bile duct ligation = BDL) in broilers. The results showed that BDL induced bile congestion, fibrosis, proliferation of Ito cells and intrahepatic bile ductules. Immunohistochemistry demonstrated that Ito cells were scattered throughout the fibrotic areas, and larger in size with more extensive immunoreactivity than those in normal livers. Ultrastructural study demonstrated that Ito cells were closely associated with the production of extracellular collagen fibers. Ito cells actively react against hepatocytic injuries, especially in fibrogenesis of cholestatic livers.   Key words: Bile duct, ito cell, broile

    The use of probiotic in the diet to improve carcass characteristics and meat quality of sheep

    An experiment has been carried out to investigate the effect of probiotic and ruminally less-degradable protein supplementation on the growth and efficiency of feed utilization in sheep. Thirty two male sheep with an average liveweight of 23 kg were divided into 4 groups of 8 individuals to test four dietary treatments. The dietary treatments were a) basal concentrate diet, b) basal concentrate diet supplemented with ruminally less degradable protein in proportion of 90:10%, c) basal concentrate supplemented with probiotic at 0.5%, and d) basal concentrate diet supplemented with ruminally less degradable protein and probiotic in proportion of 89.5:10:0.5% consequtively. The concentrate was fed at 1.5% of the liveweight, and adjusted every week after weighing the individuals. Napier grass was fed ad libitum after being chopped into 5 cm length. Drinking water was available adequately. The animals were individually caged. The experiment was carried out for 7-day adaptation to the dietary treatments and 42-day period of feeding trials. The parameters being observed include weekly liveweight changes, slaughter weight, carcass weight and carcass characteristics. The results indicated that the liveweight changes were relatively the same for all treatment groups, which ranged from 89 to 94 g/d. However, the dry matter intakes ranged from 856 to 925 g/d. The slaughter weight ranged from 26.9 to 27.3 kg with carcass weight ranged from 12.0 to 12.8 kg indicating that the carcass percentage were in the range from 55.8 to 57.1% of the empty body weight. Probiotic supplement resulted in reduction of fat content in the meat from these observation, it is reasonable to assume that the use of probiotic in the diet reduced the carcass fat without affecting the daily weight gain.   Key words: Sheep, probiotics, ruminally less-degradable protein, productivity

    The detection antibody of porcine reproductive and respiratory syndrome (PRRS) in pig sera in Eastern part of Indonesia

    Since the early 1980's, a new and highly contagious disease syndrome of pigs, porcine reproductive and respiratory syndrome (PRRS), has been spreading through major pig producing areas in the world . It is suspected that the disease has occurred in Indonesia.To confirm the presence of PRRS infection in Indonesia, a serological survey in pig was conducted between 1993 and 1995 in some areas in Eastern Indonesia to detect antibodies against PRRS virus using ELISA test . The results indicated that a total of 822 pig sera were tested and 8% of these sera had antibodies against PRRS virus . Pig sera which were collected from slaughter house in Jakarta showing the highest prevalence (30%) of reactor. However, there was no antibody was detected from pig sera originated from Irian Jaya since 1993 .   Key words: PRRS, prevalence, ELISA test, antibod

    Characterisation of protein antigen from Fasciola gigantica of different age

    The protein antigens extracted from adult fluke Fasciola gigantica, 3, 6 and 9 weeks old and newly excysted juvenile (NEJ) were identified using SDS-PAGE and immunoblotting techniques. Sera from fat-tailed sheep which artificially infected with the metacercariae of F. gigantica were used for immunoblotting. The results showed that the protein antigen profile of adult fluke, 6 and 9 weeks old flukes had similar. Bands with molecular weight between 24 kDa to 114 kDa. Protein bands with molecular weight 24 kDa and 198 kDa were also detected from the adult fluke. The use of immunoblotting technique, there were two antigenic protein molecules identified from adult fluke, NEJ and 3, 6, and 9 weeks old fluke with the molecular weight 46 kDa and 47 kDa. The protein band with molecular weight 198 kDa shown thicker on the NEJ than that of adult fluke, and 6 and 9 weeks old flukes. The role of protein with molecular weight of 46 and 47 kDa were the interested findings need to be evaluatedfor serological analysis.   Key words : Protein antigen, Fasciola gigantica, SDS-PAGE, immunoblottin

    Genetic polymorphism of the β-lactoglobuline gene in Friesian Holstein cows

    Milk protein in dairy cattle consists of casein and whey. Whey in milk of dairy cattle is about 20% with the main component is β-lactoglobulin (7-12%). Polymorphism of the β-lactoglobulin gene affects protein and whey production in milk. Selection at a molecular base to improve whey and protein in milk requires information on genetic diversity of the β-lactoglobulin gene, besides other protein genes. This study was aimed to identify genetic polymorphism of the β-lactoglobulin gene in 88 heads of Friesian Holstein (FH) cows at Cikole Dairy Cattle Breeding and Improvement Station (Cikole DCBIS), Lembang. Genotyping was done using Polymerase Chain Reaction - Restriction Fragment Length Polymorphism (PCR-RFLP) method, and a HaeIII restriction enzyme. Genotype frequency, allele frequency, and heterozygosity value were calculated by Nei method. Hardy-Weinberg equilibrium was calculated by chi-square test. Result showed that the β-lactoglobulin gene in the observed HF cattle was polymorphic, in which there were two alleles (A and B) and three genotypes (AA, AB and BB). Frequencies of the A and B alleles were 0.40 and 0.60 respectively; while those of the AA, AB, and BB genotypes were succesively 0.10; 0.60 and 0.30. Heterozygosity value was obtained for 0.483. The β-lactoglobulin gene was in Hardy - Weinberg equilibrium (χ2cal χ2table). It is concluded that polymorphic of the β-lactoglobulin gene can be used as an initial information for molecular selection on milk protein composition in FH cows.     Development of spermatogenic cells and sperm quality after administration of pegagan extract (Centella asiatica)

    The purpose of this study was to determine the development of spermatogenic cells and sperm quality after administration extracts of pegagan (Centella asiatica) in various doses and duration of administration. The research was carried out with complete randomized design (CRD), consist of 16 combinations of dose and duration of treatment. Parameters measured consist of population of spermatogenic cells (spermatogonia, primary spermatocytes, late spermatids) and sperm quality (concentration, motility, abnormality). The data were processed using analysis of variance (ANOVA), and differences between treatments followed by Duncan test. The results show that both the dose and duration have very significantly (p 0.01) affect on decreasing of late spermatid population, sperm motility and concentration, but not for population of spermatogonia, primary spermatocytes and sperm abnormalities. The decrease of population and quality may due to antifertility effect of pegagan, eventhough still in the normal range. It is concluded that spermatogenic cells development and sperm quality reduce after administration of pegagan extract, although infertility is not yet found up to the dose of 450 mg/kg BW for 49 days duration of administration.
