    The Genetic Diversity of TLR4 MHC-DRB Genes in Dairy Goats Using PCR-RFLP Technique

    This research was aimed at evaluating the genetic polymorphism of TLR4 and MHC-DRB genes in dairy goats [(Saanen, Etawah Grade-Saanen Crossbred (PESA), and Etawah Grade (PE)] using PCR -RFLP. The two genes are involved in immunity where they play a crucial role in pathogens recognition and presentation to T-cells and CD4 cells. PCR was used to amplify genomic DNA for TLR4 (382 bp) and CaLA-DRB (285 bp) genes fragments. Genetic polymorphism was detected by digesting TLR4 amplimer with AluI while DRB amplimers were digested with PstI and TaqI in two separate reactions. The results showed that TLR4|AluI was monomorphic and fixed with allele T in all three breeds while DRB|TaqI and DRB|PstI loci were found polymorphic for all breeds. Heterozygosity expected (He) and PIC were found low at both DRB|TaqI and DRB|PstI loci in PE and Saanen. Χ2 results showed that DRB|PstI in PE and DRB|TaqI in PESA were not in H-W equilibrium and did not display homozygous recessive genotype. The results declared that TLR4|AluI was not a good for marker for diseases resistance whereas DRB|TaqI and DRB|PstI gave hope for resistance based on their PIC

    Morphometric and Phylogenic Analysis of Six Population Indonesian Local Goats

    The research objectives were to characterize morphometric and genetic distance between populations of Indonesian local goats. The morphological discriminant and canonical analysis were carried out to estimate the phylogenic relationship and determine the discriminant variable between Benggala goats (n= 96), Marica (n= 60), Jawarandu (n= 94), (Kacang (n= 217), Muara (n= 30) and Samosir (n= 42). Discriminant analysis used to clasify body weight and body measurements. In the analysis of variance showed that body weight and body measurement (body length, height at withers, thorax width, thorax height, hert girth, skull width and height, tail length and width, ear length and width) of Muara goats was higher (P<0.05) compared to the other groups, and the lowest was in Marica goats. The smallest genetic distance was between Marica and Samosir (11.207) and the highest were between Muara and Benggala (255.110). The highest similarity between individual within population was found in Kacang (99.28%) and the lowest in Samosir (82.50%). The canonical analysis showed high correlation on canon circumference, body weight, skull width, skull height, and tail width variables so these six variables can be used as distinguishing variables among population. The result from Mahalonobis distance for phenogram tree and canonical analysis showed that six populations of Indonesian local goats were divided into six breed of goats: the first was Muara, the second was Jawarandu, the third was Kacang, the fourth was Benggala, the fifth was Samosir and the sixth was Marica goats. The diversity of body size and body weight of goats was observed quite large, so the chances of increasing productivity could be made through selection and mating programs

    Changes of Physico–Chemical Properties of Pig Slurry During Storage

    This study was aimed to determine changes of the characteristics of raw pig slurry as liquid organic fertilizer at various storage times. A completely randomized design was used in this research. The treatments were storage times, i.e.: 0, 15, 30, 45, and 60 days. Variables observed were loss of the slurry, degree of acidity (pH), electrical conductivity (EC), total solid (TS), volatile solid (VS), total chemical oxygen demand (tCOD), soluble chemical oxygen demand (sCOD), total nitrogen (TN), ammonia-nitrogen (NH3-N), nitrate-nitrogen (NO3-N), total phosphate (TP), and dissolve reactive phosphate (DRP). The results showed that storage time significantly affected all the observed variables, except the concentration of NO3-N and total phosphate content. The pH, TS, VS, DRP, and losses of slurry lost during storage times increased, while EC, TN, NH3-N, tCOD, and sCOD decreased.     Live Weight Estimation by Chest Girth, Body Length and Body Volume Formula in Minahasa Local Horse

    Study was conducted in the regency of Minahasa to estimate horse live weight using its chest girth, body length and body volume formula (cylinder volume formula) represented by animal chest girth and body length dimensions, particularly focused in Minahasa local horses. Data on animal live weight (LW), body length (BL), chest girth (CG) and body volume were collected from 221 stallions kept by traditional household farmers. Animal body volume was calculated using cylinder volume formula with CG and BL as the components of its formula. Regression analysis was carried out for LW with all the linear body measurements. The data were classified on the basis of age. Age significantly (P < 0.05) influenced the body measurements except animal body length (P>0.05). Animal live weight was predicted by simple regression models using dependent variable (Y) of the animal live weight and independent variable (X) of the animal body measurement, either body length, chest girth, or body volume. The correlations between all pairs of measurements were highly significant (P < 0.01) for all age groups. Regression analysis showed that live weight could be predicted accurately from body volume (R2= 0.92) and chest girth (R2= 0.90). Simple regression model that can be recommended to predict horse live weight based on body volume with their age groups ranging from 3 to ≥10 years old was as follow: Live weight (kg)= 5.044 + 1.87088 body volume (liters). The analyses of data on horse chest girth, body length and body volume formula provided quantitative measure of body size and shape that were desirable, as they enable genetic parameters for these traits to be estimated and also included in breeding programs

    Physiological Adaptation and Biomass Production of Macroptilium bracteatum Inoculated with AMF in Drought Condition

    The objective of this experiment was to evaluate the effect of drought stress and mycorrhizal inoculation on physiological adaptation and biomass production of Macroptilium bracteatum. This experiment was arranged in completely randomized design with four treatments: M0 (no AM + watered), M1 (AM + watered), M2 (no AM + drought), and M3 (AM + drought) with three replicates. The observed variables were soil water content, leaf water potential, leaf relative water content, leaf proline, leaf water soluble carbohydrate (WSC), root and shoot dry weight. Data were analyzed by ANOVA and differences between treatments were tested by DMRT. Drought treatments (M2 and M3) significantly (P < 0.05) decrease soil water content, leaf water potential, leaf relative water content and increased the leaf proline content. The result in root and shoot dry weight appear that M1 treatment was significantly different (P < 0.05) with treatment M0, M2, and M3. For leaf WSC, M0 and M2 treatments were significantly different (P < 0.05) with treatment M1 and M3. It is concluded that mycorrhiza inoculation was more effective on M. bracteatum, in drought stress. One mechanism of drought resistance of M. bracteatum is the accumulation of osmotic compounds proline. Therefore, proline can be used as an indicator of drought resistance in leguminous plants

    The Application of Planned Behavior Theory to Predict the Consumption of Processed Body-Shaping Milk

    The research aims were to identify the influence of attitude, subjective norms and perceived behavioral control on intention to buy, and effect of intention to buy and perceive behavioral control over buying behavior on processed body-shapping milk which added by supplement for body shaping. Research was conducted in Sleman Regency and design used was survey. Sampling was taken by using judgmental sampling method with criteria that were female, aged 18-25 yr old and was categorized as normal to overweight appearance. Respondents used were 100 undergraduate female students. Questionnaire was used as an instrument in order for data collecting to measure research variable that had been tested on validity and reliability. Analyses used were single and multiple linier regression. The results showed that only attitude and perceived behavioral control influenced (P < = 0.05) intention to buy in normal-weight group, while in over-weight group attitude, subjective norm and perceived behavioral control affected (P < = 0.001) attitude. Then, attitude and perceived behavioral control influenced buying behavior significantly both in normal and overweight. Therefore, it can be concluded that prediction capability of planned behavior theory are better when product relevancy to respondents was closed

    Deer Frozen Semen Quality in Tris Sucrose and Tris Glucose Extender with Different Glycerol Concentrations

    In order to improve Timor deer (Cervus timorensis) frozen semen quality, the influence of sugar and glycerol concentration on semen characteristics of sperm was investigated.  The semen was collected from five sexually mature Timor deer using an electroejaculator. The semen was evaluated and divided into six equal tubes and diluted with Tris sucrose glycerol 10% (TSG10); Tris sucrose glycerol 12% (TSG12); Tris sucrose glycerol 14% (TSG14); Tris glucose glycerol 10% (TGG10); Tris glucose glycerol 12% (TGG12); and Tris glucose glycerol 14% (TGG14). The diluted semen was packed in 0.3 ml minitub straw, equilibrated at 5 oC for 4 hours and frozen on liquid nitrogen vapor for 10 minutes. The total of forward motility, viability, acrosome integrity and membrane integrity were assessed in fresh, after equilibration and after thawing. Results demonstrated that the percentage of sperm motility in TSG10was higher (P (63.93±7.23%). The sperm in TSG10 and TSG14  extender were superior in acrosome as well as in membrane integrity. It was concluded that Tris Sucrose with 10% glycerol protected Timor deer sperm better than other combinations

    The Incidence of Repeat Breeding in Dairy Cows under Tropical Condition

    The objective of this study was to investigate the incidence of repeat breeding in dairy cows under tropical condition. This was a preliminary study conducted in Sinjai Regency, Indonesia. A total of 82 Holstein Friesian lactating cows from five dairy farmer groups were used in the present study. Of the 82 cows, 75.6% eventually became pregnant after repeated inseminations (AI). The incidence of repeat breeding in this area was very high (62%). Days in milk (DIM) at first AI, first AI conception rate, and calving to conception interval were 62.5±19.3 days, 0%, and 202.8±150.0 days, respectively. There was no difference in DIM at first AI between repeat breeders and normal fertility cows (60.4±15.2 days vs 68.3±28.6 days).  However, normal fertility cows required only 123.3±52.9 days to conceive and 2.4±0.8 inseminations per pregnancy, whereas repeat breeders required significantly more days to conceive (222.9±134.1 days) and more inseminations per pregnancy (4.8±0.9). In conclusion, repeat breeder dairy cows under tropical condition had very poor and reduced reproductive performance

    Feeding Diets Containing Different Forms of Duckweed on Productive Performance and Egg Quality of Ducks

    The present experiment was undertaken to study the feeding effect of diets containing different forms of duckweed for local ducks on their productive performance and egg quality or egg yolk pigmentation. A total of 90 birds of 24 wk old ducks were randomly divided  into 18 experimental units of 2.0 x 1.0 m2 of cages. The experiment was assigned in a completely randomized design (3 treatments with 6 replicates, 5 birds each). There were 3 dietary treatments, namely P1= ducks fed a complete diet containing 20 % of dried duckweed and given in the form of dry-mash; P2= a complete diet in P1 but it was offered in wet form (slurry); and P3= ducks were offered  basal diet in the form of dry-mash and fresh duckweed was offered separately ad libitum. Diets were formulated to have similar nutritional contents. Feed consumption, feed conversion ratio (FCR), and egg yolk pigmentation were measured. The result of the study showed that these three parameters were affected by the feeding different forms of duckweed. Feeding diet with fresh duckweed brought about the best pigmentation than did the dry one. Fresh duckweed offers a promise as a potential feedstuff for ducks and has a good implication in reducing feed processing cost

    Determination of in Vitro Digestibility of Tropical Feeds Using Cattle Faeces as Rumen Fluid Alternative

    The research was conducted to compare the ability of faeces dissolved in distilled water (P1), saline solution (P2), artificial saliva (P3), and rumen fluid (RF) as sources of inoculant  in  in vitro organic matter digestibilities (IVOMD) of rice straw, corn stover, napier grass, and pangola grass. The rumen fluid was collected from two rumen fistulated Ongole Crossbred Cows of 306 and 333 kg body weight (BW). The cows were fed 3% of BW consisted of 70% napier grass and 30% concentrate. At the end of  30 days  feeding, faecal solution was made out of 350 g fresh faeces dissolved in 1 l of each solvent, homogenized using blender for 30 second, while rumen liquor were collected directly from fistula. After straining with four layers of cheesecloth both faecal solution and rumen liquid were mixed with artificial saliva (1:4 v/v). Fifty ml of each inoculants was pipetted  into each incubator tube (100 ml) containing  500 mg sample. The tubes were then  incubated at 39 oC for 48 h. Value of  IVOMD of napier grass, rice straw, corn stover, and pangola grass did not differ among the faecal solvents, but significantly lower (Pin vitro organic matter to predict digestibility of fibrous feed determination. However, the faecal solvent as inoculant produced lower in vitro digestibility than that of rumen fluid
