    Isolasi Bakteri Pendegradasi Hidrokarbon di Tanah Tercemar Lokasi Perbengkelan Otomotif

    The research related to Isolation of the Hydrocarbon Degrading Bacteria in the Contaminated Soil of the Automotive Workshop was conducted from May to October 2014 at Laboratory of Microbiology, Biology Department, Faculty of Mathematics and Natural Sciences, Andalas University. The aimed of this study was to find  the bacteria able to degradate the hydrocarbon from the lubricating oil contaminated soil and to descripe characterization of isolates. This study used a survey method in isolation and characterization of hydrocarbon degrading bacteria.  The result obtained that the bacteria able to degradate the hydrocarbon from the lubricating oil contaminated soil about 39.02% (1.6x105 cfu/g).     The research about callus induction Artemisia vulgaris L. by giving several concentration 2,4-Dichlorophenoxyacetic acid (2,4-D), has been done from May to August 2015 in Plant Physiology Laboratory and Tissue Culture, Department of Biology, Faculty of Mathematics and Natural Science, University of Andalas. The aim of this studywas found the effective concentration of 2,4-D to induce callus of A. vulgaris. The research used Completely Randomized Design (CRD) with 7 treatments and 4 replications. The treatments were : without 2,4-D (control); 0.25 mg/L 2,4-D; 0.50 mg/L 2,4-D; 0.75 mg/L 2,4-D; 1.00 mg/L 2,4-D; 1.25 mg/L 2,4-D; 1.5 mg/L 2,4-D. The result showed that 0.25-1,5 mg/L 2,4-D were able induction callus of A. vulgaris, withcompactuntilthefriabletexture, color ofthe resultingcallusisyellowish green, brownish-green, yellow-brown, whiteyellowishandgreenishwhite. 2,4-D 1.5mg/Lwas the best concentrationto increase fresh weight of callus

    Multiplikasi Tunas Tetrastigma rafflesiae Miq. pada Media Tanam Murashige-Skoog dengan Penambahan 6-Benzylaminopurine dan 1-Naphtaleneacetic Acid secara In Vitro

    The study about in vitro shoot multiplication of Tetrastigma rafflesiae miq. on murashige-skoog media with addition of 6-Benzylaminopurine and 1-Naphtaleneacetic Acid had purpose to determine physiological responses of Tetrastigma rafflesiae nodes with addition of 6-Benzylaminopurine (BAP) and 1-Naphtaleneacetic Acid (NAA) in several concentrations on shoot multiplication stages. This experiment used randomised complete design in 11 treatments and three replications. The treatment were the addition of BAP (1.0-5.0 mg/l) and its combination with NAA (0.1 mg/l) on MS medium. The results showed that nodus successfully producing shoot and callus. The addition of BAP was more effective than its combination with NAA in shoot multiplication. MS medium with addition of 2.0-4.0 mg / l BAP showed a better media for shoot multiplication (2.3-2.7 shoot each explant). Generally, MS media with combination of BAP and NAA produced a higher shoot although it did not significantly differed with BAP treatments. The result indicated that addition of BAP in in vitro medias have a high possibility of success for shoot multiplications in Rafflesia’s host plant conservation.     The research about effect of fungicide (Dithane M-45) on growth of corn (Zea mays L.) and spore density of arbuscular mycorrhizal fungi (AMF) had been done from August to November 2013 in a wire house at Laboratory of Plant Physiology, Departement of Biology, Faculty of Sciences, Andalas University. The research used Completely Randomized Design (CRD) with 7 treatmens and 4 replications. The treatments were varions doses of Dithane M-45; A (0 g/l), B (1,5 g/l), C (2,0 g/l), D (2,5 g/l), E (3,0 g/l), F (3,5 g/l), G (4,0 g/l). The results showed that Dithane M-45 decreased height and number of leaves of corn plants, leaf dry weight and spore density of AMF.     An experimental study on growth of Hopea mengarawan Miq. seedlings under different light intensity was conducted from June to October 2012 at open field. This experiment aimed to find out optimal light intensity for the growth of H. mengarawan seedlings. The study used completely randomized design with four treatments and six replications.     Kepadatan Populasi dan Distribusi Ukuran Cangkang Kerang Lokan (Rectidens sp.) di Perairan Tanjung Mutiara Danau Singkarak, Sumatera Barat

    The study about population density and size distribution of clam (Rectidens sp.) at Tanjung Mutiara Singkarak Lake, West Sumatra has been conducted from June to July 2013. The clams were collected using stratified purposive sampling method. Sampling site was decided into three location based on human activities and three depth strata in each location (<5m, 5-10m and >10-15m). The clams were collected using “dauah†(traditional tool to cath shellfish). There were 43 individu of clams that found at Tanjung Mutiara Singkarak Lake. The highest average population density were found at location III (1.444 ind./m2). The highest average population density were found at strata 3 (1.082 ind./m2). Size distribution of clam showed that at each location and strata have different size. The size of the clam between 24.37-88.54 mm. Medium size clam were the most frequent found in comparison with the small size and large clam. Small clam distributed at strata 2 (5-10 m) locations I and III and strata 3 (> 10-15 m) location III. While large clam only distributed at strata 3 (> 10-15 m) locations II and III.     A study about species of plant consumed by Ungko (Hylobates agilis) at The Biological Education and Research Forest, Andalas University, has been done from July to November 2012. We used ad libitum observation to the gibbon’s feeding behavior and sampled those plants they ate. The samples were processed at Herbarium Andalas University (ANDA), Padang. Seventeen species of plants which belong to eleven families have been recognized as food resources of the gibbons. We described each species of the plants based on morphological characters primerily the part eaten by the gibbon. The results showed that most of the fruit eaten by ungko were fleshy, succulence, and easy to digest. The fruit types were bacca, legume, syconium, capsule and drupe.     A study on dung beetle (Coleoptera: Scarabaeidae) at Lembah Harau Nature Reserve was conducted from July to September 2013. The aim of this study was to identify dung beetles species at the Nature Reserve. We used pitfall trap to collect the beetle along three tracking paths from 500 to 800 asl. We successfully collected 539 individual dung beetles those belong to 18 species, eight genera, six tribes and two subfamilies. Eleven species (61,1 %) of the beetles was not reported in previous study in Indonesia.     Analisis Histologis Hati Ikan Asang (Osteochilus hasseltii C.V.) di Danau Maninjau dan Danau Singkarak, Sumatera Barat

    Histological analysis of liver of hard-lipped barb (Osteochilus hasseltii C.V.) was conducted from March to October 2013 at the Laboratory of Animals Structure and Development, Department of Biology, Faculty Sciences, Andalas University, Padang. Those fish samples were collected form Maninjau and Singkarak Lakes, West Sumatra. The liver tissue were isolated, fixated, dehydrated, planted on paraffin and colored by Haematoxilyn-eosin. Cell Structure and forms were described based on Microscopic tissue study. Results found that the histological damage of fish liver from Maninjau showed three storeys of Hypertrophy, lysis, and necrosis, while Singkarak showed two storeys of hypertrophy and lysis. Those results may indicate that hard-lipped barb from the two lakes had been conminated by some pollutants.Keywords: Osteochilus hasseltii, histopatology, liver, Singkarak Lake, Maninjau Lake

    Karakterisasi Bakteri Termofilik Penghasil Enzim Protease Netral

    A study on the characterization of thermophilic bacteria in producing neutral protease enzyme has been conducted in the Laboratory of Microbiology, Biology Department, Andalas University from October 2013 to March 2014. The bacteria were isolated from a hot spring at Sungai Medang, Kerinci. Water samples  were collected purposively based on gradient temperature at the hot spring which ranged from 450C to 880C. Colony of the bacteria were inoculated using nutrient agar (NA). We obtained 10 isolates of neutral protease enzyme from the thermophilic bacteria with proteolytic index ranged from 0.27 to 3.89. The MIV-14 isolate has the highest proteolytic index which collected from 830C of the hot spring. Characterization of macroscopic, microscopic and biochemical tests were reported. Generally, the colonies have round shape, wavy edge, white, producing endospores and motile. Keywords: characterization, neutral protease, thermophilic bacteria
