eDNA and specific primers for early detection of invasive species e A case study on the bivalve Rangia cuneata, currently spreading in Europe
Intense human activities facilitate the successful spread and establishment of non-indigenous aquatic
organisms in marine and freshwater ecosystems. In some cases such intrusions result in noticeable and
adverse changes in the recipient environments. In the Baltic Sea, the discovery and rapid initial spread of
the North American wedge clam Rangia cuneata represents a new wave of invasion which may trigger
unpredictable changes of the local benthic communities. In this study we present a species-specific DNAbased
marker developed in silico and experimentally tested on environmental samples. Marker specificity
and sensitivity were assessed in vitro from water samples containing different mixtures of the
target species and other five bivalves currently present in the region: the native Cerastoderma glaucum,
Macoma balthica and Mytilus trossulus, the invasive Dreissena polymorpha and the cryptogenic Mya
arenaria. Cross-species amplification was not found in any case. The method allows to detecting at least
0.4 ng of R. cuneata DNA per ml, and 0.1 g of tissue per liter of water. Finally, the marker performance was
assessed in water samples from the Baltic Sea and Vistula Lagoon. The coincidence between independent
visual observations of R. cuneata and positive PCR amplification of the marker from the water samples
confirmed the efficiency of this highly reproducible, fast, and technically easy method. R. cuneata traces
can be detected from environmental DNA even when the population is sparse and small, enabling rapid
management responses and allowing to track the invasion dynamics

Simulating the germination response to diurnally alternating temperatures under climate change scenarios: comparative studies on Carex diandra seeds
Background and Aims Environmental temperature regulates plant regeneration by seed in several superimposed ways, and this complex regulation will be disrupted by climate change. The role of diurnally alternating temperatures (ΔT) in terminating dormancy will be a major player in this disruption, as its effects on seed germination are immediate.
Methods We modelled the effect of ΔT on seed germination comparing two populations of the wetland sedge Carex diandra. We fitted a cardinal-temperature model to germination results in a thermal gradient plate, and used the model to simulate changes in germination under two representative concentration pathways scenarios (RCP2.6 and RCP8.5).
Key Results Increasing ΔT decreased the base temperature for germination and the thermal time required for germination. RCP2.6 projected moderate increases in average temperatures and ΔT, whereas RCP8.5 projected higher warming and ΔT. The higher ΔT effect together with the warmer temperatures increased seed germination in both scenarios. By considering the effects of ΔT, the model projects considerable increases in germination.
Conclusions Carex diandra germination will be very responsive to potential changes in ΔT as a consequence of climate change. This research highlights the role of ΔT in seed responses to climate change. Comprehensive cardinal-temperature models, encompassing the different effects of temperature on seed germination, are needed to understand how climate change will affect plant regeneration

The Bullwhip Effect in Water Demand Management: Taming It Through an Artificial Neural Networks-Based System
The Bullwhip Effect refers to the amplification of the variance of orders and inventories along the supply chain as they move away from the customer. This is considered as the main cause of inefficiencies in the management of a traditional supply chain. However, the Bullwhip Effect is not relevant in the classic system of water distribution, based on long-term supply management. Nevertheless, current circumstances have drawn a new context, which has introduced the concept of Water Demand Management (WDM), in which efficiency and sustainability are of great importance. Then, the time horizon of management has decreased enormously and the supply time takes on an important role. Therefore, the Bullwhip Effect must be considered, as it significantly raises the costs of management. On the one hand, this paper brings evidence that Bullwhip Effect appears in a system of real-time management of water demand. On the other hand, it proposes the application of Artificial Intelligence techniques for its reduction. More specifically, an advanced forecasting system based on Artificial Neural Networks (ANNs) has been used. The Bullwhip Effect is heavily damped.Severo Ochoa. Ref BP13011
Different metabolic features of Bacteroides fragilis growing in the presence of glucose and exopolysaccharides of bifidobacteria
Bacteroides is among the most abundant microorganism inhabiting the human intestine. They are saccharolytic bacteria able to use dietary or host-derived glycans as energy sources. Some Bacteroides fragilis strains contribute to the maturation of the immune system but it is also an opportunistic pathogen. The intestine is the habitat of most Bifidobacterium species, some of whose strains are considered probiotics. Bifidobacteria can synthesize exopolysaccharides (EPSs), which are complex carbohydrates that may be available in the intestinal environment. We studied the metabolism of B. fragilis when an EPS preparation from bifidobacteria was added to the growth medium compared to its behavior with added glucose. 2D-DIGE coupled with the identification by MALDI-TOF/TOF evidenced proteins that were differentially produced when EPS was added. The results were supported by RT-qPCR gene expression analysis. The intracellular and extracellular pattern of certain amino acids, the redox balance and the ot-glucosidase activity were differently affected in EPS with respect to glucose. These results allowed us to hypothesize that three general main events, namely the activation of amino acids catabolism, enhancement of the transketolase reaction from the pentose-phosphate cycle, and activation of the succinate-propionate pathway, promote a shift of bacterial metabolism rendering more reducing power and optimizing the energetic yield in the form of ATP when Bacteroides grow with added EPSs.
