Detecting nuisance species using NGST: Methodology shortcomings and possible application in ballast water monitoring
Detecting the presence of potential invasive species in ballast water is a priority for preventing their
spread into new environments. Next generation sequencing technologies are being increasingly used for
exploring and assessing biodiversity from environmental samples. Here we apply high throughput
sequencing from DNA extracted from ballast water (BW) samples employing two different platforms, Ion
Torrent and 454, and compare the putative species catalogues from the resulting Operational Taxonomic
Units (OTU). Water samples were taken from the RV Polastern ballast tank in five different days between
the second and the twentieth navigation day. Pronounced decrease of oxygen concentration and increase
of temperature occurred in the BWduring this time, coincident with a progressively higher proportion of
unassigned OTU and short reads indicating DNA degradation. Discrepancy between platforms for species
catalogues was consistent with previously published bias in AT-rich sequences for Ion Torrent platform.
Some putative species detected from the two platforms increased in frequency during the Polarstern
travel, which suggests they were alive and therefore tolerant to adverse conditions. OTU assigned to the
highly invasive red alga Polysiphonia have been detected at low but increasing frequency from the two
platforms. Although in this moment NGST could not replace current methods of sampling, sorting and
individual taxonomic identification of BW biota, it has potential as an exploratory methodology especially
for detecting scarce species
The role of reading fluency in children's text comprehension
Understanding a written text requires some higher cognitive abilities that not all children have. Some children have these abilities, since they understand oral texts; however they have difficulties with written texts, probably due to problems in reading fluency. The aim of this study was to determine which aspects of reading fluency are related to reading comprehension. Four expositive texts, two written and two read by the evaluator, were presented to a sample of 103 primary school children (third and sixth grade). Each text was followed by four comprehension questions. From this sample we selected two groups of participants in each grade, 10 with good results in comprehension of oral and written texts, and 10 with good results in oral and poor in written comprehension. These 40 subjects were asked to read aloud a new text while they were recorded. Using Praat software some prosodic parameters were measured, such as pausing and reading rate (number and duration of the pauses and utterances), pitch and intensity changes and duration in declarative, exclamatory and interrogative sentences and also errors and duration in words by frequency and stress. We compared the results of both groups with ANOVAs. The results showed that children with less reading comprehension made more inappropriate pauses and also intersentential pauses before comma than the other group and made more mistakes in content words; significant differences were also found in the final declination of pitch in declarative sentences and in the F0 range in interrogative ones. These results confirm that reading comprehension problems in children are related to a lack in the development of a good reading fluency.Programa Severo Ochoa (FICYT): BP14-038

Quantitative evaluation of cellular uptake, DNA incorporation and adduct formation in cisplatin sensitive and resistant cell lines: Comparison of different Pt-containing drugs
The use of Pt-containing compounds as chemotherapeutic agents facilitates drug monitoring by using
highly sensitive elemental techniques like inductively coupled plasma mass spectrometry (ICP-MS).
However, methodological problems arise when trying to compare different experiments due to the high
variability of biological parameters. In this work we have attempted to identify and correct such
variations in order to compare the biological behavior of cisplatin, oxaliplatin and pyrodach-2 (a novel
platinum-containing agent). A detailed study to address differential cellular uptake has been conducted
in three different cell lines: lung adenocarcinoma (A549); cisplatin-sensitive ovarian carcinoma (A2780);
and cisplatin-resistant ovarian carcinoma (A2780cis). The normalization of Pt results to cell mass, after
freeze-drying, has been used to minimize the errors associated with cell counting. Similarly, Pt
accumulation in DNA has been evaluated by referencing the Pt results to the DNA concentration, as
measured by 31P monitoring using
flow-injection and ICP-MS detection. These strategies have permitted
to address significantly lower Pt levels in the resistant cells when treated with cisplatin or oxaliplatin as
well as an independent behaviour from the cell type (sensitive or resistant) for pyrodach-2. Similarly,
different levels of incorporation in DNA have been found for the three drugs depending on the cell model
revealing a different behavior regarding cell cisplatin resistance. Further speciation experiments (by
using complementary HPLC–ICP-MS and HPLC–ESI-Q-TOF MS) have shown that the main target in DNA is
still the N7 of the guanine but with different kinetics of the ligand exchange mechanism for each of the

The motor-semantic meanings of verbs generated by Parkinson's disease patients on/off dopamine medication in a verbal fluency task
The human motor cortex is involved in the planning and execution of movements as well as in the processing of action language, and it is impaired in Parkinson's disease (PD) as a consequence of dopamine degeneration. The aim of this study is to understand the role of dopamine in the processing of action words by looking at PD patients with and without medication. A group of 23 healthy controls and 34 non-demented PD patients participated in an action verbal fluency task on and off dopamine medication. A repeated measures 2 × 2 ANOVA was conducted with dopamine (on/off) and type of verb (high and low specificity motor verbs) as independent variables and the number of words generated as a dependent variable. Our results showed that patients on dopamine medication produced a significantly greater number of high specificity motor verbs than low ones. In contrast, patients off dopamine medication generated a similar number of verbs with high and low motor specificity. MANOVA analyses showed a significantly lower production of verbs with high motor specificity on PD patients off medication compared to healthy controls. However, performance of PD patients in the on condition was similar to that found in healthy controls. In conclusion, the dopamine network from basal ganglia to brain motor areas might play a role in retrieving action verbs with specific semantic representations

En los últimos años se asiste en España
a una rápida pérdida y desaparición de
razas de ganado de menor especialización
productiva, que se explotaban con buenos
rendimientos hasta hace pocos años, y
que están siendo desplazadas por razas
más productivas. Estas razas, aunque poco
competitivas en el plano productivo,
constituyen una importante reserva de variabilidad
genética, que sería totalmente
irrecuperable en caso de desaparición.
Además, las razas locales presentan con
frecuencia un mayor grado de adaptación
a condiciones desfavorables o extremas,
por lo que son más indicadas para el aprovechamiento
de los recursos naturales de
esas zonas, siendo además capaces de
proporcionar productos alimentarios diferenciables
de alta calidad y por tanto susceptibles
de lograr un valor añadido.
Por todo ello, existe la obligación de
conservar el patrimonio genético con todas
las herramientas disponibles, lo que
ha venido constituyendo una preocupación
constante de la FAO. Una vez identificados
y caracterizados los recursos genéticos
de un país, se pueden conservar
in situ o ex situ. Los métodos in situ mantienen
a los animales en el hábitat donde
están adaptados, mientras que los métodos
ex situ obtienen los recursos genéticos

Herida por mordedura de perro. Limpieza y sutura en Atención Primaria
Objetive:
Shorten the period of wound Healing, reduce the risk of infection getting a satisfactory
cosmetic result and improving the quality of nursing care in this type of injuries.
Methods / description of experience
A clinical case involving a wounded dog bite in a patient whom, after a review of the
literature, combined with the use of nurse methodology NANDA, NIC and NOC, primary closure is decided
Development or results:
A 55 years old woman who comes to your primary care nurse with a dog bite on her
inner forearm. Assessment of the characteristics of the wound is made. Actual and potential diagnoses are made, objetives are established and the activities needed to
achieve them are chosen. The nurse interventions planned are performed. The whole process is recorded and continuous evaluation of it is done.
The outcome was favourable in the altered needs detected, they didn´t show signs of
infection, and the lesion healed with good aesthetic outcome.
Conclusions:
It is important to make assessments of these wounds individually, taking into account
the particular characteristics of each case and work with nursing methodology to
improve patient care holistically.Objetivo:
Acortar el período de cicatrización de la herida, disminuir el riesgo de infección consiguiendo un resultado estético satisfactorio y mejorando la calidad de los cuidados de enfermería en este tipo de heridas.
Material y métodos/descripción de la experiencia:
Se describe un caso clínico relacionado con una herida por mordedura de perro en una paciente en la que, tras una revisión bibliográfica de la literatura, unido a la utilización de la metodología enfermera con lenguaje NANDA, NIC, NOC, se decide tratar mediante sutura primaria.
Desarrollo o resultados:
Mujer de 55 años que acude a su enfermera de Atención Primaria por herida por mordedura de perro en cara interna del antebrazo derecho. Se realiza valoración de las características de la herida. Se establecen los diagnósticos reales y potenciales, se fijan unos objetivos y se eligen las actividades necesarias para alcanzarlos. Se llevan a cabo las intervenciones de enfermería planificadas. Se registra todo el proceso y se realiza una evaluación continua del mismo.
La evolución fue favorable en las necesidades que se encontraron alteradas, no aparecieron signos de infección, y la lesión cicatrizó con buen resultado estético.
Conclusiones:
La sutura de heridas por mordedura de perro, es tratamiento de elección en aquellas que presentan determinados factores, por todo ello, es importante valorar estas heridas de forma individualizada, teniendo en cuenta las características particulares de cada caso y trabajar con metodología enfermera para la mejora de la atención al paciente de forma integral
Hybridization of Energy Storage Systems for Electric Transportation by Means of Bidirectional Power Electronic Converters
This paper deals with the design of a Hybrid Energy Storage System (HESSs) for electric transportation such as Electric/Hybrid Vessel and Electric/Hybrid Train. The association of more than one Energy Storage Systems (ESSs) e.g., batteries which have different dynamics permit to take the advantages of the characteristics of both ESSs obtaining simultaneously a high energy density and high power density. This yields to a decrease in terms of the size of the main ESS and the total cost and an increase in terms of life span. The emulation of the batteries and with required control algorithm for the HESS are proposed. The design and the control of the HESSs is validated with the simulation in MATLAB/SIMULINK® environment and also with the real-time emulation of batteries in a laboratory setup of a HESS. The real-time experimental results have been validated against PC simulations showing full consistency.
