    Milk production based on grazing is being promoted over cattle housed indoors, because of the advantages regarding animal welfare, milk quality and the environment. Cows’ milk is rich in miRNAs, molecules that regulate gene expression in eukaryotes. Their profiles may vary depending on environmental factors such as farm management and feeding. We hypothesize that miRNA can be used as a certification tool for dairy farms whose milk production is based on grazing. The objective is to apply an artificial intelligence algorithm to the results of miRNA expression in milk to evaluate the possibility of designing a fast and cheap traceability tool that can differentiate the milk produced in a grazing-based system from milk produced in indoor systems. Cells and fat fractions were isolated from seventy-three milk tank samples from ‘No-Grazing’ (n=47) vs ‘Grazing’ (n=26) farms. MiRNA expression was analysed in the cells and the fat fractions of the milk samples. Following miRNAs expression analysis, decision trees were built for their expression results using the C4.5 machine learning algorithm. The algorithm was not able to correctly classify each sample in its group, nor was it able to identify relevant miRNAs. We assume that the enormous internal variability (diets, botanical composition of the pastures, and grazing duration, etc.) in commercial grazing farms could be the cause of the difficulty in machine learning of how to classify milk from grazing farms.-L.     IL6 gene polymorphism association with calcific aortic valve stenosis and influence on serum levels of interleukin-6

    Aortic valve stenosis is the most frequent valve disease in developed countries and its prevalence will increase with population aging. There is still no pharmaceutical treatment nor biomarker to determine the susceptibility to develop aortic stenosis. Therefore, we analyzed the association of polymorphisms in risk loci with calcific aortic stenosis. Patients with aortic valve disease were genotyped for PALMD rs6702619, LPA rs10455872, and IL6 rs1800795 polymorphisms and circulating levels of interleukin-6 (IL-6) were measured. Calcium content of leaflets obtained in valve replacement surgeries was determined by micro-computed tomography. In the genotyping of 578 individuals, we found significant association between PALMD and IL6 polymorphisms and aortic stenosis in patients with tricuspid aortic valve, independently of other potentially confounding variables such as age and dyslipidemia. There was no association of these polymorphisms with valve calcium content, but this value correlated with the mean aortic pressure gradient (r = 0.44; P < 0.001). The CC genotype of IL6 polymorphism was associated with higher levels of serum IL-6 compared to other genotypes (23.5 vs. 10.5 pg/ml, respectively; P = 0.029). Therefore, patients carrying the CC genotype of IL6 rs1800795 polymorphism present higher levels of circulating IL-6 and this could contribute to the severity of the aortic valve stenosis. Our results agree with the identification of IL6 as a locus risk for stenosis and also with the intervention of this cytokine in aortic valve calcification. A more exhaustive follow-up of those patients carrying risk genotypes is therefore recommended

    This database contains open, harmonized, and ready-to-use global data on holdover time. Holdover time is defined as the time between lightning-induced fire ignition and fire detection. The first version of the database is composed of three data files (censored data, non-censored data, ancillary data) and three metadata files (description of database variables, list of references, reproducible examples). These data were collected through a literature review of LIW studies and some datasets were assembled by authors of the original studies, covering more than 150,000 LIW from 13 countries in five continents and a time span of a century from 1921 to 2020. Censored data are the core of the database and consist of frequency data reporting the number or relative frequency of LIW per interval of holdover time. Ancillary data provide additional information on the methods and contexts in which the data were generated in the original studies. Potential contributors to the database are encouraged to contact the corresponding author in the readme file

    Chronic Treatment with Melatonin Improves Hippocampal Neurogenesis in the Aged Brain and under Neurodegeneration

    oai:ria.asturias.es:123456789/14605Adult hippocampal neurogenesis is altered during aging and under different neuropsychi atric and neurodegenerative diseases. Melatonin shows neurogenic and neuroprotective properties during aging and neuropathological conditions. In this study, we evaluated the effects of chronic treatment with melatonin on different markers of neurodegeneration and hippocampal neurogenesis using immunohistochemistry in the aged and neurodegenerative brains of SAMP8 mice, which is an animal model of accelerated senescence that mimics aging-related Alzheimer’s pathology. Neurodegenerative processes observed in the brains of aged SAMP8 mice at 10 months of age in clude the presence of damaged neurons, disorganization in the layers of the brain cortex, alterations in neural processes and the length of neuronal prolongations and β-amyloid accumulation in the cortex and hippocampus. This neurodegeneration may be associated with neurogenic responses in the hippocampal dentate gyrus of these mice, since we observed a neurogenic niche of neural stem and progenitor/precursors cells in the hippocampus of SAMP8 mice. However, hippocampal neurogenesis seems to be compromised due to alterations in the cell survival, migration and/or neuronal maturation of neural precursor cells due to the neurodegeneration levels in these mice. Chronic treatment with melatonin for 9 months decreased these neurodegenerative processes and the neurodegeneration-induced neurogenic response. Noticeably, melatonin also induced recovery in the functionality of adult hippocampal neurogenesis in aged SAMP8 mice

    Inflammaging: Implications in Sarcopenia

    In a world in which life expectancy is increasing, understanding and promoting healthy aging becomes a contemporary demand. In the elderly, a sterile, chronic and low-grade systemic inflammation known as “inflammaging” is linked with many age-associated diseases. Considering sarcopenia as a loss of strength and mass of skeletal muscle related to aging, correlations between these two terms have been proposed. Better knowledge of the immune system players in skeletal muscle would help to elucidate their implications in sarcopenia. Characterizing the activators of damage sensors and the downstream effectors explains the inference with skeletal muscle performance. Sarcopenia has also been linked to chronic diseases such as diabetes, metabolic syndrome and obesity. Implications of inflammatory signals from these diseases negatively affect skeletal muscle. Autophagic mechanisms are closely related with the inflammasome, as autophagy eliminates stress signaling sent by damage organelles, but also acts with an immunomodulatory function affecting immune cells and cytokine release. The use of melatonin, an antioxidant, ROS scavenger and immune and autophagy modulator, or senotherapeutic compounds targeting senescent cells could represent strategies to counteract inflammation. This review aims to present the many factors regulating skeletal muscle inflammaging and their major implications in order to understand the molecular mechanisms involved in sarcopenia
