Elucidation of flow cytometry parameters for extracellular vesicles detection
No full textTato bakalářská práce se zabývá optimalizací parametrů průtokové cytometrie pro detekci extracelulárních vezikulů (EV). Cílem bylo identifikovat vhodná nastavení přístroje NovoCyte, která umožní spolehlivou detekci a charakterizaci EV izolovaných z různých buněčných linií. V práci byly testovány různé fluorescenční markery, kontrola integrity pomocí detergentu a softwarové prahy měření. Výsledky ukazují, že pečlivá volba parametrů významně ovlivňuje kvalitu dat a reprodukovatelnost analýzy. Práce přináší i praktický návod pro začínající uživatele průtokového cytometru NovoCyte a měření EV.This bachelor's thesis deals with the optimization of flow cytometry parameters for the detection of extracellular vesicles (EVs). The aim was to identify suitable settings of the NovoCyte instrument that enable reliable detection and characterization of EVs isolated from various cell lines. Various fluorescent markers, integrity control using detergent, and software measurement thresholds were tested in this work. The results show that careful selection of parameters significantly affects the quality of data and the reproducibility of the analysis. The thesis also provides a practical guide for beginner users of the NovoCyte flow cytometer and EV measurements

Tato diplomová práce se zaměřuje na analýzu faktorů výzev a příležitostí v oblasti cestovního ruchu z pohledu malých a středních podniků. Teoretická část se zaměřuje nejprve na faktory cestovního ruchu, na mezinárodní obchod spolu se současnými globálními trendy v kontextu dopadu na poptávku po cestovním ruchu a konkurenci v odvětví cestovního ruchu. Jsou také prezentovány dílčí strategie pro udržení konkurenceschopnosti a získání konkurenční výhody pro podniky v tomto odvětví s využitím konceptů brandingu, marketingových prvků a nástrojů strategického řízení. Na základě teoretických poznatků je praktická část zaměřena na případ ekosystému Santa Claus Village. Je vypracován plán strategií pro zlepšení podnikání podobných zážitkových společností. Případová studie se zaměřuje především na to, jak by tyto společnosti mohly nejlépe prodávat své zážitky, s přihlédnutím k tomu, jak nejlépe řešit globální výzvy v cestovním ruchu a jak nejlépe využít příležitostí, které světová ekonomika v této oblasti nabízí.The more globalization expands, the more people travel for tourism. This diploma thesis focuses on analysing the factors of challenges and opportunities in the field of tourism from the perspective of small and medium enterprises. The theoretical part focuses first on factors of tourism, on international business together with current global trends in the context of the impact on tourism demand and business competition in tourism sector. Sub-strategies for maintaining competitiveness and gaining a competitive advantage for businesses in this sector, utilizing branding concepts, marketing elements and strategic management tools, are also presented. Based on theoretical knowledge, the practical part is focused on the case of the Santa Claus Village ecosystem. Plan of strategies to improve business of similar experience companies is elaborated. The case study mainly focuses on how these companies could best sell their experiences, taking into account how best to tackle global challenges in tourism and how to best use the opportunities that the world economy offers in this area

No full textBakalářská práce s názvem Konkurenceschopnost výrobního podniku se zaměřuje na zhodnocení konkurenceschopnosti společnosti Valdemar Grešík - Natura s. r. o. Hlavním cílem práce je zanalyzovat schopnost tohoto podniku konkurovat a na základě výsledků jednotlivých analýz navrhnout vhodnou strategii pro posílení konkurenceschopnosti. Struktura práce je rozdělena do dvou částí, a to na teoretickou a praktickou. Teoretická část se věnuje definici klíčových pojmů, jako jsou konkurenceschopnost, konkurence, konkurenční výhoda, vybrané metody situační analýzy a růstové strategie. Praktická část se zaměřuje na charakteristiku vybraného výrobního podniku a následně prostřednictvím jednotlivých metod situační analýzy hodnotí jeho konkurenceschopnost. Na základě provedených analýz je v závěru práce navržena růstová strategie doplněná o konkrétní doporučení, jejichž cílem je zvýšit schopnost podniku konkurovat a posílit jeho pozici na trhu.The bachelor thesis entitled Competitiveness of the manufacturing company deals with the evaluation of the competitiveness of Valdemar Grešík - Natura s. r. o. The main objective of the thesis is to analyse the ability of this enterprise to compete and based on the results of individual analyses to propose an suitable strategy to increase its competitiveness. The structure of the thesis is divided into two parts, theoretical and practical. The theoretical part is devoted to the definition of key concepts such as competitiveness, competition, competitive advantage, selected methods of situational analysis and growth strategies. The practical part focuses on the characteristics of the selected manufacturing enterprise and then evaluates its competitiveness through the individual methods of situational analysis. Based on the analyses, the thesis concludes by proposing a growth strategy complemented by specific recommendations aimed at increasing the company's ability to compete and strengthening its market position

V kontextu Česka se zaměřuje především na problematiku povodní a sucha. Praktická část představuje didaktický koncept využitelný ve výuce na nižším gymnáziu, jehož hlavní součástí je simulační rolová hra zaměřená na vyjednávání o protipovodňových opatřeních. Dále jsou navrženy i další výukové aktivity tematicky související s vodními problémy.This bachelor thesis deals with environmental and socio-economic issues related to water both globally and within the Czech Republic, which are becoming increasingly relevant. The theoretical part summarizes the importance of water, growing consumption, problems affecting different parts of the world, their impacts, and possible future developments. In the context of the Czech Republic, the thesis focuses primarily on the issues of floods and droughts. The practical part presents a didactic concept suitable for lower secondary education, the core of which is a simulation role-play activity focused on negotiating flood protection measures. Electronically controlled clock concept with alternative time display
No full textBakalářská práce se věnuje návrhu a realizaci dvou odlišných konceptů elektronicky řízených hodin s alternativním zobrazením času. Hlavním cílem bylo experimentálně ověřit možnosti netradiční vizualizace časových údajů prostřednictvím současných technologií, a to jak z hlediska funkčnosti, tak i uživatelské přívětivosti a estetického ztvárnění.
Prvním řešením jsou tzv. stínové LED hodiny, které pomocí přesně řízených segmentů LED pásků vytvářejí iluzivní analogový ciferník prostřednictvím stínových efektů. Druhý koncept tvoří binární sněžítkové hodiny, kde jsou bity časových údajů reprezentovány pohybem polystyrenových částic v uzavřených komorách, řízených pomocí nízkonapěťových ventilátorů. Obě zařízení jsou řízena mikrokontrolérem a konstrukčně postavena s využitím 3D tisku.
Práce obsahuje rešerši historických i moderních metod zobrazování času, návrh hardwarové i softwarové architektury obou zařízení, výrobní dokumentaci a kritickou reflexi dosažených výsledků. Výsledné funkční prototypy představují kombinaci praktického inženýrského řešení s tvůrčím přístupem k interakci člověka s technologií. Projekt má zároveň potenciál sloužit jako demonstrační i edukativní prostředek v prostředí technicky orientovaných institucí.This bachelor's thesis focuses on the design and realization of two distinct concepts of electronically controlled clocks with alternative time visualization. The primary objective was to explore nontraditional representations of time using contemporary technologies, emphasizing both functional and aesthetic aspects as well as user interaction.
The first concept, called Shadow LED Clock, creates the illusion of an analog dial using precisely controlled LED strips that cast symbolic shadows. The second solution, Binary Snow Globe Clock, displays time in binary format using lightweight polystyrene particles suspended by low-voltage fans inside enclosed transparent columns. Both systems are controlled by a microcontroller and mechanically built using 3D printing technology.
The thesis includes a survey of traditional and modern time display techniques, detailed hardware and software design, mechanical construction documentation, and a critical evaluation of the final prototypes. The outcome demonstrates an innovative blend of engineering practice and creative design, with the potential to serve both educational and demonstrational purposes in technical institutions

Cílem práce je zhodnotit vývoj zimníhoturismu v této oblasti, identifikovat hlavní trendy a faktory ovlivňující návštěvnost, anavrhnout strategie pro další rozvoj zimního turismu. Práce bude rozdělena do dvou částí. V teoretické části bude popsána historie a současný stav zimního turismu v Orlických horách, včetně infrastruktury, turistických atrakcí a nabídky turistických služeb. Praktická část bude obsahovat analýzu dat o návštěvnosti, motivaci turistů k návštěvě Orlických hor, ekonomických přínosech a dopadech na místní obyvatele. Data budou získána pomocí dotazníkového šetření, které bude zaměřeno na návštěvníky i místní obyvatele. Výsledky této práce by měly poskytnout užitečné informace pro místní samosprávy, podnikatele a další zainteresované strany, které se podílejí na rozvoji zimního turismu v Orlických horách.This bachelor thesis focuses on the analysis and evaluation of the development ofwinter tourism in the Orlické Mountains over the past thirty years. The aim of the thesisis to assess the development of winter tourism in this region, identify the main trendsand factors influencing visitor numbers, and propose strategies for the furtherdevelopment of winter tourism. The thesis will be divided into two parts. The theoretical part will describe the history and current state of winter tourism in the Orlické Mountains, including infrastructure, tourist attractions, and the range of tourism services. The practical part will contain an analysis of visitor data, the motivation of tourists to visit the Orlické Mountains, economic benefits, and impacts on local residents. Data will be collected through a survey, which will target both visitors and local residents. The results of this thesis should provide useful information for local authorities, entrepreneurs, and other stakeholders involved in the development of winter tourism in the Orlické Mountains

No full textTato bakalářská práce se zabývá návrhem a implementací interaktivní aplikace pro zpracování EKG signálu v prostředí MATLAB App Designer. Cílem bylo vytvořit nástroj, umožňující načtení EKG dat, jejich filtraci od běžných typů rušení, detekci klíčových komponent EKG a výpočet srdeční frekvence včetně diagnostiky srdečního rytmu. Součástí aplikace je také vizualizace frekvenčního spektra před a po filtraci, která uživateli poskytuje přehled o účinnosti zvolených filtrů. Výsledná aplikace je uživatelsky přívětivá a může sloužit jako výuková pomůcka nebo jako základ pro další vývoj v oblasti automatické analýzy EKG signálů.This bachelor thesis focuses on the design and implementation of an interactive application for processing ECG signals using MATLAB App Designer. The goal was to create a tool that allows loading ECG data, filtering out common types of noise, detecting key ECG components, and calculating heart rate including rhythm diagnostics. The application also includes visualization of the frequency spectrum before and after filtering, providing the user with insight into the effectiveness of the selected filters. The resulting application is user-friendly and can serve both as an educational tool and a foundation for further development in the field of automatic ECG signal analysis

Hlavní pozornost je zaměřena na výslovnost závěrových souhlásek /b/, /g/, /d/ v různých pozicích ve slovech, stejně jako na výslovnost souhláskových skupin /sk/, /st/ na začátku slov. Cílem této práce je analyzovat nahrávky anglického textu přečteného rodilými mluvčími španělštiny s různými úrovněmi znalosti angličtiny. Předpokládá se, že pokročilejší skupina (úrovně B2-C1) bude mít přesnější výslovnost než skupina s nižší úrovní (A2-B1). K provedení této studie bude použit rozbor odborné literatury a analýza nahrávek pomocí programu Praat.This bachelor's thesis is dedicated to the topic of the difficulties that native Spanish speakers face when pronouncing consonants in English. It primarily addresses the pronunciation of plosive consonants such as /b/, /g/, and /d/ in different positions within words, as well as the pronunciation of consonant clusters /sk/ and /st/ at the beginning of words. The objective of the thesis is to record and analyze an English text read by native Spanish speakers with different levels of English proficiency. It is expected that the more advanced group (levels B2-C1) will have more accurate pronunciation than the group with levels A2-B1. To conduct this research, a review of the literature on the topic will be carried out, along with an analysis of recordings using the Praat software

Práce čerpá inspiraci z metody hutně foukaného skla do síťových forem a zároveň recykluje odpadní materiál vznikající při výrobě rokajlu. Kombinací těchto technik se zaměřuji na vytvoření tvarově zajímavých a hravých objektů.The aim of this bachelor's thesis is to create a series of light objects using a technique that combines glass with wire mesh and mass-produced seed beads that exhibit various defects and are no longer suitable for sale. The work draws inspiration from the method of hot-blown glass into mesh molds, while also recycling waste material generated during the production of seed beads. By combining these techniques, I focus on creating visually engaging and playful forms

z důvodu zhoršené soběstačnosti poprvé stávají klienty terénní pečovatelské služby. Cílem je popsat proces akceptace terénní pečovatelské služby klientem. Tento proces zahrnuje zejména fázi rozhodování o přijetí terénní pečovatelské služby a fázi adaptace na přítomnost této služby v životě seniora. V teoretické části je popsána terénní pečovatelská služba, její fungování a zasazení do systému sociálních služeb v České republice, dále je popsána problematika seniorského věku, nastupujících změn a rozhodovacích a adaptačních mechanismů ve stáří. Průzkumná část obsahuje popis použité metodologie, kterou byl kvalitativní přístup formou nestrukturovaných rozhovorů s respondenty. Následuje popis průběhu průzkumu a prezentace získaných výsledků. Průzkumem bylo zjištěno, které faktory hrají roli v procesu rozhodování
a úspěšné adaptace seniorů na terénní pečovatelskou službu. Jako klíčový faktor se ukazuje schopnost seniora zaujmout konstruktivní přístup a učinit vlastní rozhodnutí pro přijetí terénní pečovatelské služby, které je též základem budoucí úspěšné adaptace.This bachelor´s thesis focuses on the process of decision-making and adaptation in seniors who, due to impaired self-sufficiency, become clients of field care services for the first time. The aim is to describe the process of acceptance of the field care service by the client. This process includes, in particular, the phase of deciding to accept field care service and the phase of adaptation to the presence of this service in the life of a senior citizen. In the theoretical part, the field care service, its functioning and integration into the system of social care in the Czech Republic is described, followed by description of senior age, emerging changes and decision-making and adaptation mechanisms in old age. The research part contains a description of the methodology used, which was a qualitative approach in the form of unstructured interviews with respondents. Description of the research process and a presentation of the obtained results follow. The survey found out which factors play a role in the decision-making process and the successful adaptation of seniors to the field care service. The key factor is the senior's ability to take a constructive approach and make their own decision to accept the service, which is also the basis of future successful adaptation
