    Selective hydrogenation of indolizines: an expeditious approach to derive tetrahydroindolizines and indolizidines from morita–baylis–hillman adducts

    In this study, we describe the hydrogenation of indolizines derived from Morita–Baylis–Hillman adducts. We demonstrate that functionalized tetrahydroindolizines and indolizidines can be prepared selectively, at low pressure, by simply adjusting the acidity of the medium.     Push-out bond strength of MTA HP, a new high-plasticity calcium silicate-based cement

    This study was designed to investigate the resistance to dislodgment provided by MTA HP, a new high-plasticity calcium silicate-based cement. Biodentine and White MTA Angelus were used as reference materials for comparison. Three discs 1 ± 0.1 mm thick were obtained from the middle third of the roots of 5 maxillary canines. Three 0.8-mm-wide holes were drilled on the axial surface of each root disc. Standardized irrigation was performed. Then the holes were dried with paper points and filled with one of the three tested cements. The filled dental slices were immersed in a phosphate-buffered saline (PBS) solution (pH 7.2) for 7 days before the push-out assessment. The Kruskal-Wallis test was applied to assess the effect of each endodontic cement on the push-out bond strength. Mann-Whitney with Bonferroni correction was used to isolate the differences. The alpha-type error was set at 0.05. All specimens had measurable push-out values and no premature failure occurred. There were significant differences among the materials (p <0.05). The Biodentine specimens had the highest push-out bond strength values (p < 0.05). MTA HP had significantly higher bond strength than White MTA (p < 0.05).     Oropouche virus (OROV) is a member of the Orthobunyavirus genus in the Bunyaviridae family and a prominent cause of insect-transmitted viral disease in Central and South America. Despite its clinical relevance, little is known about OROV pathogenesis. To define the host defense pathways that control OROV infection and disease, we evaluated OROV pathogenesis and immune responses in primary cells and mice that were deficient in the RIG-I-like receptor signaling pathway (MDA5, RIG-I, or MAVS), downstream regulatory transcription factors (IRF-3 or IRF-7), beta interferon (IFN-β), or the receptor for type I IFN signaling (IFNAR). OROV replicated to higher levels in primary fibroblasts and dendritic cells lacking MAVS signaling, the transcription factors IRF-3 and IRF-7, or IFNAR than in wild-type (WT) cells. In mice, deletion of IFNAR, MAVS, or IRF-3 and IRF-7 resulted in uncontrolled OROV replication, hypercytokinemia, extensive liver damage, and death, whereas WT congenic animals failed to develop disease. Unexpectedly, mice with a selective deletion of IFNAR on myeloid cells (CD11c Cre+ Ifnar f/f or LysM Cre+ Ifnar f/f) did not sustain enhanced disease with OROV or a selective (flox/flox) deletion La Crosse virus, a closely related encephalitic orthobunyavirus. In bone marrow chimera studies, recipient irradiated Ifnar −/− mice reconstituted with WT hematopoietic cells sustained high levels of OROV replication and liver damage, whereas WT mice reconstituted with Ifnar −/− bone marrow were resistant to disease.     Effect of annatto seed and coriander leaves as natural antioxidants in fish meatballs during frozen storage

    The effects of annatto (0.1 g/100g) and coriander (0.5 g/100g) were assessed against lipid oxidation in white hake meatballs cooked in boiling water (95 ± 1 °C) for 30 min and stored at −18 °C for 120 d. The fatty acids (FA) and the nutritional quality, cholesterol, cholesterol oxides, thiobarbituric acid reactive substances (TBARS) values, and conjugated dienes were analyzed to follow the course of oxidation. Annatto and coriander were efficient in the control of lipid oxidation, also preserving the essential FA. At 120 d of storage, the eicosapentaenoic acid (EPA) concentration decreased respectively by 43%, 32%, 12%, and 9% in the control, coriander, annatto, and annatto + coriander patties. For docosahexaenoic acid (DHA), these concentrations decreased, respectively, 44%, 30%, 11%, and 7%, revealing a probable synergistic effect among the antioxidant compounds present in both spices.     Scaphoid fracture nonunion: correlation of radiographic imaging, proximal fragment histologic viability evaluation, and estimation of viability at surgery

    The purpose of this study was to correlate the pre-operative imaging, vascularity of the proximal pole, and histology of the proximal pole bone of established scaphoid fracture non-union. This was a prospective non-controlled experimental study. Patients were evaluated pre-operatively for necrosis of the proximal scaphoid fragment by radiography, computed tomography (CT) and magnetic resonance imaging (MRI). Vascular status of the proximal scaphoid was determined intra-operatively, demonstrating the presence or absence of puncate bone bleeding. Samples were harvested from the proximal scaphoid fragment and sent for pathological examination. We determined the association between the imaging and intra-operative examination and histological findings. We evaluated 19 male patients diagnosed with scaphoid nonunion. CT evaluation showed no correlation to scaphoid proximal fragment necrosis. MRI showed marked low signal intensity on T1-weighted images that confirmed the histological diagnosis of necrosis in the proximal scaphoid fragment in all patients. Intra-operative assessment showed that 90 % of bones had absence of intra-operative puncate bone bleeding, which was confirmed necrosis by microscopic examination. In scaphoid nonunion MRI images with marked low signal intensity on T1-weighted images and the absence of intra-operative puncate bone bleeding are strong indicatives of osteonecrosis of the proximal fragment391677

    Utero body sarcomas : characteristics in diagnosis, treatment and survival

Apesar de agressivos, muitos são diagnosticados em estágios iniciais. Dentre sua diversidade os principais tipos histológicos são: carcinossarcoma (CCS), leiomiossarcoma (LMS), sarcoma do estroma endometrial (SEE) e adenossarcoma (ADS). Há poucas informações sobre esses tumores no Brasil. Objetivos: Avaliar o perfil no diagnóstico, tratamento e evolução dos casos de sarcoma de corpo de útero. Metodologia: Foi realizado um estudo coorte retrospectivo com analise de prontuários dos casos registrados no Hospital da Mulher Prof. Dr. J. A. Pinotti, CAISM, Unicamp, no período entre 2001 e 2016, totalizando 122 casos de sarcomas, considerando CCS, LMS, SEE, ADS e outros. Foram consideradas informações clinicas, das neoplasias, do tratamento e seguimento. Na analise estatística foram utilizados os testes Qui-Quadrado ou Fisher, avaliação de tendência do número de casos ao longo do tempo pelo teste Qui-Quadrado de tendência, análise regressão logística politômica, univariada e multivariada, com critério stepwise de seleção de variáveis para avaliar a relação entre os fatores associados com os tipos histológicos de sarcomas, montadas curvas de sobrevida de Kaplan-Meier e analisadas pelo teste log-rank. Foi considerado p<0,05 para significância. Resultados: O principal sinal ou sintoma que propiciou o diagnóstico dos sarcomas avaliados foi sangramento uterino anormal em 77%. As principais formas de diagnóstico foram curetagem uterina e cirurgia, 35% e 34% respectivamente. Foi observada uma tendência de aumento significativo nos registros de sarcomas com +1,2 casos (p=0,044) a cada 2 anos. A proporção carcinomas/sarcomas se manteve relativamente estável. Entre os sarcomas, 47% eram CCS, 22% LMS, 16% SEE e 14% ADS. 62% ocorreram a partir de 60 anos de idade, 77% eram pós-menopausa, 61% dos casos tinham IMC até 29,9 e 71% apresentavam alguma comorbidade. Na análise de regressão logística univariada, faixas etárias maiores, pacientes menopausadas e com comorbidades apresentaram maior risco de tipos histológicos com pior desfecho (LMS e CCS). Na regressão multivariada, apenas o estado menopausal manteve significância com odds ratio de 5,45 (p<0,001) para apresentar LMS ou CCS. Os sarcomas estavam em estádio I em 44% dos casos e 22% em estádio IV. A cirurgia foi o primeiro tratamento em 78% com 79% realizando terapia adjuvante e 22% não operaram. A sobrevida global foi de 76% em 12 meses e 33% em cinco anos, melhores para SEE e ADS que para CCS e LMS (p=0,003). Resposta completa ocorreu em 55 casos e 20 recidivaram no seguimento, com SLD de 66%, melhor tempo para SEE e ADS (p=0,022). Ao final, 25% permaneceram vivas sem doença e 57% haviam morrido pela doença. Conclusões: Há uma tendência de aumento na taxa de sarcomas de útero no período 2001-2016. Mais da metade foram diagnosticados em estádios avançados. 22% não realizaram cirurgia. Na análise de regressão logística multivariada apenas a menopausa apresentou associação com tipos histológicos de pior prognóstico. A sobrevida global foi de 33% após cinco anos com SLD de 66%, melhores para SEE e ADS. Although aggressive, many are diagnosed in the early stages. Among its diversity, the main histological types are: carcinosarcoma (CCS), leiomyosarcoma (LMS), endometrial stromal sarcoma (ESS) and adenososarcoma (ADS). There is little information on these tumors in Brazil. Objectives: To evaluate the profile in the diagnosis, treatment and evolution of cases of uterine body sarcoma. Methodology: A retrospective cohort study was carried out with the analysis of medical records of the cases registered at the Women's Hospital. Dr. J. A. Pinotti, CAISM, Unicamp, in the period between 2001 and 2016, totaling 122 cases of sarcomas, considering CCS, LMS, SEE, ADS and others. Clinical information, neoplasms, treatment and follow-up were considered. Statistical analysis was performed using Chi-Square or Fisher tests, trend evaluation of the number of cases over time by the Chi-Square trend test, polyvalent logistic regression analysis, univariate and multivariate, with a stepwise selection of variables to evaluate the relationship between the factors associated with the histological types of sarcomas, assembled Kaplan-Meier survival curves and analyzed by the log-rank test. It was considered p <0.05 for significance.Results: The main sign or symptom that led to the diagnosis of the sarcomas evaluated was abnormal uterine bleeding in 77%. The main forms of diagnosis were uterine curettage and surgery, 35% and 34%, respectively. A trend of significant increase in sarcomas was observed with +1.2 cases (p = 0.044) every 2 years. The proportion of carcinomas / sarcomas remained relatively stable. Among the sarcomas, 47% were CCS, 22% LMS, 16% SEE and 14% ADS. 62% occurred after 60 years of age, 77% were postmenopausal, 61% had BMI up to 29.9 and 71% had some comorbidity. In the univariate logistic regression analysis, older age groups, menopause patients and comorbidities presented higher risk of histological types with worse outcome (LMS and CCS). In the multivariate regression, only the menopausal status remained significant with an odds ratio of 5.45 (p <0.001) to present LMS or CCS. Sarcomas were in stage I in 44% of cases and 22% in stage IV. Surgery was the first treatment in 78% with 79% performing adjuvant therapy and 22% did not operate. Overall survival was 76% at 12 months and 33% at 5 years, better for SEE and ADS than for CCS and LMS (p = 0.003). Complete response occurred in 55 cases and 20 relapsed at follow-up, with SLD of 66%, best time for ESA and ADS (p = 0.022). At the end, 25% remained alive without disease and 57% had died from the disease. Conclusions: There is a trend towards an increase in the rate of uterine sarcomas in the period 2001-2016. More than half were diagnosed in advanced stages. 22% did not undergo surgery. In the multivariate logistic regression analysis only menopause was associated with histological types of worse prognosis. Overall survival was 33% after five years with SLD of 66%, better for SEE and ADS.     Roasting process affects the profile of diterpenes in coffee

    There is no consensus in the literature regarding the decrease of kahweol and cafestol contents during coffee roasting, but it has been reported that these compounds can undergo dehydration under heat. Kahweol and cafestol were quantified in Arabica and Robusta coffees with different roasting degrees (2, 4, 6, 8 and 10 min at 230 °C). The structures of the diterpenes and the presence of derivative compounds were determined by liquid chromatography with UV–Vis and mass spectrometry detection. In the dark roast samples, dehydro derivatives were found. The roasting process influenced the level of diterpenes in both species of coffee, but the effect was dependent on the intensity of the process. Cafestol and kahweol were degraded (general losses from 60 to 75 % on a lipid basis) to dehydrocafestol and dehydrokahweol, respectively, after 8 min of process, which corresponds to the commercial roasting degree.     Low-sodium fish burgers : Sensory profile and drivers of liking

    The aims of this study were to determine the descriptive sensory profile and drivers of liking of eight samples of low-sodium Serra Spanish Mackerel (Scomberomorus brasiliensis) fish burgers. The sensory profile was determined by 12 trained assessors, and acceptance testing was performed using 243 consumers. Partial least-squares regression analysis was used to identify the drivers of liking and rejection of fish burgers (i.e., “Mackerel burger”). The results of the sensory descriptive profile indicated that the attributes appearance, aroma, and flavor were stronger for the samples prepared from whole minced fish. Acceptance was also higher for formulations containing whole minced fish. The descriptive terms that drove consumer preference and determined the acceptance were fish shape, salty taste, umami taste, fish flavor, herb flavor, and residual artificial flavor, and the term fish shape positively affected the results.
