Efficient Fault Location and Characterization in Cable Networks with Known and Unknown Topologies
This thesis addresses the identification of defects in transmission lines, emphasizing the importance of cable network reliability in critical industries like telecommunications, automotive, and aerospace. Cable faults are classified into hard and soft faults, both of which can lead to significant safety risks and financial losses. Effective fault prediction and localization are essential.
We explore methods like time-domain reflectometry (TDR) and frequency-domain reflectometry (FDR) for fault location, noting their limitations in networks with parallel branches and unknown topology, particularly regarding soft faults.
The thesis introduces three innovative cable diagnosis methods:
1. A cost-effective approach for detecting multiple hard faults in harnesses, using low-cost hardware that detects faults at 1 Hz, outperforming traditional reflectometry techniques.
2. A distributed reflectometry method for bus-shaped networks, employing analytical modeling and an adjacency matrix to resolve reflections and locate faults accurately.
3. A novel TDR algorithm for networks with unknown topology, reconstructing the topology iteratively through reflection analysis using a complex mathematical model. This method significantly enhances processing speed and accuracy.
These methods address various industrial scenarios, ranging from low-risk environments to high-risk industries that require accurate soft fault location, establishing a foundation for future improvements in cable network diagnostics.:1 Introduction

Naturschutzarbeit in Sachsen
Die Broschüre behandelt sieben Themen aus der behördlichen und ehrenamtlichen Naturschutzarbeit. Ausführlich beschrieben werden der Zustand der Arten und Lebensraumtypen zur FFH-Richtlinie in Sachsen im Zeitraum von 2007 bis 2012 und die Naturschutzförderung seit dem Jahr 2015. Weitere Schwerpunkte sind die Wildnisgebiete und Naturwaldentwicklung sowie die Grundlagen und Auswahlkriterien zur Revitalisierung der Moore. Erfahrungsberichte ehrenamtlicher Naturschützer kommen aus den Altkreisen Löbau, Zittau, Weißwasser, Niesky und der Stadt Görlitz. Außerdem wird über das 20. Heulager im Bärensteiner Bielatal und über Änderungen bei den sächsischen Naturschutzgebieten berichtet.

Traces of the Sarvāstivādins in the Buddhist Monasteries of Kucha
The innovative approach to the study of Kuchean Buddhism presented in this book examines the material remains in order to identify the type of Buddhism practiced in the region, which left its traces in the rock- and surface monasteries. In addition to drawing upon as yet unpublished data from in situ fieldwork, this study also undertakes a detailed reading of the records and photographs of early 20th century international expeditions, and the artefacts which were taken from the caves and are now held in museums around the world.
The interdisciplinary collaboration of the two main authors has permitted the identification of four successive phases of the Kuchean Sarvāstivāda tradition based on the archaeological and art-historical analysis of the sites, districts, cave groups, cave types, layouts of the décor, the narrative contents and iconography therein, and the source texts. The results of this study are supported and further extended upon by the contributions of two guest scholars, the authors of appendices focusing respectively on the Vinaya texts unearthed in the Kucha region and the materials and techniques of the wall paintings.
This book focuses on a single phase of the Kuchean Sarvāstivāda tradition, which is herein referred to by the unbiased term ‘Tradition A’. This tradition can be clearly distinguished from other traditions present in the caves of Kucha. Part One offers a comprehensive catalogue of ‘Tradition A’ caves, while Part Two presents a typological analysis based on the ‘sense of place’ of each of the caves, as well a study of the features of their primary and secondary décor, before moving to the contextualization of the spatial context of these caves within the monasteries. Based on these data, ‘Tradition A’ emerges as an early phase of Kuchean Sarvāstivāda Buddhism that is closely related to the Shisong lü, which is the Sarvāstivāda-vinaya preserved in Chinese. This phase was then followed by ‘Tradition B’, another Sarvāstivāda phase which became the dominant tradition in the region after ‘Tradition A’, which contains more elements related to the Mūlasarvāstivāda-vinaya.:Foreword by the Editors

An automated single-molecule FRET platform for high-content, multiwell plate screening of biomolecular conformations and dynamics
Single-molecule FRET (smFRET) has become a versatile tool for probing the structure and functional dynamics of biomolecular systems, and is extensively used to address questions ranging from biomolecular folding to drug discovery. Confocal smFRET measurements are amongst the widely used smFRET assays and are typically performed in a single-well format. Thus, sampling of many experimental parameters is laborious and time consuming. To address this challenge, we extend here the capabilities of confocal smFRET beyond single-well measurements by integrating a multiwell plate functionality to allow for continuous and automated smFRET measurements. We demonstrate the broad applicability of the multiwell plate assay towards DNA hairpin dynamics, protein folding, competitive and cooperative protein–DNA interactions, and drug-discovery, revealing insights that would be very difficult to achieve with conventional single-well format measurements. Nutlin-3a-aa: Improving the Bioactivity of a p53/MDM2 Interaction Inhibitor by Introducing a Solvent-Exposed Methylene Group
Nutlin-3a is a reversible inhibitor of the p53/MDM2 interaction. We have synthesized the derivative Nutlin-3a-aa bearing an additional exocyclic methylene group in the piperazinone moiety. Nutlin-3a-aa is more active than Nutlin-3a against purified wild-type MDM2, and is more effective at increasing p53 levels and releasing transcription of p53 target genes from MDM2-induced repression. X-ray analysis of wild-type MDM2-bound Nutlin-3a-aa indicated that the orientation of its modified piperazinone ring was altered in comparison to the piperazinone ring of MDM2-bound Nutlin-3a, with the exocyclic methylene group of Nutlin-3a-aa pointing away from the protein surface. Our data point to the introduction of exocyclic methylene groups as a useful approach by which to tailor the conformation of bioactive molecules for improved biological activity
