Cloning and Expressing of Interleukine 2 in Amniotic Membrane-Derived Mesenchymal Stem Cells, as a Potent Feeder Layer
The application of mesenchymal stem cells (MSCs) is rapidly expanding due to their unique
properties in cell therapy, especially as the feeder layer in the ex-vivo expansion of immune
cells. Also, Interleukin 2 (IL-2) is an essential human cytokine in the expansion of
hematopoietic precursors and progenitors, i.e., NK cells and T cells, while there is no
endogenous expression of IL-2 in MSCs. This study aimed to examine the potency of amniotic
membrane (AM)-MSCs as the IL-2 secretory cells. IL-2-containing pCMV3-C-GFPspark
shuttle vector was transformed in E.coli DH5-alpha. After cloning, the plasmid DNA was
extracted and transfected in isolated AM-MSCs, by lipofectamine-2000. Then, the RNA and
protein expression levels of exogenous IL-2 were evaluated 3 to 15 days after transfection,
using ELISA and qRT-PCR. Fluorescent microscopy and flowcytometry assays were used for
evaluating the GFP-positivity of transfected AM-MSCs, as IL-2 expression control. There was a
significant increase in RNA expression of exogenous IL-2 in transfected AM-MSCs in 3 to 15
days after transfection. (p<0.001) Also, IL-2 concentration released in the medium was
increased in 3rd day after transfection (611 pg/ml). However, the RNA and protein expression
of IL-2 was reduced through passing the time. The results show AM-MSC is a suitable host for
the expression and secretion of IL-2 as a critical cytokine in the ex-vivo expansion of
hematopoietic precursors and progenitors, i.e., NK cells and T cells. Also, the survival time of
IL-2 expression in AM-MSCs was long enough for use as a feeder layer
Association of COVID-19 infection and juvenile stroke: A case series
Ischemic stroke has been increasingly reported as a consequence of COVID-19 infection. However, the
underlying etiology is not well determined. The objective of this study is to discuss association of
juvenile stroke with COVID-19 infection. We analyzed 5 COVID-19 positive and stroke patients
with a mean age of 41.2 years-old. Three patients developed large vessel occlusion, one small vessel
occlusion and one PRES with superimposed lobar ICH, respectively. The mean initial NIHSS of our
patients was 11.6. Except the one with massive cerebellar infarct, a desirable outcome occurred with
a mean mRS 2.6 at discharge. The mean ESR and CRP level was elevated to 30.4 ml and 32 mg/dl.
The severity of COVID-19 infection was considered mainly as mild. COVID-19 infection has the

The effect of yoga on the delivery and neonatal outcomes in nulliparous pregnant women in Iran: a clinical trial study
Background: Yoga can reduce the risk of preterm delivery, cesarean section (CS), and fetal death. The aim of the
present study was to investigate the effects of Yoga on pregnancy, delivery, and neonatal outcomes.
Methods: This was a clinical trial study and using the random sampling without replacement 70 pregnant women
entered Hatha Yoga and control groups according to the color of the ball they took from a bag containing two
balls (blue or red). The data collection tool was a questionnaire pregnancy, delivery, and neonatal outcomes. The
intervention in this study included pregnancy Hatha Yoga exercises that first session of pregnancy Yoga started
from the 26th week and samples attended the last session in the 37th week. They exercised Yoga twice a week
(each session lasting 75 min) in a Yoga specialized sports club. The control group received the routine prenatal care
that all pregnant women receive.
Results: The results showed that yoga reduced the induction of labor, the episiotomy rupture, duration of labor,
also had a significant effect on normal birth weight and delivery at the appropriate gestational age. There were
significant differences between the first and second Apgar scores of the infants.
Conclusion: The results of the present study showed that Yoga can improve the outcomes of pregnancy and
childbirth. They can be used as part of the care protocol along with childbirth preparation classes to reduce the
complications of pregnancy and childbirth
Effect of Different Disinfecting Agents on Dental Impressions Contaminated with Candida albicans
Detection and Molecular Characterization of Potentially Pathogenic Free-Living Amoebae from Recreational and Public Soils in Mazandaran, Northern Iran
Background: Free-living amoeba (FLA) belonging to Acanthamoeba spp., Naegleria, and Balamuthia
mandrillaris are the soil-born protozoa. This study aimed to survey the occurrence of
FLA, including Acanthamoeba spp., B. mandrillaris, Vermamoeba spp., and Naegleria spp., in soil
samples collected from various districts of Mazandaran Province (Northern Iran) from July to
December 2018.
Methods: Overall, 118 soil samples from the recreational and public places were surveyed for
the existence of Acanthamoeba spp., Vermamoeba, Naegleria, and B. mandrillaris using both morphological
key and molecular tools with genus-specific primers of JDP1, NA, ITS1, and Bal,
respectively. To verify the taxonomic status of isolated amoeba, the phylogenetic tree was
made based on sequences of 18S rRNA by MEGA (5.05) software with the maximum likelihood
model.
Results: Overall, 61/118 samples (51.6%) were contaminated with FLA, and based on the
sequencing data, 29 isolates were successfully sequenced. Among the samples, all isolated
Acanthamoeba (52.4%) belonged to the T4 genotype with amplification of the DF3 region (18S
rRNA gene). Internal transcribed spacer (ITS) sequencing revealed the presence of one strain
of Naegleria americana. Twenty-eight V. vermiformis were also confirmed based on Nuclear SSU
rDNA. Morphological survey and PCR assay did not show any positive samples for B. mandrillaris.
Conclusion: The present study indicates the occurrence of FLA in soil sources of the recreational
and public places in Mazandaran province that it can be a severe risk to human health.
Thus, more studies are expected to survey the infection source in patients with FLA-related
diseases
