In vitro comparison of two photostimulable phosphor plate systems for early detection of occlusal dentin caries with and without a sharpening filter
Electrospun poly(L-lactide) nanofibers coated with mineral trioxide aggregate enhance odontogenic differentiation of dental pulp stem cells
A combination of bioceramics and nanofibrous scaffolds holds promising potential for
inducing of mineralization in connective tissues. The aim of the present study was to
investigate the attachment, proliferation and odontogenic differentiation of dental pulp
stem cells (DPSC) on poly(L-lactide) (PLLA) nanofibers coated with mineral trioxide
aggregate (MTA). Polymeric scaffolds were fabricated via the electrospinning method
and their surface was coated with MTA. DPSC were isolated from dental pulp and their
biological behavior was evaluated on scaffolds and the control group using MTT assay.
Alkaline phosphatase (ALP) activity, biomineralization and the expression of odontogenic
genes were analyzed during odontogenic differentiation. Isolated DPSC showed
spindle-shaped morphology with multi-lineage differentiation potential and were positive
for CD73, CD90 and CD105.MTA-coated PLLA (PLLA/MTA) exhibited nanofibrous
structure with average fiber diameter of 756 ± 157 nm and interconnected pores and
also suitable mechanical properties. Similar to MTA, these scaffolds were shown to be
biocompatible and to support the attachment and proliferation of DPSC. ALP activity
transiently peaked on day 14 and was significantly higher in PLLA/MTA scaffolds than
in the control groups. In addition, increasing biomineralization was observed in all groups
with a higher amount in PLLA/MTA. Odontogenic-related genes, DSPP and collagen
type I showed a higher expression in PLLA/MTA on days 21 and 14, respectively. Taken
together, MTA/PLLA electrospun nanofibers enhanced the odontogenic differentiation
of DPSC and showed the desired characteristics of a pulp capping material
Effect of Washing and Cooking on Nitrate Content of Potatoes (cv. Diamant) and Implications for Mitigating Human Health Risk in Iran
The nitrite and nitrate in foodstuff and their associated harmful effects are of great
concern for human health. More than 80% of human nitrate intake is attributed to the
consumption of vegetables and fruits, and for this reason, more attention should be paid
to the consumption of such foodstuffs and reduction of nitrate intake. Therefore, this
study aimed to determine the variation of nitrate and nitrite content in potato during
home preparation, process, and cooking methods, and assessed their health risk in
different age groups for the first time in Iran. The nitrite content in all of the samples
was lower than the apparatus detection limit so results are reported for nitrate. A total of
471 tubers of potato cultivar Diamant were assessed. The nitrate content was measured
based on the tuber weight and also in different parts of potato. The impact of different
washing and baking methods on nitrate contents were also assessed. Ion chromatography was employed to determine the nitrate content. The US Environmental Protection
Agency (EPA) method covering the determination of nitrate in food was used to assess
the health risk associated with potato consumption. The results showed that the average
nitrate content was 123.26 mg/kg which was lower than the safe limit of nitrate. The
results also showed that the total amount of nitrate in potato peel was 35% higher than
in its central parts. Different washing methods could reduce the nitrate amount by 7.79
to 14.73%. Boiling potato could significantly reduce the amount of nitrate by 59.7%,
while frying increased it by 52%. There were significant differences among different
potato cooking methods in terms of changes in the nitrate content (P ˂ 0.001). This
means that exposure in all age groups was less than the standard limits and potato
intake was considered safe to all age groups. As there were significant changes in the
nitrate content, it was concluded that washing and cooking methods, except frying,
were suitable methods to reduce nitrate intake

The gene expression profile and DNA methylation pattern of CDH1 and DNMT1 genes in acute promyelocytic leukemia (APL)
Background:DNA methylation is an epigenetic modification that has the ability to alter gene expression and
function. These epigenetic changes have been associated with the development of cancer. Previous research has
found that DNA methylation patterns can predict disease prognosis for patients with Acute Promyelocytic
Leukemia (APL). The role of DNMT1 and CDH1 in regulating the extension of cells are studied in this study.
Methods: DNA was extracted from peripheral blood samples of APL patients and treated with bisulfite.
DNMT1 and CDH1 gene promoter methylation was subsequently analyzed using methylation-specific PCR
(MSP). Real-time PCR was used to measure the expression level of DNMT1 and CDH1 genes.
Results: Partial methylation of the CDH1 gene promoter was detected in 20% of APL patients and an
unmethylated status was detected in 80% of patient samples. Additionally, an unmethylated status in the DNMT1
gene promoter was detected in 100% of APL patient samples.
Conclusions: Our study found the CDH1 gene promoter to be unmethylated in almost all APL patients, while
the DNMT1 promoter was unmethylated in all APL patients. Furthermore, we observed an increase in both
CDH1 and DNMT1 gene expression in APL patients compared to healthy controls. These findings suggest that
DNMT1 may not have a specific role in inhibiting CDH1 gene expression in APL. Applying higher resolution
techniques would help to better uncover the DNA methylation patterns in patients with APL. Further research
is required to determine the role of DNA methylation and CDH1 and DNMT1 gene expression in APL
