Isolasi Penghambat Aktivitas Enzim Pengubah Angiotensin Dari Protein Susu
ABSTRACT. Three kinds of samles (whey protein containing casenoglycopeptide, whey protein removedcasenoglycopeptide and cheese whey powder digested with 7 kinds of proteases at 370C for 24 hr (trypsin, protenase-K, actinase-E, thermolysin, and papain) or 250C (pepsin and chymotrypsin). Strong inhibotory activity against the angiotensin converting enzyime (ACE, EC 3.4.15.1) was generated in all samples by 5 proteases digestion (pepsin, chymotrypsin, protinase-K, thermolysin and papain). In whey protein removed caseinoglycopeptide digestion by thermolysin induced the highest activity (95,25%). In cheese whey powder, the highest activity was derived by thermolysin (98.25%). On the other hand, week ACE inhibitory activity were derived by trypsin and actinase-E digestion. As no remarkable differences in inhibitory activity were observed between whey protein containingcasenoglycopeptideand whey protein removedcasenoglycopeptide samples, the bioactive peptides are considered to come mainly not fromcasenoglycopeptide but from cheese whey powder components
Karakteristik Semen dan Komposisi Kimia Plasma Seminalis Sapi Aceh yang Dipelihara di BIBD Saree Aceh Besar
ABSTRAK. Penelitian ini bertujuan untuk menentukan karakteristik dan komposisi plasma semen sapi aceh. Sampel semen dikoleksi dari 2 ekor sapi aceh jantan sehat berumur 3,0-3,5 tahun menggunakan vagina buatan. Koleksi semen dilakukan satu kali dalam seminggu selama enam minggu. Evaluasi karakteristik semen meliputi volume, warna, konsistensi, pH, konsentrasi spermatozoa, motilitas, viabilitas, abnormalitas spermatozoa dan komposisi kimia plasma semen. Data kualitas dan komposisi kimia plasma semen yang diperoleh dianalisa secara diskriptif. Hasil penelitian menunjukkan semen segar sapi aceh memiliki volume 3,82 0,47 ml, warna krem keputihan, konsistensi kental, pH 6,84 0,17, konsentrasi 1194 4,80 106sp/ml, persentase motilitas 77,28 3,17 % dan abnormalitas 5,98 1,77%. Plasma seminalis semen segar sapi aceh mengandung protein total sebanyak 1,51 0,21 g/100ml, Kalium 75,80 22,63 mg/100 ml dan Magnesium 32,30 17,64 mg/100 ml lebih tinggi, tetapi Natrium 170,00 81,60 mg/100 ml; kalsium 40,00 12,76 mg/100 ml, Fosfor 16,70 6,17 mg/100 ml, dan Mangan 7,08 2,48 mg/100 ml lebih rendah dibandingkan dengan sapi potong. Secara umum kuantitas dan komposisi plasma seminalis semen sapi aceh yang dihasilkan cukup baik, dan memenuhi syarat untuk diproses sebagai semen beku.(The characteristics of semen and chemical composition of the seminal plasma of aceh cattle maintaned in BIBD Saree Aceh Besar)ABSTRACT. This study aim were to determine characteristics and composition of semen plasma Aceh cattle. Semen samples were collected from two healthy Aceh bulls that aged from 3.0 to 3.5 year. Samples were yielded by using an artificial vagina. Samples collecting were conducted once a week for six consecutive weeks. The evaluation of semen characteristics includes volume, color, consistency, pH, sperm concentration, motility, viability, abnormal spermatozoa and seminal plasma chemical composition. Data of semen quality and chemical composition of seminal plasma were analyzed descriptively. The evaluation showed that Aceh Cattle semen had 3.82 0.47 ml volume, beige whitish color, thick consistency, 6.84 0.17 pH, 1194 4.80106 sp/ml sperm concentration, 77.28 3.17% motility and 5.98 1.77% abnormal spermatozoa. Plasma seminalis fresh semen of Aceh Cattle contained 1,51 0,21 g/100ml total protein; 75,80 22,63 mg/100ml potassium and 32,30 17,64 mg/100ml magnesium higher than the heifer, but 170,00 81,60 mg/100ml sodium; 40,00 12,76 mg/100ml calcium, 16,70 6,17 mg/100ml fosfor, and 7,08 2,48 mg/100ml mangan lower. It was concluded that quality and composition of seminal plasma Aceh cattle semen could be made as frozen semen
Host And Temperature Preferensce, Male Occurrence And Morfomentrics Offungivorous Nematode, Aphelenchus Avenae Isolates From Japan
ABSTRACT.Host and temperature preference, male accurrence, and morfomentrics of 39 isolates of Apehelenchus avenae were investigated. Of the 39 isolates, 33 were from 7 sistrict of Kyushu, 3 preference were investigated on 4 species of fungi; Botrytis cinerea, Rhizoctonia solani AG-4, Fusarium oxysporum f. sp. Melonis, and Pyitum aphanidermatum growing on 1/5 strength PDA medium. The nematodes were incubated on respective fungus mat for 30 days at 25 and 30 OC host fungi and temperatures gave rise to various levels of reproduction and male accurance. The isolates were divided into 5 groups based on their reproductivity on their host fungi at the 2 temperature regimes. Most of the isolates of A. avenae progated themselves at 25 0C on B. cinera and R. solani AG-4, was 60.850 given by Nagasaki isolates (NA3). The maximum multiplication 48.420 at 0C accured on R. solani byu Kagoshima isiolate (KA3). The occurrence of males was very low in all isolates and it occurred only at 30 0C. De Mans value of body dimension were measured and showed no significant variation among the isolates. Besides, There were no realtions between groupings by host preference and by body dimension
Pengaruh Penambahan Molases Terhadap Komposisi Kimia Silase Beberapa Jenis Rumput Yang Ditanam Bercampur Dengan Stylos
ABSTRACT. This study was conducted in UPTD (Unit Pelaksana Tugas Dinas) Saree Aceh Besar about 4 weeks. The matter of this study are 4 kinds of grass there are rumput benggala (panicum maximum) rumput BD (Brachiaria decumbent). The grass that use in this study ditanam mixe with stylosanthes guyanesis legume, while the additive in silage is molasses. The study was designed into Completely Randomized Design (CDR) Factorial 4x3. There were 3 replications. First factor was R1 (Rumput Benggala + Stylosanthes guyanesis), R2 (Rumput lampung+Stylosanthes guyanesis), R3 (Rumput BD + Stylosanthes guyanesis), R4 (Rumput Star grass +Stylosanthes guyanesis) and the second factor were 3 level there were M1 (0%0,M2(3%) dan M3 (5%). Invetigated variabes included pH, Dry matter, crude protein, crude fiber, ash and water containet..The data were analyzed by an analysis of variance and as a significant effect was detected, the analysis was contained by Uji Beda Nyata Terkecil (BNT), Steel and Torrie (1991). The result of this study showed that the different of several grass have hight significantly (P,0.010 of dry matter, crude protein, ash and water containing but have no significantly on pH and crude fiber silage. The diferent among molasses have significantly on pH, crude protein and crude fiber silage. From BNT test showed that molasses increased dry matter and crude protein of silage compared to no molasses added. However, th used of molasses decreased crude fiber and water contain compared to no molasses added. The quality of silage found at level of 5.5% molasses especially on star grass (R4M3). The study concluded that using different level molasses es on several grass producted better silage than without molasses
Performan Sapi Sumba Ongole (SO) yang Diberi Jerami Padi Amoniasi dan Konsentrat yang Disuplementasi dengan Tepung Daun Waru (Hibiscus Tiliaceus)
ABSTRAK. Tujuan penelitian adalah untuk mengetahui interaksi antara penambahan tepung daun waru (Hibiscus tiliaceus) (TDW) dan imbangan bahan kering (BK) jerami padi amoniasi (JPA) dan konsentrat terhadap konsumsi BK, kecernaan bahan organik (KBO) dan performan sapi Sumba Ongole (SO). Sebanyak 18 ekor sapi SO jantan dengan bobot awal 218,67 Kg 17,62. Pola faktorial 2 x 3 yang dirancang menurut Rancangan Acak Lengkap . Faktor pertama adalah imbangan BK JPA dan konsentrat masing-masing 35 : 65 (I1) dan 30 : 70 (I2). Konsentrat disuplementasi dengan TDW dengan level (ppm) 0% (W1), 0,24% (W2), dan 0,48% (W3)sebagai faktor kedua. Konsumsi BK tiap sapi adalah 3,3% dari bobot hidup. Penambahan tepung daun Waru (Hibiscus tiliaceus) dan imbangan BK JPA dan konsentrat maupun interaksinya tidak berpengaruh nyata (P0,05) terhadap konsumsi BK, KBO, performan sapi SO. KBO cenderung tinggi pada I1W2 dan performa cenderung baik level W3 baik pada I1 (1.27 kg 0.28 dan`17.21% 0.11) maupun I2 (1.26 kg 0.08 dan 19.87% 0.03). Penambahan tepung daun waru pada konsentrat tidak direkomendasikan untuk memperbaiki KBO maupun performan sapi SO.(Performances of sumba ongole cattle fed ammoniated rice straw and concentrate supplemented with waru leaf meal (Hibiscus tiliaceus))ABSTRACT. The aim of this research was to find the interaction between supplementation of Hibiscus tilliaceus leaf meal (HLM) and dry matter (DM) ratio of ammoniated rice straw (ARC) and concentrate on DM intake (DMI), organic matter digestibility (OMD) and Sumba Ongole (SO) cattle performances. Eighteen of SO male cattle with the average of 21.67 Kg 17.62 early body weight were used in this research. Completely Randomized Design with factorial pattern which consists of two factors (2 x 3) was applied. Those factors were DM ratio of ARC and concentrates of 35: 65 (I1) and 30: 70 (I2); and the concentrates that supplemented with HLM level (ppm) of 0% (W1), 0.24% (W2), and 0.48% (W3). DMI of each cattle was 3.3% of body weigh. HLM supplementation and ARC and concentrates DM ratio as well as their interaction were not significantly effected (P 0.05) on OMD, and SO cattle performances. OMD tended to increase at I1W2 and performances tended to be better at W3 both I1 (1.27 kg 0.28 and 17.21% 0.11) and I2 (1.26 kg 0.08 dan 19.87% 0.03). HLB supplementation could not be recommended to improve OMD and SO cattle performances
Penggunaan Kulit Nanas Fermentasi dalam Ransum yang Mengandung Gulma Berkhasiat Obat Terhadap Konsumsi Nutrient Ayam Broiler
ABSTRAK. Penelitian ini bertujuan untuk mengetahui penggunaan kulit nanas fermentasi ke dalam ransum yang mengandung gulma berkhasiat obat terhadap konsumsi nutrient ayam broiler. Penelitian ini menggunakan DOC jantan strain cob umur 2 hari sebanyak 200 ekor yang dipelihara selama 42 hari. Penelitian menggunakan Rancangan Acak Lengkap (RAL) dengan 5 perlakuan dan 5 ulangan. P0 = 0% tepung kulit nanas fermentasi (TKNF) dalam ransum mengandung 0% gulma obat(GO), (kontrol positif), P1 = 0% TKNF dalam ransum mengandung 2 % gulma obat (GO),(kontrol negatif), P2 = 7.5% TKNF dalam ransum mengandung 2 % gulma berkhasiat obat, P3 = 15% TKNF dalam ransum mengandung 2 % gulma berkhasiat obat, P4 = 22.5% TKNF dalam ransum mengandung 2 % gulma berkhasiat obat. Parameter yang diamati kandungan gizi kulit nanas sebelum dan sesudah fermentasi dan konsumsi nutrient. Pengaruh yang nyata terhadap parameter yang diamati dilanjutkan dengan Uji Jarak Berganda Duncan, Data yang diperoleh dari setiap parameter dianalisis ragam (ANOVA) menggunakan bantuan software SPSS 16.0. Dari hasil penelitian disimpulkan bahwa penggunaan kulit nanas yang fermentasi dengan yoghurt dalam ransum yang mengandung gulma berkhasiat obat pada taraf 15% memberikan hasil terbaik pada konsumsi nutrient ayam broiler.(Fermented pineapple peel supplementation with addition of medicinal weeds on nutrient intake consumption of broiler chicken)ABSTRACT. The aim of this study was to determine the effect of using fermented pineapple peel in the ration containing medicinal weeds on nutrient consumption and performance of broiler . Two hundred 2 days male broiler chicken cob strain were used in this study and kept for 42 days. Completely randomized design (CRD)was applied with 5 treatments and 5 replicates. Treatments consisted of P0 = 0% of fermented pineapple peels meal (FPPM) in ration containing 0% medicinal weed (MW), positive control). P1 = 0% of fermented pineapple peels meal (FPPM) in the ration containing 2 % medicinal weed (MW), negative control). P2 = 7.5% of FPPM in the ration containing 2 % medicinal weed (MW). P3 = 15% of FPPM in the ration containing 2 % medicinal weed (MW). P4 = 22,5% of FPPM in the ration containing 2 % medicinal weed (MW). The parameters measured was nutrien intake before and after fermented peel and consumption. The observed data were analyzed by statistical product and service solution (SPSS 16.0). Results showed that use of fermented pineapple peel meal up to 15% with addition medicinal weeds was the best on nutrient consumption of broiler chicken
Pengaruh Pemberian Tepung Ikan Lemuru (Sardenella Fimbriata) Terhadap Persentase Karkas Broiler
ABSTRACT. An experiment was conducted for five weeks to study the effect on feeding different levels of fish meal, Sarninella fibriata on carcass percentage of Cobb broiler strain. 120 day old Cobb chicks were randomly divided into eight expremiental groups, each consisting of 15 birds. Each groubs of 15 bird was divided into three replications of five birs, which then placed into 1x1 x 0.7 m cages at the rate of five birds per cage. Group one, two, three and four were fed 1 %, 4%, 7% and 10% of fish meal, Sardinella fibriata. Groub five, six, seven and eight were maintained with feed and water always available. The carcass percentage of teo birds of each five bird of each reflication were recorded at the end of the experiment. The average of carcass pecentage of groub one, two, three and four of fish meal. Sardinella fimbriata were 62, 67, 69, 68.40 and 69.37% resvectivaly. The average of carcass percentage of groub five, six, seven and eight of commercial fish meal were 63.22, 59.49, 64.90 and 70.22% respectively. There were significant different (P0.01) between groub six and groub eight of commercial fish meal, but there were no significant differences noted
Performa Ayam Broiler yang Mengkonsumsi Kulit Nanas yang Difermentasi dengan Yogurt dalam Ransum Mengandung Gulma Obat
ABSTRAK. Penelitian bertujuan untuk mengetahui performa ayam broiler yang mengkonsumsi kulit nanas yang difermentasi (KNF) dengan yogurt dalam ransum mengandung gulma obat (GO). Materi yang digunakan yaitu 200 ekor ayam broiler jantan umur 2 hari, ransum mengandung gulma obat dan kulit nanas yang difermentasi dengan yogurt. Rancangan yang digunakan adalah Rancangan Acak Lengkap (RAL) dengan 5 perlakuan dan 5 ulangan. Perlakuan yang diberikan yaitu T0 (ransum kontrol, ransum mengandung 0 % KNF dan 0 % GO), T1 (ransum mengandung 0 % KNF dan 2 % GO), T2 (ransum mengandung 7,5 % KNF dan 2 % GO), T3 (ransum mengandung 15 % KNF dan 2 % GO), dan T4 (ransum mengandung 22,5 % KNF dan 2 % GO). Peubah yang diamati adalah konsumsi ransum, pertambahan bobot badan (PBB), bobot akhir dan konversi ransum. Data dianalisis menggunakan ANOVA dan Uji kontras ortogonal. Hasil penelitian menunjukkan bahwa pemberian ransum yang mengandung KNF berpengaruh nyata (P0.05) terhadap konsumsi ransum, PBB, bobot akhir, dan konversi ransum. Konsumsi dan konversi ransum meningkat dengan peningkatan pemberian KNF sedangkan bobot badan menurun. Disimpulkan bahwa pemberian tepung kulit nanas setelah difermentasi dengan yogurt sebanyak 22,5 % di dalam ransum mengandung campuran gulma obat dapat menurunkan performa ayam broiler.(Broiler chicken performance fed fermented pineapple peel by yogurt in diet containing medicinal weed)ABSTRACT. Research aimed to determine broiler performance fed different level of yoghurt - fermented pineapple peel (FPPM) in the diet containing medicinal weed. Two hundred 2 day - old male broiler chicken, diet containing medicinal weed (MW) and yoghurt - fermented pineapple peel used in this study. The experimental design was a completely randomized design with 5 treatments and 5 replications. The treatments were T0 (control, diet containing 0% FPPM and 0% MW), T1 (diet containing 0% FPPM and 2% MW), T2 (diet containing 7.5% FPPM and 2% MW), T3 (diet containing 15% FPPM and 2% MW), and T4 (diet containing 22.5% FPPM and 2% MW). The variables were feed consumption, body weight gain, live weight and feed conversion ratio. Data were analyzed by ANOVA and Contrast orthogonal Test. The results showed that there was a significant effect (P0,05) of treatment on the feed consumption, body weight gain, live weight and feed conversion ratio. Feed consumption and feed conversion ratio increased with increasing level of FPPM in the diet, however, body weight decreased. It is concluded that feeding yoghurt - fermented pineapple peels up to 22,5 % in the broiler diet containing medicinal weed decreased broiler chicken performance
Dinamika Pertumbuhan Lactobacillus casei dan Karakteristik Susu Fermentasi Berdasarkan Suhu dan Lama Penyimpanan
ABSTRAK. Mutu susu fermentasi sangat dipengaruhi oleh bahan baku, proses pengolahan, proses fermentasi, dan penyimpanan. Perubahan nilai gizi dapat terjadi karena variasi dan fluktuasi suhu dan penyimpanan akan mempercepat kerusakan susu fermentasi. Penelitian bertujuan untuk mempelajari pengaruh suhu dan lama penyimpanan yang berbeda pada susu fermentasi terhadap pertumbuhan L. casei dan karakteristik susu fermentasi. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) pola faktorial yang terdiri dari 2 faktor yaitu faktor A (suhu) terdiri dari a1=4-10C, a2=10-16C. Faktor B (lama penyimpanan) terdiri b1=0 hari b2=30 hari, b3=60 hari, b4=90 hari dan dianalisis meliputi jumlah total bakteri L. casei, nilai derajat keasaman (pH), kadar asam laktat, sineresis, dan kadar alkohol. Hasil analisis menunjukkan bahwa perlakuan suhu dan lama penyimpanan berpengaruh sangat nyata (P0.01) terhadap total bakteri, sineresis, kadar alkohol, asam laktat, tetapi pada pH terjadi interaksi antara suhu dan penyimpanan. Disimpulkan bahwa suhu dan lama penyimpanan dapat mengakibatkan pertumbuhan L. casei tidak stabil dan terjadinya perubahan karakteristik fisik susu fermentasi L. casei.(Growth dynamics of Lactobacillus casei and characteristics of milk fermentation based on temperature and storage time)ABSTRACT. Fermented milk quality is strongly influenced by raw materials, processing, fermentation and storage. Changes in nutritional value may occur due to variations and fluctuations in temperature and storage will accelerate damage fermented milk. The research aimed to study the effect of temperature and different storage length of fermented milk on the growth of L. casei and characteristics of fermented milk. The study used completely randomized design (CRD) with factorial pattern which are consisted of two factors. Factor A (storage temperature) they were a1=4-10C, a2=10-16C. Factor B (storage time) they were b1=0 days, b2=30 days, b3=60 days, b4=90 days and analyzed include the total number of bacteria L. casei, the value of the degree of acidity (pH), lactic acid levels, syneresis, and alcohol content. The results showed that the temperature and storage time was highly significant (P0.01) effect the total bacteria, syneresis, alcohol level, lactic acid level, and there was interaction between temperature and storage time on pH. It can conclude that the temperature and storage time affect the growth of L. casei unstable and changes in the physical characteristics of L. casei fermented milk
Hubungan Antara Konsumsi Serat Kasar dan Lemak Kasar dengan Kadar Total Solid dan Lemak Susu Kambing Peranakan Ettawa
ABSTRAK. Penelitian ini bertujuan mengetahui hubungan antara konsumsi serat kasar dan lemak kasar dengan total solid dan lemak susu kambing Peranakan Ettawa. Penelitian ini menggunakan metode survei dan purposive sampling. Sampel yang digunakan pada penelitian ini adalah 35 ekor kambing Peranakan Ettawa dengan kriteria bulan laktasi 4-5, periode laktasi 2-3 dan frekuensi pemerahan 2 kali. Analisis data dilakukan menggunakan analisis regresi linier sederhana dan non linier kuadratik dan korelasi. Variabel bebas (variabel X) pada penelitian ini adalah konsumsi serat kasar dan lemak kasar, sedangkan variabel terikat (variabel Y) pada penelitian ini adalah total solid dan lemak susu. Hasil penelitian menunjukkan bahwa terdapat hubungan linier positif yang lemah antara konsumsi serat kasar dengan total solid dan lemak susu (r = 0,185 dan r = 0,160). Hubungan antara konsumsi lemak kasar dengan total solid dan lemak susu pada penelitian ini menunjukkan adanya hubungan kuadratik yang lemah (r = 0,316 dan r=0,320). Kesimpulan dari penelitian ini adalah terdapat hubungan yang lemah antara konsumsi serat kasar dan lemak kasar dengan total solid dan lemak susu. Semakin tinggi konsumsi serat kasar akan meningkatkan kandungan total solid dan lemak susu, tetapi konsumsi lemak kasar yang semakin tinggi tidak selalu meningkatkan kandungan total solid dan lemak susu.(Correlations between crude fiber and crude fat intake with total solid and milk fat contents in ettawa crossbreed goat)ABSTRACT. This study was aimed to determine the correlations between crude fiber and crude fat intake with total solid and milk fat contents. Samples used in this study were 35 Ettawa Crossbreed goats with lactation month of 4-5, lactation period of 2-3 and twice daily milking. Simple linear regression and non linear quadratic regression analysis were used in this study with crude fiber and crude fat intake as independent variables (X), meanwhile total solid and milk fat as dependent variables (Y). Simple Linear Regression showed that there was low positive correlation between crude fiber with total solid and milk fat contents (r = 0,185 and r = 0,160). Moreover, Non Linear Quadratic Regression analysis showed that there was low quadratic correlation between crude fat intake with total solid and milk fat contents (r=0,316 dan r = 0,320). In conclusion, there was alow correlation between crude fiber and crude fat intake with total solid and milk fat. High intake of crude fiber would increase total solid and milk fat contents, but high intake of crude fat would not increase total solid and milk fat contents
