    Evaluasi Produktivitas Padi Transgenik Rojolele yang Potensial Tahan Penggerek Batang Padi Kuning Scirpophaga incertulas Wlk

    Rice yellow stem borer (YSB, Scirpophaga incertulas Wlk) is one of the most damaging rice pests in Indonesia, especially in rice growing centers in West Java. Because there is no source of resistance genes in rice or its close relatives, resistant improvement by genetic engineering is one strategy to obtain resistant rice cultivar. Previously, the cryIB::cryIAa fusion gene from Bacillus thuringiensis has been introduced into the Rojolele variety to obtain 6 transgenic rice events resistant to YSB (X22, U10, W3, Y7, Q20, and P8) and has been tested for resistance in greenhouses. This study was aimed to evaluate the transgenic rice productivity carried out in a confined field trial in field station belonging to the Center for Rice Research in Sukamandi, Subang, West Java. The procedure for the field testing followed the 1998 Guidelines for Biosafety Testing of Agricultural Biotechnology Products: Plant Seri by the Ministry of Agriculture. The parameters observed were agronomic characters (plant height, number of tillers, and number of panicles) and yield components (number of filled grains per panicle, number of empty grains per panicle, panicle length, and weight of 1000 grains). The yield was also measured by dry grain yield. The results indicated that the performance were varied among the 6 events tested. Rice events X22, W3, Y7, and P8 showed agronomic characteristics and yield components resembled Rojolele varieties, and performed well under field condition.     Kajian Hubungan Allometrik dan Biomassa Lamun Thalassia hemprichii sebagai Bioindikator Lingkungan

    Seagrass has long been known to be very sensitive to environmental changes, especially caused by human activities (anthropogenic disturbance) and has been used as a bioindicator for environmental condition. This research aimed to study interaction of Thalassia hemprichii measured and derived variables (metrics), at two locations i.e., inhabited island (high anthropogenic location) and uninhabited island (low anthropogenic location). Confirmative approach was conducted using a developed conceptual model based on preliminary studies, the conceptual model was analyzed in multivariate context using data from field observation. The result showed that Thalassia hemprichii dominated and have higher above ground biomass value in inhabited area which indicated high nutrient input, meanwhile uninhabited area showed a higher below ground biomass value. Above ground of Thalassia hemprichii that have direct effect on its biomass and below ground metrics was leaf width, meanwhile leaf length of Thalassia hemprichii have indirect effect on its biomass and below ground metrics. The result also indicated some different metric covariations which were influenced by the degree of Thalassia hemprichii density. Leaf length, leaf width, density, and percent cover of Thalassia hemprichii potentially could be used as components in multimetric index and bioindicator for environmental quality.   Keywords: anthropogenic disturbance, path analysis, structural equation modeling, Thalassia hemprichi

    Pengaruh Suhu Tinggi terhadap Perkembangan Organ Reproduksi Jantan pada Cabai (Capsicum annum L.) cv. Tanjung-2

    The development of male reproductive organ in plants is seriously affected by the adverse abiotic stresses. In this study, we investigated pollen development, particularly at microsporogenesis and micro gametogenesis stages of chili pepper (Capsicum annuum) cv. Tanjung-2 upon exposure with high temperature (day/night = 36/33°C). For this objective, anther of different flower sizes ranging from <2.5 mm; 3-4.5 mm; 4.5-7 mm; to 7-11 mm from treated and non-treated plants were collected. The results revealed that the early microsporogenesis stage was highly sensitive to high temperature indicated by a low progression of cells into subsequent process in meiotic division. This result was followed by the inhibition of tetrad formation up to 2%. Consequently, plant produced smaller microscpores and pollens with thin exin that resulted in the decreased pollen viability to 90%. In conclusion, high temperature has negative impact on the development of male reproductive program in chili pepper, including microsporogenesis and micro gametogenesis. Extending approach should be allocated to overcome this problem so that such environmental stress would not decrease the productivity of chili pepper.     Formulasi Pasta Buah Merah (Pandanus conoideus Lamk) dan Tepung Ubi Jalar Mikmak (Ipomea batatas L.) untuk Produksi Saus Buah Merah Pedas

    Red fruit (Pandanus conoideus Lamk) and sweet potatoes (Ipomea batatas L) are the endemic plants that grow in the plains of Wamena, Papua. The processing of red fruit is very necessary to diversify food and increase its economic value. This study aims to determine the best formulation between red fruit paste, red chili pepper, and sweet potato flour to obtain spicy red fruit sauce product with a good nutritional quality and organoleptic properties. The formulation of red fruit paste and sweet potato flour was done with six levels i.e., A1 (100%:0%), A2 (95%:5%), A3 (85%:15%), A5 (80%:20%), and A6 (75%:25%). The quality of red spicy fruit sauce tested was physical quality (viscosity and total dissolved solids), chemical quality (and pH value), and organoleptic quality (hedonic test and hedonic quality with color, taste, flavor and consistency parameters). Based on the research, the best formulation was found in A2 treatment with the ratio of red fruit paste and sweet potato flour of 95%:5%. Formulation A2 has viscosity of 1733 cP, total dissolved solids of 21° Brix, pH value of 5.13, organoleptic value of color of 7.6, taste of 7.4, flavor of 7.4, and consistency of 6.6. The product of spicy red fruit sauce is in conformity with Standard National Indonesian (SNI 01-2976-2006) about chili sauce quality requirement on parameters of pH, viscosity, total dissolved solids, flavor, taste, color, and consistency.     Phytochemical Contents of Torbangun (Coleus amboinicus Lour) from Fractionation of Pressurized Liquid Extraction

    Coleus amboinicus (Lour) (torbangun) has been used traditionally as a breast milk stimulant, flavoring agent in many cuisines, and reported to possess many pharmacological values. The aim of this study was to explore the utility of the Pressurized Liquid Extraction (PLE) technique to identify the phytochemical contents of torbangun. In this study, total phenolic content and flavonoid in torbangun were determined using spectrophotometric method. The GC-MS analysis was used to identify the chemical constituents of torbangun, which was sequentially extracted with a wide range of solvent or solvent mixture from nonpolar-moderately polar and finally polar solvent. Total phenolic and flavonoid contents in the methanol extract of torbangun were 42.17±2.96 mg GAE/g and 11.20±0.58 mg QE/g, respectively. The identified chemical constituents in torbangun were sugar, hydrocarbon, ketone, terpenes, phenolics, fatty acid, fatty alcohol, steroids, alkaloids, and others, whereas the dominant constituents were phenolic compound, alkane, and sugar. In conclusion, this study demonstrated the effectiveness and rapid extraction of the PLE technique. Many valuable compounds in torbangun were extracted using gradual composition of solvent and were able to identify certain compounds in different polarities of solvents that are important in functional food preparation, pharmaceutical, and metabolomics research.     Peningkatan Pembungaan dan Hasil Biji Bawang Merah Varietas Bima melalui Vernalisasi dan Aplikasi GA3

    The low production of true seed of shallot (TSS) is mostly caused by a low flowering rate. The research was aimed to increase the flowering rate and seed yield in lowland (240 masl) and highlands (1250 masl) by vernalization and submersion of the seed bulbs in GA3 before planting. The research was arranged in a completely randomized block design with 2 factors and replicated four times. The first factor was vernalization, i.e. without and with vernalization at 10±2°C for 30 days, and the second factor was the submersion of the seed bulbs following vernalization in GA3 at the concentrations of 0, 100, and 200 ppm for 1 h. Parameters measured were time of 50% flowering (days after planting), percentage of flowering plants, number of umbel per plant, number of floret per umbel, fruitset, seed weight per umbel and per plant, as well as the percentage of seed germination. The result suggested that when production of TSS of var. Bima is to be carried out in the lowland then the seed bulbs had to be vernalized at 10±2°C for 30 days which will increase the flowering percentage and seed yield. If the production of TSS is to be carried out in the highland then vernalization at 10±2°C for 30 days will increase the flowering percentage and seed yield. However, if for any reason vernalization is impossible to proceed then the submersion of the seed bulbs in 100 ppm GA3 for 1 h was sufficient to increase the flowering percentage and seed yield.     Pengaruh Asam Humat terhadap Produktivitas dan Serapan Nitrogen pada Tanaman Kangkung Darat (Ipomoea reptans Poir.)

    An uncertain climate and infertile soils are factors that cause fluctuations in crop yields. In addition, improper application of fertilizers causes kangkong plants to not absorb nutrients optimally. The application of humic acid as a biostimulant can be used to improve soil structure and optimize the productivity of kangkong through the absorption of nutrients from fertilizers or elements around plants. This study aims to study the effect of application of humic acid on growth, productivity, and nitrogen uptake in kangkong (Ipomoea reptans). This research was carried out with completely randomized design (CRD) with a single factor including 7 treatments, namely control, application of humic acid through leaves at a dose of 5 mg.L-1, 20 mg.L-1, and mg.L-1 and through roots a dose of 5 mg.L-1, 20 mg.L-1, and 35 mg.L-1. Planting media used without the addition of basic fertilizers to see the most effective effect of the application of humic acid. The results showed that application of humic acid dose of 20 mg / L through leaves and roots caused an increase in plant height, stem diameter, number of leaves, biomass and nitrogen uptake of kangkong. The highest chlorophyll level of kangkong plants was produced in the treatment of humic acid dose of 35 mg.L-1 through leaves.  Based on the research it can be concluded that the application of humic acid through leaves and roots increase growth, productivity and nitrogen uptake in kangkong.     Daya Saing Bawang Merah di Provinsi Maluku dan Upaya Peningkatannya

    Shallot is one of horticultural commodities with a high demand. Majority of shallots in Maluku are bought from Surabaya and Makasar. However, the government continuously does horticultural development in the center of shallot production. The objective of this study was to analize the competitiveness and comparativeness of farming between shallots which are cultivated by local farmer and shallots which are imported. There were three locations of research i.e., Maluku Tenggara, Seram Bagian Barat, and Maluku Tengah. Primary data were collected by quessioner and used purposive sampling method. Primary data were analyzed using policy analysis matrix. The results showed that shallot farming in Maluku has comparative and competitive advantages compared with imported shallots. Agricultural government policy effectively increases local farmer insentive. However, it shows a decreasing trend while increasing productivity of shallot. Shallot farming having production higher than 8 ton/ha need output subsidy to give price guarantee. Effort to increase shallot competitiveness is conducted by increasing productivity and developing a farmer institution to make a sustainable agricultural development.   Keywords: shallot, competitiveness, competitive and comparativ

    The synthesis of various local wisdom values in the Tengger society in terms of utilization of ethnobotany resources as herbal products for health has long been existed in the scope of the Tengger locality and surrounding areas. The comparative advantage of Tengger ethnobotany certainly has potential that can be developed through certain strategies in order to be able to further contribute to the operationalization of the research, development, and application of science and technology (RISBANGRAPIPTEK) in the Field of Health and Medicine 2005–2025 towards a state of adequacy of raw materials and preparations of standardized herbal medicinal products and Indonesia branded for the Indonesian people. Therefore, the research question raised is the formulation of a strategy that must be immediately created and can be used as a reference for the activities of Tenggerese etnobotanical resource development to become a superior branded Indonesian biotechnology commercial product. The sampling method in this study was done by purposive sampling. The method of data collection was done by observation, interviews which included primary data and literature studies which were secondary data. The research method used was descriptive and analytic. The results showed that: 1) the perception map of the Tengger Society towards medicinal plants was classified as a moderate and the relationship of the characteristics of the Tengger Society did not have a close correlation to herbal medicines with a value of r less than 0.8. 2) Determinants that influence the influence of society on traditional medicine are age (X1), income (X3), and education (X4), while number of family dependents (X2) not significant effect.   Keywords: determinant factors, ethnobotany, perceptio

    Karakter Morfo-fisiologi dan Hasil Padi Gogo Toleran Kekeringan

    The objective of this study was to obtain varieties with high yield along with proper morpho-physiological characters under drought condition. Eleven varieties of upland rice i.e., Gilirang, Situ Patenggang, Kalimutu, Aek Sibundong, Batutegi, Towuti, Sunggal, Danau Gaung, Way Apo Buru, Danau Tempe, and Situ Bagendit were characterized based on morphology, physiology, and yield. Non factorial Complete Randomized Block Design was used with three times repetition. Parameters measured were morphological characters, physiological characters, and yield components. Morphological characters measured were plant height, number and area of leaf, shoot dry biomass, tiller number, and total root length. Physiological characters measured were plant growth rate, net assimilation rate, and relative growth rate. Yield components measured were number and length of panicle, number of seeds per panicle, 1000 seeds weight, weights of seed per panicle, weights of seed per plot, and weights of seed per hectare. The results showed that Danau Tempe variety had better characters against drought stress compared to the other varieties with higher yields supported by high plant growth rate, relative growth rate, chlorophyll content and leaf area with low accumulation of proline content and short root length.   Keywords: drought, morphological character, upland rice variety, yiel
