Effects of Applicator Type on the Safety and Efficacy of Commercial Minoxidil Products
It is important when developing effective and safe pharmaceutical products to select adequate active components and formulations as well as to design their containers and devices. Minoxidil was developed as a direct over-the-counter (OTC) drug in 1999 and has long been marketed only by one original company. Recently, many generic products have been marketed. However, the shape and type of the containers for these generic products differ from that of the original product, leading to a possibility that the amount of minoxidil applied by patients may differ. In the present study, the original and three typical generic products with 5% minoxidil external formulations were evaluated for their filling amounts, single application amounts, extent of distribution on application, ability to reach the scalp, and dripping properties. As a result, statistically significant differences were found in the filling amounts and ability to reach to the scalp between the products due to differences in the nozzle shape of the container. In addition, a large variation was observed in some of the generic products in terms of the drug liberation. These results indicate that it is important to suitably design and adequately evaluate the container for OTC products such as minoxidil formulations, which can exhibit toxicity from overuse, and that are easily purchased by consumers
Role of IGF-I in the intracellular signaling pathway of growth hormone-stimulated hepatocyte proliferation
Growth hormone (GH) is known to stimulate liver regeneration in 70% partially hepatectomized rats in vivo. Because complex involvement by multiple factors is unavoidable in in vivo experimental systems, clear interpretation becomes almost impossible when attempting to study the detailed mechanism of action. Therefore, we used a simpler, in vitro experimental system of primary cultured liver parenchymal cells to study the proliferation-stimulating effect of GH and its associated intracellular signal transduction mechanism from the GH receptor (GHR) to the nucleus. Many small molecular-targeted (specific signal transduction inhibitors) and large molecular-targeted agents (biological products and monoclonal antibodies) have now become available as pharmacological tools for research, and such agents enable a very detailed analysis of the mechanism underlying GH-induced stimulation of hepatic parenchymal cell proliferation. Consequently, we discovered that GH stimulates the proliferation of primary cultured mature rat liver parenchymal cells. We also found that the GH-induced stimulation of hepatic parenchymal cell proliferation is mediated by insulin-like growth factor-I (IGF-I), which is secreted by an autocrine mechanism.第一著者表記変更 Kurihara, Kazaki→Kurihara, Kazuk
The Effects of Gold Kiwifruit Intake Timing with or without Pericarp on Postprandial Blood Glucose Level
The purpose of this study was to examine how gold kiwifruit pericarp (pericarp is defined as the skin of the fruit) consumption and the timing thereof affect the postprandial blood glucose profile. The study was conducted on twelve healthy volunteers (six men and six women). According to our results, the simultaneous intake of gold kiwifruit with bread and the prior intake of gold kiwifruit evidently suppressed the postprandial blood glucose elevation compared with exclusive bread intake. There was no significant difference in postprandial blood glucose changes between the ingestion of gold kiwifruit pericarp and pulp and that of gold kiwifruit pulp only. The highest postprandial blood glucose elevation was suppressed by 27.6% and the area under the blood glucose elevation curve by 29.3%, even with the exclusive ingestion of gold kiwifruit pulp. We predicted that the ingestion of both the pericarp and pulp of gold kiwifruit would reduce the postprandial blood glucose elevation to a greater extent than that of gold kiwifruit pulp only; however, there was no significant difference between the two. These results indicate that gold kiwifruit consumption significantly suppresses the postprandial blood glucose elevation regardless of pericarp presence or absence and the timing of ingestion.Article number: 2103 This article is an open access article distributed under the terms and conditions of the Creative Commons Attribution (CC BY) license (https://creativecommons.org/licenses/by/4.0/)
