Get PDF本研究では、切迫流・早産の治療薬（±）- リトドリンを妊娠中期のマウスに尾静脈内投与した際の、母体内薬物動態及び母体から胎児への移行性を解析した。その結果、鏡像異性体間で投与直後より母体内血中濃度に差の生じることや、AUCinf、Vdαにも有意差があること等、薬物動態学的特性に違いのあることを明らかにした。一方で、胎児内リトドリン量の経時変化を解析したところ、母体血中とは異なるパターンを示すことが明らかとなった。これらの知見は、より低用量で治療可能な製剤、胎児の有害事象発現リスクを低下させる製剤、あるいはその両方を実現する新規リトドリン製剤の開発の一助になるものと期待される。In this study, we analyzed the intrapartum pharmacokinetics and maternal-to-fetal transfer of (±) - ritodrine, a drug for the treatment of impending miscarriage and preterm delivery, when administered intravenously by tail vein to mice in mid-pregnancy. The results showed that there were differences in pharmacokinetic properties, such as differences in maternal blood concentrations immediately after administration between enantiomers and significant differences in AUCinf and Vdα. Meanwhile, analysis of changes in fetal ritodrine levels over time revealed a pattern that differs from that in maternal blood.
