Gene knockdown in HaCaT cells by small interfering RNAs entrapped in grapefruit-derived extracellular vesicles using a microfluidic device
Small interfering RNAs (siRNAs) knockdown the expression of target genes by causing mRNA degradation and are a promising therapeutic modality. In clinical practice, lipid nanoparticles (LNPs) are used to deliver RNAs, such as siRNA and mRNA, into cells. However, these artificial nanoparticles are toxic and immunogenic. Thus, we focused on extracellular vesicles (EVs), natural drug delivery systems, for the delivery of nucleic acids. EVs deliver RNAs and proteins to specific tissues to regulate various physiological phenomena in vivo. Here, we propose a novel method for the preparation siRNAs encapsulated in EVs using a microfluidic device (MD). MDs can be used to generate nanoparticles, such as LNPs, by controlling flow rate to the device, but the loading of siRNAs into EVs using MDs has not been reported previously. In this study, we demonstrated a method for loading siRNAs into grapefruit-derived EVs (GEVs), which have gained attention in recent years for being plant-derived EVs developed using an MD. GEVs were collected from grapefruit juice using the one-step sucrose cushion method, and then GEVs-siRNA-GEVs were prepared using an MD device. The morphology of GEVs and siRNA-GEVs was observed using a cryogenic transmission electron microscope. Cellular uptake and intracellular trafficking of GEVs or siRNA-GEVs to human keratinocytes were evaluated by microscopy using HaCaT cells. The prepared siRNA-GEVs encapsulated 11% of siRNAs. Moreover, intracellular delivery of siRNA and gene suppression effects in HaCaT cells were achieved using these siRNA-GEVs. Discrimination of Sayamakaori and Yabukita Which Are Original Plant Source of Japanese Green Tea and Identification of Specific Compounds for the Former by Nuclear Magnetic Resonance Metabolomics Techniques Combined with Isolation by Chromatography Methods
Camellia sinensis leaves are one of the most economically important crops, and various tea cultivars are available in Japan. In the present study, we compared the constituents of Camellia sinensis var. sinensis cv. Yabukita (“Yabukita”), a standard tea cultivar, and those of C. sinensis var. sinensis cv. Sayamakaori (“Sayamakaori”), grown in Saitama Prefecture, using proton nuclear magnetic resonance (1H-NMR) metabolomics technology. Principal component analysis and hierarchical cluster analysis of 1H-NMR spectroscopic data of the MeOH extracts of C. sinensis leaves revealed differences in the metabolic profiles of each tea cultivar. Chromatographic separation based on 1H-NMR spectral data showed that kaempferol 3-O-β-D-glucopyranosyl-(1 → 3)-α-L-rhamnopyranosyl-(1 → 6)-β-D-glucopyranoside was a distinct component of “Sayamakaori”. 本稿は,オープンキャンパスにおける体験授業のあり方についての試案を呈示するものである。まず,オープンキャンパスの目的を,大学,来場者,在学生の3つの観点から記述し,整理した。次に,オープンキャンパスの目的に適った体験授業とするためには,体験授業で「何を」めざすのか,「どのように」伝えると効果的か,「どのような教員が」伝えるかについて論じた。最後に,教員が,体験授業というタスクの内に,広報ではなく教育機会としての意義を見出すことが,心的葛藤の緩和と授業の質の向上につながる可能性に言及した。研究ノート / Research Not
A Study on the Disparity between Urban-Rural Areas in Compulsory Education in China

Satisfaction Level of Participants with the Content Health Promotion Seminars Organized by Community Pharmacies in Chiba, Japan
Aim: This study aimed to identify the level of satisfaction and needs of the content of the “health promotion seminars” to support health promotion and improve healthy life expectancy provided by community pharmacies to community residents. Methods: An awareness survey was conducted among seminar participants. Anonymous survey questionnaires were distributed to the participants of the six sessions of the “health promotion seminars” held from December 2019 to February 2020 were recruited. Out of 136 participants, 116 responses were obtained (response rate: 85.3%; female: 114, male: 1, unanswered: 1) from the target population. Results: The level of satisfaction with the seminar was reported at 77.5% (n=90) for the first run and 68.5% (n=80) for the second run. Currently, the “Health Promotion Seminar” consists of an active (participation) lecture in the first lecture and a passive (classroom) lecture in the second lecture, conducted by pharmacists and dietitians. The level of understanding (84.5%) and active participation (86.2%) in the first course were higher than those in the second course (understanding: 80.2%, active participation: 80.2%). Participants reported anxiety issues in their everyday life stemming from the items “Forgot to take medicine” and “frequency of using the toilet increased night”. Conclusions: Health promotion seminars are viewed by community residents to be effective in disseminating health promotion information.
