    1970年ごろになると,計算を得意とするコンピュータが経理業務の合理化に導入され始めていった。これは企業にとって莫大な予算の投入であった。事務処理の自動化,合理化は次世代への必須アイテムであるといわれ多くの企業で改革が進められて来たのである。これもDX(デジタルトランスフォーメーション)の1つといえよう。また,同時に大きく変化したこととして,高校進学率があげられるが,約9割の中学生が高校へ進学したころでもある。また,商業高校学習指導要領にはタイプライターの学習項目があり,盛んに事務機械教育が進められていることがわかる。2020年に入り,GIGAスクール構想の効果もあり,日本では学校現場において一人一台タブレットPCの導入が進んでいる。本研究では,情報化時代に添った商業高校の簿記教育の指導法を調査し,教育効果の育成を検証する。研究ノート / Research Not

    Role of Hepatocyte Growth Regulators in Liver Regeneration

    We have studied whether growth factors, cytokines, hormones, neurotransmitters, and local hormones (autacoids) promote the proliferation of hepatic parenchymal cells (i.e., hepatocytes) using in vitro primary cultured hepatocytes. The indicators used for this purpose include changes in DNA synthesis activity, nuclear number, cell number, cell cycle, and gene expression. In addition, the intracellular signaling pathways from the plasma membrane receptors to the nucleus have been examined in detail for representative growth-promoting factors that have been found to promote DNA synthesis and cell proliferation of hepatocytes. In examining intracellular signaling pathways, the effects of specific inhibitors of presumed signaling factors involved have been pharmacologically confirmed, and the phosphorylation activities of the signaling factors (e.g., RTK, ERK, mTOR, and p70 S6K) have been evaluated. As a result, it has been found that there are many factors that promote the proliferation of hepatocytes (e.g., HGF, EGF, TGF-α, IL-1β, TNF-α, insulin, growth hormone (GH), prostaglandin (PG)), and serotonin (5-HT)), while there are very few factors (e.g., TGF-β1 and glucocorticoids) that inhibit the effects of growth-promoting factors. We have also found that 5-HT and GH promote the proliferation of hepatocytes via different autocrine factors (e.g., TGF-α and IGF-I, respectively).     Usefulness of direct intratumoral administration of doxorubicin hydrochloride with an electro-osmosis–assisted pump

    Patients receiving chemotherapy by intravenous (i.v.) or oral administration of anticancer drugs often experience side effects. In this study, an electro-osmotic flow (EO) pump was used for the direct administration of an anticancer drug with minimum side effects. Doxorubicin hydrochloride (DXR) was used as an anticancer drug, and its antitumor effect and toxicity were evaluated in comparison with i.v. administration. Balb/c femalemice were subcutaneously transplanted with a breast cancer cell line (4T1/Luc) stably expressing luciferase, and 20 μL of DXR solution (1.0mg/mL)was administered intratumorally (i.t.) at a slowrate (0.6 μL/min) using an EO pump or rapidly using a syringe. For comparison, 100 μL of DXR solution was injected through the tail vein at the same dose and a 5-times higher dose. A tumor growth inhibitory effect without significant weight loss was observed with direct i.t. administration of DXR using an EO pump. On the other hand, no suppressive tumor growth effect was observed with i.v. administration of DXR at the same dose. Although there was no significant difference in the suppression effect on tumor growth between i.t. administration with EO pump and syringe, the peripheral skin concentration of DXR were decreased after slow administration with EO pump compared with that after rapidly administration with a syringe. These results indicated that direct i.t. administration of DXR with lower dosing using an EO pump at slower administration rate may be useful for exhibiting antitumor effects and suppressing systemic side effects. In addition, the blood concentration and the peripheral skin concentration of DXR after administration at lower rate with EO pump were decreased compared with those after the rapidly administration with a syringe.p.8 This is an openaccess article distributed under the terms of the Creative Commons Attribution License (CC BY).     Collagen-Derived Dipeptide Pro-Hyp Enhanced ATDC5 Chondrocyte Differentiation under Hypoxic Conditions

    Chondrocytes are surrounded by a lower oxygen environment than other well-vascularized tissues with higher oxygenation levels. Prolyl-hydroxyproline (Pro-Hyp), one of the final collagen-derived peptides, has been previously reported to be involved in the early stages of chondrocyte differentiation. However, whether Pro-Hyp can alter chondrocyte differentiation under physiological hypoxic conditions is still unclear. This study aimed to investigate whether Pro-Hyp affects the differentiation of ATDC5 chondrogenic cells under hypoxic conditions. The addition of Pro-Hyp resulted in an approximately 18-fold increase in the glycosaminoglycan staining area compared to the control group under hypoxic conditions. Moreover, Pro-Hyp treatment significantly upregulated the expression of SOX9, Col2a1, Aggrecan, and MMP13 in chondrocytes cultured under hypoxic conditions. These results demonstrate that Pro-Hyp strongly promotes the early differentiation of chondrocytes under physiological hypoxic conditions. Therefore, Pro-Hyp, a bioactive peptide produced during collagen metabolism, may function as a remodeling factor or extracellular matrix remodeling signal that regulates chondrocyte differentiation in hypoxic cartilage.p.7 Article number: 4664 Licensee MDPI, Basel, Switzerland.     Potentiation of Anticancer Activity of G2/M Blockers by Mild Hyperthermia

    Background/Aim: Hyperthermia (HT), combined with chemotherapy, has been used to treat various types of cancer. This study aimed to investigate the HT-sensitivity of malignant and non-malignant cells, and then evaluate the combination effect of docetaxel (DTX) and a newly synthesized chromone derivative (compound A) with HT. Materials and Methods: The number of viable cells was determined using the MTT method. Cell cycle distribution was analyzed using a cell sorter, and DNA fragmentation pattern was detected using agarose gel electrophoresis. Results: Among 12 cultured cells, oral squamous cell carcinoma (OSCC), especially Ca9-22 cells, and myelogenous leukemia cells showed higher sensitivity to HT than lung carcinoma and glioblastoma cell lines, while normal oral cells were the most resistant. Cytotoxicity of DTX on Ca9-22 cells was maximum at 41-42°C and 45~60 min exposure to HT. DXT, compound A, and HT induced G2/M arrest of Ca-22 cells. Mild HT enhanced the DTX- and compound A-induced subG1 arrest, in a synergistic fashion.     Development of a depot formulation with an in situ non-lamellar liquid crystal-forming system with phospholipids

    Non-lamellar liquid crystal (NLLC) structures have gained increasing attention for the controlled release of entrapped drugs. In the present study, an in situ NLLC structure-forming depot formulation through contact with water was developed using a ternary mixture system of soya phosphatidyl choline (SPC), 1, 2-dioleoyl-sn-glycero-3-phosphoglycerol sodium salt (DOPG), and sorbitan trioleate (Span 85), and the long-term release of an entrapped model drug, leuprolide acetate (LA), was investigated using evaluation of in vitro release and in vivo blood concentration–time profiles. Polarized images and small angle X-ray scattering analysis were used to confirm the presence of NLLC structures by contacting the prepared formulation with water. In addition, LA release and blood concentration–time profiles were investigated using in vitro and in vivo experiments, respectively. In situ NLLC constructed formulations by contacting water were achieved using a ternary mixture of SPC, DOPG, and Span 85. In particular, negative curvature was increased with an increase in the amount of Span 85 in the formulation, and an Fd3m structure was obtained with a sustained release of LA. A maintained blood concentration of LA over 21 days was confirmed by subcutaneous (s.c.) administration of the formulation. No retained administered formulation at the injection site was confirmed 28 days after administration without any signs of irritation, inflammation, or other apparent toxicity confirmed by visual observation. This result may be helpful for the development of a lipid-based formulation of peptides and proteins with sustained drug release.This is an open-access article distributed under the terms of the Creative Commons Attribution License (CC BY).     Screening for crude drug extracts that regulate spike protein cleavage in SARS-CoV-2

    COVID-19の原因ウイルスであるSARS-CoV-2は2019年12月に発生以降,変異を繰り返し,現在に至るまで世界的な猛威を振るい続けている.本研究課題ではウイルス膜タンパク質であるSARS-CoV-2スパイクタンパク質(Sタンパク質)のプロセシングを抑制する生薬エキスの探索を行った。Sタンパク質に存在するFurin認識配列(FCS)の切断活性化を標的として、130種の生薬エキスを用いてin vitroによるスクリーニングやmRNA発現解析を実施し、ジャショウシやOstholeを始めとしたFCS開裂抑制作用を有する生薬や化合物を同定した。The spike (S) protein of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) contains a furin recognition site (FCS) for furin-like enzymes at the boundary of the S1/S2 subunits. The cleavage of the site is essential for S protein activation and virus entry. We screened the inhibitory effects of crude drugs on in vitro furin-like enzymatic activities and mRNA expression analysis.     A Study on the Trial of China's Long-Term Care Insurance System and Japan's Efforts in Healthcare and Welfare Export Strategy

    本研究は、主に以下の3つの研究成果をあげた。1つ目は、中国版介護保険制度の試行の過程において、日本と同様に「救貧型福祉」から「普遍型福祉」へと政策転換していることを確認し、その転換プロセスにおいて民間の役割、外国資本と海外のノウハウを活用している特徴を明らかにした。2つ目は、各地で試行されている介護保険制度の仕組みを解明し、相違点をまとめ、制度設計や事業内容における多様性と、介護分野における医療と福祉の混合を指摘した。3つ目は、日本の医療福祉輸出戦略と関連するが、両国における介護人材不足の問題を分析し、win-win体制のできる「循環型定着」という人材育成と人材の相互移動の仕組みを提案した。This study has found out three main findings. Firstly, it confirmed that in the process of implementing the Chinese version of the long-term care insurance system, the policy shift from a "welfare for the impoverished" approach to a "universal welfare" approach, similar to Japan, has taken place. It also revealed the utilization of the private sector's role, foreign capital, and overseas experience in this transition process. Secondly, it clarified the mechanisms of the long-term care insurance system in various regions, summarized the differences, and highlighted the diversity in system design and service provisions, as well as the integration of healthcare and welfare in the field of long-term care.
