A wareness and prevalence of locomotive syndrome in urban areas
都市部におけるロコモティブシンドロームの認識度,その割合ならびに運動習慣とロコモティブシンドロームの関係性を明らかにすることを目的とした。調査内容は,性別,年代,運動習慣の有無,ロコモティブシンドロームの認知度等とロコモ度であった。健康イベントへの参加者は男性 38名,女性 99名であった。このうち 21歳以上の男性 29名,女性 63名を分析の対象とした。ロコモ度のテストは,立ち上がりテスト,2ステップテスト,ロコモ 25であり,それぞれのロコモ度数のうち最も高い度数を総合のロコモ度とした。ロコモティブシンドロームの認知度は男性 45%,女性 46%,男女全体で46%であった。ロコモ度 1以上の割合は男性で 45%,女性で 48%であった。ロコモ度 2以上の割合は男性で 10%,女性で 10%であった。ロコモ度に至った 3つのテストのうち,質問紙のロコモ 25のテストが最も多かった。運動習慣とロコモ度には関連性が認められなかった。本研究の結果から,都市部におけるロコモティブシンドロームの認知度は,全国平と同程度であったが,ロコモティブシンドロームの割合は多かった。加えて,国民健康・栄養調査で定義する「1回 30分以上の運動を,週 2日以上実施し,1年以上持続していますか」という運動習慣では,ロコモティブシンドロームを抑制するには十分でない可能性が示唆された。The purpose of this study was to reveal the degree of awareness of locomotive syndrome in urban areas, its prevalence, and its relationship with exercise habits. We measured gender, age, exercise habits, awareness of locomotive syndrome, and prevalence of locomotive syndrome. The stage of the prevalence of locomotive syndrome was determined by the highest stage achieved in the stand-uptest, 2-step test, and 25-question geriatric locomotive function. Awareness of locomotive syndrome was 46%(male: 45%; female: 46%). The prevalence of locomotive syndrome in stage 1 or higher was 45% in male and 48% in female. Also, the prevalence of locomotive syndrome in stage 2 or higher was 10% in male and 10% in female. Between those with an exercise habit and those without, the percentages of individuals with locomotive syndrome at stage 1 or higher were 53% and 55%, respectively, with no significant difference between the two groups. No relationship was found between exercise habits and degree of locomotive behavior. The degree of awareness of locomotive syndrome was similar to the 45% reported in the second final report of Health Japan 21. The finding suggests that the exercise habit of "Have you been exercising for at least 30 minutes at a time, two or more days a week, and have you continued this for over a year?" may not be able to sufficient to prevent locomotive syndrome.論文 / Article
Preparation, Characterization, and Antioxidant Capacity of Xanthone-Urea Complex
Xanthones are a group of polyphenolic compounds widely known to have antitumor, anti-inflammatory, antibacterial, antifungal, antiviral, and antioxidant properties. To fully utilize their therapeutic potential, this study aimed to enhance the solubility of a poorly soluble xanthone by preparing a 1:1 molar ratio of xanthone–urea complex utilizing a cogrinding method via a vibration rod mill. DSC analysis revealed the disappearance of the characteristic endothermic peaks of xanthone (177 °C) and urea (136 °C) in the ground mixture (GM), along with the appearance of a new endothermic peak at 185 °C, indicating potential complexation. Additionally, new peaks were observed in the PXRD patterns of the GM at 9.1°, 12.0°, 14.0°, 18.6°, 19.6°, and 24.6°, suggesting structural changes that were also observed in SEM morphology. FTIR spectroscopy revealed significant shifts in the -NH and C=O peaks of xanthone and urea, as well as the disappearance of a -CN peak. Altered diffusion coefficients for both xanthone and urea were measured using DOSY-NMR, accompanied by notable improvements in solubility and dissolution profiles. The GM exhibited nearly a 2-fold increase in solubility, reaching 88.08 ± 1.25 µg/mL at 24 h and 90.97 ± 0.98 µg/mL at 72 h, alongside a 2-fold and 5-fold increase in dissolution at 0.21 µg/mL and 0.51 µg/mL for the physical mixture (PM) and GM, respectively. Furthermore, an enhanced antioxidant capacity was observed, as demonstrated in the calculated Trolox equivalent (TE) value, which increased from 1.48 ± 1.12 for xanthone alone to 1.65 ± 1.03 in the xanthone–urea complex. Administration of Ascorbic Acid Alleviates Neuronal Damage afterCerebral Ischemia in ODS Rats
Reactive oxygen species (ROS) contribute to cerebral damage in transient cerebral ischemia, making their elimination a key therapeutic target. Osteogenic disorder Shionogi (ODS) rats, which lack endogenous L-ascorbic acid (AA) synthesis, serve as a useful model for investigating AA’s protective effects against ischemic brain injury. ODS rats were given an AA-free diet (0% AA), 0.1% AA, or 1% AA in drinking water for two weeks before undergoing middle cerebral artery occlusion and reperfusion (MCAO/Re). The 0% AA group exhibited pronounced damage following MCAO/Re, characterized by the induction of lipid peroxidation, O2− production, inflammation-related gene expression, and extensive infarct formation. In contrast, the 1% AA group showed reductions in these markers, along with fewer TUNEL-positive cells and a smaller infarct volume. Notably, sodium-dependent vitamin C transporter 2 (SVCT2) expression increased in both two AA-supplemented groups, although the 0.1% AA group did not exhibit sufficient improvement in post-ischemic damage. A two-week intake of AA significantly alleviated MCAO/Re-mediated injuries associated with oxidative stress and inflammation in ODS rats. Sufficient AA intake is thus supposed to mitigate ischemic damage, possibly through SVCT2 upregulation and enhanced AA availability, leading to the suppression of oxidative stress and inflammation.Article No.773 Licensee MDPI, Basel, Switzerland. Effect of Betulinic Acid and Ursolic Acid on IL-17-Induced CCL20 Release in Normal Human Epidermal Keratinocytes
Psoriasis is a chronic inflammatory skin disease characterized by erythema, infiltration, and scaling, which is mainly caused by interleukin (IL)-17. The use of molecular targeted drugs in specific therapies offers high efficacy; however, high medical costs and a significant risk of side effects highlight the need for novel therapeutic agents. We previously observed that Morus alba extract (MAE) suppressed IL-17-induced CCL20 mRNA expression in normal human epidermal keratinocytes (NHEKs). In this study, we focused on the IL-17 signaling pathway and investigated the effects of pentacyclic triterpenoids, betulinic acid (BA), and ursolic acid (UA), which are present in MAE, on NHEK cells. Real-time reverse transcription polymerase chain reaction (RT-PCR) and enzyme-linked immunosorbent assay (ELISA) revealed that both BA and UA suppressed CCL20 expression, while only UA alone inhibited CCL20 release. ELISA using specific inhibitors demonstrated that both the p38 and extracellular-signal-regulated kinase 1/2 (ERK1/2) pathways were crucial for IL-17-induced CCL20 release in NHEK. UA effectively suppressed ERK1/2 nuclear localization and moderately affected p38 phosphorylation.
