    トランス位に配位部位である4-ピリジル基を有するポルフィリン11種を合成し、遷移金属イオンとの溶媒熱合成を行うことにより、一次元細孔を有する多孔性配位高分子(PCP)11種類を得た。得られたPCPの一次元細孔内部には溶媒分子が一次元配列していることが単結晶X線構造解析により明らかになった。熱測定により合成したPCPの骨格が350 ℃まで安定に存在すること、空孔内部には水分子が可逆的に吸脱着し、その吸脱着挙動が一次元細孔内部の環境により異なることを明らかにした。Porous Coordination Polymer (PCP) is a material with a porous coordination network structure composed of metal ions and organic ligands, and is expected to be applied to gas adsorption, separation technology, catalysts, etc. Porphyrins form stable complexes with many metal ions and can introduce an interaction site such as a coordination bond in a peripheral portion, thus they are used as a component of various PCPs. we synthesized eleven porphyrins having two pyridyl groups at 5, 15-position as ligating sites. The reaction of Mn(II) with these porphyrins under solvothermal conditions produced PCPs.In all cases, the single crystal X-ray structural analysis revealed these PCPs had isostructural hexagonal type networks possessing channels occupied by solvents molecules. Thermogravimetric analysis of PCPs revealed the guest molecules were liberated below 100 °C and the crystal lattice was stable up to 350 °C.     Study on non-invasive cutaneous vaccination using ultrasound as physical stimulus

    ラットおよびマウスの皮膚に、20 kHz、1および3 MHz超音波を異なる強度で適用したとき、皮膚免疫細胞であるランゲルハンス細胞を活性化し、その活性化の程度を強度および適用時間で制御できる可能性があることを示した。マウスにおけるオボアルブミンをモデル抗原に用いた繰り返し経皮免疫実験では、皮下および皮内投与単独条件に比べて、20 kHz, 0.332 W/cm2 超音波の10 min照射を併用したときの血中IgG量は増加した。これらの結果は、超音波照射は皮内免疫応答を惹起し、経皮投与した抗原に対する特異的抗体産生を亢進させる可能性があることを示唆している。In rats and mice, applications of 20 kHz, 1 and 3 MHz ultrasounds (US) to skin activated Langerhans cells and the extent of activation depended on the intensity and the application period of US. It shows that it might potentially be able to control intradermal immune response by conditioning of US parameters. Furthermore, production of IgG induced by repeated subcutaneous or intracutaneous injections of ovalbumin was increased by the combination of 20 kHz US treatment at an intensity of 0.332 W/cm2 for 10 min.     Mechanisms of decline of cognitive functions by chronic renal failure involving neurogenesis

    慢性腎不全により認知機能の低下するリスクが高まる機構として、慢性腎不全による認知機能低下に脳内の神経新生の低下が関与するのではないかと仮説を立て検証した。慢性腎不全モデルラットでは、認知機能が低下し、抑うつ症状が認められた。脳の組織学的解析の結果、記憶や学習に関係のある海馬でミクログリアが活性化され、神経新生が抑制されていた。尿毒症物質を定量したが、血漿や腎臓では増加が認められたが、脳ではほとんど検出されなかった。今後さらなる原因物質の探索が必要である。As a mechanism that increases the risk of cognitive decline due to chronic renal failure, we hypothesized that decrease of neurogenesis in the brain of hippocampus might be involved in cognitive decline due to chronic renal failure. In the rat model of chronic renal failure, cognitive function were decreased and depressive symptoms were observed. As a result of histological analysis, microglia, involved in inflammation, were activated and neurogenesis was suppressed in the hippocampus, related to memory and learning. Uremic toxins were increased in plasma and kidney, but hardly detected in brain.     Rapid identification of Colchicum autumnale based on loop-mediated isothermal amplification (LAMP) assay

    Purpose Colchicum autumnale (Colchicaceae) exhibits toxicity, and severe poisoning cases due to ingestion of this plant have been reported in Japan. Identifying the cause of poisoning is important for emergency medical treatment, and a rapid and simple detection technique is required for the identification of the cause of poisoning. In the present study, we developed a rapid and simple method for the detection of C. autumnale based on a loop-mediated isothermal amplification (LAMP) assay, which is 100- to 1000-fold higher than the conventional polymerase chain reaction method. Methods Specific LAMP primers for C. autumnale were designed based on the trnL-trnF intergenic spacer region. Using the LAMP primers, the LAMP assay was performed at an isothermal reaction temperature of 63 ℃. Results The LAMP reaction was shown to be specific and highly sensitive to C. autumnale, given that the assay can be used for 10 pg of purified DNA. Using a simple protocol for on-site detection, the entire procedure from pretreatment to evaluation required approximately 1 h. Moreover, the LAMP method using a microfluidic device detected multiple genes, including C. autumnale-specific DNA regions, at a time. Conclusions The currently proposed protocol exhibits good potential as a screening method for C. autumnale poisoning in emergency medical care
