Syntheses and Evaluation of 2- or 3-(N-Cyclicamino)chromone Derivatives as Monoamine Oxidase Inhibitors
A series of 2-(N-cyclicamino)chromone derivatives (1a–4c) and 3-(N-cyclicamino)chromone derivatives (5a–8c) were synthesized, and their monoamine oxidase (MAO) A and B inhibitory activities were studied as part of a structure–activity relationship investigation. Compounds 1a–4c showed no remarkable inhibition for MAO-A or MAO-B, whereas compounds 5a–8c (with a few exceptions) showed significant and selective inhibition of MAO-B. Of these compounds, 7c, 7-methoxy-3-(4-phenyl-1-piperazinyl)-4H-1-benzopyran-4-one inhibited MAO-B the most potently and selectively, having IC50 of 15 nM and an MAO-B selectivity index of more than 6700; c.f, 50 nM and 2000, respectively, for safinamide. The mode of inhibition of 7c to MAO-B was competitive and reversible. Considering the IC50 values and selectivity indices of the other synthetic compounds, the presence of the methoxy group on the chromone ring (R2) of 7c seemed to increase MAO-B inhibition. Molecular docking analysis also supports this hypothesis. Our results suggest that 3-(N-cyclicamino)chromones are useful lead compounds for the development of MAO-B inhibitors

Issues and the Expansion of Policies on Accepting Foreign Care Workers into Japan
ここ数年、高齢者介護業界における人材不足の問題が注目されている。厚生労働省が発表した資料によると、2025 年時点の介護人材の需給ギャップは約37.7 万人と推計されている。政府は介護分野の人手不足の問題を解決するために、外国人介護人材の受け入れに関する政策を急速に進めている。2019 年末現在、外国人介護人材の受け入れに関しては、4 つの制度・政策が存在している。本稿では、まずは介護業界における介護人材不足の背景と実態を把握したい。そのうえで、外国人介護人材の受け入れ政策の展開のプロセスを整理し、それぞれの政策における課題を検討する。最後に、留学生に対する福祉教育の支援と促進による外国人介護人材の受け入れは、わが国の介護業界における人手不足の問題を解決するための最善案であることを示唆する。In recent years, the problem of a labor shortage in the elderly care industry in Japan has been attracting attention. According to a data from the Ministry of Health, Labor and Welfare, it is estimated that the supply-demand gap for care workers in 2025 will be about 377,000. Accordingly, the Japanese government has begun promoting several policies on accepting foreign care workers to solve the labor shortage issue in the area of long-term care, and by the end of 2019, four methods and policies for accepting foreign care workers had been implemented. In this paper, the author first grasped the background and actual situation of the shortage of care workers in the long-term care industry. The policy process for accepting foreign care workers the issues pertaining to each were then examined. It is suggested that encouraging support and the promotion of welfare education among foreign students is the best way to deal with the care-worker labor shortage problem.論
Collaborating with Japanese University Students on a Computer Laboratory Project in a Nairobi Slum School
著者は2019 年の夏に城西大学現代政策学部生の9 名と共にケニア共和国のナイロビ市を訪れた。学生たちと2016 年に創設したスラム学校でのパソコン・ラボを訪ねパソコンの台数を増幅し、ラボの改善、パソコン点検など彼らにも協力してもらった。さらに、スラム学校で開催されたシンポジウムにも参加し、豊田通商トヨタケニアアカデミーにも訪問した。また、The Management University of Africa [アフリカ経営大学] の学生たちとの文化交流会に参加し、その他様々な活動を行った。今回この執筆では、研修を開催する理由、背景などを理論的に提示し、現地で学生たちの活躍、活動を詳しく述べ、帰国後学生たちの声も紹介する。今回のケニア研修の経験を通じ、苦悩している現地のスラム学校の子供たちや地域コミュニティに対して少しでも貢献出来たかもしれない学生たちの一人一人には貴重な体験であり、一生記憶に残る体験であったと思われる。In the summer of 2019, the author accompanied a group of nine students from the Faculty of Contemporary Policy Studies, Josai University to Nairobi to partake in a project to develop and expand a computer laboratory that he had established there in 2016. During the trip, the group also took part in a number of other activities, including a symposium, a visit to Toyota Kenya, and cultural exchange activities with African students at the Management University of Africa. This paper will outline the background and rationale for the trip, and the activities that the students took part in, before briefly discussing their feedback.教育実践報
Contribution of Pyk2 pathway and reactive oxygen species (ROS) to the anti-cancer effects of eicosapentaenoic acid (EPA) in PC3 prostate cancer cells
Background: n-3 polyunsaturated fatty acids (n-3 PUFAs), including eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA), are thought to exert protective effects in cardiovascular diseases. In addition, n-3 PUFAs have demonstrated anti-cancer effects in vitro and in vivo. Objective: We investigated the anti-cancer effects and mechanism of action of EPA on PC3 prostate cancer cells in vitro. Methods: PC3 cells were treated with various concentrations of EPA, and cell survival and the abilities of migration and invasion were evaluated. The time course of the growth inhibitory effect of EPA on PC3 cells was also assessed. The mechanism underlying the anti-cancer effects of EPA was investigated by human phosphokinase and human apoptosis antibody arrays, and confirmed by western blot analysis. We also examined the contribution of reactive oxygen species (ROS) to the effects of EPA using the ROS inhibitor N-acetyl cysteine. Results: EPA decreased the survival of PC3 cells in a dose-dependent manner within 3 h of application, with an effective concentration of 500 μmol/L. EPA inhibited proline-rich tyrosine kinase (Pyk)2 and extracellular signal-regulated kinase 1/2 phosphorylation as determined by western blotting and the antibody arrays. The growth of PC3 cells was inhibited by EPA, which was dependent on ROS induction, while EPA inhibited Pyk2 phosphorylation independent of ROS production. Conclusions: Inhibition of Pyk2 phosphorylation and ROS production contribute to the anticancer effects of EPA on PC3 cells

Synthesis and pharmacological evaluation of childinin E and several derivatives as anti-hyphal formation inhibitors against Candida albicans
The natural highly substituted benzophenone childinin E (1) was previously isolated from the fungus Daldinia childiae. Here we describe the total synthesis of childinin E and several derivatives using a linear seven-step sequence. The antifungal properties of the synthesized compounds against Candida albicans were evaluated by an anti-hyphal formation test. Childnin E and two derivatives exhibited anti-hyphal formation activity.4p. Article number: 15258
