Role of CD44 and CD8 in Colorectal Adenocarcinoma metastatic
Colorectal adenocarcinoma is one of the most prevalent malignancies worldwide. The presence of regional lymph node metastases is an important prognostic indicator in colorectal adenocarcinoma. CD44 is a transmembrane protein and a cancer stem cell biomarker that plays a role in tumor proliferation, metastasis, and recurrence. The tumor microenvironment has a vital role in the stage of tumor development. CD8 is one of microenvironment's component which kills the tumor cells. This study aimed to analyze the expression of CD44 and CD8 and investigated their association with colorectal adenocarcinoma metastatic. This study's benefit is that the resulting study can be used as a prognostic factor in colorectal adenocarcinoma. This study included 56 paraffin blocks of patients diagnosed as colorectal adenocarcinoma during January 2017-December 2018 in Anatomic Pathology Laboratory Dr. Soetomo Hospital Surabaya. The paraffin blocks were sliced and stained with immunohistochemistry for CD44 and CD8. The parameters were a positive percentage of tumor cell and lymphocyte immunoreactivity against CD44 and CD8 antibodies. The percentage of CD44 is higher in the metastatic group, and the percentage of CD8 is higher in the non-metastatic group. There was a significant negative correlation between CD44 and CD8 expression in regional lymph nodes metastatic. These results indicated that CD44 and CD8 have a vital role in the incidence of metastasis
The Effects of Hyperbaric Oxygen Therapy on Blood Glucose Levels and Lipid Profile in Diabetes Mellitus Patients
Diabetes mellitus and its complications are still a major health problem in the world. Diabetes mellitus is closely related to the lipid profile, especially concerning the cardiovascular disease. A method that can be used to treat diabetes mellitus is hyperbaric oxygen therapy (HBOT). This study aims to observe changes in blood glucose levels and lipid profiles in patients with diabetes mellitus with hyperbaric oxygen therapy. The determination of blood glucose levels and lipid profiles is done by photometry. A total of 30 patients with diabetes mellitus examined for their blood glucose level and lipid profile before and after hyperbaric oxygen therapy. In the Paired T-test, there were significant decrease in blood glucose levels (p= 0,000), total cholesterol (p= 0,000), and triglycerides (p= 0,000) after hyperbaric oxygen therapy. The conclusion of this study shows there is a significant decrease in blood glucose level and lipid profile (total cholesterol, and triglycerides) after hyperbaric oxygen therapy
The Effect of Dehydroepiandrosterone on Ovarian Reserve in Ovarian Damage Caused by Methotrexate
To determine the possible detrimental effects of multiple methotrexate doses has on the ovarian reserve and to determine the beneficial effects of dehydroepiandrosterone supplementation. The rats (n:24) divided into three groups; Group 1: control group, Group 2: dehydroepiandrosterone and methotrexate group (6mg/kg dehydroepiandrosterone dissolved in 0.1 ml sesame seed oil subcutaneously for ten days and 1mg/kg intramuscular methotrexate at the 1st, 3rd, 5th and 7th days) and Group 3: methotrexate group (1mg/kg intramuscular methotrexate at the 1st, 3rd, 5th and 7th day). The groups compared in regards to their histopathological ovarian damage scores and AMH values. It established that multiple methotrexate applications had a considerable effect on reducing vascular congestion in the ovarian tissue. Both in groups 2 and 3 AMH values found to be significantly lower. When this decline in the ovarian reserve examined comparatively; while both the group 2 and 3 reported having a considerable and continuous reduction in the AMH levels correlative to the control group; the primordial, primary and total follicle counts shown to stay statistically the same in the group 2 (
The Effect of Oral Contraceptive Pills on The Gene Mutation of Factor V Leiden among Sudanese Women
Oral contraceptive pills are problems for women, often have many effects, and may cause several diseases. The purpose of this research was to determine the impact of oral contraceptive pills on factor V sufferers. This case-control study conducted in Khartoum Sudan during the period from April to November 2018. The study included 50 women who used oral contraceptive as a case and 50 women who did not use oral contraceptive pills as a control, all of whom were verbally informed of the study and approved for participation. The PCR do for each sample. The results obtained from cases show that the mean age is 30±5.5 and divided into three groups less than 20 with a lower frequency of 4 % (2/50), (20-35) with a higher incidence of 80 % (40/50) and a higher rate of 16 % (8/50) for more than 35 years. Most cases use the oral contraceptive pill for more than one year at a frequency of 60% (30/50) with a mean of 2±0.8. The most frequent oral contraceptive pill use was levonorgestrel 88% (44/50), followed by desogestrel 12% (6/50). The study concludes that there is no significant difference in gene mutation between case and control. There was also an insignificant association between the mutation and demographic data
Complete Blood Cells Findings Among Covıd-19 In-Patients at Covıd-19 Reference Hospitals, West Nusa Tenggara
Covid-19 virus still attacks various worldwide sectors, especially health. The current problem is that the Covid-19 vaccine is still in progress, so the best strategy to deal with this problem is to prevent this disease's spread. In the laboratory sector, complete blood cells tend to use as a screening method for disease, especially infectious disease. This study describes the CBCs findings among Covid-19 patients at Covid-19 reference hospitals in West Nusa Tenggara. This study will enhance the data to provide early diagnosis for patients with Covid-19. This was a retrospective study conducted from Mei to July 2020. A total of four hospitals have access to 85 medical records of Covid-19 patients. The patient’s data included in this study should have (1) initials; (2) Gender; (3) Age; (4) Patients' disease other than Covid-19; and (5) The results of the first CBCs examination. Incomplete data excluded from the study. The CBCs data collected from Hospital Management Information Systems (HMIS) and Laboratory Information Systems (LIS). The data were analyzed descriptively. The results showed that 5.88% of patients with comorbidities or another disease reported abnormal CBCs results. In comparison, another 94.12% of patients without another disease than Covid-19 tend to present normal results. In conclusion, most of the CBC's findings among Covid-19 patients in West Nusa Tenggara show normal value among patients with no disease other than Covid-19
Stability of Routine Hematology Sample Using The Medonic M-Series Analyzer
Routine hematology tests (Hb, Hct, RBC, WBC, PLT) generally done by automation methods using a hematology analyzer. Ideally, the examinations should be done as soon as possible, although, in some circumstances, it can be delayed. Based on the literature, the sample for routine hematology testing should be processed within 4 – 8 hours of venesection. Some studies revealed that the sample could be stored for up to 48 hours, and it can be influenced by the technology applied to the hematology analyzer. Studies conducted to see the stability of the samples using a hematology analyzer with impedance technology. Tests performed on whole blood samples collected from 12 ostensibly healthy individuals, immediately after collection (fresh sample
The Results of Toxoplasma gondii Examination IgM Antibodies Using ICT Method and ELISA Method in Pregnant Women
Toxoplasmosis is a disease that can infect animals and zoonotic in humans. In pregnant women (congenital toxoplasmosis), the risk and severity of symptoms of congenital toxoplasmosis are more severe if infection occurs early in pregnancy and asymptomatic. The only way to determine disease is by serological screening. Serological diagnosis techniques for toxoplasmosis include ELISA (Enzyme-linked Immunosorbent Assay) and ICT (Immunochromatography Assay Test). However, until now, research on toxoplasmosis with the ICT method is still minimal. This study aims to analyze the results of the IgM antibody examination Toxoplasma gondii using the ICT method with the ELISA method and calculate the sensitivity and specificity of the ICT method and the ELISA method in pregnant women. The research design was used analytic (cross-sectional), with the number of samples is 33 serums. Data from the ELISA method obtained from secondary medical record data and the remaining serum samples were examined by the ICT method by researchers at the Harapan Kita Hospital Laboratory. In the ICT method, two samples (6.1%) were positive, and 31 samples (93.9%) were negative. In the ELISA method, results obtained similar to the ICT method, two samples (6.1%) positive and 31 samples (93.9%) with negative results. Toxoplasmosis examination can be done using the ICT method as the initial serological screening stage, but it not recommended as a diagnosis enforcer
Renal Function Evaluation in Secondary Amyloidosis According to Location of Deposition
It aimed to evaluate the renal functions according to the accumulation site of amyloid in patients diagnosed with secondary amyloidosis by renal biopsy. Fifty patients diagnosed with secondary amyloidosis, who referred to the nephrotic syndrome outpatient clinic included in the study. According to the findings of renal biopsy, only-glomerular amyloid deposition (group- 1) detected in 9 patients and glomerular+ tubulointerstitial deposition (group- 2) identified in 41 patients. Serum creatinine, creatinine clearance, and total proteinuria levels compared between groups at the time of presentation and on 3rd, 6th, 12th, and 24th months. Initial and subsequent serum creatinine, creatinine clearance, and total proteinuria levels not found to be significantly different between groups (p>0,05 for all). Among subjects with only-glomerular deposition, statistically significant descents in total proteinuria, when compared to initial levels, were observed from the beginning of the 6th month to the end of the 24th month (
Manual and Automated Comparatıve Analysıs in Retıculocyte Countıng Method
The traditional method of reticulocyte count is based on microscopic evaluation of peripheral blood films. Automated processes have replaced the manual method of reticulocyte counts in most laboratories. They are readily available and give more precise results. The present study was conducted to compare the manual and automated methods of reticulocyte estimation. The study was conducted for six months, from January to June 2019, at a tertiary care center in Dibrugarh, Assam. One hundred twenty-six venous blood samples of patients were analyzed, of whom 27 were males and 99 females. Student t-test between the two methods showed a p-value of 0.41. Pearson's coefficient between the two methods showed r value =0.992. Scatter plot and Passing &Bablock regression analysis showed a positive correlation between the two methods. The present study showed a good correlation between manual and automated methods. The manual method can use the manual method of counting reticulocytes in situations of limited cost. In contrast, the automatic method can be an option when fast results and large sample sizes are required
Early Relapse After Autologous Stem Cell Transplantation in Multiple Myeloma is Still Prognostic in The Era of Novel Agents
Significant improvements in the prognosis of Multiple Myeloma(MM) have recently observed in the era of novel agents. Induction treatment, including new agents followed by conditioning regimen and upfront autologous stem cell transplantation(ASCT), has been accepted as the standard treatment approach for newly diagnosed eligible MM patients. Despite novel agents, upfront ASCT is still superior to conventional chemotherapy alone. Previous studies revealed that the duration between ASCT and relapse had predicted overall survival(OS), and meantime, it was widely used to determine the potential benefit from a second ASCT. However, the majority of the data collected reflects the treatment modalities before novel agents. In this study, we aimed to investigate the impact of post-transplantation early relapse(ER) on survival in the era of novel agents. The results of 155 MM patients that underwent ASCT at our center between January 2010 and May 2018 were analyzed retrospectively. The median follow-up duration was 20 months in the ER group, 27 months in the non-ER group, and 24 months in all patients. 33.3% of patients in the ER group and 71.4% of patients in the non-ER group were alive at the time of analysis. Median OS was 20.77±3.66 months in the ER group and 40.89±4.21 months in the non-ER group. We found a statistically significant relationship between the ER and the poor OS (p
