    Video Rate Adaptation Based on QoS and QoE information in Wireless Access Networks

    Streaming applications are becoming more and more popular in the mobile world, especially with the new developments in wireless data networks. Promises of maximum throughput speeds of a 100Mbps along with the appearance of multimedia tablets opened the path to a diverse range of streaming services. Signal degradation and user mobility make wireless networks a challenging environment for real-time applications like streaming, due to frequent bandwidth variations. Hence, to assure a continuous service and to maximise the user experience, it is necessary to automatically adapt the streaming rate of the session, based on the network state. This report presents a layered QoS and QoE based rate adaptation algorithm that tackles both delivery and playback problems. It uses measurements of the RTT variation, an original probing technique and playback quality reports to estimate user experience and react as fast and as accurate as possible. The adaptation strategy parameters have been tuned for optimal performance in different wireless environments, like WiFi, WiMAX and LTE.Les applications de streaming video sont de plus en plus populaire dans le monde mobile, en particulier avec les nouveaux développements dans les réseaux sans fil. Les vitesses de débit maximale de 100Mbps ainsi que l'apparition de teblettes multimédia ouvert la voie à une gamme diversifiée de services de streaming. La dégradation du signal et la mobilité des utilisateurs des réseaux sans fil font un environnement difficile pour les applications en temps réel comme le streaming, dues aux variations de bande passante fréquentes. Par conséquent, pour assurer un service continu et à maximiser l'expérience utilisateur, il est nécessaire d'adapter automatiquement la qualité d'encodage video, sur la base de l'état du réseau. Ce rapport présente un algorithme d'adaptation basé sur les parametres de QoS et de QoE qui corrige les problèmes de livraison et de lecture. Il utilise les mesures de la variation de RTT, une technique originale de sondage reseax et de rapports sur la qualité de lecture pour estimer l'expérience utilisateur et de réagir aussi vite et aussi précis que possible.     Characterization of PdhS functions, an essential and polarly localized histidine kinase, in Brucella abortus

    Several bacteria divide asymmetrically, one of the key processes leading to cellular differentiation. A huge number of complex shapes are observed among bacteria, where functionalities can be attributed to specialized cell types as observed in the prokaryotic model Caulobacter crescentus life cycle where a stalked and proliferative competent cell generates a motile flagellated cell able to find favorable replication niches. We are working on Brucella abortus, a facultative intracellular class III pathogen that is responsible for a worldwide zoonosis called Brucellosis, humans being accidental hosts. B. abortus divides asymmetrically, and following an old pole marker called PdhS, it has been suggested that a differentiation event was taking place during B. abortus cell cycle growing in bacteriological medium. PdhS, an essential histidine kinase, is involved in B. abortus cell cycle regulation and displays atypical features such as its large sensory domain. In order to better characterize PdhS, we used a domain mapping approach and identified minimal domains for protein-protein interaction (with PdhS itself or with a partner called FumC), for polar localization of for altered morphologies generation in B. abortus. We hypothesized that the slight morphological asymmetry observed in B. abortus could reflect a functional asymmetry generated after cytokinesis completion. We thus attempted to label the two different sibling cells using chemical compound and molecular markers. We showed that a mother cell, proliferative and non-invasive generates a virulent daughter cell that is blocked in its cell cycle. This shows that a differentiation event is taking place during a cellular invasion by B. abortus. Interestingly, the generation of the invasive daughter cell is observed at each division, using polar marker in growing bacteria. The generation of specialized cell types just after cytokinesis and despite the inheritance of an identical genetic material shows that clonal population of bacteria are more heterogeneous than previously thought. Responsive and stochastic events can trigger phenotypic heterogeneity and we propose that programmed differentiation along the B. abortus cell cycle is another way to generate phenotypic diversity.(DOCSC03) -- FUNDP, 201

    Feature-based Configuration: Collaborative, Dependable, and Controlled

    A growing share of products expose sophisticated configurability to handle variations in user and context requirements. Configurators usually rely on variability models, like feature models (FMs), that structure and constrain the available options. However, most feature-based configuration techniques do not scale well to collaborative contexts. They offer limited mechanisms to determine responsibilities and access rights, to schedule configuration tasks, and to resolve conflicts. Fragments of solutions already exist but a unified and formal foundation for collaborative feature-based configuration is still missing. To provide enhanced control and guidance, we specify responsibilities and rights with views on the FM. Views establish insulated spaces in which users can safely configure the part of the FM assigned to them. The configuration of these views is regulated by a workflow that defines the configuration process. The result of that combination is a new formalism called feature-based configuration workflow. Finally, to handle conflicts between user decisions, we develop a range fix generation algorithm. These concepts and their properties are integrated into a sound mathematical framework. Our contribution is motivated and illustrated through several real-world applications: a product line of meeting management applications (PloneMeeting), a product line of communication protocols used in the aerospace industry (CFDP), the Linux kernel, and an operating system for embedded applications (eCos). The definitions and algorithms are implemented in a toolset that extends SPLOT, an open source configuration environment, and YAWL, a comprehensive workflow management environment.     Self-organised luminescent materials templated by carbon nanotubes scaffolds

    The core of this doctoral dissertation concerns the application of the concepts of self-assembly and self-organization for the preparation of highly luminescent materials obtained upon controlled and directed functionalisation of CNTs with luminescent lanthanide complexes (LnCs). Particularly, both confinement within the hollow cavity of CNTs and exohedral decoration have been pursued, allowing efficient preparation of performing luminescent hybrids. Finally from the work accomplished aiming at the development of novel emissive CNT-templated materials, two side projects stemmed, which will also be considered during this dissertation. The first regards the preparation of an azobenzene (AB) based supramolecular polymer employed for the photo-resposive solubilisation of multi-walled CNTs (MWCNTs).     Towards the thermally reversible adhesion between metallic and polymeric surfaces functionalized by atmospheric plasma polymerization and gas phase reactions.

    The industry shows a growing interest for the reversible adhesion property due to their extremely high potential in microelectronics, automotive or food packaging as they can be easily disassemble or selectively remove pieces for repair, maintenance or recycling. For example, it is estimated that 95% of computer parts could be reused or recycled if the reversible adhesives were used for the electronic packaging. But how to achieve this property? In this study it was demonstrated that thermally reversible adhesion between two functionalized surfaces can be achieved thanks to the thermally reversible interfacial Diels-Alder reaction and the functionalization can be performed by a two-steps dry process. First, the atmospheric plasma copolymerization aimed at deposition of the reactive anhydride functions was performed. Then, the gas phase reaction with commercially available precursors was realized in order to graft diene and dienophile groups enabling the Diels-Alder reaction at a solid interface. The chemistry and morphology of the functionalized surfaces were thoroughly studied using WCA, FT-IR, XPS, SIMS, SEM, AFM in order to ensure high concentration of the diene and dienophile groups and smooth morphology of the layer. The assembling of the diene functionalized Kapton and dienophile functionalized aluminium led to a peel strength of 0.5 N/mm, while assembling of native substrates or surfaces coated by plasma layer only led to that of ~0.02 N/mm. By performing heating and cooling cycles during a continuous peel test measurement, the thermal response adhesion was determined. A detailed characterization of the peeled surfaces using SEM, AFM, WCA, FT-IR, ToF-SIMS and XPS allowed for the determination of the locus of failure.(DOCSC01) -- FUNDP, 201

    Diffusion élastique d'électrons lents par des structures hélicoïdales: application au microscope à projection de Fresnel

    We develop several methods for computer simulation of low energy electron scattering on three-dimensional nanostructures. We apply these formalisms to the simulation of the Fresnel projection microscope. One of these formalisms uses the Lippmann-Schwinger integral equation and another uses the technique of transfer matrices. These formalisms are based on cylindrical partial waves decomposition of the stationary diffusion state. This approach allows the decoupling of the scattering problem in the case of structures with axial or helical symmetry. We propose a new method for calculating the matrices 't' and 's'. This method is based on solving the Lippmann-Schwinger equation on a single layer of the structure at a time. It is more stable and faster than the available methods. For structures infinite according to one dimension of the space, we introduce the new concept of effective scattering width instead of the scattering cross section. Among other things, our simulations concern the scattering by nanotubes and the B-DNA molecule. To this end, we determine effective atomic potentials based on the total and differential atomic cross sections. We also propose a new algorithm for reconstruction of the structure from the scattered intensity in the far field considered as an electronic hologram.Nous développons plusieurs méthodes pour la simulation sur ordinateur de la diffusion électronique de faible énergie sur des nanostructures tridimensionnelles. Nous appliquons ces formalismes à la simulation du microscope à projection de Fresnel. Un formalisme exploite l'équation intégrale de Lippmann-Schwinger et un autre utilise la technique des matrices de transfert. Ces formalismes sont basés sur la décomposition en ondes partielles cylindriques de l'état stationnaire de diffusion. Cette approche permet le découplage du problème de diffusion dans le cas de structures possèdant des symétries axiales ou hélicoïdales. Nous proposons une nouvelle méthode pour le calcul des matrices 't' et 's'. Cette méthode se base sur la résolution de l'équation de Lippmann-Schwinger sur une seule tranche de la structure à la fois. Elle est plus stable et plus rapide que les méthodes disponibles. Nous introduisons le nouveau concept de largeur efficace de diffusion en lieu et place de celui de section efficace lorsque la structure est infinie selon une des dimensions de l'espace. Nos simulations portent entre autres sur la diffusion par des nanotubes et par la molécule d'ADN-B. A cette fin, nous déterminons des potentiels atomiques effectifs basés sur les sections efficaces atomiques totales et différentielles.     Synthèse et caractérisation fondamentale de nanocristaux de silicium et germanium synthétisés par implantation ionique pour le développement de capteurs photovoltaïques à haut rendement - Développement de l'ionoluminescence

    Solar energy, which is renewable, readily available and accessible to all, seems to possess all the qualities to become tomorrow's energy. However, the improvement of photovoltaic technologies is crucial to promote solar cells as a major source of electricity. The goal of the so-called "third generation" cells, is twofold: affordability (abundant material, production techniques), and maximum efficiency. A way to achieve this goal is the insertion of semiconductor nanocrystals in a dielectric material by ion implantation, followed by annealing. Such systems should allow the generation of multiple excitons per incident photon, and thus promote cell efficiencies. Amongst the materials considered, silicon nanocrystals (Si-nc) embedded in silica are favorites, and germanium nanocrystals (Ge-nc) are full of advantages in terms of solar absorption. In this fundamental work, we have investigated the synthesis parameters and mechanisms of formation of these nanocrystals. We have combined ion beam analysis with physicochemical characterizations by XPS and μ-Raman, electron microscopy, optical characterizations by ellipsometry and photoluminescence spectroscopy, and have approached the study of photovoltaic properties of these materials. The promise delivered by Si nanocrystals was initially evaluated. We have implanted Si into SiO2 at fluences of 6 - 20 x 1016 Si.cm-2, followed by different annealing times in N2 at 1100 ° C. The formation of Si-nc whose size depends on the local implanted concentration was observed by TEM and PL, and measuring their IV response provided evidence of a photovoltaic effect in these structures. Depth profiling of Si was made possible by the development of an ion beam analysis method, based on BS and NRA, in order to profile easily Si ions implanted in a matrix containing Si. The turn of Ge nanocrystals in silica came in a second time : we have joined ion beam analysis to SEM and μ-Raman spectroscopy to reveal the desorption of Ge during annealing ( up to 65%), as well as the formation of Ge-nc and large spherical nanocavities at temperatures above the melting point of Ge, of diameters between 4 and 35 nm. The mechanisms of formation of these structures were revealed. A lower desorption of Ge was observed in thin films of SiO2/Si characterized by RBS and XPS. Migration of Ge to preferential sites at the interface was revealed. Third, co-implantation of Si prior to Ge provided a solution of prestige to the desorption of Ge. We have demonstrated, by RBS characterizations, a trapping of Ge by Si atoms introduced in excess, with a linear increase in the amount of Ge retained with the fluence of implanted Si, far as to retain the amount of Ge measured before annealing. In parallel to these characterizations, we have explored a new way for the characterization of Si-nc and nc-Ge: ionoluminescence (IL), or the study of light emissions in the ion beam. This method has revealed a strong potential for the characterization of damage to nanostructures and transfer of carriers in these materials in the case of Si-nc, and has revealed numerous agreements with μ-Raman and XPS characterizations for Ge-nc, allowing also a study of the charge transfer mechanisms in PL of these structures.L'énergie solaire, renouvelable, aisément disponible et accessible à tous, semble posséder toutes les qualités pour devenir l'énergie de demain. Cependant, l’amélioration des technologies photovoltaïques est un point crucial pour faire des cellules solaires une source majeure d’électricité. L’objectif pour les futures cellules, dites « de 3e génération » est double : un coût abordable (matériau abondant, techniques de production), et un rendement maximal. Une voie pour y parvenir est l’insertion de nanocristaux de semi-conducteurs dans un matériau diélectrique par implantation ionique, suivie d’un recuit. De tels systèmes devraient permettre la génération des plusieurs excitons par photons incident, et promouvoir ainsi l’efficacité des cellules créées. Parmi les matériaux envisagés, les nanocristaux de silicium (Si-nc) enrobés dans la silice sont favoris, et les nanocristaux de germanium (Ge-nc) regorgent d’atouts en termes d’absorption solaire. Dans ce travail fondamental, nous avons investigué les paramètres de synthèse et les mécanismes de formation de ces nanocristaux. Nous avons combiné l’analyse par faisceau d’ions aux caractérisations physico-chimiques par XPS et µ-Raman, à la microscopie électronique, aux caractérisations optiques par ellipsométrie et spectroscopie de photoluminescence, et approché l’étude des propriétés photovoltaïques de ces matériaux. La promesse livrée par les nanocristaux de Si a dans un premier temps été évaluée. La formation de Si-nc dans le SiO2 implanté et recuit, dont la taille dépend de la concentration locale implantée, a été observée par MET et PL, et la mesure de leur réponse I-V a fourni les preuves d’un effet photovoltaïque dans ces structures. Le profilage en profondeur de Si a été rendu possible par la mise au point d’une méthode d’analyse par faisceau d’ions, basée sur le BS et la NRA, pour profiler aisément en profondeur des ions de Si implantés dans une matrice contenant du Si. Le tour des nanocristaux de Ge dans la silice est ensuite venu : nous avons pour la première fois joint l’analyse par faisceau d’ions au MEB et à la spectroscopie µ-Raman pour mettre en évidence la désorption de Ge au cours du recuit (jusqu’à 65%), ainsi que la formation de Ge-nc et de larges nanocavités sphériques de diamètres compris entre 4 et 35 nm à des températures supérieures au point de fusion du Ge. Les mécanismes de formation de ces structures ont été révélés. Une moindre désorption de Ge a été observée dans des couches minces de SiO2/Si caractérisées par RBS et XPS. La migration de Ge sur des sites privilégiés à l’interface a été révélée. Troisièmement, la co-implantation de Si préalablement au Ge a fourni une solution de prestige à la désorption du Ge. Nous avons mis en évidence, par caractérisations RBS, un piégeage du Ge par les atomes de Si introduits en excès, avec une croissance linéaire de la quantité de Ge retenu avec la fluence de Si implanté, jusqu’à retenir la quantité de Ge mesurée avant le recuit. En parallèle à ces caractérisations, nous avons exploré une nouvelle voie pour la caractérisation de Si-nc et Ge-nc : l’ionoluminescence, ou l’étude des émissions lumineuses sous faisceau d’ions. Cette méthode a révélé un fort potentiel pour la caractérisation de l’endommagement des nanostructures et du transfert des porteurs de charge dans ces matériaux dans le cas de Si-nc, et a révélé de nombreux accords avec les caractérisations µ-Raman et XPS pour des Ge-nc, permettant également une étude des mécanismes de transfert de charge dans la PL de ces structures.(DOCSC01) -- FUNDP, 201

    Etude spectroscopique de mélanges à base d'acétylène : expérience et modélisation.

    This work is devoted to the study of gaseous mixtures based on acetylene diluted in other gases. Two complementary aspects are developed: an experimental study of the spectral parameters of infrared absorption lines and a theoretical study which is able to model the intermolecular collisions. The experimental part concerns the study of the temperature dependence of the collisional broadening and shift coefficients of infrared absorption lines of self-perturbed acetylene and diluted in hydrogen, nitrogen or xenon. These measurements are carried out by diode-laser spectroscopy at very high resolution, for absorption lines in the n4+n5 ro-vibrational band, located around 1330 cm−1, and under conditions of very different pressures (between 20 mbar and 400 mbar) and temperatures (between 170.0 K and 500.0 K). The determinations of the collisional broadening widths and shifts are obtained by fitting to the experimental profiles the theoretical lineshapes (Voigt, Rautian-Sobel'Man and Galatry). The temperature dependencies of the collisional shift coefficients of lines of self-, N2- or Xe-perturbed C2H2 are analyzed for the first time in the n4+n5 ro-vibrational band. The measurement of these coefficients has required the development of an experimental technique allowing to have a great accuracy. The theoretical part is devoted to the modeling of interactions between surrounding molecules. The intermolecular collisions are described by a semiclassical model developed by Robert and Bonamy. The latter considers that the molecule energy levels are quantified but supposes that their trajectories are governed by the classical mechanics laws. An ab initio potential is implemented within the numerical version of this model developed few years ago. Indeed, the description of the intermolecular interaction potential is very important because of its effect on the molecular collisions and, in addition, its isotropic part has a great influence on the molecules trajectories. Taking into account an ab initio potential, one avoids some approximations and it's especially an honest way to test the semiclassical model. Two molecular systems are concerned by these calculations : N2-H2 and C2H2-H2. Finally, these two aspects bring different but complementary informations. In one way, a numerical simulation has to be corroborated by exprimental results in order to validate its results. In an other way, an experimental setup is always limited by its technical characteristics and thus, the modeling allows to explore conditions which are very difficult to access by experiments.Ce travail porte sur l'étude de mélanges à base d'acétylène mis en présence d'autres gaz. Deux aspects complémentaires sont développés : une étude expérimentale de paramètres spectraux des raies d'absorption infrarouge et une étude théorique permettant de modéliser les collisions intermoléculaires. La partie expérimentale porte sur l'étude de la dépendance en température des coefficients d'élargissement et de déplacement collisionnels de raies d'absorption infrarouge d'acétylène autoperturbé ou dilué dans l'hydrogène, l'azote ou le xénon. Les mesures sont réalisées par spectroscopie diode-laser à très haute résolution, pour des raies d'absorption de la bande vibrationnelle n4+n5, située vers 1330 cm−1, et dans des conditions de pression (entre 20 mbar et 400 mbar) et de température (entre 170.0 K et 500.0 K) très différentes. Les largeurs et les déplacements collisionnels sont déterminés par ajustements de profils théoriques (Voigt, Rautian-Solbel'Man et Galatry) sur les profils expérimentaux des raies. Les dépendances en température des coefficients de déplacement collisionnel de C2H2 autoperturbé et mis en présence de N2 ou de Xe, sont étudiées pour la première fois dans la bande n4+n5. La détermination expérimentale de ces coefficients a nécessité le développement d'une méthode de mesure permettant d'atteindre une grande précision. La partie théorique porte sur la modélisation des interactions entre les molécules mises en présence. Les collisions intermoléculaires sont décrites par un modèle semiclassique développé par Robert et Bonamy. Celui-ci considère que les niveaux d'énergie des molécules sont quantifiés mais suppose que leurs trajectoires sont régies par les lois de la mécanique classique. Un potentiel intermoléculaire ab initio est intégré à la version numérique de ce modèle développée il y a quelques années. En effet, la description du potentiel d'interaction moléculaire est très importante puisqu'elle régit la collision entre les molécules en tant que telle et, de plus, sa partie isotrope influe directement sur leurs trajectoires. Intégrer un potentiel ab initio, c'est s'affranchir d'éventuelles approximations mais c'est aussi et surtout une manière honnête de tester le modèle semiclassique. Dans ce contexte, deux systèmes moléculaires sont étudiés : N2-H2 et C2H2-H2. Finalement, ces deux aspects apportent des informations distinctes mais complémentaires. D'une part, une simulation numérique doit être corroborée par des résultats expérimentaux afin que ses résultats soient validés. D'autre part, un dispositif expérimental est toujours limité de par ses considérations techniques et la modélisation permet alors d'explorer des conditions difficilement accessibles par l'expérience.(DOCSC01) -- FUNDP, 201

    The MAGED1 gene is located on the X chromosome, it belongs to the group/category of type II MAGE genes , which are expressed in various tissues during embryonic development and in adult organism. Among the type II MAGE genes, NECDIN and MAGEL2 are probably the most studied because of their putative involvement in the etiology of the Prader-Willi syndrome. Indeed, these two genes are located in the 15q11-q13 region that is absent in most people with the Prader-Willi syndrome. The most striking feature of individuals suffering from Prader-Willi syndrome is their hyperphagic behavior which leads to the development of morbid obesity if their food intake is not controlled. To investigate the function of Maged1 in vivo, Maged1- mice were previously obtained by gene targeting in our laboratory. Curiously, as it is observed in people with the Prader-Willi syndrome, the Maged1- mutants exhibit hyperphagia and develop obesity, although this obesity is milder and appears later compared to what is observed in individuals suffering from Prader-Willi syndrome. Maged1- knockout mice have also behavioral defects similar to those observed in autistic individuals. Interestingly 25% of the people with Prader-Willi syndrome also develop autism. To characterize the autistic-like behaviors of the Maged1- knockout mice, we submitted these mice to a battery of tests. The Maged1- knockout mice exhibited a reduced sociability in the test of reciprocal social interactions. In the three chamber experiment, the Maged1- mutants remained at distance of the mouse stimulus and a concomitant reduction in direct social interactions was observed. Finally in the social memory test, the Maged1- knockout mice were unable to discriminate a new partner from a familiar one. The Maged1- mutants also displayed reduced ultrasonic communications during courtship behavior and a severe reduction of coital activity. Besides defects in social interactions and communications, Maged1- mutants are hypoactive and display anxiety-like behaviors such as exaggerated self-grooming. This association of behavioral defects allows us to propose Maged1- knockout mice as a new model of "autistic mice". Loss of function of Maged1 gene in mice also leads to a reduced production of mature oxytocin, while the precursor and intermediate forms of this neuropeptide are normally synthesized. The lack of mature oxytocin may explain some of the phenotypes displayed by the knockout mice. Indeed, oxytocin is known to be involved in the regulation of food intake, social behavior, anxiety and sexual behavior. To demonstrate the role of oxytocin in the altered behavior of the Maged1- mice, we showed that peripheral injection of oxytocin rescued the social memory of these animals. Altogether, our results demonstrate an essential role for Maged1 in the regulation of social and sexual behaviors as well as in the control of metabolism which are in part explained by the regulation of mature oxytocin production by Maged1.Le gène MAGED1 est localisé sur le chromosome X, il appartient à la catégorie des gènes MAGE de type II qui sont exprimés dans de nombreux tissus pendant le développement embryonnaire et également aux stades adultes. Parmi les gènes MAGE de type II, NECDIN et MAGEL2 sont probablement les plus étudiés en raison de leur implication probable dans le développement du syndrome de Prader-Willi. En effet, ces deux gènes se trouvent dans la région 15q11-q13 qui est absente chez la plupart des patients atteints du syndrome de Prader-Willi. Ces patients présentent entre autres une hyperphagie qui entraîne le développement d'une obésité morbide si la prise alimentaire n'est pas contrôlée chez ces individus. Afin d'étudier la fonction de Maged1 in vivo, des souris knock-out Maged1- ont été crées dans notre laboratoire. Curieusement, comme les personnes atteintes du syndrome de Prader-Willi, les souris Maged1- présentent de l’hyperphagie et développent aussi de l'obésité bien qu'elle soit plus légère et apparaisse plus tard que chez les patients atteints du syndrome de Prader-Willi. Les mutants Maged1- présentent également des troubles du comportement semblables à ce qui est observé chez certains autistes. Or les personnes atteintes du syndrome de Prader-Willi développent également de l'autisme dans 25 % des cas. Afin de caractériser les comportements autistiques des mutants Maged1-, nous avons soumis ces souris à une batterie de tests. Ainsi, les mutants Maged1- présentent une sociabilité réduite dans le test des interactions sociales réciproques. Dans l'expérience des trois chambres, les mutants Maged1- restent à distance de la souris stimulus et présentent une réduction des interactions sociales directes mesurées par le temps d'investigation olfactive du stimulus. Enfin, dans le test de la mémoire sociale, les mutants Maged1- sont incapables de discriminer un nouveau partenaire par rapport à un partenaire familier. Les mutants Maged1- présentent également une réduction des communications vocales ultrasoniques pendant la parade nuptiale et une activité coïtale très réduite. En plus de présenter des défauts au niveau des interactions sociales et de la communication, les mutants Maged1- ont également une activité motrice spontanée réduite, une augmentation de certains comportements répétitifs (activité d’auto-toilettage exagérée) et présentent certains comportements associés à l’anxiété. L’association de ces défauts comportementaux nous permet de proposer les souris Maged1- comme un nouveau modèle de « souris autiste ». La délétion de Maged1 in vivo entraîne également une diminution de la production d’ocytocine mature tandis que les formes précurseurs et intermédiaires de ce neuropeptide sont normalement synthétisées chez les souris Maged1- . Ce défaut d'ocytocine mature peut expliquer une partie des phénotypes que nous avons observés chez les souris knock-out Maged1-. En effet, l'ocytocine est impliquée dans la régulation de la prise alimentaire, dans la régulation des comportements sociaux, de l'anxiété et du comportement sexuel. Le rôle de l'ocytocine dans l’altération des comportements sociaux, a été démontré par le rétablissement de la mémoire sociale après administration périphérique d’ocytocine. L'ensemble de ces résultats démontre un rôle essentiel de Maged1 dans la régulation des comportements sociaux et sexuels ainsi que dans le contrôle du métabolisme qui s’explique en partie par le rôle de Maged1 dans la production d’ocytocine mature.(DOCMED02) -- FUNDP, 201

    Caulobacter crescentus triggers the secretion of various proteins to adapt to stressful or changing environment. In particular, C. crescentus secretes an extracellular lipoprotein, ElpS, in response to phosphate starvation. Our results demonstrate that ElpS is partially released in the environment dependently on type II secretion system and activates an external alkaline phosphatase activity. Nevertheless, the exact role of ElpS in phosphate mobilization remains unclear. Interestingly, PcoAcc, encoding an putative protein involved in copper resistance, modulates the expression of elpS but physiological relevance of this interaction is still unknown. PcoAcc is encoded on C. crescentus chromosome in operon with a second gene, pcoBcc, which also contains several predicted copper binding domains. Although the exact mechanism is not still entirely described, PcoAcc et PcoBcc are both essential for C. crescentus survival in medium containing copper. Preliminary results suggest that PcoAcc is exported in cell surface and facilitate copper efllux. Deletion of pcoAcc and/or pcoBcc in C. crescentus results in enhanced copper accumulation in bacteria leading to cell death. Reciprocally, mutant strains for pcoAcc are able to remove high amount of copper from contaminated aqueous solutions and can be consequently be used as biosorbents in water treatment.La sécrétion de protéines chez Caulobacter crescentus est primordiale dans la réponse à différents stress environnementaux. C. crescentus module ainsi la production et la sécrétion d’une lipoprotéine de surface, ElpS, en réponse à une carence en phosphate inorganique dans son milieu de culture. Différents résultats montrent qu’ElpS est partiellement relarguée dans l’espace extracellulaire de manière dépendante du système de sécrétion de type II où elle stimule une activité phosphatase alcaline externe. Le rôle exact d’ElpS dans la mobilisation du phosphate extracellulaire reste néanmoins à déterminer. De manière intéressante, PcoAcc, annotée en tant qu’oxydase capable de fixer du cuivre, influence la transcription d’elpS sans que les détails physiologiques de cette régulation ne soient élucidés. De manière intéressante, PcoAcc est encodée sur le chromosome de C. crescentus sous forme d’opéron avec un second gène, pcoBcc, qui possède également plusieurs domaines putatifs de liaison au cuivre. Bien que les fonctions exactes de PcoAcc et PcoBcc restent à élucider, les deux protéines sont essentielles simultanément à la survie de C. crescentus en présence de cuivre. Des résultats préliminaires indiquent que PcoAcc serait exportée en surface bactérienne et pourrait ainsi faciliter l’export d’ions métalliques hors de la cellule. La délétion de pcoAcc et/ou de pcoBcc chez C. crescentus conduit à une accumulation importante de cuivre dans les cellules qui induit une mort cellulaire importante. Réciproquement, des souches mutantes pour pcoAcc sont capables d’absorber de grandes quantités de cuivre depuis des échantillons liquides et peuvent par conséquent être utilisées en bioremédiation des métaux pour le traitement d’effluents contaminés. Nos premières analyses indiquent également que ces souches peuvent être régénérées après absorption de cuivre par incubation dans du milieu frais sont ainsi potentiellement ré-utilisables.(DOCSC03) -- FUNDP, 201
