    Immunohistochemical expression of heat shock proteins in the mouse periodontal tissues due to orthodontic mechanical stress

    松本歯科大学博士(歯学)2011甲第98号application/pdfThe histopathology of periodontal ligament of the mouse subjected to mechanical stress was studied. Immunohistochemical expressions of HSP27 and pHSP27 were examined. Experimental animals using the maxillary molars of ddY mouse by Waldo method were used in the study. A separator was inserted to induce mechanical stress. After 10 minutes, 20 minutes, 1 hour, 3 hours, 9 hours and 24 hours, the regional tissues were extracted, fixed in 4% paraformaldehyde and 0.05 M phosphate-buffered fixative solution. Paraffin sections were made for immunohistochemistry using HSP27 and p-HSP27. In the control group, the periodontal ligament fibroblasts expressed low HSP27 and p-HSP27. However, in the experimental group, periodontal ligament fibroblasts expressed HSP27 10 minutes after mechanical load application in the tension side. The strongest expression was detected 9 hours after inducing mechanical load. p-HSP27 was also expressed in a time-dependent manner though weaker than HSP27. The findings suggest that HSP27 and p-HSP27 were expressed for the maintenance of homeostasis of periodontal ligament by the activation of periodontal ligament fibroblasts on the tension side. It also suggests that these proteins act as molecular chaperones for osteoblast activation and maintenance of homeostasis.doctoral thesi

    application/pdfPanax notoginseng is used as a hemostatic drug to treat the dental extraction socket. However, the mechanisms remain to be elucidated. Therefore, the effects of Panax notoginseng on the blood coagulation system were examined in this study. Extraction of Panax notoginseng with phosphate-buffered saline (PBS) or PBS + bovine serum albumin (BSA) prolonged activated partial thromboplastin time (APTT), but had no effect on prothrombin time (PT). APTT by treatment with PBS+BSA-extraction was shorter than that with PBS-extraction. Heat treatment of PBS-extraction had no effect on APTT, but heat treatment of PBS+BSA-extraction reduced the effect of shortening of APTT compared to the control. Fifty percent of ethanol extraction of Panax notoginseng shortened APTT, while 100% ethanol extraction prolonged APTT. When 50% ethanol extraction (Fr.I) was fractionated, the ethyl acetate-soluble fraction (Fr.II) shortened APTT, but the n-butanol-soluble fraction (Fr.III) and the water-soluble fraction (Fr.IV) had no effect on APTT. These findings suggested that water-soluble components inhibit the intrinsic coagulation system, that the lipid-soluble components facilitate, and that both components were heat-stable. Moreover, the components facilitating the intrinsic coagulation system were present in Fr.I and in particular Fr.II. Therefore, both Fr.I and Fr.II may be applicable as a hemostatic drug.journal articl

    applicationFifteen prefectures in Japan have established regulations on dental health. They stipulate the objectives of the regulations, roles and responsibilities of prefectural and municipal governments and residents, collaboration across the fields of health, medicine, welfare, and education, roles of insurers and employers, development of dental health plans by prefectures, implementation of measures (collection and provision of health-related information, prevention of dental caries, life-long health maintenance, involvement of relevant people and enhancement of their qualifications, surveys on the status of dental health, and research promotion), and securing of financial resources. To improve the dental health of residents, the dental care community should become involved in the development and implementation of dental health plans. The regulations were established with the concerted efforts of people involved in dental health as a backdrop. Their activities, including the promotion of the "8020" campaign and "Healthy Japan 21" project, received widespread praise and markedly influenced the legislature and residents. In recent years, dental science studies have clarified the relationship between periodontal diseases and general health or disorders, increasing public awarenessof the importance of dental health. The Community Health Act, Health Promotion Law, Maternal and Child Health Act, School Health and Safety Act, and Industrial Safety and Health Act include provisions regarding dental health because it is required to establish a system to provide a life-long dental health services in a comprehensive and effective manner. For this reason, "the regulations on the promotion of dental health" were established as basic principles or laws to address specific issues.journal articl
