    ウサギ葉状乳頭ホスファターゼの生化学的研究 : I.葉状乳頭遠心分画の各種ホスファターゼ活性

    application/pdfHistochemical studies have revealed that taste organs in mammals have high activities of several phosphatases, but the nature and roles of these phosphatases have not yet been elucidated. The present study was undertaken to find a suitable material for biochemical study on phosphatases in mammalian taste organ. Each experiment was carried out with six foliate papillae of rabbit dissected thoroughly under a stereoscopic microscope. The foliate papillae were homogenized and centrifuged to obtain P_1 (900×g, 10min), P_2 (5,000×g, 10min), P_3 (8,000×g, 20min), P_4 (105,000×g, 60min) and S_4. The highest specific activities of Ca^-ATPase, Mg^-ATPase and Ca^, Mg^ ATPase were obtained in P_4 fractions. The specific activity of Mg^-ATPase in the P_4 fraction of the foliate papillae of rabbit was several hundred times higher than that obtained in bovine circumvallate papillae by Lum & Henkin. This indicates that the material used in the present study is suitable for the enzymatic study of ATPase in mammalian taste organ.journal articl

    application/pdfThe effect of the Nerve Growth Factor (β-subunit) antiserum to submandibular glands and the surperial servical sympathetic ganglions was investigated using the mouse of different developmental stages. The intra-abdominal injection of the antiserum to a newbron mouse did not affect to the structure or the development of the gland, although antiserum massively destroyed it's sympathetic ganglion. The nervous ganglion cells were decreased in population and size. In 50-day-old mouse a severe morphological alternation of the granular tubular portion of the gland was observed. The granular tubular portion appeared to reduce in number and size. The epithelial cells lining the wall of the tubuli showed marked sings of degeneration, and there was shrinking and vacuolation of the cytoplasm, and depleted of secretory granules. No significant alternations were detected in the acinar portion. The surperial servical sympathetic ganglions of the 50-day-old mouse were undergone a moderate atrophy.journal articl

    application/pdfThe effects of cholinergic agents on fluoride absorption through the rabbit intestine were investigated both in situ and in vitro. The results obtained are summarized as follows: 1) When the concentration of the fluoride solution applied in an intestine was increased until lO ppm, the rate of intestinal absorption of fluoride was inversely decreased. 2) Acetylcholine caused an increase in the rate of fluoride absorption.and the reduction of it occurred due to addition of atropine. 3) Bethanechol markedly increased the absorption rate of fluoride and methylscopolamine reduced it. Those effects were observed in the both agents with a narrow range of concentrations. 4) Barium chloride reduced the fluoride absorption and papaverine slightly increased it. From these results, the following suggestion was discussed: The intestinal absorption of fluoride may be caused not only by simple diffusion, but also by facilitated diffusion and may be under the control of parasympathetic nerves.journal articl

    application/pdfIt was found by the authors that there was an activity which inhibited oral Bacteroides sp. and Propionibacterium acnes in the cultured cells of dental plaque. This inhibitory activity is derived from the bacteriocin-like activity producing microorganisms in dental plaque, but they have not been identified as yet Twenty strains of bacteriocin-like activity producing microorganisms, isolated from adult dental plaques, were investigated their biological characteristics. The bacteriocin-like activity producing strains proved to be Gram positive Streptococci. They grew well anaerobically, and were catalase nagative. They produced α-hemolysis on blood agar, and grew on the Mitis-Salivarius agar and 40% bile blood agar, while they failed to grow in 6.5% saline broth, at 10℃ and pH 9.6 broth. However, nine strains of them grew in either medium at 45℃. All strains produced ammonia from arginine. Twelve strains of them extracellularly synthesized polysaccharides in a 5% sucrose-containing broth. These strains fermented various carbohydrates, but all of strains failed to ferment mannitol, sorbitol and glycerol. Most of strains fermented inulin, while only few of strains fermented starch and raffinose. From these biological characteristics, the bacteriocin-like activity producing strains were identified as Streptococcus sanguis. Streptococcus sanguis is a predominant species of dental plaque flora. From the bacteriocin-like activity of this species which inhibits the anaerobes important to the etiology of periodontal diseases, the possible role and significance of Streptococcus sanguis in the oral cavity was discussed.journal articl

    application/pdfThe effects of sodium fluoride (NaF) on drug-metabolizing enzymes (aminopyrine demethylase-AD, Hexobarbital oxidase-HO and aniline hydroxylase-AH) in the rat liver were studied in vivo and in vitro. Prior to the study, preliminary investigations were carried out on procedures of the enzyme assay concerning storage stability of these enzymes in a frozen 9,000×g supernatant fraction from the rat liver homogenate and whether nicotinamide should be involved in the incubation mixtures. Results are summarized as follows: 1) When a 9,000×g supernatant fraction was stored at -20℃, AD activity was decreased in 5 days to approx. 40% of the level of the first day, whereas both activities of AH and HO remained unchanged during the same period. 2) AD, AH and HO activities were all inhibited by the addition of nicotinamide when a higher concentration of NADP was maintained during the assay. In this study.nicotinamide was excluded from incubation mixtures. 3) In the in vitro study, when 0.1, 1, 10 and 100 mM of NaF was added to the incubation mixtures, AD and HO activities were both decreased In contrast, AH activity was slightly increased. 4) In the in vivo study, when rats ingested NaF (10, 38, 76 and 152ppm as F) in their drinking water for a month, it was observed that the sleeping time was prolonged by hexobarbital sodium, the fluoride was accumulated in the liver of these rats and HO activity was inhibited, but activities of AD and AH were both similar to those of control rats.journal articl
