Notch signaling acts on cell differentiation and acquisition of tumor-specific character of oral neoplasms
application/pdfHistopathological findings of oral neoplasm cell differentiation and metaplasia suggest that tumor cells induce their own dedifferentiation and re-differentiation and may lead to the formation of tumor-specific histological features. Notch signaling is involved in the maintenance of tissue stem cell nature and regulation of differentiation and is responsible for the cytological regulation of cell fate, morphogenesis, and/or development. In our previous study, immunohistochemistry was used to examine Notch expression using cases of odontogenic tumors and pleomorphic adenoma as oral neoplasms. According to our results, Notch signaling was specifically associated with tumor cell differentiation and metaplastic cells of developmental tissues. Notch signaling was involved in the differentiation of the ductal epithelial cells of salivary gland tumors and ameloblast-like cells of odontogenic tumors. However, Notch signaling was also involved in squamous metaplasia, irrespective of the type of developmental tissue. In odontogenic tumors, Notch signaling was involved in epithelial–mesenchymal interactions and may be related to tumor development and tumorigenesis. This signaling may also be associated with the malignant transformation of ameloblastomas. Overall, Notch signaling appears to play a major role in the formation of the characteristic cellular composition and histological features of oral neoplasms, and this involvement has been reviewed here.journal articl

Implant placement is rarely difficult in the anterior maxilla due to insufficient bone volume below the maxillary sinus.The indications and methodology of implant treatment for patients are dependent on the planning of endosseous implant treatment in the esthetic zone with implant therapy and should be based on careful diagnosis.Knowledge and understanding of maxillary sinus floor pneumatization is important to achieve an accurate diagnosis and realize appropriate treatment and management.journal articl
筋突起骨折を含む下顎骨骨折の 1 例
application/pdfThe incidence of mandibular fractures is the highest among facial bone fractures. Addi-tionally, most of mandibular fractures occur in the mandibular angle and condylar process. On the other hand, the incidence of fracture of coronoid process is extremely low. We experienced a case of mandibular fractures involving mandibular body, condylar process as well as the coronoid process in a man aged 3₉–years–old who had received strong direct external force to the mandible. Mandibular fractures usually occur in the condylar process and mandibular angle because direct external force is more likely to transmit to these re-gions. Based on the classification of mandibular fractures, the incidence of mandibularfractures involving coronoid process increases with an increased number of fractures lines that means complicated fracture. At the viewpoint of anatomical portion, direct external force dose not transmit to coronoid process; however, it is possible that direct external force may transmit coronoid process in the case of complicated fracture. In this case report, we considered the potential mechanism of fracture of coronoid process by using a three–dimensional finite element model of a human mandible stress distribution analysis.journal articl
The Role of Transient Receptor Potential (TRP) Channels in the Transduction of Dental Pain
application/pdfDental pain is a common health problem that negatively impacts the activities of daily living. Dentine hypersensitivity and pulpitis-associated pain are among the most common types of dental pain. Patients with these conditions feel pain upon exposure of the affected tooth to various external stimuli. However, the molecular mechanisms underlying dental pain, especially the transduction of external stimuli to electrical signals in the nerve, remain unclear. Numerous ion channels and receptors localized in the dental primary afferent neurons (DPAs) and odontoblasts have been implicated in the transduction of dental pain, and functional expression of various polymodal transient receptor potential (TRP) channels has been detected in DPAs and odontoblasts. External stimuli-induced dentinal tubular fluid movement can activate TRP channels on DPAs and odontoblasts. The odontoblasts can in turn activate the DPAs by paracrine signaling through ATP and glutamate release. In pulpitis, inflammatory mediators may sensitize the DPAs. They could also induce post-translational modifications of TRP channels, increase trafficking of these channels to nerve terminals, and increase the sensitivity of these channels to stimuli. Additionally, in caries-induced pulpitis, bacterial products can directly activate TRP channels on DPAs. In this review, we provide an overview of the TRP channels expressed in the various tooth structures, and we discuss their involvement in the development of dental pain.journal articl
Analysis of the Effect of Parathyroid Hormone (PTH) Treatment on Osteoblastogensis from Leptin Receptor-Positive Mesenchymal Stem Cells
松本歯科大学博士(歯学)2018甲第211号application/pdfBone marrow mesenchymal stem and progenitor cells (BM-MSPCs) maintain homeostasis of bone tissue by providing osteoblasts. Although studies have suggested that MSPCs are observed as leptin receptor (LepR)-positive cells in BM, these cells still contain non-MSPC populations. On the other hand, genetic lineage tracing approaches revealed that intermittent parathyroid hormone (iPTH) treatment induced osteoblastic differentiation from BM-MSPCs. However, little is known about the mechanistic details of these process in vivo. Here, we demonstrate that the LepR+MSPC population contains Runx2-GFPlow sub-population, which possesses higher stem cell activity, than the Runx2-GFP− stromal population. In response to iPTH treatment, LepR+MSPCs differentiate into Type I collagen a (Col1)+ mature osteoblasts. Along with osteoblastogenesis, the number of Col1+ mature osteoblasts increased around the bone surface, although most were characterized as quiescent cells. However, the number of LepR+MSPCs also increased in the vicinity of bone tissue, and the cell cycle was accelerated in these cells by iPTH treatment. The expression levels of osteoblastic markers were increased in the LepR+MSPC population in response to iPTH treatment. In contrast, the expression levels of markers for adipocytes decreased in this population. Altogether, our results indicate that Runx2 is weakly expressed in the LepR+MSPC population without osteoblastic commitment, and iPTH treatment expanded the LepR+MSPC population and skewed their lineage differentiation from adipocytes toward osteoblasts through cell cycle withdrawal.doctoral thesi
成長期の反対咬合に対して筋機能訓練装置の影響と効果を認めた 1 例
application/pdfAlthough functional jaw orthopedic appliances have been used to treat anterior cross bite during the growth and development period, its functional effect for severely anterior cross bite has not been clarified. Cases treated with a myofunctional training device for deciduous tooth anterior cross bite have been reported, but normally, these devices are not used to improve occlusion of patients with severly anterior cross bite. The patient was an 8-year and 4-month-old boy with Skeletal 3 anterior cross bite, Terminal plane Mesial step type with -3 mm over jet and +6 mm over bite, and ANB of -3.0° and U1 to SN of 86°. We proposed the conventional treatment method, but active treatment was rejected due to various background factors, and consent could not be obtained. Thus, we proposed the use of a myofunctional training device and consent was obtained. The attached device guided the abnormal pressure of various muscles of the oral cavity in an appropriate direction and promoted forward development of the upper jaw. ANB improved to +1.0°, curvature of Spee was flattened, and labial inclination of the upper teeth and inferior backward growth of the mandible were observed, which improved coverage of the upper tooth region that the patient most strongly desired.journal articl
Effects of shokyo (Zingiberis Rhizoma) and kankyo (Zingiberis Processum Rhizoma) on prostaglandin E2 production in lipopolysaccharide-treated mouse macrophage RAW264.7 cells
application/pdfWe previously reported that shokyo and kankyo, which are water-extracted fractions of ginger, reduced LPS-induced PGE2 production in human gingival fibroblasts. In this study, we examined the effects of these herbs on LPS-treated mouse macrophage RAW264.7 cells. Both shokyo and kankyo reduced LPS-induced PGE2 production in a concentration-dependent manner. Shokyo and kankyo did not inhibit cyclooxygenase (COX) activity, nor did they alter the expression of molecules in the arachidonic acid cascade. In addition, these herbs did not alter NF-κB p65 translocation into nucleus, or phosphorylation of p65 or ERK. These results suggest that shokyo and kankyo inhibit cPLA2 activity. Although 6-shogaol produced similar results to those of shokyo and kankyo, the concentration of 6-shogaol required for the reduction of PGE2 production were higher than those of 6-shogaol in shokyo and kankyo. Therefore, several gingerols and shogaols other than 6-shogaol may play a role in the reduction of LPS-induced PGE2 production. Thus, 6-shogaol, and other gingerols and shogaols inhibit cPLA2 activity and reduce LPS-induced PGE2 production via a different mechanism from traditional anti-inflammatory drugs. Moreover, kampo medicines that contain shokyo or kankyo are considered to be effective for inflammatory diseases.journal articl
