    application/pdfMetal binding agents, 2,3-dimercaptosuccinic acid (DMSA) and 2,3-dimercapto-1-propanesulfonic acid, Na salt (DMPS) were studied on the effectiveness for treatment of arsenic poisoning in mice. All mice which were given a toxic dose of arsenic trioxide (0.2 mmol As/kg, s. c.) survived by the treatment immediately after arsenic, with either one of these two agents (0.4 mmol/kg,i.p.). DMPS showed also the therapeutic effects against the acute poisoning with sodium arsenate and sodium monomethylarsonic acid.journal articl

    application/pdfCausative agents of oral infectious diseases are normal residents in the oral cavity. From the point of view that one of the conceivable factors which controls the ecosystem of normal flora is bacteriocin (or a bacteriocin-like substance), we have studied biological properties of some bacteriocins elaborated by the members of indigenous oral bacteria. Author report here an outline of the properties of several bacteriocins. (i) Bacteriocins of dental plaque bacteria Strains of St. sanguis, P. acnes, and B. matruchotii which produced bacteriocins, were isolated from dental plaque. M. W. of a bacteriocin from St. sanguis was 280,000. It inhibited the growth of Bacterodes, P. acnes, and Actinomyces bacteriostatically. A bacteriocin from P. acnes composed of five subunits. M. W. of the native from was 60,000. It's inhibitory spectrum was very narrow. B. matruchotii produced an extracellular bacteriocin by shaking cultivation. This purified bacteriocin contained two components. However, there was no significant difference in the inhibitory spectrum between the two components. (ii) Bacteriocin of gingival crevice bacteria About a half of B. melaninogenictis strains isolated from gingival crevice deposits exhibited antibacterial activity and the bacteriocin was obtained from cell extract of this organism. The purified bacteriocin was 105,000 in M. W. and inhibited the growth of Bacteroides, Actinomyces, St. mitis, and St. salivarius. It was also revealed that hematin accumulated within the cells of this organism was able to inhibit many gram positive bacteria including St. mutans. (iii) Bacteriocin of saliva bacteria In the culture supernatant of S. aureus obtained from saliva, a relatively low M. W. (6,000) bacteriocin was found It was basic polypeptide with a pi value of 10.0, and a high content of lysine was characteristic. This bacteriocin killed St. salivarius, P. acnes, A. israelii, and nonbacteriocinogenic 5. aureus. On the basis of this information, the possible ecological role of the bacteriocins or bacteriocin-like substances in oral microorganisms on the flora is discussed.journal articl

    Comparative Studies of Acid and Alkaline Phosphatases from Bacteroides melaninogenicus

    application/pdfAcid and alkaline phosphatases from an oral strin of Bacteroides melaninogenicus were purified to homogeneity from cell extracts and their properties were compared. Molecular weights of acid and alkaline phosphatases were estimated to be 62,000 and 160,000, respectively. Michaelis constant for p-nitrophenylphosphate of the acid phosphatase was 0.17 mM and that of the alkaline enzyme was 0.23 mM. The alkaline phosphatase was more stable than the acid phospatase when they were heated at 60℃. The activity of the acid phosphatase was largely reduced by Cu^ or fluoride and the alkaline phosphatase was quite sensitive to inhibition by Zn^, thiol compounds, or EDTA. The inactivation by EDTA of the alkaline phosphatase was restored with Ca^ or Mg^. The acid phosphatase hydrolyzed α-D-glucose 1.6-diphosphate, p-nitrophenylphos-phate, D-glucose 6-phosphate, and D-fructose 6-phosphate. On the other hand, p-nitro-phenylphosphate was the most suitable substrate for the alkaline phosphatase. Nucleoside triphosphates were also hydrolyzed by the alkaline phosphatase.journal articl

    application/pdfA multicentric tumor was spontaneously occurred in a 10-week-old female Wistar rat. It consisted of six, soft and dark-reddish-colored tubercles which ranged from soybean- to hen egg-size. They were so located near the mammillas that the tumor was suspected, of mammary gland origin. The animal was killed and the tubercles were examined by light and electron-microscopy. The tumor consisted of atypically proliferated cells forming a gland-like pattern which contained secreted materials. In some gland-like structures the tumor cells projected in papillary fashion into the spaces. Histochemistry on the materials revealed that these secreted materials contained both lipids and proteins. Based upon the above mentioned facts, this tumor was diagnosed as carcinoma papillotubulare which originated from the mammary glands. Electron microscopically the tumor cells were composed of gland cells and myoepithelial-cells. The former cells were rich in rough surfaced endoplasmic reticulum and contained many secreting granules which could be divided into electron-sparse large ones and electron-dense small ones.journal articl

    application/pdfThis survey was made on 1428 wisdom teeth of 581 undergradute students of Matsumoto Dental College (♂497, ♀84) who received dental clinic trainings in the years 1980, 1981, and 1982, respectively. 1) The eruption of wisdom teeth was observed in 1063 teeth of 500 students out 1328 teeth of 581 examinees. 2) The eruption of all 4 wisdom teeth was observed in 106 cases, while that of only the lower 2 in 34 cases. 3) In cases with all 4 wisdom teeth, most of the root apices completed their formation while in the impacted cases root apices were incompleted. 4) As to the inclination of the wisdom teeth examined, the following distribution was observed; vertical-889, mesical or horizontal-418, distal-72, cheek-tongue-49. 5) During the examinees' clinic practice, 278 wisdom teeth out of 1428 teeth were extracted (1150 teeth remaining). The number of wisdom teeth that had received surgical treatment was 284.
