    Quantitative Characterization of α-Synuclein Aggregation in Living Cells through Automated Microfluidics Feedback Control

    Aggregation of α-synuclein and formation of inclusions are hallmarks of Parkinson's disease (PD). Aggregate formation is affected by cellular environment, but it has been studied almost exclusively in cell-free systems. We quantitatively analyzed α-synuclein inclusion formation and clearance in a yeast cell model of PD expressing either wild-type (WT) α-synuclein or the disease-associated A53T mutant from the galactose (Gal)-inducible promoter. A computer-controlled microfluidics device regulated α-synuclein in cells by means of closed-loop feedback control. We demonstrated that inclusion formation is strictly concentration dependent and that the aggregation threshold of the A53T mutant is about half of the WT α-synuclein (56%). We chemically modulated the proteasomal and autophagic pathways and demonstrated that autophagy is the main determinant of A53T α-synuclein inclusions’ clearance. In addition to proposing a technology to overcome current limitations in dynamically regulating protein expression levels, our results contribute to the biology of PD and have relevance for therapeutic applications

    Molecular Detection of Babesia spp. (Apicomplexa: Piroplasma) in Free-Ranging Canids and Mustelids From Southern Italy.

    Babesiosis is an emerging tick-borne disease caused by apicomplexan parasites with widespread geographical distribution and various wildlife species as reservoir hosts. The aims of this study were to investigate the prevalence and assess the role of free-ranging canids and mustelids in the maintenance of Babesia spp. in southern Italy. PCR analysis of splenic samples targeting the 18S rRNA gene revealed the presence of Babesia spp. in 36 of 82 (43.9%) red foxes (Vulpes vulpes) including 29 (58%) from Campania region and seven (21.8%) from Calabria region, in seven of 13 (53.8%) Eurasian badgers (Meles meles), and in one of 13 (7.7%) gray wolves (Canis lupus). Samples from other host species including 9 Eurasian otters (Lutra lutra), 1 stone marten (Martes foina), 1 least weasel (Mustela nivalis), and 1 European polecat (Mustela putorius) tested Babesia spp. negative. Sequence analysis of the 18S rRNA gene demonstrated the presence of B. vulpes in the red fox and two sequence types of badger-associated Babesia spp. in the Eurasian badger. The Babesia sp. sequence detected in the gray wolf was identical to a badger-associated Babesia sp. This study shows that the number of Babesia spp. infecting free-ranging carnivores in Italy is higher than currently believed, and suggests that these hosts may play an important role in the maintenance of the sylvatic cycle of these parasites. It is the first report of badger-associated Babesia spp. in Italy and in a gray wolf

    Gegenstand der vorliegenden Bachelorarbeit sind amtliche Veröffentlichungen in elektronischer Form. Es wird die Frage beantwortet, wie diese Publikationen an regionalen Pflichtexemplarbibliotheken in Deutschland behandelt werden. Die Arbeit basiert auf der Auswertung einschlägiger Fachliteratur und Internetressourcen sowie auf der von der Verfasserin durchgeführten Umfrage von ausgewählten Pflichtexemplarbibliotheken. Es gilt unter anderem folgende Fragen zu klären: Werden bereits in allen Bundesländern amtliche Veröffentlichungen in elektronischer Form gesammelt? Wann begann die Sammlung amtlicher Veröffentlichungen in elektronischer Form? Wie wird bei der Sammlung vorgegangen? Welche Formate werden gespeichert und wo? Die Ergebnisse zeigen, dass bereits in fast allen Bundesländern amtliche Veröffentlichungen in elektronischer Form gesammelt werden, die Vorgehensweise variiert jedoch von Bundesland zu Bundesland.The subject of the present bachelor thesis are official publications in electronic form. In particular, the different practices of regional depository libraries are examined. The thesis is based on the analysis of relevant literature and internet resources as well as on the author's survey of selected depository libraries. Among other things, the following questions need to be answered: Are official publications already collected in electronic form in all federal states? When did the collection of official publications begin in electronic form? How does the collection work? Which formats are saved and where? The results show that official publications are already collected in electronic form in almost all federal states, but the procedure varies from state to state

    Die vorliegende Arbeit beschäftigt sich mit dem Metadatenmanagement in Bibliotheken. Es wird untersucht, inwiefern die Programme KNIME und Catmandu geeignet sind, Bibliotheken bei typischen Aufgaben des Metadatenmanagements zu unterstützen. Die technischen Entwicklungen im Bereich Metadaten sind aufgrund der Vielzahl an Formaten, Schnittstellen und Anwendungen komplexer geworden. Um die Metadaten entsprechend aufbereiten und nutzen zu können, werden Informationen über die Eignung verschiedener Programme benötigt. KNIME und Catmandu werden sowohl theoretisch analysiert als auch praktisch getestet. Dazu wird unter anderem untersucht, wie die Dokumentation gestaltet ist und welche Datenformate und Schnittstellen unterstützt werden. Im Anschluss werden verschiedene Szenarien aus den Bereichen Filtern, Analyse, Ergänzen von Inhalten und Anreicherung von Daten getestet. Die Arbeit zeigt, dass beide Programme unterschiedliche Stärken und Schwächen haben. Catmandus Stärke ist ein leichterer Einstieg in das Programm und vielfältige Optionen, bibliothekarische Datenformate und Schnittstellen zu nutzen. Ein Vorteil von KNIME ist, dass nach einer gewissen Einarbeitung viele Probleme schnell gelöst werden können und für zahlreiche Fälle spezielle Funktionen zur Verfügung gestellt werden.This thesis deals with metadata management in libraries. It examines to what extent the tools KNIME and Catmandu can be used to support libraries in typical tasks of metadata management. The technical developments in the field of metadata have become more complex due to the multitude of formats, interfaces, and applications. In order to prepare and use metadata, information about the suitability of different programs is needed. KNIME and Catmandu are both theoretically analyzed and practically tested. For this purpose it is examined, among other things, how the documentation is designed, and which data formats and interfaces are supported. Typical tasks like filtering, analysis, content enhancement, and data enrichment will be tested. The work shows that both tools have different strengths and weaknesses. Catmandu's strength is an easier introduction into the program and a variety of options for using library data formats and interfaces. An advantage of KNIME is that after an initial familiarization many problems can be solved quickly and special features are made available for numerous cases

    Cohesive law identification of adhesive layers subject to shear load – An exact inverse solution

    An exact general solution of the following inverse problem is obtained: given a couple of beams adhe- sively bonded along a surface parallel to their axes, subject to traction / flexure load, find the unknown interface shear force. This solution is used to identify the cohesive law that characterizes the behaviour of adhesive layers subject to shear load. In order to generate data to be used to evaluate the identifica- tion methodology, three decohesion virtual tests are performed by means of detailed FE simulations. The series of tests is related to standard test configurations, and shows the correctness of the inverse solution and the efficiency of the proposed methodology, as it is more precise than the data reduction method- ologies provided by literature. If ad hoc test configurations are designed, this identification methodology can be applied to adhesion between different materials and to adhesive layers with variable behaviour

    Advancing integration of data on food microbiome studies: FoodMicrobionet 3.1, a major upgrade of the FoodMicrobionet database

    We present a new version of FoodMicrobionet, a database for the exploration of food bacterial communities. The database, available as an app built with the Shiny package of R, includes data from 44 studies and 2234 samples (food or food environment), covering dairy, meat, fruit and vegetables, cereal based and ready-to-eat foods. The interactive interface allows exploration of data, access to external resources (on line versions of the studies, sequence data on SRA, taxonomic databases), ltering samples on the basis of a number of criteria, aggregation of samples and bacterial taxa and export of data in a variety of formats. FoodMicrobionet is the largest collection of data on food bacterial communities and, due to the structure of sample metadata, largely derived from the European Food Safety Agency FoodEx2 classi cation, makes comparison and re-analysis of data from published and unpublished studies easy. Data exported from FoodMicrobionet can be readily used for graphical and sta- tistical meta-analyses using open-source software (Gephi, Cytoscape, CoNet, and R packages and apps, such as phyloseq and Shiny-Phyloseq) thus providing scientists, risk assessors and industry with a wealth of information on the structure of food biomes

    Nel presente saggio si mira ad esaminare la teoria giusnaturalistica di Hans Welzel, che egli formula negli anni del secondo dopoguerra, al fine di arginare quel positivismo giuridico, che aveva predominato nel periodo del nazionalsocialismo. Più in particolare, il giusnaturalismo welzeliano si incentra sull’idea del diritto quale ordine valorativo, basato sull’autonomia morale dell’individuo. Tale idea è confortata anche dagli studi che Welzel conduce della teoria giusnaturalistica pufendorfiana e specialmente della sua distinzione fra etica e diritto
