An Innovative Medium-Altitude Long-Endurance Unmanned Aircraft System with Advanced Flight Management and Navigation Features
This paper presents preliminary activities in the framework of the research and development project entitled "Geared unmanned Aircraft with Novel Navigation unit and Engine on Tail - GANNET". It is a project funded by the Italian Ministry of Economic Development that involves two Italian aerospace companies such as Dallair and Wind Aeronautics. They are supported by University of Naples for the research activities. This project aims at developing an innovative Medium-Altitude Long-Endurance remotely piloted aircraft. The most important issue related to this project is the reduction of overall system costs both in terms of production, operation, and maintenance. Therefore, an approach similar to the one adopted for the development of manned ultralight aircraft has been adopted. In particular, two terms have been addressed, such as airframe and on-board system development. The first one was realized with a large use of advanced composite materials. Regarding onboard systems, an original Inertial Measurement Unit have been developed to provide accurate and high-integrity navigation measurement. This option is very important when strapdown payloads are adopted, such as Synthetic Aperture Radars. The resulting system is suitable for frequent surveillance missions over large areas, such as sea monitoring. Combined Blockade of Activating ERBB2 Mutations and ER Results in Synthetic Lethality of ER+/HER2 Mutant Breast Cancer.
PURPOSE:
We examined the role of ERBB2-activating mutations in endocrine therapy resistance in estrogen receptor positive (ER+) breast cancer.
EXPERIMENTAL DESIGN:
ERBB2 mutation frequency was determined from large genomic databases. Isogenic knock-in ERBB2 mutations in ER+ MCF7 cells and xenografts were used to investigate estrogen-independent growth. Structural analysis was used to determine the molecular interaction of HER L755S with HER3. Small molecules and siRNAs were used to inhibit PI3Kα, TORC1, and HER3.
RESULTS:
Genomic data revealed a higher rate of ERBB2 mutations in metastatic versus primary ER+ tumors. MCF7 cells with isogenically incorporated ERBB2 kinase domain mutations exhibited resistance to estrogen deprivation and to fulvestrant both in vitro and in vivo, despite maintaining inhibition of ERα transcriptional activity. Addition of the irreversible HER2 tyrosine kinase inhibitor neratinib restored sensitivity to fulvestrant. HER2-mutant MCF7 cells expressed higher levels of p-HER3, p-AKT, and p-S6 than cells with wild-type HER2. Structural analysis of the HER2 L755S variant implicated a more flexible active state, potentially allowing for enhanced dimerization with HER3. Treatment with a PI3Kα inhibitor, a TORC1 inhibitor or HER3 siRNA, but not a MEK inhibitor, restored sensitivity to fulvestrant and to estrogen deprivation. Inhibition of mutant HER2 or TORC1, when combined with fulvestrant, equipotently inhibited growth of MCF7/ERBB2 V777L xenografts, suggesting a role for TORC1 in antiestrogen resistance induced by ERBB2 mutations.
CONCLUSIONS:
ERBB2 mutations hyperactivate the HER3/PI3K/AKT/mTOR axis, leading to antiestrogen resistance in ER+ breast cancer. Dual blockade of the HER2 and ER pathways is required for the treatment of ER+/HER2 mutant breast cancer

Approcci affermativi e rilevanza del minority stress nel counseling psicologico con persone LGBT: una revisione della letteratura internazionale
L’acronimo LGBT viene utilizzato per indicare una vastissima gamma di individui – lesbiche, gay, bisessuali, transgender – che, nonostante intuibili differenze legate a diversi orientamenti sessuali e identità di genere, possono essere considerati come classe omogenea in ragione delle specifiche esigenze e delle peculiarità dei percorsi di sviluppo contrassegnati spesso da vissuti di stigmatizzazione e discriminazione. Negli ultimi anni è sensibilmente aumentata la richiesta di aiuto psicologico da parte delle persone appartenenti a tale fascia della popolazione, e ciò rende tanto più necessaria la definizione di obiettivi e strategie di intervento da considerare in tali casi. Il presente contributo intende evidenziare aspetti peculiari, criticità e contesti operativi connessi all’intervento psicologico-clinico affermativo con persone LGBT, con particolare riferimento all’ambito professionale del counseling

Clinical evolution and epilepsy outcome in three patients with CDKL5-related developmental encephalopathy
Aims. To further characterise CDKL5-related disorder, previously classified as an early-onset seizure variant of Rett syndrome, which is currently considered a specific and independent early-infantile epileptic encephalopathy.Methods. We describe the epileptic phenotype and neurocognitive development in three girls with CDKL5 mutations showing severe neurodevelopmental impairment, with different epileptic phenotypes and severity.Results. The patients differed regarding age at epilepsy onset, seizure frequency, duration of honeymoon periods, as well as EEG features. The honeymoon period, defined as a seizure-free period longer than two months, represented, in our case series, a good indicator of the epilepsy outcome, but not of the severity of developmental impairment. However, even during the honeymoon period, the interictal EEG showed epileptiform abnormalities, slowing, or a disappearance of physiological pattern. The natural history of CDKL5 disorder was compared between the three girls, focusing on the relationship between electroclinical features and neurological development.Conclusion. Our findings suggest that CDKL5 mutations likely play a direct role in psychomotor development, whereas epilepsy is one of the clinical features associated with this complex disorder
