Skripsi dengan judul Waris Beda Agama Di Kampung Pancasila Perspektif Maqashid Syari’ah (Studi Kasus di Desa Pogalan Kecamatan Kabupaten Trenggalek) yang ditulis oleh Muhammad Malik Farhan, NIM 126102212170, dengan pembimbing Dr. Hj. Nur Fadhilah, M.H.I.

In a previous study, the Na-P1 type zeolites were synthesized from aluminosilicate residues using an efficient and cost-effective process, exhibiting an excellent adsorption capacity for Ca2+ in comparison to commercial zeolites 13X and A. Building upon this, the current study evaluates their performance for the adsorption of various elements, including Ca2+, Mg2+, and NH₄⁺. The objective was to evaluate the performance of the Na-P1 type zeolites for the adsorption of various elements, including Ca2+, Mg2+, and rare earth elements, with a particular emphasis on the adsorption kinetics and water hardness removal in comparison to commercial zeolite A. The results demonstrated that the Na-P1 zeolite exhibited a satisfactory sorption capacity for Ca2+ and NH₄⁺ ions (66 mg/g), while displaying a relatively lower effectiveness for the sorption of Mg2+ ions (5.6 mg/g). The Langmuir model is particularly well suited to the sorption of Ca2+, while the Freundlich model is more appropriate for Mg2+. Both models demonstrated satisfactory representation of NH₄⁺ sorption. Moreover, the pseudo-second-order kinetic model provides an excellent description of the Ca2+ and Mg2+ sorption processes, while both models effectively describe the NH₄⁺ adsorption kinetics. Additionally, Na-P1 zeolite was observed to effectively reduce water hardness from 322 to 63 mg CaCO₃/L at temperatures of 10, 20, and 38 °C, and to 18 mg/L at 58 °C. These findings suggest that Na-P1 zeolite has promising potential for applications as a water softening agent. Regarding metals and rare earths, the Na-P1 zeolite demonstrated noteworthy sorption efficiencies for Cd2+ (138 mg/g), Ce3+ (209 mg/g), Cr3+ (56.2 mg/g), and Cu2+ (60.5 mg/g). However, it demonstrated lower sorption efficiencies for Co2+, Mn2+, Ni2+ and Dy3+ (below 16 mg/g). The findings illustrate that Na-P1 zeolites are effective for the adsorption of diverse elements, offering a promising avenue for the sustainable transformation of industrial waste into valuable materials for environmental applications

An estrogenic municipal effluent decreased fathead minnow reproduction to a near stop.

Municipal wastewater treatment plants (WWTPs) release endocrine disrupting chemicals (EDCs) which can impact aquatic species’ reproduction (e.g., decrease fecundity). As bioassays can quantify the cumulative effect of mixtures, such as effluents, this study proposes a two-Tier bioassay-based approach to assess EDCs in WWTP effluents. The first Tier is composed of the human receptor transactivation assay for estrogen (hERα) and androgen (hAR), and the Tier 2 comprises an abbreviated 7-day fish short-term reproduction assay (FSTRA; OECD TG 229) in fathead minnow (Pimephales promelas). The approach was assessed on a municipal effluent (12.5, 25 and 50% v/v) and compared to the standard 21-day FSTRA and chemical analysis. The first Tier detected mainly estrogenic activity at 6.7–51.5 ng/L of estrogen equivalent (EEQ). The 7-day FSTRA of Tier 2 was able to capture the same arrest in reproduction in the 50% v/v treatment as the 21-day FSTRA. Moreover, around halfway of the 21-day FSTRA, the 12.5% v/v treatment led to a reproduction arrest similarly to the positive control (12.5% v/v + 40 ng/L of 17α-ethinylestradiol (EE2)). During the 21-day experiment, bisphenols, nonylphenol and 19 pharmaceuticals were detected. Altogether, the results indicate the Tier 1 methodology was robust to identify potential endocrine disruptive activity of the effluent, while the Tier 2 was able to detect reproduction arrest without a clear mechanism of action. Overall, this two-Tier approach provides an initial framework to assess EDCs levels in WWTP effluents, which will help inform on the risk of complex mixtures to protect aquatic ecosystems

Pharmacological characterization of cannabidiol as a negative allosteric modulator of the 5-HT2A receptor

Promising clinical evidence suggests that psychedelic compounds, like lysergic acid diethylamide (LSD), have therapeutic value for treatment of psychiatric disorders. However, they often produce hallucinations and dissociative states, likely mediated by the serotonin (5-HT) receptor 5-HT2A, raising challenges regarding therapeutic scalability. Given the reported antipsychotic effects of cannabidiol (CBD) and its promiscuous binding at many receptors, we assessed whether CBD could modulate 5-HT2A signalling. Activation of the 5-HT2A intracellular signalling events were assessed using resonance energy transfer- or fluorescence-based biosensors in HEK 293 cells and in rat primary cortical neurons. In 5-HT2A-transfected HEK 293 T cells, CBD antagonized LSD-mediated Gq activation in a saturable way, while leaving β-arrestin2 recruitment unaffected. CBD decreased Gq activation mediated by the 5-HT2A-specific agonist DOI as well as LSD-mediated activity in primary rat neonatal cortical neurons. Using Site Identification by Ligand Competitive Saturation (SILCS) simulations, we also predicted that the putative binding site of CBD overlapped with that of oleamide, a positive allosteric modulator of 5-HT2A, and could displace the binding of orthosteric ligands toward the external binding pocket. Based on these findings, we propose that CBD acts as a negative allosteric modulator of 5-HT2A.</br

TGFβ1 Restores Energy Homeostasis of Human Trophoblast Cells Under Hyperglycemia In Vitro by Inducing PPARγ Expression, AMPK Activation, and HIF1α Degradation

Elevated glucose levels at the fetal-maternal interface are associated with placental trophoblast dysfunction and increased incidence of pregnancy complications. Trophoblast cells predominantly utilize glucose as an energy source, metabolizing it through glycolysis in the cytoplasm and oxidative respiration in the mitochondria to produce ATP. The TGFβ1/SMAD2 signaling pathway and the transcription factors PPARγ, HIF1α, and AMPK are key regulators of cell metabolism and are known to play critical roles in extravillous trophoblast cell differentiation and function. While HIF1α promotes glycolysis over mitochondrial respiration, PPARγ and AMPK encourage the opposite. However, the interplay between TGFβ1 and these energy-sensing regulators in trophoblast cell glucose metabolism remains unclear. This study aimed to investigate whether and how TGFβ1 regulates energy metabolism in trophoblast cells exposed to normal and high glucose conditions. The trophoblast JEG-3 cells were incubated in normal (5 mM) and high (25 mM) glucose conditions for 24 h in the absence and the presence of TGFβ1. The protein expression levels of phosphor (p)-SMAD2, GLUT1/3, HIF1α, PPARγ, p-AMPK, and specific OXPHOS protein subunits were determined by western blotting, and ATP and lactate production by bioluminescent assay kits. JEG-3 cells exposed to 25 mM glucose decreased ATP production but did not affect lactate production. These changes led to a reduction in the expression levels of GLUT1/3, mitochondrial respiratory chain proteins, and PPARγ, coinciding with an increase in HIF1α expression. Conversely, TGFβ1 treatment at 25 mM glucose reduced HIF1α expression while enhancing the expression levels of GLUT1/3, PPARγ, p-AMPK, and mitochondrial respiratory chain proteins, thereby rejuvenating ATP production. Our findings reveal that high glucose conditions disrupt cellular glucose metabolism in trophoblast cells by perturbing mitochondrial oxidative respiration and decreasing ATP production. Treatment with TGFβ1 appears to counteract this trend, probably by enhancing both glycolytic and mitochondrial metabolism, suggesting a potential regulatory role of TGFβ1 in placental trophoblast cell glucose metabolism.</br

Natural organic matter dynamics in permafrost peatlands: Critical overview of recent findings and characterization tools.

    No full text
Rising temperatures are destabilizing permafrost in northern latitudes, leading to the mobilization, transformation and cycling of natural organic matter (NOM), nutrients, and contaminants into newly formed aquatic systems. Analyzing the chemical composition of organic matter is crucial for understanding the biogeochemical processes at play. Furthermore, it is essential to investigate how seasonal variations and anoxic conditions influence these processes, as well as their effects on microbial activity and NOM composition. This review provides an overview of northern peatlands, terminal electron acceptor species, and key analytical techniques used to characterize organic matter: UV/Vis and Fluorescence Spectroscopy, FTIR, FT-ICR-MS, and Nuclear Magnetic Resonance. Rather than focusing on the theoretical aspects of these techniques, we emphasize the type of information they offer about NOM and how to interpret these data within the context of biogeochemical transformations in permafrost-affected systems
