In recent years, several severe global environmental problems are proceeding and one of the 
solutions is a shift to a sustainable economic system utilizing biotechnology. Biotechnology 
utilizing valuable microorganisms can continuously produce food and chemical compounds using 
bioproducts. Among the production process of bioproducts, the scale-up examination is important 
for production on an industrial scale. To increase the yield of useful material produced in large 
cultivations, monitoring the internal environment of a large cultivation tank (jar fermenter) using 
various sensors in real-time and controlling optimum cultivation conditions based on the results of 
monitoring are effective. There are various types of indicators for monitoring the jar fermenter. 
Since microorganisms respond to the extracellular environment and the influence is reflected in 
the microbial morphology, the microbial morphology may be utilized as a more useful indicator of 
microbial cultivation than the other conventional monitoring indicator (pH, dissolved oxygen, etc.). 
However, a trade-off exists between speed and image quality when acquiring morphological 
information, which it was difficult for the microbial morphology to be acquired at high-definition 
continuously as well as other sensors. In this thesis, the author aimed to develop a real-time 
monitoring system in a jar fermenter utilizing microbial morphology. 
 Chapter 1 describes the background of this research, especially the monitoring method for 
a large-scale cultivation of microorganism, and mentions the significance of this study. 
Chapter 2 describes a module that can acquire microbial morphology in high-definition 
from one-batch cultivation in the jar fermenter by using a microscope, a developed microfluidic 
device, and a high-speed camera. By using this module, the following points were demonstrated: 
1) Images could be acquired with a sufficiently high resolution to observe organelles of cell in the 
flowing cultivation medium through the microfluidic device. 2) A large number of images could be 
captured at high speed, and more than 10,000 cell images could be acquired in 2.2 seconds in one 
take. This image acquiring rate was comparable to cell sorter. The developed module could acquire 
morphological parameters of microbes in real time like the other general monitoring sensors. 
Chapter 3 describes the estimation of the culture state (growth or materials productivity) 
through analysis of images acquired using the developed real-time monitoring system. For image 
analysis, a deep learning was applied. Oleaginous yeasts were used for the demonstration since 
these yeasts are some of the most capable for industrial use and insights in the relationship between 
their morphology and their growth or lipid-productivity has not been clarified. The deep learning 
image analysis with training data showed a high correlation between the specific cell image and 
the growth/lipid productivity of the microorganisms. In addition, image analysis using deep 
learning without training data also confirmed a high correlation between cell image and

growth/lipid productivity. Moreover, there seemed to be species-dependent differences in the 
relationship between the morphology and the state of cells.  
Finally, in chapter 4, the results presented in this thesis are summarized and discussed the 
usefulness of these developed modules and future issues.

Nowadays, energy harvesting methods are being developed from natural renewable 
energy resources owing to the demand for sustainable energy. Especially, piezoelectric, 
triboelectric, and pyroelectric nanogenerators that convert thermal and mechanical energies 
could be produced by adding a small-scale physical change into electricity with the usage of 
digitally enhanced applications.  
A novel electro-thermodynamic power generation method that uses Diode and two 
Switches circuit (DSW circuit) with higher power generation is more efficient than the Olsen 
electro-thermodynamic cycle method. The temperature differences and the behavior of the 
external electric field are very important parameters to optimize the amount of generating 
power. Therefore, the purpose of this study is to improve pyroelectric power generation by 
using waste heat energy as a renewable energy source in combination with pyroelectric 
nanogenerators and an external pulse electric field. Under optimal conditions, the maximum 
power can be accumulated by applying different pulses of the external electric field at 
temperature variations on the surface of the pyroelectric body since there is no reported 
pyroelectric power generation experiment in any materials. The experimental process is 
performed for measuring and calculating to power generation of the pyroelectric body using 
a newly developed experimental setup based on the modified DSW circuit.  
A standard Fuji ceramic C-9 sample was employed to generate pyroelectric power in 
an environment with continuous temperature variation from low (120 °C) to high (140 °C), 
with a temperature difference of 20 °C. The continuous temperature variation frequency was 
0.05 Hz, and the pulsed electric field was applied when the temperature rose. The pulse 
width of the electric field 10, 20, 50, 100, and 200 ms pulses were applied to the sample 
under single pulse and multi-pulse conditions. And the amplitude of the pulses was 250, 500, 
1000, and 1500 V/mm. Finally, the maximum power generation condition was evaluated by 
comparing single-pulse and multi-pulse trains.  
 The maximum power generated through the application of an external pulsed 
electric field under the above conditions was evaluated. Finally, a technique for generating 
pyroelectric energy was devised by applying a pulsed electric field using a pyroelectric body. 
This power production system had the highest power density of 0.204 mJcm-2°C-1kV-1 in 
the C-9 sample. Also, for the lowest input power, the maximum power generation condition 
was a 10 ms pulse width and an amplitude of 250 Vmm-1 in the applied electric field. When 
applying the multi-pulse to the sample, the power generation density was maximum for the

highest number of pulses at the low voltage (250 V/mm) input of the pulse electric field. 
Furthermore, all the results showed that the power generation rate decreased as the pulsed 
electric field input voltage rose. The greatest power generating capability ratio (η) value for 
a 250 V/mm pulse electric field was 7.834 at the highest number of pulses. The results show 
that the η ratio is greatest for a high number of pulses in a low voltage (250, 500 V/mm) 
input pulsed electric field. Finally, it was determined that a multi-pulse electric field might 
enhance the performance of the pyroelectric power generating system. 
Consequently, this condition might be used to power smart sensor modules, Internet 
of Things devices, automobiles, and other waste heat energy applications. When the sample 
is subjected to nano-pulse electric field application, the input power may be lowered to its 
lowest level depending on the net-producing power. As a result, new researchers can exploit 
the net-generation power efficiency to develop a large-scale power generation source

Improvement of Power Recovery by Applying Pulse Electric Field of Thermoelectric Cycle Power Generation

opera t ion in  harsh env ironments  needs to  be  accura te ly  assessed .  In  par t icula r,  
re-evaluat ing the ro le  of  so lute  e lements  in  the mater ia ls  used for  RPV is  the key  
to  the  decis ion  to  extend the ir  opera t ion  t ime.  However,  the  da tabase  on  the  
harden ing  and  embri t t lement p rocess  of  RPV mater ia ls  i s  s t i l l  lack ing.  In  o rder  to  
cont inue improving the model  of  p rocesses  occurr ing in  RPV materia ls  such as:  
format ion  mechanism of  so lu te  c lusters ,  in teract ion be tween  so lute  and dis locat ion  
loop  (DL),  inf luence of  solu te  c lus te r  and DL on ag ing  of  mater ia ls ,  i ron model  
a l loys  i r rad ia ted  by Fe 2 +  ion were  se lec ted .  The condit ions wi th  higher  tempera ture  
(400 oC) and h igher  dose (2  dpa  in  average)  than the ac tual  work ing  condi t ions  in  
RPV were chosen  to  accelera te  the p rocesses  occurr ing in  the mater ia l  and  
increases  the s ize  o f  the so lute  c lus te rs  as  well  as  d is locat ion  loop .  
( I)  Three-d imens iona l  a tom probe  tomography (APT) i s  an  excel lent  
technique for  iden ti fy ing nanometer -s ized so lute  a tom enrichment regions ,  but  the 
relat ionship  be tween  these nano -fea tures  and  c rys ta l  o r ientat ion  has not  been  wel l  
analyzed,  except  for  in  some semiconductor  mater ia ls .  In  the f i rs t  par t  of  th is  thes is ,  
a  method that  combines crysta l  o r ienta t ion analys is  of  f ie ld  desorp tion map and  
geometr ica l  ca lcu la t ions to  improve  the  recons truc t ion parameters  for  APT 
analysis  was app lied .  Spher ica l  Cu -r ich c lus te rs  and p lanar  Mn and Ni  segrega t ions  
on d is loca t ion loops in  Fe model  a l loys  af te r  ion i rradiat ion  were  used as  ind ica tors  
for  th is  ana lys is .  The matr ix  a toms were  well  recons tructed.  In  add i t ion,  
Cu-enr iched  regions  formed as  d iscre te  c luste rs ,  whi le  Mn - and  Ni-enr iched  
regions tended to  be connected as  part  of  a  torus.  The Cu -rich clusters  had an ova l  
shape wi th  the smal le r  dimens ion a long the  z -ax is ,  which may be due to  the  
diffe rence in  the f ie ld  evapora t ion be tween Cu(I)  and Fe( II ) ,  so  cal led “ loca l  
magnif ica t ion e ffect” .  The dis t r ibut ion of  solu te  a toms in  the i ron la t t ice  was  
consis ten t  with  our  knowledge in  prev ious s tudies ,  conf i rming the val idi ty  of  th is  
method for  analyzing  minori ty  e lements  dis t r ibu ted  in  meta l  a l loys .  
( II )  In  reac tor  pressure  vessel  mater ia ls ,  the  format ion of  Mn - and  Ni- r ich  
nanoclus ters  i s  a  major  cause of  neu t ron ir rad ia t ion embr i t t lement.  The segrega tion  
of  these solu te  a toms into  d is locat ion  loops has a t t rac ted  a t tent ion  as  a  mechanism 
to accele ra te  solu te  c luste r ing.  In  the second part ,  the  behaviors  o f  solu te  Mn and  
2 
Ni atoms in  Fe -0.6wt.%Ni,  Fe -1.4wt.%Mn, and Fe -1.4wt.%Mn-0.6wt.%Ni a l loys  
i r rad iated a t  400  C up to  3  dpa were ana lyzed using  th ree -dimensiona l  a tom probe  
tomography.  So lu te  a tom clus ters  were observed in  a l l  mate r ia ls ,  and  the ir  shapes 
were spher ica l ,  f la t ,  and torus in  FeNi,  FeMn,  and FeMnNi,  respec t ively.  In  te rnary  
al loy FeMnNi,  Mn and Ni a toms were concentrated in  the sample in  the form of  a rcs ,  
and the o r ien tat ion  of  the plane conta in ing  the arcs  was est imated by compar ing  
f ie ld  desorp t ion images .  The  s ize ,  number dens i ty,  and  or ien ta t ion of  this  s tructure  
were found to  be in  good agreement with  those of  both types of  d is locat ion loops,  
namely,  b=1/2<111> and b=<100>,  ident i f ied in  a  previous s tudy using the same 
mater ia l .  The  pos i t ions of  Ni and Mn enrichment  d id  no t  fu l ly  overlap .  Ni  a toms 
tended to  be concentra ted more in  the inner  part  o f  the loop than the Mn atoms.  Mn 
atoms were enr iched  on ly in  the v ic ini ty  of  the dis loca t ion loops,  whereas Ni a toms 
showed a  h igher  concentra t ion ins ide  the d is locat ion loops than in  the bulk.  
( II I)  Las t ly,  the cor rela t ion be tween  microst ruc ture  and  mechanica l  
propert ies  o f  RPV materia ls  has  been  modeled.  In  RPV mater ia ls ,  the  most  common 
types  o f  defects  tha t  can lead  to  i r rad ia t ion - induced hardening  a re  po int  defec ts  and  
dis loca t ion loops (DLs).  However,  the ir  respect ive  con tr ibu tions to  the overa l l  
harden ing  of  RPV materia ls  have no t  been clearly  de termined.  The s tudy employed  
transmission e lec tron microscopy (TEM),  a tom probe tomography  (APT),  and  
nanoindentat ion  to  assess  the  roles  of  SCs  and  DLs in  con tr ibu t ing  to  the harden ing  
behavior  of  RPV mater ia ls .  The resul ts  ind ica ted  that  bo th so lute  c luste rs  (SCs) ,  
which a re  formed by  the aggregat ion  of  po int  defec ts ,  and DLs play equa lly  
important  ro les  in  the harden ing behavior  of  RPV mater ia ls .  However,  an in -dep th  
analysis  of  three -d imensiona l  images us ing the APT method revea led that  there  may  
have been e rrors  in  evalua t ing SCs and DLs,  leading to  double  count ing when  
assess ing the  impac t  o f  i r rad iat ion -induced  defec ts  on the  hardening behavior  of  
RPV mater ia ls .  These  f indings h igh l igh t  the need  for  improved  accuracy in  
evalua t ing these defec ts  to  be t ter  unders tand the behavior  o f  RPV mater ia ls  under  
i r rad iat ion .  
Overal l ,  th is  thesis  addressed the c r i t ica l  issue  of  assess ing the integr i ty  of  the RPV 
in nuclear  power  p lan ts ,  par t icular ly  regard ing the ro le  o f  so lute  e lements  in  RPV 
mater ia ls  and the ir  impac t  on long - te rm opera t ion in  harsh env ironments .  By  
conduct ing  experiments  us ing acce lerated condi t ions,  we a imed to  improve  the  
unders tand ing of  the processes  occurr ing in  RPV mater ia ls  and enhance the ex is t ing  
knowledge of  solute  c lus te r ing,  solu te  a tom dis tr ibu tion ,  and  the behavior  of  
defec ts  under  i r rad iat ion.  The f indings prov ided va luab le  ins igh ts  into  solu te  
c lus te r ing,  solu te  a tom dis t r ibu tion ,  and the ro le  o f  defec ts ,  enhancing the

knowledge  base necessary  fo r  accurate ly  assess ing  the in tegr i ty  and po ten tia l  
extension of  the operat iona l  l i fespan of  RPVs.  The outcomes of  th is  research have  
the poten t ia l  to  inform decis ion -making processes  regard ing  the sa fe  and e ff ic ien t  
opera t ion of  nuc lear  power p lan ts .  Further  inves t igat ions bui ld ing upon  this  
research wi l l  be crucia l  in  cont inual ly  improving the unders tand ing and assessment  
of  RPV materia ls  fo r  the fu ture .

本研究では、酸化物系全固体ナトリウム電池の創製をターゲットにしている。酸化物全
固体電池では、活物質と固体電解質の間に緻密で抵抗の小さな界面を形成することが困難
であり、現在様々な手法が検討されている。その中で、本研究ではレーザー照射を利用し
た新しい界面形成プロセスを提案し、従来技術と比較しその優位性を実証した。活物質に
は、安価で優れた電池性能を示すことが報告されているリン酸鉄ナトリウム正極活物質を
選択し、レーザーの特徴を利用した結晶の構造変化や活物質層の緻密化により活物質の電
池性能の向上を報告している。学位論文の構成は以下のとおりである。  
1 章では、現行の二次電池の課題と次世代の二次電池の必要性を述べた。理想的な次世
代の二次電池として全固体電池を挙げ、全固体電池の研究進捗について記述した。また、
本研究の目的および着想に至った経緯について述べ、具体的な論文の構成を示した。  
2 章では本研究で用いた装置、解析手法および実験方法についてまとめている。  
3 章では、Na2FeP2O7 にレーザー照射した際の構造変化の評価と活物質 /固体電解質間の
界面形成について評価している。Na2FeP2O7 結晶はナトリウムイオン二次電池（Sodium ion 
battery, SIB）の正極活物質として優れた電池特性を示す材料であり、近年世界で初めて報
告されたオール酸化物ナトリウム全固体電池の活物質としても利用された。固体電解質上
の Na2FeP2O7 にレーザー照射することで、活物質 /固体電解質の間に緻密な接合界面を形成
することができた。また、レーザー照射した Na2FeP2O7 はガラス化していることが明らか
となり、レーザー照射による加熱後の冷却速度は再結晶化を妨げるほどの急冷が達成でき
ていることを明らかとした。レーザー照射によりガラス化した Na2FeP2O7 は再度結晶化温
度で熱処理することで可逆的に Na2FeP2O7 が析出することを確認した。還元剤や導電助剤
の役割を持つグラファイトを添加した Na2FeP2O7 にレーザー照射すると、加熱時に還元剤
として機能し、レーザーの到達距離に影響を及ぼすことを明らかとした。レーザーの照射
条件と構造変化領域の厚さにも相関があることを見出し、活物質層の厚さを制御できるこ
とがわかった。  
4 章では、SIB の正極活物質として不活性であることが知られているマリサイト NaFePO4
に着目している。この結晶は、155 mAhg-1 という高い理論容量を有しているが、ナトリウ
ムイオン伝導を阻害する孤立した PO4 ユニットが周期的に存在するため、正極活物質とし
ては不活性である。そこでこの章では、マリサイト NaFePO4 にレーザー照射し、その構造
を変化（特にガラス化）し、高いナトリウムイオン伝導性を有するガラス状（非晶質）
NaFePO4 正極材料を新たに作製し、相変化と電気化学的評価を行った。また、ガラス状の
NaFePO4 を固体電解質基板上に形成する。実際にマリサイト NaFePO4 にレーザー照射を行
うと、マリサイト NaFePO4 からガラス化した NaFePO4 を形成できることを見いだした。こ
の結果から、一般的な溶融急冷法と比較して、レーザー照射による加熱後の冷却速度は、
より広い範囲の組成でリン酸鉄ナトリウムをガラス化できることを実証した。また、ガラ

ス化した NaFePO4 中に Na+イオンの 3 次元伝導経路が形成されていることがわかり、NIB
の正極材料としての可能性が示された。さらに、固体電解質上のマリサイト NaFePO4 に固
体電解質と導電助剤を添加した電極合材にレーザー照射したところ、緻密な界面形成が達
成されただけでなく、活物質層で固体電解質とガラス化した NaFePO4 の間に大きな接触界
面が形成されていることがわかった。  
５章では、ガラスフォーマーを一切含まないβ-NaFeO2をSIBの正極活物質として選択し、
粒界抵抗を低減してNa+イオン伝導性を向上させることを目的に、レーザーにより溶融急
冷し、緻密化を行った。レーザー照射によりβ-NaFeO2を溶融急冷すると、メルトプールか
ら試料表面方向に向かって結晶が異方的に成長していることがわかり、さらに粒界が減少
して緻密化され、ナトリウムイオン伝導性が向上することを明らかとした。また、レーザ
ー照射したβ-NaFeO2は結晶性が低くなり一部非晶質化している可能性が高く、再加熱処理
によりα-NaFeO2およびNa3Fe5O9結晶を選択的に得られることがわかった。β-NaFeO2を非晶
質化したような報告はこれまでなく、非常に興味深い結果である。さらに、固体電解質基
板上の遷移金属酸化物であるβ-NaFeO2をレーザー照射で溶融固化することで固体電解質
に無加圧で接合でき、緻密な界面と活物質層を得ることができた。一般的に焼結に高温・
長時間の焼成が要される遷移金属酸化物を活物質として選択した場合でも、容易に固体電
解質上に緻密な活物質層を形成できることを証明する結果となった。この結果は、今後の
全固体電池の研究において活物質の材料選択の幅を広げ、焼成では困難な界面抵抗の低減
が期待される。  
6章では本論文を総括している。

世界中のあらゆるものがインターネットにつながる IoT（ Internet of Things）技術の発展
により，さまざまな分野での安全性，生産性や品質の向上が期待されている．酸素センサ
は産業，医療，安全，科学といった多くの分野で利用されているが，比較的大型で高価な
ものが多く，IoTのような多点計測の用途では使用が限られている．そこで新たなセンシン
グ方式として，ホットスポット現象を利用した酸素センサの実現が期待されている．当該
センサは極めて単純な構造であり，小型で安価なセンサが実現できる可能性がある．しか
しながら，実用化にあたり主な課題が二点あげられる．一つ目は耐湿性である．センサ材
料であるREBa2Cu3Oy（RE：希土類元素）は水蒸気に弱く，40 ℃ /93 %RH 雰囲気で50 h以
内に材料の劣化が生じてしまう．二つ目は省電力化である．現状の当該センサの消費電力
は約0.9 Wと大きく，携帯して使用する際にはバッテリ容量が問題となる．そこで本研究で
は、ホットスポット式酸素センサの耐湿性向上および省電力化を目的とした。本論文は，
以下の７章から構成される．  
第１章では，本研究の社会的背景と必要性，酸素センサにおける各検知方式の特徴を整
理するとともに，研究対象であるホットスポット式酸素センサの特長，先行研究を説明し，
当該センサの課題と本研究の目的を述べた．  
第２章では，本研究で用いた共通する実験方法についてまとめた．まず，材料の合成方
法，線材試料の作製方法について説明した．次に，材料の構造，電気的特性，熱的特性，
化学的特性に関する評価，分析方法について説明した．最後に，BVS（Bond Valence Sum）
による結晶構造の解析方法，FEM（Finite Element Method）による電気的および熱的特性の
計算方法について説明した．   
第３章では，BaサイトをNdで置換した組成Nd1+xBa2−xCu3Oyに着目し，耐湿性への影響を
調査した．その結果，BaサイトをNdで置換することで，耐湿性が大きく向上することを見
出した．Nd置換による結晶構造への影響について，BVSを用いて調査した．これにより，
Ndで置換されたBaサイトは，他のサイトに比べ周囲のギャップが大きい可能性が示された． 
Nd3+はBa2+に比べて高原子価であるため，Ndで置換されたBaサイトはOH−のトラップサイ
トとして作用し，水蒸気によるBaの劣化反応を抑制している可能性を明らかにした．また，
Ndよりもイオン半径の大きなBaイオンが，表面近傍のサイトを占めている可能性を示した．  
第４章では，Nd1+xBa2−xCu3Oy基セラミックスの線材を作製し，ホットスポット式酸素セ
ンサとしての特性を調査した．その結果，BaサイトをNdで置換することは，耐湿性向上と
センサ特性の維持を両立できることを明らかにした．Nd置換がガス感度に影響をおよぼさ
なかった理由は，酸素分圧によるホール密度の変化から説明した．また，Nd置換による電
流–電圧特性への影響や，酸素分圧が0.1 kPa以下の際に電流が流れなくなり酸素応答性が
失われる現象のメカニズムを明らかにした．

第５章では，Nd1+xBa2−xCu3Oy基セラミックスのセンサ層を断熱層で挟んだサンドイッチ
構造の線材を作製し，その省電力効果とセンサ特性を調査した．その結果，当該構造はガ
ス感度を損なわずに消費電力を約26%低減できることを実験的に示した．この理由は，サ
ンドイッチ構造におけるセンサ層内の電力密度は従来構造よりも高いため，同電力であっ
てもサンドイッチ構造の方がホットスポット部の内部温度が高く保たれるためと考えられ
る．加えて，当該構造は低電力であっても従来構造と同等のガス応答時間が得られること
を実証し，その理由がセンサ材料中での酸化物イオンの自己拡散係数に関係していること
を示した．  
第６章では，FEMによる当該センサの計算手法を確立し，サンドイッチ構造にくびれを
設けたネック構造による省電力化と低電圧化のポテンシャルを示した．その結果，ネック
構造は，サンドイッチ構造よりもさらに省電力化，低電圧化が可能であることを明らかに
した．当該構造は，同ガス感度において電力は46%低減，電圧は34%低下できると見積も
られる．加えて，ネック構造に類似したサンドイッチ構造に切り欠きを設けた構造におい
て，9 V，0.27 Wでセンシング可能なセンサを実証した．これは，角型9 V電池（340 mAh）
で11 h動作可能な消費電力に相当する．当該構造においても計算結果と実験値はよく一致
し，計算の妥当性が示された．   
第７章は総括であり，本研究で明らかになった結果および今後の課題をまとめた．

本論文は、医薬品の原料 として有用であ り、 ヒ トを含む哺乳動物に対 して有益な生理作
用が報告 されている D‐ アスパ ラギン酸 (D‐Asp)の 乳酸菌を用いた発酵生産法 と新規な機
能性食品の開発を 目的 とし、乳酸菌培養液における D―Aspの酵素学的定量法を開発 して食
品か ら D‐Aspを 多量に分泌生産す る乳酸菌を単離す るとともに、その D‐Asp高 生産機構に
ついて解析 した一連の研究成果をまとめている。
序章では、 D―Aspを含む D‐ ア ミノ酸の乳酸 菌を含む微生物や動物における生理機能 、生
理作用や生合成機構について概説す るとともに、D―Aspを 含む D‐ ア ミノ酸の検出・定量法、
生産方法や応用的な利用について も概説 し、本研 究の 目的 を示 している。
第 1章 では、 D―Aspに 高い特異性 を有す る酵 母 C′ッ′′οcο cc″s力″″たο′α UJl株 由来の D‐ ア
スパ ラギン酸 オキシダーゼ (ChDDO)を 用いて、乳酸菌培養液の D―Aspの 簡便かつハイス
ループ ッ トな定量法を開発 している。発色基質 として T00Sと 4-aminoantipyrineを 用いた、
ChDDOと 西洋 ワサ ビペルオキシダーゼの共役反応による、乳酸菌培養液の D―Aspを 簡便
かつ迅速に検 出・ 定量可能 な方法が開発 され ている。 さらに、新潟県内の野菜・ 果物や 発
酵食品か ら単離 された 628株 の乳酸菌株 のなかか ら、開発 した D‐Asp検出・ 定量法を用い
て、既知の D‐Asp高生産乳酸菌の約 2.6倍 の D‐Asp生産能 を有す る乳酸菌 WDN19株 の取
得に成功 している。 また、乳酸菌の D―Asp生産能は属や種ではな く、株 レベルで異なるこ
とを明 らかに している。
第 2章 では、WDN19株 を同定 し、近縁株の ια′〃ar′οb“″′″sο″″ッαr″s DSM 20019株 との
比較解析か ら、WDN19株の D‐AsP高生産機構 について解析 している。WDN19株のゲノム
配列 と生化学的な表現形の解析か ら、WDN19株は ια′′α
`′
οbασ′′″s属 に属する乳酸菌であ
り、ι.c″′να′″sに最 も近縁であることを明 らかに している。 さらに、ι.`″″να′″s基準株 で
ある DSM 20019株 の比較解析か ら、D―Aspは アスパラギン酸 ラセマーゼ (RacD)に より L―
AsPか ら生産 され ること、WDN19株における RacD活 性は DSM 20019株 よ りも高い こと、
また この理由 として WDN19株における RacD発 現量が高いことに加 え RacDそ の ものの
活性が高いことに起因す ることを明 らかに している。加 えて、WDN19株におけるアスパ ラ
ギナーゼ活性 が DSM 20019株 よりも高いこと、WDN19株のゲノム上のアスパ ラギン酸ア
ミノ トランスフェラーゼ遺伝子が トランスポ ゾン挿 入に よ り破壊 され ている ことによる
D‐Asp前駆体 となる L―Aspの RacDへ の高い供給量 も WDN19株 の高い D―Asp生 産能に寄 与
す ることを明 らかに している。
よって、本論文は工学上及び工業上貢献す るところが大きく、博士 (工学)の学位論文
として十分な価値 を有す るもの と認める。
