工業分野において，機械的強度，高温耐酸化性，耐食性に優れるセラミックス材料の需要が増
加している．しかしながら，セラミックス材料の加工性の低さは，材料の適用範囲を制限する
重大な問題である．セラミック材料は硬くて脆いため，切削によって複雑な形状に加工するこ
とは困難である．さらに，セラミックス材料の加工面にはクラックが導入されやすい．セラミ
ックス部品の機械的強度や信頼性を確保するためには，加工後に研磨などによって表面損傷を
除去する必要があり，加工コストが高いという問題もある．そのような加工に関する問題が発
生しない高強度なマシナブルセラミックスとして，MAX 相セラミックスの一つである Ti2AlC セ
ラミックスが注目されている．Ti2AlC セラミックスは層状 3 元素化合物であり，金属結合と共
有結合を有している．そのため，金属的な特性（金属並みの電気伝導，熱伝導，高い熱衝撃抵
抗）とセラミックス的な特性（高弾性率，耐酸化性，耐化学侵食性）を有している．また，セ
ラミックス的な性質を持っているにも関わらず汎用超硬合金製工具で切削加工が可能である．
それに加えて，高温環境下に置かれると自動的に傷が治癒する自己治癒能力を有しているため
再使用が容易である．このような優れた特性を有する Ti2AlC セラミックスは，高温用途におけ
る様々な機械部品への利用が期待されているだけでなく，新たな産業を創生しながら生産・消
費・廃棄のそれぞれの段階で資源を循環させる仕組みであるサーキュラー・エコノミー（ CE）
構築に資することも期待されている．しかしながら，Ti2AlC セラミックスは材料特性の制御が
困難であるため，高温環境製品として社会実装が進んでおらず，技術成熟度レベル(TRL)が低
いという問題がある．高温環境材料として使用する際には耐酸化性が重要となる．しかしなが
ら，Ti2AlC セラミックスは高い耐酸化性を有しているという報告もあるが，悪いという報告も
ある． Ti2AlC セラミックスの高温部品への普及を促進するためには， Ti2AlC セラミックスの
耐酸化性を安定化させることが必要である．それに加えて，機械的強度も高温耐熱部品にとっ
て重要な特性であるが，Ti2AlC セラミックスの機械的強度の報告値はばらつきがある．Ti2AlC
セラミックスを構造部材として用いる場合，機械的強度のばらつきの原因を突き止め，高い機
械的強度を有したサンプルを作製する必要がある．また，MAX 相セラミックスの切削加工は報
告されているが，切削面の損傷メカニズムや切削ダメージの影響については明らかにされてい
ない．Ti2AlC セラミックス製品を切削加工を用いて生産する際には，切削損傷が機械的強度に
与える影響を系統的に評価することが必要である．これらの問題を解決して，高温材料にとっ
て特に重要な特性に関する材料設計指針を構築することにより，Ti2AlC セラミックスの TRL を
上昇と社会実装の促進を行うことが可能となる．本論文の主な目的は， Ti2AlC セラミックス
の材料設計指針を確立することである．高温部品に対する材料設計指針を確立するために，組
織制御と組成調整が高温耐酸化性，機械的強度，被削性に及ぼす影響について検討した．そし
て，その材料設計指針を高温ねじ部品に適用し，その特性を評価した．耐酸化性に関しては高

Al 濃度の Ti2AlC が良好な耐酸化性を有し，Nb 添加により低 Al 濃度の Ti2AlC の非保護性酸化
を抑制可能であることを明らかにした．この結果は，Ti2AlC セラミックスの耐酸化性を安定化
させるために有用である．また，機械的強度に関しては TiC を制御することにより曲げ強度が
向上すること，耐酸化性の向上に有効な Nb 添加は機械的強度に影響を与えないことを明らか
にした．この結果は，Ti2AlC セラミックスの機械的強度を向上させるために有用である．被削
性に関しては，Ti2AlC セラミックスをグラファイトや SKD61 と同じ条件で切削加工できるこ
と，切削加工したままの Ti2AlC セラミックス試験片の曲げ強さが大きく低下しないことを明
らかにした．これらの結果から，Ti2AlC セラミックスは，加工時に曲げ強度が大きく低下しな
いため，その後の研磨を必要としない可能性があり，実用性の高いマシナブルセラミックスで
あることが示された．以上の議論を踏まえて，高温部品に適用するための Ti2AlC セラミックス
に関する材料設計指針を作成した．その材料設計指針に基づき Ti2AlC セラミックスボルト・ナ
ットを作製して特性評価を実施した．その結果，Ti2AlC セラミックスは一般的なマシナブルセ
ラミックスであるマイカよりも被削性に優れていることを明らかにした．また，Ti2AlC セラミ
ックスボルト・ナットは市販の耐熱ねじ部品である，アルミナボルト・ナットよりも高い機械
的強度を有していた．これらの結果は，今回構築した材料設計指針に沿って作製した Ti2AlC セ
ラミックスは耐熱ねじ部品用の材料に適しており，従来のセラミックスねじ部品よりも優れた
特性を有することが確認された．したがって，本論文で構築された高温部品用材料設計指針は

Ti2AlC MAX相セラミックスは組成域が狭いため，様々な異相と共存する．その異相のばらつ
きが耐酸化性に影響を与えているのではないか．また，強度がバラついている原因は結晶構
造に影響があるのではないかと考える．加工性に関しても，加工したという報告はあるが，
表面粗さや，加工後の強度低下についての報告はない．以上の知見を組み合わせることによ

Taxis are an essential means of transportation for addressing the movement requirements of the citizens 
of the local area. Additionally, effective transportation infrastructure is required to lower CO2 
emissions in the context of climate change. Inefficien t taxi services result in increased idle time as well 
as increased CO2 emissions. So, optimizing taxi allocation can help reduce CO2 emissions and improve 
the sustainability of suburban transportation. The main objective of this thesis is to develop a 
mathematical model which would be used for shared taxi routing problems and allocation. The shared 
taxi problem model deals with passenger demand, which is to find the shortest route of taxi routing 
decisions that can minimize the customers' total trip cos t. On the other hand, the objective of the taxi 
allocation model is to maximize allocation based on idle time costs, and passenger waiting time while 
minimizing CO2 emissions.  
Firstly, this study focuses on taxi demand prediction for areas in Nagaoka city using Machine Learning 
algorithms. To do so, several ride-related parameters have been studied such as the month of the year, 
day of the week, and time of the day. Moreover, weather-related variables, such as temperature, and 
weather conditions are used as predictors to estimate the demand level for taxis in urban areas. A 
suggested approach for predicting taxi demand in suburban areas involves the use of four well -known 
machine learning algorithms: Linear Regression, Decision Tree, Random Forests, and Grad ient 
Boosting. Additionally, a Hybrid Machine Learning method has been proposed for this purpose.  
Secondly, a nonlinear shared taxi routing model explores the concept of a shared taxi to minimize the 
total cost for passengers and the number of taxis utiliz ed, while maximizing customer satisfaction 
thereby increasing the taxi demand and driver’s income. The problem is solved using the Branch and 
Cut algorithm employed for estimating the parameters. The computational results obtained by the 
Gurobi solver showed that the proposed mathematical model can find the shortest route of taxi routing 
decisions that can minimize the customers' total cost. Furthermore, the sensitivity of significant 
parameters such as time window, initial fare, and travel distance in the fixed fare is analyzed by using 
taxi data of Sanjo city, Japan.  
Lastly, this study proposes a mathematical model of the taxi allocation optimization problem for 
minimizing taxi driver idle time costs, which helps to reduce CO2 emissions in suburban context s. For 
solving the proposed model, we used instead of proposing three heuristic algorithms: greedy, Simulated 
annealing, and dynamic greedy. For the case study, we first identified the taxi travel hotspots as 
potential locations for taxi spots by investiga ting the pickup and drop-off locations and times by 
analyzing GPS taxi data of Nagaoka, Japan. Dynamic greedy heuristic was used to obtain excellent 
solutions quickly. The number of taxis through data analysis could potentially reduce CO2 emissions 
in the transportation sector by approximately 81.84%.  The study recommends that cities prioritize taxi 
location optimization to achieve sustainable transportation and validates the model with sensitivity 
analysis. 


Development of Mathematical Models for Allocation and Routing Problem of the Shared Taxi with Heuristic Algorithm

In recent years, the application of cardiomyocytes differentiated from human iPS cells (hiPSCs) has emerged as 
a promising avenue for advancing treatments targeting cardiac disorders. Induced pluripotent stem cells (iPSCs) 
are pluripotent stem cells generated from somatic cells. HiPSCs are used to elucidate disease mechanisms and 
develop new drugs, making them a good research tool in the medical field. In particular, the transplantation of 
hiPSCs-derived cardiomyocytes is attracting attention because it is necessary to repair damaged areas for 
treatment of hearts, which have the poor ability of proliferation. However, although an empirical induction 
method of cardiac differentiation from hiPSCs has been developed, the identification and functions of the factors 
required for cardiac differentiation are unclear completely. Furthermore, there is no good animal model for 
experimental system that can quickly and easily confirm the safety and performance of induced human 
cardiomyocytes in vivo. 
This study encompasses two primary objectives employing cardiac linage cells derived from hiPSCs. First, to 
elucidate the cardiac differentiation mechanism, a comprehensive exploration into the role of auto/paracrine 
morphogen was conducted using a optimized induction method of cardiac differentiation from 201B7 cell line at 
an initial low cell density. Specifically, the preculture medium was changed to Stem fIt medium from F9a medium, 
and differentiation was induced at a higher concentration of CHIR. Co-culture with high density cells promoted 
cardiac differentiation of low density cells, suggesting cardiac differentiation required not cell-cell contact 
signaling but auto/paracrine morphogen secreted from high density cells. Early Wnt signaling inhibition also 
promoted cardiac differentiation. In addition, several morphogens that inhibit Wnt signaling were detected from 
the medium at the early stage of cardiac differentiation induction, suggesting the cells secreted these morphogens. 
Therefore, I concluded that auto/paracrine effect of Wnt inhibitor morphogen at early timing of differentiation 
promoted cardiac differentiation. 
Second, I aimed to develop a humanized cardiac zebrafish model with human cardiac lineage cells in the heart. 
HiPSCs transfected with fluorescent protein genes enabled noninvasive tracking of injected cells in embryos. To 
establish a humanized heart model, I transplanted cardiac linage cells derived from hiPSCs in the vicinity of the 
area proximal to the outflow tract of heart (heart resource) of zebrafish embryos. One day after injection, the 
injected cells migrated to the embryo heart and moving like heartbeat-like. Those results suggested that I succeed 
human cardiac lineage cells engrafted into the zebrafish embryonic heart. This model can be used to verify the 
effects of drugs on human cardiac lineage cells. In addition, the model can be useful to assess the viability and 
efficacy of the cell grafts before transplantation in patients. 
The findings of this research have notable implications for future studies in cardiovascular medicine, 
contributing to the development of therapeutic methods leveraging human cardiac cell lines obtained from hiPSCs.

 The fast expansion of the world economy comes with sustainable development have gathered much 
attent ion in recent years. Since the 1st of January 2016, the 17 Sustainable Development Goals (SDGs) 
officially came into force. From the industrial perspective, recycling or treating the excess amount of waste , 
which contains valuable metals, generated from manufactur ing activi ties can be a problem. Ultrasound (US) 
is an exceptional technology with a wide range of applicat ions such as cleaning, welding, sterilizing, food 
processing, and so on. With the mentioned points, this study was dedicated to the apply US technology to 
treat industr ial waste, especially for washing arsenic (As) contaminated soil and extracting silica from 
copper slag. 
 In the polluted soil treatment research, the soil was donated by a construction company in the form 
of wet soil. After receiving, the soil was air-dried, crushed, and sieved to the desired state before the washing 
process. The US system with 28 kHz and 1200 W was used as the main washing mechanism combined with 
NaOH and HCl; also, a shaker was used as the conventional method for the comparison. The result shows 
that, after 1-hour washing with US at 1200 W combined with NaOH 2 M and HCl 2 M, nearly 98% of A s 
was eliminated from the initial concentration of 9.12 ppm. However, at the same condition, As remained in 
the soil after washing using shaking was 6 and 6.8 ppm for NaOH and HCl, respect ively. The efficiency of 
the US washing comes from the fact that, after the washing process, the soil particle was severely fragmen t ed, 
facilitat ing more interaction between the pollutants and washing additives. Furthermore, XRD and FTIR 
results from treated soil indicated almost no difference before the treatment, indicat ing that the treatmen t  
did not alter the original state of the soil. 
 In the case of copper slag (CS), silica was successfully extracted from pyrometal lurgical CS under 
acidic conditions, with the aid of high-power US. The application of US irradiation proved effective in 
inhibi ting the formation of silica gel, thereby facilitat ing the removal of impurit ies from the obtained silica . 
During US exposure, the CS particles underwent efficient refinement, while the Si remained in an ionized 
state. However, when the irradiat ion stopped, the dissolved Si value experienced a signifi cant drop, lead ing 
to a rapid increase in particle size due to the gelation of silica. This is the newly found effect of the US, 
and there was no research before to extract silica from CS. The study also highlighted the crucial role of 
the chosen acid in the extraction process, as it strongly influenced the purity of the final product. When 
assisted by US, the utilization of 6 M hydrochloric acid (HCl) resulted in a 90% extraction yield with an 
iron (Fe) impurity level of 0.08%. On the other hand, employing 3 M sulfuric acid (H2SO4) in the same 
system yielded a lower Fe content of 0.04%, but with only an 80% recovery rate. Notably, the HCl 6 M 
system without US irradiation achieved a higher yield of 96%. However, the final product contained a higher  
Fe impurity of 0.5% compared to the US-assisted system. These results demonstrate that the application of 
US technology has advanced the production of purer silica and serves as a valuable tool for control l ing 
gelation.

is vital for waste characterizing, determine burn u-up level and appropriate disposal strategies.  
mutual separation of actinides has been studied extensively, to develop the nuclide separation 
methods as pre-treatment for highly precise and accurate analyses of actinides in spent fuels and 
nuclear fuel debris by using mass-spectrometry such as inductively coupled plasma-mass 
spectrometry (ICP-MS). ICP-MS is an analysis technique for trace multi elemental and isotopic 
concentration. ICP-MS allows determination of elements by filtering ions based on mass to charge 
ration (m/z). Therefore, the mutual separation of actinides is required to remove the isobaric 
interference (238U/238Pu,241Pu/241Am,243Am/243Cm). in this study, actinides separation was 
conducted by using several types of resin, which are, impregnated resin Tributyl phosphate (TBP-
resin) for Uranium (U) and Plutonium (Pu) separation, synthesized tertiary pyridine resin for 
separation trivalent actinides (AnIII) fron lanthanides (Ln),  and separation of americium from 
curium by using octyl phenyl-N,N-di-isobutyl carbamoyl phosphine oxide (TRU-resin ) and 
N,N,N’,N’-tetra-n-octyldiglycolamide (TODGA-resin). TBP-resin, DGA-resin and TRU-resin is 
commercially supplied by EiChrom and Triskem International. 
Generally, there are two types of experiments were conducted. First, batch experiment to 
observe the absorption behavior of actinides on the resins in different condition of solution.  By 
using TBP, CMPO and TODGA, the behavior of An(III) was observed in hydrochloride acid (HCl) 
and nitric acid (HNO3) solution. We obtained the distribution coefficient of trivalent actinides and 
lanthanides from batch experiment in 0.5M to 10M of HCl and HNO 3 solutions. Next, 
chromatograph separation to evaluate the separation behavior of actinides. The separation 
experiments were carried out in the column by ~8.5 mL resin packed in Muromac  mini column L 
with the ~10 cm column height. 0.5 mL feed solution was injected from above of column and eluted 
by desired concentration of acid solutions. Nuclides in collected 1mL fractions eluted from the 
column was detected and measured by using Gamma-ray spectrometer, alpha spectrometer, and 
ICP-MS/MS, Agilent 8900.  
In chapter II, separation of Pu from U by using TBP-resin is discussed. As the first step, Pu and 
U can be separated from An(III) and Ln in high concentration of HNO3 and Iron(II) oxide. The 
same resin also used to separate Pu from U in HCl system in the next step. And then, low 
concentration of HNO3 used to recover U from the resin, with recovery rate around 96%-98%. The 
effect of Fe(II) as reduction agent towards Pu(VI) and reduce oxidation state of Pu to Pu(IV), is 
also discussed on this study. The result confirmed that separation of actinides based on oxidation 
state is successfully done by combining HCl and HNO 3 system on TBP resin.

Chapter III discuses about An(III) and Ln’s behavior on the tertiary pyridine in sodium nitrate 
(NaNO3) aqueous solution. It is known that tertiary pyridine resin has ability to perform as ion 
exchange resin and N soft donor ligand in the An(III) and Ln separation.  The resin was proposed 
for separation of An (III) from Ln, before separation of americium (Am) from curium (Cm). The 
experiment was conducted to observe the complex between An(III), Ln, and N soft donor ligand in 
high pH of NaNO3 solution. In addition, from the behavior of An(III) and Ln in NaNO3 solution, 
we found that high pH will suppressed protonation in the system, avoid bonding between nitrogen 
and H+. As a result, separated peak of Am as an(III) from europhium (Eu) and neodymium (Nd) as 
lanthanide elements, is achieved with elution order . we confirmed that tertiary pyridine resin could 
work as a soft donor ligand in pH 11.  
Separation of Am from Cm by using TRU-resin and TODGA-resin was discussed in Chapter IV. 
Based on distribution coefficient (Kd), Am, Cm, and Ln elements are strongly absorbed into TRU-
resin in HNO3 solution. affinity between Am, Cm and Lns are decreasing in low concentration of 
HCl. Therefore, chromatograph separation was designed to use TRU-resin for extraction of An(III) 
and Ln in HNO3 solution and low concentration is used for recovery of the elements from TRU-
resin. As the last step, separation Am from Cm in 1M HCl by using TODGA-resin. We confirmed 
that Am and Cm is successfully separated in 1M HCl acid in DGA resin. The recove ry of Am and 
Cm reached 100% and 96.5%, respectively.  
 In summary, separation actinides elements are achieved and a new route of mutual nuclide 
separation method is successfully designed. By performing actinides separation from Pu/U 
separation by using TBP-resin until Am/Cm separation by using TODGA resin, as a pretreatment 
before the analysis of actinides by using ICP-MS, the isobaric interference such as of 238U/238Pu, 
241Pu/241Am, and 243Am/243Cm can be removed.

加硫機構の解明は、力学物性に優れる加硫天然ゴムの開発に道を切り開く重要な課題で
ある。なぜなら、加硫機構を解明することにより副反応に影響を及ぼす因子を特定し、こ
の因子を制御することにより、cis-1,4-イソプレン単位の含有率が高い加硫天然ゴムを調
製できるようになるからである。加硫機構は、種々の配合処方や加硫条件で調製される加
硫天然ゴムの構造から逆合成解析することにより検討できるため、加硫天然ゴムの構造を
精緻に解析することにより解明できると考えられる。例えば、核磁気共鳴（ＮＭＲ）法を
適用することにより、加硫天然ゴムの構造は解析できるため、加硫機構は解明できる可能
性がある。しかしながら、これまでの研究では、加硫天然ゴムは、既存の溶液ＮＭＲ法や
固体ＮＭＲ法により構造解析が困難であっため、その構造は実証的に解析されることはな
かった。最近、ゴムＮＭＲ法が開発され、加硫天然ゴムの一次構造は実証的に解析できる
ようになった。このゴムＮＭＲ法では、約20 kHzの高速マジック角回転により双極子双極
子相互作用の効果を無視できるほど小さくし、高分解能スペクトルを得ることができる。
さらに、定量的な解析が可能であり、溶液NMR法と同様に二次元NMR測定を行うことが
できる。それ故、ゴムNMR法により、種々の配合処方や加硫条件で調製される加硫天然
ゴムの構造を解析し、加硫機構を解明できると考えられる。本研究では、加硫ゴムの材料
設計を科学的根拠に基づいて行う道を切り拓くため、ゴムNMR法により、種々の条件で
調製した加硫天然ゴムの構造を解析し、加硫機構を解明する検討を行った。  
第一章「序論」では、本研究の背景について述べ、硫黄加硫の加硫機構、ゴムNMR
法、架橋点の構造解析に関する過去の研究成果を紹介した後、問題点と解決策および本研
究の目的を述べた。  
第二章「ゴムNMR法による天然ゴムの加硫における加硫促進剤の効果に関する研究」
では、ラジカル機構を促進すると報告されている2-メルカプトベンゾチアゾール亜鉛
（ZMBT）、ジブチルジチオカルバメート亜鉛（ZDEC）、ジエチルジチオカルバメート亜
鉛（ZDBC）を加硫促進剤とし、促進硫黄加硫により得られた加硫天然ゴムの架橋点の構
造をゴムNMR法により解析した。 13C-NMRスペクトルに現れたシグナルを既報に従って
帰属し、Distorsionless Enhancement by Polarization Transfer（DEPT）測定およびAttached 
Proton Test（APT）測定により帰属を検証した。さらに、加硫促進剤の種類と架橋点の構
造を比較することにより、加硫促進剤が架橋点の構造に及ぼす影響を検討した。  
第三章「ゴムNMR法による加硫天然ゴムの架橋点の定量的解析」では、ゴムNMR法に
よる構造解析を、促進硫黄加硫天然ゴムと非促進硫黄加硫天然ゴムの二種類に対して行っ
た。加硫後、 1H-NMRスペクトルに現れたシグナルは、 13C-NMR法（DEPT、APT、
HMQC）を通して、架橋点、ペンダントスルフィド基およびその隣接基に帰属された。さ

らに、Nuclear Overhauser Effect Spectroscopy（NOESY）を適用することによって架橋点と
ペンダントスルフィド基の識別を試みた。  
第四章「高立体規則性加硫天然ゴムの調製と物性」では、異なる温度で調製した加硫天
然ゴムの構造を、逆ゲート付きデカップリングを用いたゴム NMR 法により解析した。シ
グナル強度から見積もった cis-1,4-イソプレン単位、 trans-1,4-イソプレン単位および架橋
点の割合と加硫天然ゴムの力学物性との関係を検討した。  
第五章「高立体規則性加硫天然ゴムの構造と物性におけるタンパク質の効果」では、異
なる温度で調製した加硫脱タンパク質化天然ゴム（V-DPNR）の構造を、逆ゲート付きデ
カップリングを用いたゴム NMR 法により解析し、加硫天然ゴムの構造と比較した。さら
に、シグナル強度から見積もられた cis-および trans-1,4-イソプレン単位と架橋点の割合
と、V-DPNR の力学物性を加硫天然ゴムと比較することにより、加硫天然ゴムの立体規則
性および力学物性におけるタンパク質の効果を明らかにした。  
第六章「総括」では、各章で得られた結論を述べ、総括した。  
 以上より、これまで、勘と経験を頼りに行われてきた加硫天然ゴムの材料設計を科学的
根拠に基づいて行う道を切り拓いた。
